Fe3O4@Cu/C and Fe3O4@CuO Composites Derived from Magnetic Metal–Organic Frameworks Fe3O4@HKUST-1 with Improved Peroxidase-Like Catalytic Activity

Huang Yan-Feng,Zhang Lu,Ma Li,Li Ying,Zhong Chongli

Abstract Fe 3 O 4 @Cu/C and Fe 3 O 4 @CuO composites were achieved using nanoparticle-encapsulated MOFs Fe 3 O 4 @HKUST-1 as both a precursor and a self-sacrificing template. Fe 3 O 4 @HKUST-1 was thermally converted into magnetic Fe 3 O 4 @Cu/C and Fe 3 O 4 @CuO composites in different atmosphere (N 2 and air). The resulting composites not only kept superparamagnetic characteristics, but also exhibited improved peroxidase-like catalytic activity and high stability when compared to the precursor Fe 3 O 4 @HKUST-1. The morphology, crystal structure, magnetic and porous properties of Fe 3 O 4 @Cu/C and Fe 3 O 4 @CuO composites were characterized, and the kinetics parameters and the influence factors on the peroxidase-mimicking activity such as temperature, pH and H 2 O 2 concentration were evaluated. On account of the excellent peroxidase-like activity, the as-prepared Fe 3 O 4 @Cu/C and Fe 3 O 4 @CuO composites were successfully used for the catalytic removal of methylene blue (MB) dye with H 2 O 2 oxidant. This work provides an effective way to fabricate highly reactive MOFs-derived biomimetic catalysts, which have potential applications in bioassays and pollutant degradation. Graphic Abstract Fe 3 O 4 @Cu/C and Fe 3 O 4 @CuO composites were achieved using Fe 3 O 4 nanoparticle-encapsulated HKUST-1 as self-sacrificial templates and demonstrated excellent peroxidase-like activity.

Self-assembly directed synthesis of Au nanorices induced by polyaniline and their enhanced peroxidase-like catalytic properties

Wei Song,Maoqiang Chi,Mu Gao,Bing Zhao,Ce Wang,Xiaofeng Lu

The self-assembly of noble metal nanoparticles into new-fashioned one, two and three-dimensional structures is very important due to their superior properties in optics, electrics, catalysis, and chemical sensing compared with individual nanoparticles. Here, we first report a facile one-step approach that allows the fabrication of assembled Au nanorices induced by polyaniline (PANI). During the synthesis, a complex of HAuCl4-(3-aminopropyl)triethoxysilane (APTES) acts as both a soft template and an oxidant; then the reduction of HAuCl4 is accompanied by the oxidation of aniline, resulting in the formation of Au nanorices within a PANI matrix. The application of the prepared Au/PANI nanorices as enzyme mimics toward the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) is demonstrated. The resultant Au/PANI nanorices display an enhanced peroxidase-like catalytic activity compared with individual Au nanospheres and PANI nanofibers alone, revealing a synergistic effect between Au and PANI components in the Au/PANI nanorices. The Au/PANI nanorices are also used as efficient surface-enhanced Raman scattering (SERS) substrates for in situ monitoring of the oxidation process of TMB during the peroxidase-like catalytic reaction. On the basis of the SERS technique, a discernible detection level of H2O2 as low as 10−8 M is obtained. We envision that such a system may show more potential applications in biocatalysis, disease diagnosis and environmental monitoring.

A Facile synthesis of superparamagnetic Fe3O4 nanofibers with superior peroxidase-like catalytic activity for sensitive colorimetric detection of l-cysteine

Sihui Chen,Maoqiang Chi,Yun Zhu,Mu Gao,Ce Wang,Xiaofeng Lu

Superaramagnetic Fe 3 O 4 nanomaterials are good candidates as enzyme mimics due to their excellent catalytic activity, high stability and facile synthesis. However, the morphology of Fe 3 O 4 nanomaterials has much influence on their enzyme-like catalytic activity. In this work, we have developed a simple polymer-assisted thermochemical reduction approach to prepare Fe 3 O 4 nanofibers for peroxidase-like catalytic applications. The as-prepared Fe 3 O 4 nanofibers show a higher catalytic activity than commercial Fe 3 O 4 nanoparticles. The steady-state kinetic assay result shows that the Michaelis–Menten constant value of the as-obtained Fe 3 O 4 nanofibers is similar to that of horseradish peroxidase (HRP), indicating their superior affinity to the 3,3′,5,5′-tetramethylbenzidine (TMB) and H 2 O 2 substrate. Based on the outstanding catalytic activity, a sensing platform for the detection of l- cysteine has been performed and the limit of detection is as low as 0.028 μM. In addition, an excellent selectivity toward l- cysteine over other types of amino acids, glucose and metal ions has been achieved as well. This work offers an original means for the fabrication of superparamagnetic Fe 3 O 4 nanofibers and demonstrates their delightful potential applications in the fields of biosensing, environmental monitoring, and medical diagnostics.

Engineering Perovskite Hydroxide as a Cold-Adapted Oxidase Mimic for Construction of the Robust Low-Temperature Adaptive Biosensors

Yufeng Liu,Jing Zhang,Yuxin Wan,Cong Li,Shuai Cui,Xuejiao J. Gao,Hui Wei,Dongzhi Yang

Traditional biological detection methods rely on signal amplification strategies such as enzymatic catalysis or nucleic acid amplification. However, their efficiency decreases in low-temperature environments, compromising their detection sensitivity. To break the loss of enzyme catalytic activity at low temperatures, research on cold-adaptive nanozymes has attracted much attention. Till now, only a few nanozymes have been reported to have cold-resistant catalytic properties. Here, a new type of cold-adapted nanozyme was constructed by engineering a perovskite hydroxide. The nanozyme not only boosted the oxidase-like catalytic activity by 2 orders of magnitude but also retained excellent catalytic performance at 0 °C. This enhanced activity may be attributed to the increase in manganese content, vacancy oxygen, and tetravalent manganese. Then, a robust low-temperature adaptive biosensor was established with a cold adaptive nanozyme. Notably, the detection of sulfide ion, ascorbic acid, alkaline phosphatase, and cellular glutathione by the cold-adapted probe was less affected by the temperature reduction, and the detection sensitivity of the probe for ALP at 0 °C is better than that of a commercial kit. Finally, the cold-adapted nanozyme was further used to construct a paper-based H2S gas colorimetric probe. This study develops a new cold adaptive nanozyme and broadens the application scenarios of the nanozymes.

Colorimetric Chemosensor Based on Fe3O4 Magnetic Molecularly Imprinted Nanoparticles for Highly Selective and Sensitive Detection of Norfloxacin in Milk

Maiquan Li,Lingli Luo,Jiayin Li,Yingzi Xiong,Ling Wang,Xia Liu

Long-term use of norfloxacin (NOR) will cause NOR residues in foods and harm human bodies. The determination of NOR residues is important for guaranteeing food safety. In this study, a simple, selective, and label-free colorimetric chemosensor for in situ NOR detection was developed based on Fe3O4magnetic molecularly imprinted nanoparticles (Fe3O4MMIP NPs). The Fe3O4MMIP NPs showed good peroxidase-like catalytic activity to 3,3′,5,5′-tetramethylbenzidine (TMB) and selective adsorption ability to NOR. The colorimetric chemosensor was constructed based on the Fe3O4MMIP NPs-H2O2-TMB reaction system. The absorbance differences were proportional to the concentrations of NOR in the range of 10–300 ng/mL with a limit of detection at 9 ng/mL. The colorimetric chemosensor was successfully applied to detect NOR residue in milk. The recovery range was 78.2–95.81%, with a relative standard deviation of 2.1–9.88%. Together, the proposed colorimetric chemosensor provides a reliable strategy for the detection of NOR residues in foods.Keywords:Fe3O4MMIP NPs;peroxidase-like catalytic activity;colorimetric chemosensor;norfloxacin detection

A reusable colorimetric assay based on mixed valence state Ce-MOF@Pt nanoparticles for highly sensitive detection of visfatin

Yan Liu,Lianying Zhang,Qinge Li,Han Dai,Ting Xiang,Gangyi Yang,Ling Li

In this work, a colorimetric assay for visfatin detection is described. The mixed valence state Ce-MOF (MVCM) modified by platinum nanoparticles (Pt NPs) is used as a novel catalyst. MVCM exhibits excellent intrinsic peroxidase-like catalytic activity due to the spontaneous recycling of the Ce(III)/Ce(IV) system. Pt NPs serve not only as a carrier of the –NH 2 -modified single strand DNA (S1) but also as a synergistic catalyst of MVCM. The capture probe (S2) attached to the streptavidin-modified magnetic beads (Mag-SA) could combine with the aptamer to form the Mag-SA/S2/aptamer complex. When in the presence of the target visfatin, aptamer specifically combines with the visfatin, which induces the release of Mag-SA/S2 from the Mag-SA/S2/aptamer complex. At this time, the [email protected] /S1 complex connects with the released Mag-SA/S2, which quickly catalyzes the 3,3,3′,3′-tetramethylbenzidine (TMB), leading to a color change. Under optimal conditions, the absorbance increases linearly when the concentration ranges from 1 ng mL −1 to 100 ng mL −1 , and the detection limit is as low as 0.11 ng mL −1 . Furthermore, Mag-SA/S2 can be reused at least five times by using the uracil-DNA glycosylase (UDG) and an external magnetic field. The proposed method shows satisfying reproducibility, stability, specificity, and sensitivity, and it was successfully applied to detect visfatin in spiked human serum samples. Thus, it has great potential for clinical research, detection, and catalytic applications.

Discovery of FeP/Carbon Dots Nanozymes for Enhanced Peroxidase-Like Catalytic and Antibacterial Activity

Jiaxin Dong,Guanxiong Liu,Yuri V. Petrov,Yujie Feng,Dechang Jia,Vladimir E Baulin,Aslan Yu Tsivadze,Yu Zhou,Baoqiang Li

Iron phosphide/carbon (FeP/C) serving as electrocatalysts exhibit excellent activity in oxygen reduction reaction (ORR) process. H 2 O 2 catalyzed by peroxidase (POD) is similar to the formation of new electron transfer channels and the optimization of adsorption of oxygen-containing intermediates or desorption of products in ORR process. However, it is still a challenge to discover FeP/C with enhanced POD-like catalytic activity in the electrocatalytic database for biocatalysis. The discovery of FeP/carbon dots (FeP/CDs) nanozymes driven by electrocatalytic activity for enhanced POD-like ability is demonstrated. FeP/CDs derived from CDs-Fe 3+ chelates show enhanced POD-like catalytic and antibacterial activity. FeP/CDs exhibit enhanced POD-like activities with a specific activity of 31.1 U mg −1 that is double higher than that of FeP. The antibacterial ability of FeP/CDs nanozymes with enhanced POD-like activity is 98.1%. The antibacterial rate of FeP/CDs nanozymes (250 µg mL −1 ) increased by 5%, 15%, and 36% compared with FeP, Fe 2 O 3 /CDs, and Cu 3 P/CDs nanozymes, respectively. FeP/CDs nanozymes will attract more efforts to discover or screen transition metal phosphide/C nanozymes with enhanced POD-like catalytic activity for biocatalysis in the electrocatalytic database.

A label-free colorimetric platform for DNA via target-catalyzed hairpin assembly and the peroxidase-like catalytic of graphene/Au-NPs hybrids

Chaohui Chen,Ningxing Li,Jingwen Lan,Xinghu Ji,Zhike He

A target-catalyzed hairpin assembly (CHA) and graphene/Au-NPs hybrids-based platform has been developed for the determination of DNA . This new sensor not only avoided any labeling but also reduced the background signal. In the absence of target, the assembly of H1 and H2 couldn't be triggered. The catalytic activity of graphene/Au-NPs hybrids was inhibited by adsorption of H1 and H2, leading to the “inactive” hybrids unable to catalyze the oxidation reaction of 3,3′,5,5′-tetramethylbenzidine (TMB). However, with the addition of target DNA, the target-catalyzed hairpin assembly was initiated and produced plenty of H1–H2 duplex, which had a weak binding affinity with the graphene/Au-NPs. Thus, the protected interface of graphene/Au-NPs hybrids became active and catalyzed the oxidation reaction of TMB accompanied with a colorless to-blue color change. This approach exhibited good sensitivity and specificity for target DNA with a detection limit of 5.74 × 10 −11  M, and realized the assay of target DNA in human serum samples. Besides, this sensor could be further expanded to detect viruses or proteins by adapting the corresponding aptamers, showing great potential in biochemical detections.

Efficient in situ growth of enzyme-inorganic hybrids on paper strips for the visual detection of glucose

WanYun Li,ShiYu Lu,ShuJuan Bao,ZhuanZhuan Shi,Zhisong Lu,ChangMing Li,Ling Yu

A visual colorimetric microfluidic paper-based analytical device (μPAD) was constructed following the direct synthesis of enzyme-inorganic hybrid nanomaterials on the paper matrix. An inorganic solution of MnSO 4 and KH 2 PO 4 containing a diluted enzyme (glucose oxidase, GOx) was subsequently pipetted onto cellulose paper for the in situ growth of GOx@Mn 3 (PO 4 ) 2 hybrid functional materials. The characterization of the morphology and chemical composition validated the presence of hybrid materials roots in the paper fiber, while the Mn 3 (PO 4 ) 2 of the hybrid provided both a surface for enzyme anchoring and a higher peroxidase-like catalytic activity as compared to the Mn 3 (PO 4 ) 2 crystal that was synthesized without enzyme modulation. This new approach for the in situ growth of an enzyme-inorganic hybrid on a paper matrix eliminates centrifugation and the dry process by casting the solution on paper. The sensing material loading was highly reproducible because of the accuracy and stability of pipetting, which eventually contributed to the reliability of the μPAD. The self-assembled natural and artificial enzyme hybrid on the μPADs specifically detected glucose from a group of interferences, which shows great specificity using this method. Moreover, the colorimetric signal exhibited detection limitation for glucose is 0.01   mM, which lies in the physiological range of glucose in biological samples.

Preparation of Well-Constructed and Metal-Modified Covalent Organic Framework Nanoparticles for Biosensing Design with Cascade Catalytic Capability

Tianci Zhou,Ying Deng,Xinyu Qu,Lin Wang,Haojie Xie,Yetao Xu,Lizhou Sun,Jie Yang,Genxi Li

Uniform covalent organic framework nanoparticles (COF NPs) with a well-defined pore structure may provide a robust platform for scaffolding enzymes. Herein, bipyridine-based spherical COF NPs have been successfully prepared in this work through the Schiff base condensation reaction. Moreover, they are functionalized by metal modification and are further used for biosensor fabrication. Experimental results reveal that the metal-modified COF NPs also display impressive peroxidase-like catalytic activities, while they can load enzymes, such as glucose oxidase (GOx) and sarcosine oxidase (SOx), to develop a cascade catalysis system for design of various kinds of biosensors with very well performance. For example, the optimized GOx@Fe-COFs can achieve a sensitive detection of glucose with a low limit of detection (LOD) of 12.8 μM. Meanwhile, the enzymes also exhibit a commendable preservation of 80% enzymatic activity over a span of 14 days under ambient conditions. This work may pave the way for advancing cascade catalysis and the analysis of different kinds of biological molecules based on COF NPs.

Xinyuan Yan,Juan Chai,Luohao Yuan,Hongfeng Yin

Glutathione (GSH), as an significant intracellular antioxidant, plays a vital role in preserving the balance of cellular redox homeostasis. The accurate and sensitive detection of GSH is essential for understanding the cellular redox status and for the diagnosis and treatment of many diseases. Herein, a core–shell structure composed of glucose oxidase (GOx), platinum (Pt) nanoparticles and a zeolitic imidazolate framework (ZIF-8) shell was constructed. Pt NPs in the core–shell showed high peroxidase-like catalytic activity. The as-prepared Pt-GOx@H-ZIF-8 can be utilized for colorimetric detection of GSH, exhibiting a broad linear range from 0.5 to 90 μM, and a low limit of detection (LOD) of 0.4 μM. Pt-GOx@H-ZIF-8 showed excellent sensitivity, selectivity and stability towards glutathione detection, making it a promising candidate for intracellular antioxidants.

Engineering efficient artificial nanozyme based on chitosan grafted Fe-doped-carbon dots for bacteria biofilm eradication

Ting Pan,Huanhuan Chen,Xiang Gao,Zeyu Wu,Yingwang Ye,Yizhong Shen

Bacterial biofilms have evoked worldwide attention owing to their serious threats to public health, but how to effectively eliminate bacterial biofilms still remains great challenges. Here, we rationally designed a novel and vigorous chitosan grafted Fe-doped-carbon dots ( [email protected] /CDs) as an efficient artificial nanozyme to combat rigid bacterial biofilms through the selective activation of Fenton-like reaction-triggered peroxidase-like catalytic activity and the synergistic antibacterial activity of CS. On the one hand, the peroxidase-like catalytic activity made [email protected] /CDs catalyze H 2 O 2 for producing hydroxyl radicals (•OH), resulting in efficient cleavage of extracellular DNA (eDNA). On the other hand, CS was capable of binding with the negatively charged cell membrane through electrostatic interaction, changing the cell membrane permeability and causing cell death within bacterial biofilms. Based on their synergistic effects, the fragments of bacterial biofilm and exposed bacteria were persistently eradicated. Remarkably, [email protected] /CDs-based nanozyme not only enabled the effective destroying of gram-positive Staphylococcus aureus ( S. aureus ) biofilms, but also completely eliminated gram-negative Pseudomonas aeruginosa ( P. aeruginosa ) biofilms, showing great potential as a promising anti-biofilm agent against bacteria biofilms. This proposed synergistic strategy for bacterial biofilm eradication might offer a powerful modality to manage of bacterial biofilm fouling in food safety and environmental protection.
