Motility is finely regulated and is crucial to bacterial processes including colonization and biofilm formation. There is a trade-off between motility and growth in bacteria with molecular mechanisms not fully understood. Hypermotile Escherichia coli could be isolated by evolving non-motile cells on soft agar plates. Most of the isolates carried mutations located upstream of the flhDC promoter region, which upregulate the transcriptional expression of the master regulator of the flagellum biosynthesis, FlhDC. Here, we identified that spontaneous mutations in clpX boosted the motility of E. coli largely, inducing several folds of changes in swimming speed. Among the mutations identified, we further elucidated the molecular mechanism underlying the ClpXV78F mutation on the regulation of E. coli motility. We found that the V78F mutation affected ATP binding to ClpX, resulting in the inability of the mutated ClpXP protease to degrade FlhD as indicated by both structure modeling and in vitro protein degradation assays. Moreover, our proteomic data indicated that the ClpXV78F mutation elevated the stability of known ClpXP targets to various degrees with FlhD as one of the most affected. In addition, the specific tag at the C-terminus of FlhD being recognized for ClpXP degradation was identified. Finally, our transcriptome data characterized that the enhanced expression of the motility genes in the ClpXV78F mutations was intrinsically accompanied by the reduced expression of stress resistance genes relating to the reduced fitness of the hypermotile strains. A similar pattern was observed for previously isolated hypermotile E. coli strains showing high expression of flhDC at the transcriptional level. Hence, clpX appears to be a hot locus comparable to the upstream of the flhDC promoter region evolved to boost bacterial motility, and our finding provides insight into the reduced fitness of the hypermotile bacteria.

Regulating the swimming motility of bacteria near surfaces is essential to suppress or avoid bacterial contamination and infection in catheters and medical devices with wall surfaces. However, the motility of bacteria near walls strongly depends on the combination of the local physicochemical properties of the surfaces. To unravel how nanostructures and their local chemical microenvironment dynamically affect the bacterial motility near surfaces, here, we directly visualize the bacterial swimming and systematically analyze the motility ofEscherichia coliswimming on ZnO nanoparticle films and nanowire arrays with further ultraviolet irradiation. The results show that the ZnO nanowire arrays reduce the swimming motility, thus significantly enhancing the trapping ability for motile bacteria. Additionally, thanks to the wide bandgap nature of a ZnO semiconductor, the ultraviolet irradiation rapidly reduces the bacteria locomotion due to the hydroxyl and singlet oxygen produced by the photodynamic effects of ZnO nanowire arrays in an aqueous solution. The findings quantitatively reveal how the combination of geometrical nanostructured surfaces and local tuning of the steric microenvironment are able to regulate the motility of swimming bacteria and suggest the efficient inhibition of bacterial translocation and infection by nanostructured coatings. Keywords:ZnO nanowire arrays;bacteria motility;antibacterial;particle tracking

Abstract The genus Citrobacter is commonly found in environmental and industrial settings, some members of which have been used for bioremediation of heavy metals owing to the absorption ability of their biofilms. Although our previous studies have found that the outer membrane protein A (OmpA) contributes to the process of Citrobacter werkmanii biofilm formation, the underlying mechanisms remain elusive. Therefore, we deleted ompA from the genome of C. werkmanii and investigated its phenotypes in comparison to the wild type strain (WT) and the complementary strain using biochemical and molecular techniques including RNA-Seq. Our results demonstrated that the deletion of ompA led to an increase in biofilm formation on both polystyrene and glass surfaces due to upregulation of some biofilm formation related genes. Meanwhile, swimming ability, which is mediated by activation of flagellar assembly genes, was increased on semi-solid plates in the ∆ompA strain when compared with WT. Additionally, inactivation of ompA also caused increased 1,2-benzisothiazolin-3-one (BIT) resistance, differential responses to Ca 2+ stress, curli protein expression and cellulose production. Finally, ∆ompA caused differential expression of a total of 1470 genes when compared with WT, of which 146 were upregulated and 1324 were downregulated. These genes were classified into different Gene Ontology (GO) and KEGG pathways. In summary, ompA in C. werkmanii contributes to a variety of biological functions and may act as a target site to modulate biofilm formation. Key points
• ompA is a negative regulator for biofilm formation by C. werkmanii.
• ompA inhibits swimming motility of C. werkmanii.
• ompA deletion causes different expression profiles in C. werkmanii.

Vibrio parahaemolyticusis a primary seafood-associated pathogen that could cause gastroenteritis. It can attach to various surfaces and form a biofilm, which poses serious threats to food safety. Hence, an effective strategy is urgently needed to control the biofilm formation ofV. parahaemolyticus. Laurel essential oil (LEO) is used in food, pharmaceutical and other industries, and is commonly used as a flavoring agent and valuable spice in food industries. The potential antibiofilm effects of LEO againstV. parahaemolyticuswere examined in this study. LEO obviously reduced biofilm biomass at subinhibitory concentrations (SICs). It decreased the metabolic activity and viability of biofilm cells. Microscopic images and Raman spectrum indicted that LEO interfered with the structure and biochemical compositions of biofilms. Moreover, it also impaired swimming motility, decreased hydrophobicity, inhibited auto-aggregation and reduced attachment to different food-contact surfaces. RT-qPCR revealed that LEO significantly downregulated transcription levels of biofilm-associated genes ofV. parahaemolyticus. These findings demonstrate that LEO could be potentially developed as an antibiofilm strategy to controlV. parahaemolyticusbiofilms in food industries.Keywords:V. parahaemolyticus;laurel essential oil;biofilm formation;gene expression;motility

Xanthomonas albilineans (Xal) is a gram-negative bacterial pathogen responsible for developing sugarcane leaf scald disease, which engenders significant economic losses within the sugarcane industry. In the current study, homologous recombination exchange was carried out to induce mutations within the virB/D4 -like type IV secretion system (T4SS) genes of Xal. The results revealed that the virB11 -deletion mutant (Δ virB11 ) exhibited a loss in swimming and twitching motility. Application of transmission electron microscopy analysis further demonstrated that the Δ virB11 failed to develop flagella formation and type IV pilus morphology and exhibited reduced swarming behaviour and virulence. However, these alterations had no discernible impact on bacterial growth. Comparative transcriptome analysis between the wild-type Xal JG43 and the deletion-mutant Δ virB11 revealed 123 differentially expressed genes (DEGs), of which 28 and 10 DEGs were notably associated with flagellar assembly and chemotaxis, respectively. In light of these findings, we postulate that virB11 plays an indispensable role in regulating the processes related to motility and chemotaxis in Xal.

Resveratrol (trans-3,4′,5-trihydroxystilbene, RES) is a stilbenoid naturally present in a variety of plants. Although there are several reports about its anti-fatigue activity, its impact on intensive exercise-induced fatigue and the underlying mechanisms are yet not well understood. In the present study, we established a swimming exercise protocol in mice that is similar to the fatigue condition induced by a long period of intensive exercise and explored the effect of RES on fatigue and the mechanisms from the perspective of intestinal injury and gut microbiota. The results revealed that RES significantly prolonged exhaustive swimming time in fatigued mice and improved the serum indexes associated with fatigue, including serum glucose, lactic acid (LA), urea nitrogen (BUN), lactate dehydrogenase (LDH), creatine kinase (CK), catalase (CAT), glutathione peroxidase (GSH-Px), and glycogen storage in liver and muscle. Meanwhile, RES increased the expressions of ZO-1, Occludin, and Claudin-1, thereby enhancing intestinal barrier integrity and inhibiting mRNA expressions of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), and interleukin-1β (IL-1β) in the colon, thereby improving the pathological injury in the colon. Importantly, RES modified gut microbiota dysbiosis by increasing the diversity of gut microbiota, regulating microbiota associated with inflammation and fatty acid metabolism at the phylum ( Bacteroidetes and Firmicutes ), family ( Erysipelotrichaceae , Enterobacteriaceae, and Prevotellaceae ), and genus ( Brevundimonas diminuta , Coprobacillus , Megasphaera, and Lactobacillus ) levels, respectively. The results supplemented the anti-fatigue mechanism for RES from the perspective of intestinal injury and gut microbiota. The detailed mechanisms and associated metabonomics analysis remain for further study.

Aeromonas hydrophilais a Gram-negative bacterium that widely exists in various aquatic environments and causes septicemia in fish and humans. Resveratrol, a natural polyterpenoid product, has potential chemo-preventive and antibacterial properties. In this study, we investigated the effect of resveratrol onA. hydrophilabiofilm formation and motility. The results demonstrated that resveratrol, at sub-MIC levels, can significantly inhibit the biofilm formation ofA. hydrophila, and the biofilm was decreased with increasing concentrations. The motility assay showed that resveratrol could diminish the swimming and swarming motility ofA. hydrophila. Transcriptome analyses (RNA-seq) showed thatA. hydrophilatreated with 50 and 100 μg/mL resveratrol, respectively, presented 230 and 308 differentially expressed genes (DEGs), including 90 or 130 upregulated genes and 130 or 178 downregulated genes. Among them, genes related to flagellar, type IV pilus and chemotaxis were significantly repressed. In addition, mRNA of virulence factors OmpA, extracellular proteases, lipases and T6SS were dramatically suppressed. Further analysis revealed that the major DEGs involved in flagellar assembly and bacterial chemotaxis pathways could be regulated by cyclic-di-guanosine monophosphate (c-di-GMP)- and LysR-Type transcriptional regulator (LTTR)-dependent quorum sensing (QS) systems. Overall, our results indicate that resveratrol can inhibitA. hydrophilabiofilm formation by disturbing motility and QS systems, and can be used as a promising candidate drug against motile Aeromonad septicemia. Keywords:resveratrol;Aeromonas hydrophila;biofilm formation;motility;RNA-seq;quorum sensing

Two-component systems (TCS) of Salmonella enterica serovar Enteritidis are composed of a histidine kinase and a response regulator (RR) and represent a critical mechanism by which bacteria develop resistance to environmental stress. Here, we characterized the functions of RRs in TCS in the formation of stress tolerance, motility and biofilm using twenty-six S . Enteritidis RR-encoding gene deletion mutants. The viability results unraveled their essential roles in resistance to elevated temperature (GlrR), pH alterations (GlrR, TctD, YedW, ArcA and YehT), high salt (PhoB, BaeR, CpxR, PhoP, UvrY and TctD), oxidative stress (PhoB, YedW, BaeR, ArcA, PhoP, UvrY, PgtA and QseB) and motility (ArcA, GlnG, PgtA, PhoB, UhpA, OmpR, UvrY and QseB) of S . Enteritidis. The results of the crystal violet staining, microscopy observation and Congo red binding assays demonstrated that the absence of ArcA, GlnG, PhoP, OmpR, ZraR or SsrB in S . Enteritidis led to a reduction in biofilms and an impairment in red/dry/rough macrocolony formation, whereas the absence of UvrY exhibited an increase in biofilms and formed a brown/smooth/sticky macrocolony. The results indicated the regulatory effects of these RRs on the production of biofilm matrix, curli fimbriae and cellulose. Our findings yielded insights into the role of TCSs, making them a promising target for combating S . Enteritidis.

The motility of bacteria is crucial for navigating competitive environments and is closely linked to physiological activities essential for their survival, such as biofilm development. Precise regulation of bacterial motility enhances our understanding of these complex processes. While optogenetic tools have been used to control and investigate bacterial motility, the excitation light in most existing systems are limited to the visible light spectrum. Here, we introduce a new type of bio-microrobot comprising genetically engineered E. coli cells and orthogonally emissive upconversion nanoparticles that can respond to both 980 nm and 808 nm NIR light. This system allows toggling of bacterial states between tumbling and swimming via simply alternating the NIR light between different wavelengths. It is believed that the use of NIR light with deeper tissue penetration suggests potential applications for these bio-microrobots in areas like targeted drug delivery.

The N-Acy-L-homoserine lactones (AHLs) mediated quorum sensing (QS) system exhibited important ecological significance in bacterial biofilm formation. However, the previous studies mainly focused on indigenous AHLs while the role of exogenous AHLs has remained unclear. This study evaluated the roles of exogenous AHLs on the biofilm formation of Pseudomonas aeruginosa . Both the C6-HSL and C8-HSL promoted the biofilm formation of P. aeruginosa with an enhancement of 2.47 and 1.88 times, respectively. Further analysis showed that exogenous AHLs contributed greatly to the adhesive ability instead of growth rate. Also, the bacterial motility and metabolic activities were significantly improved by AHLs. Moreover, the microbial functional genes ( i.e. lasI , lasR , rhlI and rhlR ) involved in regulating the biofilm formation were highly expressed in AHLs reactors. These findings expanded the knowledge of AHLs functions in mediating biofilm formation, and provided insightful guidance on the biofilm regulation in the wastewater treatment via biofilm technology.

BACKGROUND Given the escalating challenges posed by antimicrobial resistance in phytobacterial infections, which are exacerbated by the suboptimal efficacy of existing bactericides, limited curative options, and mounting environmental concerns, there is a pressing need to develop innovative bactericidal agents with novel molecular architectures and distinctive modes of action. RESULTS To identify novel molecular scaffolds for bactericide development, we systematically devised a wide series of imidazo[1,2- a ]pyridine derivatives incorporating benzylpiperazinyl moieties, followed by evaluating their antibacterial activities. Bioassay results manifested that compound C 19 exhibited remarkable antibacterial efficacy against Xanthomonas oryzae pv. oryzae ( Xoo ) and Xanthomonas axonopodis pv. citri ( Xac ), with EC 50 values of 2.16 μg mL −1 and 4.64 μg mL −1 , respectively. Transcriptomic analysis initially revealed that compound C 19 involved significant downregulation of genes associated with biofilm, type III secretion system (T3SS), and flagella assembly. Subsequent multiple validations via biofilm formation assays, bacterial ultrastructure analysis, motility assays, virulence evaluation, and hypersensitive response (HR) assays, collectively indicated that compound C 19 both inhibited biofilm formation and suppressed swimming ability while reducing the expression of hrp family genes in T3SS, ultimately attenuating Xoo pathogenicity. Notably, Pot experiment demonstrated that compound C 19 exhibited superior control efficacy (protective activity: 42.74%; curative activity: 44.14%), surpassing commercial BT and TC. CONCLUSION Given its excellent antibacterial potency and the mechanism of action elucidated through transcriptome analysis and ample biochemical assays, compound C 19 represented a promising candidate for developing novel bactericides via targeting bacterial virulence to combat refractory plant bacterial diseases. © 2025 Society of Chemical Industry. Graphical This study successfully identified the benzylpiperazinyl-decorated imidazo[1,2- a ]pyridine derivative C 19 as a novel antibacterial alternative featuring a unique molecular scaffold and low toxicity for controlling plant bacterial diseases through virulence-targeting mechanisms.

Citrobacter werkmanii , an aerobe and mesophilic Proteobacterium, is universal in industrial putrefaction, coastal water, and human blood. Our previous studies have discovered that outer membrane protein X (OmpX) of C. werkmanii is involved in calcium response, but the underlying mechanisms and its molecular characteristics remain elusive. To that end, the ompX gene was deleted from the genome of C. werkmanii and its phenotypic variations were thoroughly investigated in conjunction with the wild type (WT) and complementary strains using biochemical and molecular techniques such as RNA-Seq, respectively. The results demonstrated that deleting ompX reduces biofilm formation on polystyrene and glass surfaces. Meanwhile, Δ ompX ’s swimming ability but not for its twitching or swarming abilities, was also reduced on semi-solid plates compared with WT, which was caused by inhibition of flagellar assembly genes, such as flgC , flhB , and fliE , etc. Furthermore, ompX inactivation altered susceptibility to various bactericide classes, as well as responses to Ca 2+ and Mg 2+ stress. In addition, when compared to WT, Δ ompX captures a total of 1,357 deferentially expressed genes (DEGs), of which 465 were up-regulated and 892 were down-regulated, which can be enriched into various GO ontology and KEGG pathway terms. Furthermore, ompX , as well as ompD and ompW , can be modulated at the transcriptional levels by rbsR and tdcA . Overall, the ompX gene contributed to a variety of biological functions in C. werkmanii and could be served as a targeted site for controlling biofilm formation and developing new bactericides.
