Capsaicin alleviates abnormal intestinal motility through regulation of enteric motor neurons and MLCK activity: Relevance to intestinal motility disorders

Dapeng Chen,Yongjian Xiong,Yuan Lin,Zeyao Tang,Jingyu Wang,Li Wang,Jihong Yao

Scope Capsaicin is an active component of chili peppers, having diverse effects. However, the effects of capsaicin on intestinal motility are still controversial. The present study aimed to investigate the effects of capsaicin on intestinal motility disorder and uncover related mechanisms. Materials and results A rat model with intestinal motility disorder was established in vitro through adding different stimuli into tissue bath; in vivo using constipation and diarrhea model, respectively. Capsaicin exerted dual effects on intestinal motility, i.e. the relaxation and contraction of jejunum induced by corresponding stimulus were, respectively, regulated to be normal contraction by capsaicin. The mechanisms underlined capsaicin-induced dual effects were investigated using Western blotting, qRT-PCR, and whole-cell patch clamp, respectively. Results showed that cholinergic excitatory nerves, adrenergic nerves, and neurons containing nitric oxide synthase, which are the main muscle motor neurons in enteric nervous system (ENS), are involved in capsaicin-induced dual effects. The competition for regulation of Ca 2+ influx by capsaicin induced the interaction with components of the ENS. Capsaicin significantly increased myosin light chain kinase (MLCK) expression and myosin phosphorylation extent in jejunal segments of constipation-prominent rats and significantly decreased MLCK expression and myosin phosphorylation extent in jejunal segments of diarrhea-prominent rats. Conclusion In summary, capsaicin alleviates abnormal intestinal motility through regulating enteric motor neurons and MLCK activity, which is beneficial for the treatment of gastrointestinal motility disorders.

Correlation between the translational and rotational diffusion of rod-shaped nanocargo on a lipid membrane revealed by single-particle tracking

Liangna He,Yiliang Li,Lin Wei,Zhongju Ye,Hua Liu,Lehui Xiao

Revealing the diffusion dynamics of nanoparticles on a lipid membrane plays an important role in a better understanding of the cellular translocation process and provides a theoretical basis for the rational design of delivery cargo. However, most studies focus on the investigation of the positional fluctuations of the nanocargo on the fluidic membrane, ignoring the contribution from orientational variation. In particular, less is known about the correlation between the rotational freedom and translational movability of a particle surveying a lipid membrane. In this work, the ligand–receptor interaction (by using streptavidin (SA) and biotin as the model)-modulated diffusion dynamics of rod-shaped nanocargo (i.e., gold nanorods, GNRs) on an artificial lipid membrane was explored with dark-field (DF) optical microscopy. A correlation between translational and rotational motion was observed whereby the freedom of rotational motion could be released intermittently. The conformational entropy release is usually associated with the promotion of translational diffusion, where large step surveying on the lipid membrane takes place subsequently. These new messages might afford valuable kinetic information for the design of nanocargo with appropriate surface functionality to achieve satisfactory cellular uptake efficiency.

A reliable, battery-free and implantable magnetic sensing system for wireless monitoring of spinal motion in rats and humans

Yunsong Shi,Sihan Tang,Weifeng Zhang,Chunchi Lei,Xiaobo Feng,Kun Wang,Yamei Yue,Qi Wang,Annan Chen,Jin Su,Hu Zhao,Chunze Yan,Yujia Tian,Kun Zhou,Jian Lu,Cao Yang,Bin Su

Continuous monitoring of spinal and joint motions in patients allows surgeons to optimize patient rehabilitation. However, the current monitoring approaches are achieved via tethered devices, which are inconvenient and associated with a risk of infections. In this study, we developed and validated a new type of reliable wireless, battery-free, and implantable sensing system (RWBS) for the real-time monitoring of physiological bending of spines and joints. The RWBS exploits the unique movement-sensing capabilities of an implantable flexible magnetic strip, the signals from which are detected with an external receiver. We implanted this magnetic strip beneath the skins of Sprague Dawley rats and human cadavers and demonstrated that in combination with the receiver, the magnetic strip enabled real-time in vivo and wireless monitoring of motions in both rats (anesthetized/awake) and human cadavers. The adaptability of RWBS in other human bones has been verified by sensing the motions of elbow joints and knee joints. The simple design of the RWBS allowed it to continue functioning in vivo after the magnetic strip had been broken into two or more pieces, indicating its high reliability. Thus, the RWBS is biocompatible, exhibits excellent functionalities, and can be conveniently implemented in both rats and humans. This device will help to facilitate the post-surgery monitoring of spinal and joint movements of human patients.

A novel non-invasive method for measuring the spatial kinematic behavior of cardiomyocytes regulated by mechanical cues

Si Tang,Huiyao Shi,Jialin Shi,Zhenghua Wang,Dingyi Wang,Chanmin Quanmin Su,Lianqing Liu

The intact heart undergoes complex and multiscale mechanical remodeling processes. Measuring rhythmic spatial contraction of the myocardium is crucial for assessing mechanical durability and the ability to mount coordinated responses to pressure, electrical, and hemodynamic signals. However, current cardiomyocyte measurement platforms typically focus on action potentials and XY-plane contractions. Therefore, effective evaluation methods for studying the influence of mechanical cues on the spatial dynamic contraction of cardiomyocytes are still lacking. In this study, we developed a topographic guiding combined with an optical spatial motion tracking method to provide controllable mechanical stimulation for inducing directed contraction of cardiomyocytes and obtaining spatial motion information in vitro . We first performed a detailed investigation of cell connections and cytoskeleton orientations by combining the proposed method with immunofluorescence. Next, spatial constrictive modes, features, and key parameters of microgroove-guided cardiomyocytes were studied. Finally, the three-dimensional (3D) motions of the cardiomyocytes at different positions on the structure were compared. We found that the XY-plane contraction of cardiomyocytes typically has only one direction and shows a significant phase delay compared to the axial motion. In addition, cardiomyocytes located near the edges of the microgrooves were restricted by stronger mechanical forces, resulting in a significant height change reduction. These results provide new perspectives for structural and functional research on cardiomyocytes under long-term mechanical regulation. Overall, this study provides a highly precise and convenient method for evaluating the 3D cardiomyocyte motion under mechanical induction. This method is expected to enhance understanding of cardiomyocyte development and be useful for research on cardiac mechanics and functions.

Self-powered locomotion of a hydrogel water strider

Hong Zhu,Borui Xu,Yang Wang,Xiaoxia Pan,Zehua Qu,Yongfeng Mei

Hydrogels are an exciting class of materials for new and emerging robotics. For example, actuators based on hydrogels have impressive deformability and responsiveness. Studies into hydrogels with autonomous locomotive abilities, however, are limited. Existing hydrogels achieve locomotion through the application of cyclical stimuli or chemical modifications. Here, we report the fabrication of active hydrogels with an intrinsic ability to move on the surface of water without operated stimuli for up to 3.5 hours. The active hydrogels were composed of hydrophobic and hydrophilic groups and underwent a dynamic wetting process to achieve spatial and temporal control of surface tension asymmetry. Using surface tension, the homogeneous active hydrogels propelled themselves and showed controlled locomotion on water, similar to common water striders.

Soft Crawling and Flipping Robots Based on Liquid Metal-Liquid Crystal Elastomer Composites

Yuan Ling-Xin,Liu Chang-Yue,Yang Ji-Ping,Wang Zhi-Jian

Soft robots have shown great advantages with simple structure, high degree of freedom, continuous deformation, and benign human-machine interaction. In the past decades, a variety of soft robots, including crawling, jumping, swimming, and climbing robots, have been developed inspired by living creatures. However, most of the reported bionic soft robots have only a single mode of motion, which limits their practical application. Herein, we report a fully 3D printed crawling and flipping soft robot using liquid metal incorporated liquid crystal elastomer (LM-LCE) composite as the actuator. With the application of voltage, liquid metal works as the conductive Joule heating material to induce the contraction of the LCE layer. The bending angle of the LM-LCE composite actuator highly depends on the applied voltage. We further demonstrate that the soft robot can exhibit distinct moving behaviors, such as crawling or flipping, by applying different voltages. The fully 3D printed LM-LCE composite structure provides a strategy for the fast construction of soft robots with diverse motion modes.

Striatal CDK5 Regulates Cholinergic Neuron Activation and Dyskinesia-like Behaviors through BK Channels

Chu Tong,Peng-Xiang Min,Qian Zhang,Ru-Xin Gu,Yao-Hai Wen,Yi Shi,Yu-Huan Bao,Xiang Chen,Yi-Xuan Zhang,Xing-Feng Mao,Hao-Yang Yuan,Xiu-Xiu Liu,Takuya Sasaki,Li Zhang,Feng Han,Ying-Mei Lu

Disturbance of the cholinergic system plays a crucial role in the pathological progression of neurological diseases that cause dyskinesia-like behaviors. However, the molecular mechanisms underlying this disturbance remain elusive. Here, we showed that cyclin-dependent kinase 5 (Cdk5) was reduced in cholinergic neurons of midbrain according to the single-nucleus RNA sequencing analysis. Serum levels of CDK5 also decreased in patients with Parkinson’s disease accompanied by motor symptoms. Moreover, Cdk5 deficiency in cholinergic neurons triggered paw tremors, abnormal motor coordination, and motor balance deficits in mice. These symptoms occurred along with cholinergic neuron hyperexcitability and increases in the current density of large-conductance Ca2+-activated K+ channels (BK channels). Pharmacological inhibition of BK channels restrained the excessive intrinsic excitability of striatal cholinergic neurons in Cdk5-deficient mice. Furthermore, CDK5 interacted with BK channels and negatively regulated BK channel activity via phosphorylation of threonine-908. Restoration of CDK5 expression in striatal cholinergic neurons reduced dyskinesia-like behaviors in ChAT-Cre;Cdk5f/f mice. Together, these findings indicate that CDK5-induced phosphorylation of BK channels involves in cholinergic-neuron-mediated motor function, providing a potential new therapeutic target for treating dyskinesia-like behaviors arising from neurological diseases.

Magnetic Shaftless Propeller Millirobot with Multimodal Motion for Small-Scale Fluidic Manipulation

Yaozhen Hou,Shihao Zhong,Zhiqiang Zheng,Jiabao Du,Ruhao Nie,Qing Shi,Qiang Huang,Huaping Wang

Magnetic miniature robots have shown great potential in biomedical applications in recent years. However, a challenge remains in which it is difficult for magnetic miniature robots to achieve balanced capabilities for multimodal locomotion and fluidic manipulation in various environments. Here, we report a magnetic shaftless propeller-like millirobot (MSPM) that possesses the capabilities of rotating-based multimodal 3-dimensional motion and cargo transportation with untethered manipulation. The MSPM utilizes the propulsion and pumping capabilities of the propeller structure to achieve fluidic manipulation. The shaftless propeller structures are designed to achieve omnidirectional locomotion through rolling, propelling, and tumbling. Additionally, the shaftless 3-blade propeller is used to perform a pumping function to achieve controllable transportation of fluids and particles. We anticipate that the MSPM holds great potential as a minimally invasive device for thrombosis treatment and targeted medicine delivery.

Exposure to amitriptyline disturbs behaviors in adult zebrafish and their offspring via altering neurotransmitter levels

Jie Tang,Anqi Liu,Kun Chen,Yanhong Shi,Xuchun Qiu

Amitriptyline (AMI), one of the widely used tricyclic antidepressants (TCAs), has become a pharmaceutical contaminant frequently detected in aquatic ecosystems. However, the impacts of AMI exposure and underlying mechanisms on fish are still limited. In this study, adult zebrafish (F0) were exposed to AMI at 0 (control), 0.8, and 8 μg/L for 14 days. Subsequently, the exposed zebrafish were paired for spawning, and their offspring (F1) were reared in an AMI-free medium until 5 days post-fertilization (dpf). This study aimed to assess variations in behaviors and neurotransmitter levels in both the F0 (at the end of the 14-day exposure) and F1 generations (at 5 dpf). As a result, waterborne AMI exposure significantly reduced the locomotor activity, frequency of body contact, and duration of chase in F0 zebrafish, and resulted in notable changes in monoamine neurotransmitter levels in their brains. Parental exposure to AMI significantly elevated the heart rate and eye movement but reduced the locomotor activity in the F1 zebrafish, also along with significant changes in monoamine neurotransmitters and acetylcholine. Furthermore, significant correlations between the changes in behavioral traits and neurotransmitter levels were identified in both F0 and F1 generations. Our findings confirm the critical role of monoamine modulation in the neurobehavioral toxicity of AMI on zebrafish and their offspring, and emphasize the importance of paying attention to its multigenerational effects on fish.

Polysaccharides from Millettia speciosa Champ. Ameliorates Excessive Exercise-Induced Fatigue by Regulating Gut Microbiota

Jinfeng Zhang,Wenkai Zhang,Longhui Yu,Qinru Yang,Wenjing Wang,Xinru Hu,Jingen Li,Guodong Zheng

This study aimed to explore the ameliorative effect of Millettia speciosa Champ. polysaccharides (MSP) on excessive exercise-induced fatigue (EF) mice. The physicochemical characterization and in vitro antioxidant activities were analyzed. The fatigue-related biochemical indexes were determined, and gut microbiota was analyzed by 16S rDNA sequencing. The results showed that MSP primarily consists of total carbohydrates, uronic acids, and proteins. It was mainly composed of glucose, arabinose, and galactose. Additionally, MSP showed good scavenging effects on free radicals. In the experiment of EF mice, MSP intervention effectively extended the pole-climbing and swimming time, significantly increased the levels of muscle and liver glycogen, blood glucose, adenosine triphosphate, glutathione peroxidase, and superoxide dismutase, while significantly decreased the levels of fatigue-related lactate dehydrogenase, creatine kinase, and urea nitrogen. Furthermore, MSP intervention reduced harmful bacteria (Helicobacter, Anaerotruncus, Erysipelatoclostridium, and Lachnospiraceae_FCS020_group), and enriched the beneficial bacteria (Lactobacillus, Alistipes, Ruminococcaceae, and Roseburia). Therefore, MSP may utilize the gut microbiota as a target to alleviate fatigue caused by excessive exercise.

Interaction between the oil droplet in water and wetted wall: Force model and motion law

Rong Feng,He Limin,Lǚ Yuling,Lu Xiaolei

To investigate the force model and motion law of oil droplets in water near the wetted wall, oil droplets with R 1  = 0.29–0.62 mm and oil films with R 2  = 1–6 mm are solved numerically. In addition to buoyancy, flow resistance, and added mass force, the film-induced force triggered by the wetted wall constraint is also introduced into the force model. The drainage process is described using the Stokes–Reynolds equation, and the Young–Laplace equation is used to calculate the pressure within the water film. The results show that the force model can be coupled with the Stokes–Reynolds–Young–Laplace model equation to better describe the drainage dynamics near the wetted wall. The pressure distribution law is closely related to the shape of the water film, especially when the oil–water interface is in the shape of a dimple, which can lead to the formation of negative pressure zones within the water film. The maximum pressure first grows in an exponential, then logarithmic pattern and eventually approaches the equivalent Laplace pressure. Around the critical size, the direction of the film-induced force changes and the form of action switches between driving and drag forces. The film-induced force's dominant effect is strongest when the curvature radius of the oil film is comparable to the droplet size.

Design and Motion Controllability of Emerging Hydrogel Micro/Nanorobots

Yang Liu,Ying Feng,Linlin Liu,Miao An,Huaming Yang

Micro/nanorobots (MNRs) are promising for biomedical applications due to their unconstrained nature and small enough size to pass through many tiny environments. However, the efficient movement of MNRs in liquid environments is still a challenge due to the low Reynolds number environment and the Brownian motion of particles. Herein, emerging MNRs with hydrogel-loaded magnetic particles are designed. The proposed hydrogel MNRs (HMNRs) exhibit biocompatible and controllable characteristics. The motion controllability of HMNRs is realized by applying oscillating magnetic field and customized magnetic field. Experimentally, it is demonstrated that the HMNR swarms driven by the oscillating magnetic field exhibit a faster motion speed than the MNR swarms composed of magnetic particles. The HMNRs show precise controllability of the movement in the complex pipeline under the control of customized magnetic field. This method can offer a more benign approach to the general production of HMNRs for biological applications.
