Tailored graphene oxide-doxorubicin nanovehicles via near-infrared dye-lactobionic acid conjugates for chemo-photothermal therapy

Chunzhi Huang,Xu Hu,Zhaosheng Hou,Jianbo Ji,Zhonghao Li,Yuxia Luan

Graphene oxide (GO), as a drug delivery carrier, has attracted considerable attention because of its interesting properties. However, GO tends to aggregate in aqueous solution. Amphiphilic molecules are usually necessary to stabilize GO. The introduction of these non-functional macromolecules on the one hand reduces drug loading, but on the other hand may cause unpredictable side effects. This study proposes a new strategy for stabilizing GO with a functional photothermal agent, IR820 (new indocyanine green) derivative. IR820 derivative results from the conjugation of active targeted lactobionic acid (LA) with IR820 for the formation of IR820-LA. IR820-LA features central aromatic groups that can associate with the GO basal plane through π-π interactions. The flanking moiety of hydrophilic LA and sulfonic groups thus provides steric stabilization of GO in aqueous solution. Moreover, IR820-LA endows GO/doxorubicin (GO/DOX) nanovehicles with fluorescence imaging ability and actively targeted chemo-photothermal therapy. Experimental results both in vitro and in vivo have indicated its good chemo-photothermal therapeutic effect according to its active tumor targeting ability and pH-sensitive drug release characteristics. Therefore, our GO/DOX/IR820-LA nanohybrids can be excellent nanoplatforms for active tumor-targeted chemo-photothermal therapy with imaging guidance.

Unveiling the Excited-State Dynamics and Interfacial Interactions in Dye-Sensitized NaNdF4 Nanoparticles for Efficient Photothermal Effect

Jiacheng Gong,Wusen Zhou,Zhuo Li,Xingjun Li,Wen Yuan,Xiaobo Gu,Qianqian Niu,Yan Liu,Jin Xu,Renfu Li,Datao Tu,Shan Lu,Xueyuan Chen

Near-infrared (NIR) dyes can overcome the weak absorption of lanthanide nanoparticles (NPs) by antenna sensitization, offering new avenues to develop efficient and versatile lanthanide nanomaterials. However, current research on dye-sensitized lanthanide NPs for photothermal conversion is still preliminary, and the involved excited-state dynamics and interfacial interactions remain elusive. Herein, steady-state/transient absorption spectroscopy and theoretical calculation are used to investigate the coordination and aggregation states of cypate dyes on NaNdF 4 NPs, revealing the influence of interfacial interactions on resonant energy transfer. Synergetic heat-generation mechanism of lanthanide cross-relaxation and dye intermolecular collisions is further proposed. The photothermal conversion efficiency of cypate-NaNdF 4 nanocomposites reaches 50.4%, outperforming those of typical photothermal materials with high NIR absorption. Moreover, the intersystem crossing of cypate can be inhibited due to the depopulation of the S 1 exciton via ET, thereby improving anti-photobleaching ability. These dye-sensitized NaNdF 4 nanocomposites exhibit superior photothermal effect, stability and NIR-II luminescence, showing great potential in theranostic applications.

Albumin-based fluorescence resonance energy transfer nanoprobes for multileveled tumor tissue imaging and dye release imaging

Xianghan Zhang,Xiaohan Gao,Jialin Zhou,Zhiqing Gao,Yingdi Tang,Zuhong Tian,Pengbo Ning,Yuqiong Xia

Tumor tissue imaging and drug release imaging are both crucial for tumor imaging and image-guided drug delivery. It is urgent to develop a multileveled tumor imaging platform to realize the multiple imaging applications. In this work, we synthesized an albumin-based fluorescence resonance energy transfer (FRET) probe Cy5/7@HSA NPs containing two near-infrared cyanine dyes (CyBI5 and CyBI7) with high FRET efficiency (97 %). Excellent brightness and efficient FRET inside Cy5/7@HSA NPs enabled high-sensitive cell imaging and tumor imaging. This unique nanoprobe imaged 4T1 tumor-bearing mice with high sensitivity (TBR = 5.2) at 24 h post-injection and the dyes penetrated the tumor interior around 4 h post-injection. The release of dyes from nanoprobes was also tracked. This result shows the strong potential of this albumin-based FRET nanoprobe as multileveled tumor imaging platform for in vivo tumor imaging, drug delivery and image-guided surgery.

Dextran-polylactide micelles loaded with doxorubicin and DiR for image-guided chemo-photothermal tumor therapy

Yanmei Shi,Ronghua Tan,Chan Yu,Ying Wan

Image-guided chemo-photothermal therapy based on near-infrared (NIR) theranostic agents has found promising applications in treating tumors. In this multimodal treatment, it is of critical importance to image real-time distribution of photothermal agents in vivo and to monitor therapeutic outcomes for implementing personalized treatment. In this study, an optimally synthesized dextran-polylactide (DEX-PLA) copolymer was assembled with doxorubicin (DOX) and DiR, a kind of NIR dye, to construct desirable micelles ((DiR + DOX)/DEX-PLA) for performing image-guided chemo-photothermal therapy. These (DiR + DOX)/DEX-PLA micelles had good physical and photothermal stability in aqueous media and showed high photothermal efficiency in vivo. Based on the H22-tumor-bearing mouse model, (DiR + DOX)/DEX-PLA micelles were found to accumulate inside tumors sustainably and to emit strong fluorescence signals for more than three days. The (DiR + DOX)@DEX-PLA micelles together with NIR laser irradiation were able to highly inhibit tumor growth or even eradicate tumors with one injection and two dose-designated 5-minute laser irradiations at the tumor site during 14 days of treatment. Furthermore, they showed almost no impairment to the body of the treated mice. These (DiR + DOX)@DEX-PLA micelles have confirmative translational potential in clinical tumor therapy on account of their persistent image-guided capacity, high antitumor efficacy and good in vivo safety.

In vivo irreversible albumin-binding near-infrared dye conjugate as a naked-eye and fluorescence dual-mode imaging agent for lymph node tumor metastasis diagnosis

Wenjuan Zhang,Sichen Song,Hanxun Wang,Qiu Wang,Dan Li,Shunzhe Zheng,Zhuangyan Xu,Haotian Zhang,Jian Wang,Jin Sun

Tumor metastases account for about 90% of cancer-related death, among which lymphatic metastases play a pivotal role . Therefore, high-efficiency sentinel lymph node (SLN) identification is significant for lymph node (LN) metastasis diagnosis in clinic. Herein, a novel in vivo covalent albumin-binding near-infrared (NIR) fluorescent IR820-maleimide conjugate (IR-Mal) is firstly designed as a SLN dual-mode imaging agent. The IR-Mal conjugate exhibits bright blue appearance and its large Stokes shift (over 100 nm) increases the fluorescent imaging resolution effectively. The fluorescence intensity of covalent albumin-binding IR-Mal (BSA-IR-Mal) complex is considerably stronger than that of IR-Mal. In vivo , IR-Mal could rapidly covalently bind the tissue interstitial albumin following subcutaneous administration and BSA-IR-Mal complexes could specifically accumulate on LN, and detect both normal and metastatic SLN through naked-eye and fluorescence imaging with high resolution. Moreover, the light stability and enhanced fluorescence intensity of BSA-IR-Mal complex facilitates its diagnosis accuracy. These findings suggest that such in vivo irreversible albumin-binding fluorescence conjugates could serve as a new agent for dual-mode imaging and have a great potential to be applied in the SLNs imaging and diagnosis.

Transmissive Mode Laser Micro-Ablation Performance of Ammonium Dinitramide-Based Liquid Propellant for Laser Micro-Thruster

Baosheng Du,Yongzan Zheng,Chentao Mao,Haichao Cui,Jianhui Han,Luyun Jiang,Jifei Ye,Yanji Hong

The transmissive mode laser micro-ablation performance of near-infrared (NIR) dye-optimized ammonium dinitramide (ADN)-based liquid propellant was investigated in laser plasma propulsion using a pulse YAG laser with 5 ns pulse width and 1064 nm wavelength. Miniature fiber optic near-infrared spectrometer, differential scanning calorimeter (DSC) and high-speed camera were used to study laser energy deposition, thermal analysis of ADN-based liquid propellants and the flow field evolution process, respectively. Experimental results indicate that two important factors, laser energy deposition efficiency and heat release from energetic liquid propellants, obviously affect the ablation performance. The results showed that the best ablation effect of 0.4 mL ADN solution dissolved in 0.6 mL dye solution (40%-AAD) liquid propellant was obtained with the ADN liquid propellant content increasing in the combustion chamber. Furthermore, adding 2% ammonium perchlorate (AP) solid powder gave rise to variations in the ablation volume and energetic properties of propellants, which enhanced the propellant enthalpy variable and burn rate. Based on the AP optimized laser ablation, the optimal single-pulse impulse (I)~9.8 μN·s, specific impulse (Isp)~234.9 s, impulse coupling coefficient (Cm)~62.43 dyne/W and energy factor (η)~71.2% were obtained in 200 µm scale combustion chamber. This work would enable further improvements in the small volume and high integration of liquid propellant laser micro-thruster.Keywords:laser micro-ablation;ammonium dinitramide;near-infrared dye;ammonium perchlorate;energy deposition;energy conversion;laser micro-thruster

Enhanced photothermal-photodynamic therapy for glioma based on near-infrared dye functionalized Fe3O4 superparticles

Xiaohui Wang,Zhipeng Li,Yadan Ding,Kexin Wang,Zhenkai Xing,Xue Sun,Wanqiu Guo,Xia Hong,Xiaojuan Zhu,Yichun Liu

It is challenging and urgent in neurosurgery to improve therapeutic effect of the most common and lethal malignant glioma in central nervous system. The superadditive therapeutic effect of photothermal therapy and photodynamic therapy holds great promise. Herein, a highly efficient photothermal-photodynamic therapy nanoplatform was developed on the basis of Fe 3 O 4 -IR806 superparticles. The introduction of the near-infrared (NIR) dye IR806 and the fabrication of the self-assembled structure of the superparticles warrant 3.5 times increase in the photothermal conversion efficiency compared with the Fe 3 O 4 nanoparticles . The Fe 3 O 4 -IR806 superparticles generated reactive oxygen species (ROS) with the NIR light irradiation by virtue of the dye IR806. Both in vitro and in vivo experiments demonstrated that the Fe 3 O 4 -IR806 superparticles exhibited negligible toxicity. The Fe 3 O 4 -IR806 superparticles efficiently inhibited the cultured glioma cells via photothermal ablation and ROS cytotoxicity in vitro, as well as suppressed the bearing glioma growth in vivo with the NIR light induction. Our results highlight the promise of the Fe 3 O 4 -IR806 superparticles for improved photothermal-photodynamic therapy of cancer.

Regulating the Interaction Between Near-Infrared Dye and Endogenous Albumin for Concurrent Imaging Skin Inflammation and Neovascularization After Flap Transplantation

Yijing Du,Xue Zheng,Yanli Gao,Zetao Dang,Yuewei Zhang,Shoujun Zhu

Concurrent imaging of skin inflammation and neovascularization is crucial for diagnosing and monitoring skin conditions, especially in flap transplantation. However, current imaging modalities in the clinic are often non-intuitive, have low resolution, or lack the ability to specifically target skin inflammation. Given that albumin can serve as a biomarker for the disruption of skin-vessel barrier (SVB), probes targeting skin inflammation typically need to specifically bind to endogenous albumin, which often results in high background signals. In this study, we screen a series of near-infrared (NIR) dyes for their in vivo covalent binding capabilities with endogenous albumin, and identify the optimal dye for achieving high-contrast imaging of skin inflammation in models of SVB disruption, with minimal interference from other tissues or organs (e.g., skin and muscle). Moreover, by utilizing an albumin-targeting dye with another albumin-escaping NIR-II dye with a non-overlapping emission wavelength, this work explores the concurrent imaging of skin inflammation and neovascularization after flap transplantation, affording to simultaneously assess skin inflammation and the restoration of blood supply.

Enzyme-sensitive gemcitabine conjugated albumin nanoparticles as a versatile theranostic nanoplatform for pancreatic cancer treatment

Haijie Han,Jinhui Wang,Tingting Chen,Lichen Yin,Qiao Jin,Jian Ji

Development of gemcitabine (GEM) nanocarriers as theranostic agents for pancreatic cancer chemotherapy has received extensive attention in recent years. A novel enzyme-sensitive albumin-based GEM delivery nanoplatform was developed in this research by simple conjugation of GEM to human serum albumin (HSA) via cathepsin B cleavable peptide GFLG and then complexing with near-infrared (NIR) dye IR780, forming a HSA-GEM/IR780 complex. The successful preparation of HSA-GEM/IR780 complex was confirmed by Matrix-Assisted Laser Desorption/Ionization Time of Flight Mass Spectrometry (MALDI-TOF-MS), UV–vis–NIR absorption spectra and fluorescent emission spectra . The in vivo performance of HSA-GEM/IR780 complex was carried out on BxPC-3 pancreatic tumor xenografted mice. As revealed by in vivo NIR imaging, HSA-GEM/IR780 exhibited enhanced accumulation and long-term retention in tumor tissues compared to free IR780. Meanwhile, compared to free GEM, the deamination of GEM nanovectors into inactive 2′,2′-difluorodeoxyuridine (dFdU) can be greatly suppressed, while the concentration of the activated form of GEM (gemcitabine triphosphate, dFdCTP) was significantly increased in tumor tissue, thus exhibiting superior tumor inhibition activity with minimal side effects.

β-Lactamase-Responsive Probe for Efficient Photodynamic Therapy of Drug-Resistant Bacterial Infection

Yue Xu,Haiyan Chen,Shufen Xu,Ji Liu,Yang Chen,Lijuan Gui,Hua Li,Ruixi Li,Zhenwei Yuan,Bowen Li

Several photosensitizers have recently been proposed as novel approaches against β-lactamase-producing drug-resistant bacteria. However, these reported photosensitizers are rarely used for accurate recognition of drug-resistant bacteria. To tackle this challenge, the structurally modified photosensitizer CySG-2 based on a lipophilic cationic heptamethine indocyanine near-infrared (NIR) dye (IR-780) and an important synthesis intermediate of cephalosporin antibiotic (GCLE) not only achieved the accurate recognition of TEM-1 methicillin-resistant Staphylococcus aureus (MRSA) successfully but also achieved antimicrobial photodynamic therapy (aPDT) in animal models infected by drug-resistant bacteria. Accurate enzyme recognition and efficient photodynamic therapy capabilities allow CySG-2 to achieve one stone with two birds. In addition, CySG-2 could also promote the eradication of internalized MRSA by facilitating the autophagy process, which is synergistic with its capacity of inducing reactive oxygen species generation under NIR laser irradiation for aPDT. Collectively, it is an effective multifunctional photosensitizer with the potential ability to guide the optimal use of different antibiotics and apply them in clinical treatment.

Dictating Excited State Pathways at the Perovskite Nanocrystals–NIR Dye Interface by Thermodynamics-Governed Kinetics

Jihao Jin,Qiaoyu Zhang,Rui Hu,Guoqiang Yang,Tianjun Yu,Jinping Chen,Yi Li,Yi Zeng

Elucidating the intricate mechanisms of excited state interactions within inorganic semiconductor–organic molecule conjugates is of great scientific and practical significance, as it enables the control of energy transfer and electron transfer processes across the inorganic–organic interface. Herein, we report the distinct excited state interactions upon selective excitation of individual components in a conjugate comprising perovskite nanocrystals (NCs) and the near-infrared (NIR) dye IR808. Transient absorption investigation reveals that upon excitation of IR808 by NIR light, electron transfer from excited singlet IR808 to the NCs occurs with a time constant of 8.2 ps. This is followed by charge recombination with a time constant of 407 ps, ultimately forming triplet IR808. In contrast, selective excitation of the NCs with red photons, a one-step triplet energy transfer (kTET = 1.22 ns–1) from the excited NCs to IR808 rather than stepwise charge transfers, is observed during the triplet IR808 formation. Energy transfer outcompetes charge transfers in this case despite both processes being thermodynamically favorable because charge transfer falls deeper into the inverted region of Marcus theory than the triplet energy transfer. Thermodynamics can govern the kinetics in the excited state interactions within NCs and organic hybrids, thereby enabling excitation wavelength-controlled excited state interaction pathways. The effective generation of triplet IR808 and the broad spectral response of the NCs-IR808 conjugate enable applications in the sensitization of singlet oxygen and photochemical upconversion through visible to NIR excitation.

Covalent Organic Framework-Supported Molecularly Dispersed Near-Infrared Dyes Boost Immunogenic Phototherapy against Tumors

Shaoju Gan,Xiaoning Tong,Yue Zhang,Jinhui Wu,Yiqiao Hu,Ahu Yuan

Photodynamic therapy (PDT) mediated by near-infrared (NIR) dyes is a promising cancer treatment modality; however, its use is limited by significant challenges, such as hypoxic tumor microenvironments and self-quenching of photosensitizers. These challenges hamper its utility in inducing immunogenic cell death (ICD) and triggering potent systemic antitumor immune responses. This study demonstrates that molecular dispersion of NIR dyes in nanocarriers can significantly enhance their ability to produce reactive oxygen species and potentiate synergistic PDT and photothermal therapy against tumors. Specifically, NIR dye indocyanine green (ICG) can be spontaneously adsorbed to covalent organic frameworks (COFs) via π–π conjugations to prevent intermolecular stacking interactions. Then, ICG-loaded COFs are ultrasonically exfoliated and coated with polydopamine (PDA) to construct a new phototherapeutic agent ICG@COF-1@PDA with enhanced efficacy. In conjunction with ICG@COF-1@PDA, a single round of NIR laser irradiation can induce obvious ICD, elicit antitumor immunity in colorectal cancer, and yield 62.9% inhibition of untreated distant tumors. ICG@COF-1@PDA also exhibits notable phototherapeutic efficacy against 4T1 murine breast to lung metastasis, a spontaneous metastasis mode for triple-negative breast cancers (TNBCs). Overall, this study reveals a novel nanodelivery system for molecular dispersion of NIR dyes, which may present new therapeutic opportunities against primary and metastatic tumors.
