Oligopeptide modified ICG/Cisplatin@CaCO3 for targeted osteosarcoma NIR-II imaging guided multimodal therapy

Li-chen Ji,Jia-qing Huang,Xu-gang Zhong,Qi-hong Sun,Yong Fan,Li Yin,Qiong Zhang,Yu Cai,Zeju He

Osteosarcoma, as the most prevalent primary malignant bone tumor, has consistently shown suboptimal clinical treatment outcomes. In this manuscript, we proposed an efficacious and novel phototherapy strategy for osteosarcoma based on a photoresponsive near-infrared (NIR) material, CaCO 3 -ICG-DDP-PEG-PT (CIDP-PT), which contained calcium carbonate (CaCO 3 ) and Cisplatin (DDP) via the gas dispersion method, and indocyanine green (ICG) was encapsulated through a nanoprecipitation reaction by employing DSPE-PEG 2000 -COOH to enhance the stability and hydrophilicity. The targeting oligopeptide (PT) for osteosarcoma cells was inserted into the outer membrane of nanoparticles, thereby further enhancing the tumor-targeting capability through receptor-mediated binding. The prepared CIDP-PT nanoparticles exhibited high targeting efficiency and multifunctionality. In vitro and in vivo experiments demonstrated that CIDP-PT significantly enhanced the therapeutic efficacy for osteosarcoma via the NIR-II photoresponsive and tumor-targeting approach. It achieves efficient tumor cell eradication through multiple modalities while maintaining an exceptional level of biocompatibility. Hence, the CIDP-PT nanocarriers possess considerable practical value for future clinical applications in adjunct osteosarcoma phototherapy.

Copolymerized carbon nitride nanoparticles for near-infrared II photoacoustic-guided synergistic photothermal/radiotherapy

Wu Min,Huang Yuxin,Huang Xiaoyu,Wang Fu,Wei Xunbin

Nanotheranostic agents that integrate diagnosis and treatment are promising for precision medicine, but they encounter some obstacles such as penetration depth and efficiency. In this study, novel carbon nitride-rose bengal nanoparticles (CN-RB NPs) with a graphite carbon nitride skeleton were synthesized by one-step thermal copolymerization. The enhanced absorption in the near-infrared-II region (NIR-II) endows CN-RB NPs with an excellent photothermal effect under 1064 nm laser irradiation, as well as an obvious photoacoustic signal for imaging in vivo. Interestingly, due to the introduced iodine element, CN-RB NPs exhibit enhanced radiation therapy, indicating that CN-RB NPs can achieve ideal therapeutic outcome through collaborative photothermal/radiation therapy under the guidance of NIR-II photoacoustic imaging. Moreover, CN-RB NPs demonstrate minimal side effects and long-term biological stability after 14 days. Therefore, the proposed new multifunctional nano-platform CN-RB NPs hold great potential in the application of deep therapeutics.

Ni2+-activated MgTi2O5 with broadband emission beyond 1200 nm for NIR-II light source applications

Chuan-Jun Tang,Bo-Mei Liu,Lin Huang,Jing Wang,Qiang Tang

The rapid development of minimized near-infrared (NIR) phosphor-converted LEDs (pc-LEDs) has evoked a large demand for broadband NIR phosphors and will enable breakthrough NIR spectroscopy applications. Consequently, the search for NIR pc-LEDs which can emit in the NIR-II window (1000–1700 nm) has become increasingly urgent. Herein, an unprecedented long-wavelength NIR MgTi2O5:Ni2+ phosphor that exhibits broadband NIR emission covering 1200 to 1700 nm with a full width at half maximum (FWHM) of 245 nm is successfully synthesized. By combining the MgTi2O5:Ni2+ phosphor with an ultraviolet LED chip, a high-performance NIR pc-LED is fabricated, which has a NIR output power of 3.6 mW at 300 mA driving current. These results indicate that the fabricated NIR-II pc-LED device is a promising candidate as a broadband light source for NIR spectroscopy applications including night vision, high-penetrating imaging, and information restoration. For instance, the secret information covered by ink can be recovered with the aid of an NIR-II pc-LED. This work not only provides a new material that has great potential as a NIR light source, but also demonstrates a set of approaches to expand the application of NIR spectroscopy and imaging technology.

Ultra-homogeneous Cu2−xSe nanoparticles as near-infrared II plasmonic phototheranstics for photothermal/chemodynamic synergistic therapy of liver cancer

Bei Li,Jing Li,Yi Fu,Xiwen Ye,Maodi Xie,Lei Feng,Xiaoya Niu,Zhen You

Laser mediated photothermal/chemodynamic co-therapy is widely applied for anti-cancer treatment due to its good therapeutic effect. However, the laser wavelength for effectively targeting deep tumor tissues can decide whether or not it is a key premise of irradiation treatment. In this study, a super-homogenous and highly dispersive spherical nanoparticle Cu 2-X Se was prepared, which displayed a strong surface plasmon resonance (SPR) peak in the near-infrared region (NIR)-II and high Fenton catalytic efficiency. The results of in vitro experiments showed that Cu 2-X Se nanoparticles exhibited high photothermal conversion efficiency under the excitation of NIR-II laser irradiation, which significantly improved its ability to produce ·OH. In a tumor-bearing mouse model, the combination of Cu 2-X Se and photothermal/chemodynamic co-therapy guided by the real-time photoacoustic imaging in the NIR-II could effectively inhibit tumor growth and exhibit good in vivo/in vitro biocompatibility. In summary, this Cu 2-X Se nanoparticle-based NIR-II laser-mediated photothermal/chemodynamic co-therapy strategy is expected to provide a new option for the diagnosis and treatment of liver cancer.

Hollow semimetal PtTe2 nanorods with high performance for photothermal/photodynamic/chemodynamic therapy in the NIR-II window

Mengyao Zhang,Xin Hu,Yudai Huang

Compared to the first near-infrared (NIR-I) window (650–1000 nm), the second near-infrared (NIR-II) window (1000–1700 nm) is more valuable for photothermal treatment (PTT) due to its deeper tissue penetration and lower phototoxicity. However, the therapeutic efficacy of PTT remains suboptimal owing to the inherently low photothermal conversion efficiency (PCE) of existing materials. Therefore, the development of nanomaterials with high PCE is a promising strategy for improving anti-tumor therapy. In this study, hollow semimetal PtTe 2 nanorods were synthesized. Their unique electronic properties and hollow structure contribute to achieving high PCE under NIR-II irradiation, while simultaneously promoting reactive oxygen species (ROS) generation. Additionally, PtTe 2 catalyzed the endogenous conversion of hydrogen peroxide (H 2 O 2 ) into oxygen (O 2 ) and hydroxyl radicals (•OH), further enhancing anti-tumor effects via chemodynamic therapy (CDT). In vivo experiments confirmed the excellent biocompatibility of PtTe 2 , and laser treatment following PtTe 2 administration significantly inhibited tumor growth, demonstrating its favorable therapeutic efficacy. This study establishes a new nanoplatform for the treatment of malignant tumors through photothermal/photodynamic/chemodynamic therapy, providing valuable insights for future research on multifunctional cancer therapies.

Polyvalent Aptamer-Functionalized NIR-II Quantum Dots for Targeted Theranostics in High PD-L1-Expressing Tumors

Xin Huang,Jiawei Zhu,Chuhuang Dong,Yuqing Li,Qing Yu,Xuan Wang,Zhejie Chen,Jiabei Li,Yu Yang,Haifang Wang

Ag2S quantum dots (QDs) show superior optical properties in the NIR-II region and display significant clinical potential with favorable biocompatibility. However, inherent defects of low targeting and poor solubility necessitate practical modification methods to achieve the theranostics of Ag2S QDs. Herein, we used rolling circle amplification (RCA) techniques to obtain long single-stranded DNA containing the PD-L1 aptamer and C-rich DNA palindromic sequence. The C-rich DNA palindromic sequences can specifically chelate Ag2+ and thus serve as a template to result in biomimetic mineralization and formation of pApt-Ag2S QDs. These QDs enable specific targeting and illuminate hot tumors with high PD-L1 expression effectively, serving as excellent molecular targeted probes. In addition, due to the high NIR-II absorption of Ag2S QDs, pApt-Ag2S QDs exhibit remarkable photothermal properties. And besides, polyvalent PD-L1 aptamers can recognize PD-L1 protein and effectively block the inhibitory signal of PD-L1 on T cells, enabling efficient theranostics through the synergistic effect of photothermal therapy and immune checkpoint blocking therapy. Summary, we enhance the biological stability and antibleaching ability of Ag2S QDs using long single-stranded DNA as a template, thereby establishing a theranostic platform that specifically targets PD-L1 high-expressing inflamed tumors and demonstrates excellent performance both in vitro and in vivo.

Multispectral photoacoustic imaging of cancer with broadband CuS nanoparticles covering both near-infrared I and II biological windows

Yubin Liu,Duyang Gao,Mengze Xu,Zhen Yuan

In this study, CuS nanoparticles with optical absorption covering both near-infrared I (NIR-I) and NIR-II biological windows were prepared and served as the contrast agents for multispectral photoacoustic imaging. The physiological parameters including concentrations of deoxyhemoglobin and oxyhemoglobin as well as the water content in the tumor location were quantified based on the multispectral photoacoustic reconstruction method. More importantly, the concentration of CuS nanoparticles/drugs accumulated in the tumor was also recovered after intravenously injection, which are essential for image-guided cancer theranostics. In addition, phantom and in vivo experimental tests were performed to inspect and compare the imaging depth and signal-to-noise ratio (SNR) between the two NIR biological windows. Interestingly, we discovered that a higher SNR was obtained in the NIR-II window than that in the NIR-I window. Meanwhile, the multispectral imaging results also demonstrated that the imaging contrast and penetration depth in the NIR-II window were also significantly improved as compared to those from the NIR-I window.

Photothermal Antibacterial Therapy of Near-Infrared II Region Laser Mediated by Gold Hollow Nanorod

Yan Wang,Shuai Pan,Wenqiong Zhang,Jinfeng Cai,Weijing Han,Zhaoqin Zhu

The current traditional treatment for bacterial infections is to treat them with antibiotics, and the misuse of antibiotics can lead to an increase in bacterial resistance. In contrast, the development of new antibiotics is much slower than the speed of adaptation of drug-resistant bacteria, making it necessary to develop a drug that does not rely on antibiotics. Therefore, based on the advantages of photothermal therapy, NIR II-responsive gold hollow nanorods (GHNRs) were developed to overcome the limitation of bacterial drug resistance in conventional bacterial therapy. GHNRs can quickly respond to a 980 nm laser with a high photothermal conversion efficiency of 41.78%. The high temperature produced by GHNRs can effectively kill Staphylococcus aureus and Escherichia coli, providing a new strategy for the clinical treatment of bacterial infectious diseases without antibiotic dependence.

A hemostatic carbon foam prevents postoperative tumor recurrence and metastasis via NIR-II light triggered drug release and hyperthermia

Xiuxia Wang,Shide Dong,Xinxi Li,Min Wang,Chunjie Yan,Huaming Yang,Yuting Gao

Postoperative tumor recurrence and metastasis have long been challenging issues for cancer patients. In this work, we have developed an implantable carbon foam that uniquely integrates second near-infrared (NIR-II) light triggered drug release, photothermal therapy, and rapid hemostasis by cross-linking a drug-loaded agarose gel with porous carbon foam (DOX@AG/CF). The functionalized carbon foam possessed outstanding photothermal conversion performance, achieving a rapid temperature rise exceeding 108 °C in just 4 s under a low power intensity of 0.25 W cm −2 with 1064 nm laser irradiation, thereby minimizing the risk of severe skin damage. Furthermore, DOX@AG/CF can regulate the release of the loaded drug though the temperature-responsive agarose gel, which synergistically facilitates postoperative therapy. As a result, such all-in-one gel/carbon foam composite, integrating postoperative hemostasis, photothermal therapy and photocontrolled chemotherapy, can effectively eradicate residual tumor cells and prevent the recurrence and metastasis of cancer in vivo . More importantly, the implanted carbon foam demonstrated excellent degradability and biocompatibility, presenting great potential in clinical application.

Rare Earth-Doped Nanoparticles for Near-Infrared II Image-Guided Photodynamic Therapy

Jiajia Meng,Yanyan Cui,Yaling Wang

Image-guided disease treatment not only significantly enhances the efficacy of disease treatment but also minimizes unnecessary medication doses. Near-infrared II (NIR-II, 1000–1700 nm) bioimaging has been considered as one of the ideal bioimaging techniques in recent years due to its deep tissue penetration and low tissue fluorescence scattering. Here, we design the highly biocompatible lanthanide-doped nanoparticles NaYF4:Ce/Er@NaYbF4:Er@NaYF4:Nd-ZnPc/RB/mPEG-FA (RENPs-ZnPc/RB/mPEG-FA). Under an 808 nm excitation, the internal rare earth-doped nanoparticles (RENPs) can emit NIR-II luminescence for imaging. In addition, the generated upconversion luminescence in the visible region can be used as an excitation light source to excite the photosensitizers on the surface of the RENPs to generate singlet oxygen for antitumor photodynamic therapy. Based on the guidance of NIR-II imaging, the intratumoral injection approach is selected. During the treatment (day 7) after the second dose of RENPs-ZnPc/RB/mPEG-FA injection, the tumors are almost inhibited and eliminated, and there is no sign of recurrence in the follow-up observations. Interestingly, intravenously injected nanoparticles also enable high-resolution skeletal imaging with a high signal-to-background ratio, which holds promise for the clinical translation of RENPs-ZnPc/RB/mPEG-FA as versatile probes for imaging and therapy.

Single-molecular phosphorus phthalocyanine-based near-infrared-II nanoagent for photothermal antitumor therapy

Li-Na Zhou,Houhe Pan,Jing-Lan Kan,Qun Guan,Yang Zhou,Yu-Bin Dong

As one of the noninvasive cancer treatments, photothermal therapy (PTT) has drawn intense attention recently. In this context, an important task is to explore novel and versatile nanoscale photothermal agents (PTAs), especially those with strong NIR-II light absorption, high photothermal conversion efficiency, good photostability and biocompatibility. Phthalocyanines (Pcs), as the second-generation photosensitizers, are a promising class of candidates for PTT due to their strong NIR absorption and high photothermal conversion efficiency. However, the poor water solubility severely limited their application as PTAs in tumor treatment. Herein, we report a molecular phosphorus phthalocyanine (P-Pc)-based nanoagent via incorporation of human serum albumin (HSA) under mild conditions. The obtained nanoscale P-Pc-HSA possesses excellent photothermal conversion efficiency (64.7%) upon 1064 nm light irradiation, furthermore, it can be a highly efficient NIR-II antitumor nanoagent via photothermal treatment (PTT), which is fully evidenced by the in vitro and in vivo experiments.

NIR-II-driven and glutathione depletion-enhanced hypoxia-irrelevant free radical nanogenerator for combined cancer therapy

Zhang Li,Fan Yadi,Yang Zhe,Yang Mo,Wong Chun-Yuen

Background Though the combination of photodynamic therapy (PDT) and chemodynamic therapy (CDT) appears to be very attractive in cancer treatment, hypoxia and overproduced glutathione (GSH) in the tumor microenvironment (TME) limit their efficacy for further application. Results In this work, a smart hypoxia-irrelevant free radical nanogenerator (AIPH/PDA@CuS/ZIF-8, denoted as APCZ) was synthesized in situ via coating copper sulphide (CuS)-embedded zeolitic imidazolate framework-8 (ZIF-8) on the free radical initiator 2,2′-azobis[2-(2-imidazolin-2-yl)propane]-dihydrochloride (AIPH)-loaded polydopamine (PDA). APCZ showed promising GSH-depleting ability and near-infrared (NIR)-II photothermal performance for combined cancer therapy. Once internalized by 4T1 cells, the outer ZIF-8 was rapidly degraded to trigger the release of CuS nanoparticles (NPs), which could react with local GSH and sequentially hydrogen peroxide (H2O2) to form hydroxyl radical (·OH) for CDT. More importantly, the hyperthermia generated by APCZ upon 1064 nm laser excitation not only permitted NIR-II photothermal therapy (PTT) and promoted CDT, but also triggered the decomposition of AIPH to give toxic alkyl radical (·R) for oxygen-independent PDT. Besides, the PDA together with CuS greatly decreased the GSH level and resulted in significantly enhanced PDT/CDT in both normoxic and hypoxic conditions. The tumors could be completely eradicated after 14 days of treatment due to the prominent therapeutic effects of PTT/PDT/CDT. Additionally, the feasibility of APCZ as a photoacoustic (PA) imaging contrast agent was also demonstrated. Conclusion sThe novel APCZ could realize the cooperative amplification effect of free radicals-based therapies by NIR-II light excitation and GSH consumption, and act as a contrast agent to improve PA imaging, holding tremendous potential for efficient diagnosis and treatment of deep-seated and hypoxic tumors.Graphic abstract
