Currently, photodynamic therapy (PDT) is restricted by the laser penetration depth. Except for PDT at 1064 nm wavelength excitation, the development of other NIR-II-activated nanomaterials with a higher response depth is still hindered and rarely reported in the literature. To overcome these problems, we fabricated a nanoplatform with heterostructures that generate reactive oxygen species (ROS) and ferrite nanoparticles under a high concentration of zinc doping (ZnxFe3–xO4 NPs), which can achieve oxidative damage of tumor cells under near-infrared (NIR) illumination. The recombination of photoelectrons and holes has been markedly inhibited due to the formation of heterostructures in the interfaces, thus greatly enhancing the capability for ROS and oxygen production by modulating the single-component doping content. The efficiency of PDT was verified by in vivo and in vitro assays under NIR light. Our results revealed that NIR-II (1208 nm) light irradiation of ZnxFe3–xO4 NPs exerted a remarkable antitumor activity, superior to NIR-I light (808 nm). More importantly, the reported ZnxFe3–xO4 NPs strategy provides an opportunity for the success of comparison with light in the first and second near-infrared regions.

Photothermal therapy (PTT) has gained significant attention as a non-invasive treatment in clinical oncology. However, the translation of PTT into clinical practice remains constrained by three fundamental limitations: acquired thermal tolerance in tumor cells, restricted light penetration depth in biological matrices, and insufficient therapeutic outcomes from single-modality treatment. To address these issues, a strategy for forming in situ complexes between near-infrared-II (NIR-II) photothermal agents and proteins is developed, aimed at damaging protein conformation and enhancing PTT effectiveness. We developed a nanoplatform called PCy-SF, consisting of the NIR-II photothermal polymer (PCy) and sorafenib (SF). PCy-SF responds to the tumor microenvironment (TME), specifically releasing Cy-CHO and sorafenib from the assemblies. The released Cy-CHO covalently binds to proteins, forming Cy-Protein complexes that activate NIR-II fluorescence, facilitating NIR-II imaging-guided photothermal therapy. Concurrently, the released SF intensifies microvascular damage, synergizing with PTT for enhanced therapeutic efficacy. Notably, PCy-SF induces a strong anticancer immune response, effectively suppressing tumor recurrence and metastasis. This study introduces a promising protein deactivation strategy for achieving mild-temperature PTT, offering broader applicability of PTT and insights for sensitizing tumors to photothermal therapy. Together, this innovative approach combining NIR-II photothermal agents with protein complexation and a responsive nanoplatform enhances PTT precision and efficacy, demonstrating significant potential in the field of cancer nanomedicine.

The availability of colloidal nano-materials with high efficiency, stability, and non-toxicity in the near infrared-II range is beneficial for biological diagnosis and therapy. Rare earth doped nanoparticles are ideal luminescent agents for bio-applications in the near infrared-II range due to the abundant energy level distribution. Among them, both excitation and emission range of Er3+ ions can be tuned into second biological window range. Herein, we report the synthesis of ∼15 nm LiYF4, NaYF4, and NaGdF4 nanoparticles doped with Er3+ ions and their core-shell structures. The luminescent properties are compared, showing that Er3+ ions with single-doped LiYF4 and NaYF4 nanoparticles generate stronger luminescence than Er3+ ions with doped NaGdF4, despite the difference in relative intensity at different regions. By epitaxial growth an inert homogeneous protective layer, the surface luminescence of the core-shell structure is further enhanced by about 5.1 times, 6.5 times, and 167.7 times for LiYF4, NaYF4, and NaGdF4, respectively. The excellent luminescence in both visible and NIR range of these core-shell nanoparticles makes them potential candidate for bio-applications.

Near-infrared-II (NIR-II) luminescence, which serves as a promising spectral window for biomedical applications and spectral analysis, has numerous potential applications. In this work, the NIR-II luminescence is realized in Li4SrCa(SiO4)2:Cr4+ phosphors. Under 465 nm excitation, the emission peak of Li4SrCa(SiO4)2:3%Cr4+ is located at 1215 nm and the full width at half maximum (FWHM) is more than 230 nm, covering the ultra-long range of 950–1600 nm. The photoluminescence spectra, diffuse reflectance spectra, Raman spectra and time-resolved emission spectra confirm the NIR-II luminescence mechanism. Furthermore, a novel post-reduction strategy is proposed to realize the leap-forward enhancement of NIR-II luminescence from Cr4+. By mutually substituting Sr2+ and Ca2+, a series of Li4Sr1+zCa1−z(SiO4)2:3%Cr4+ (z = −1 to 1) samples are prepared to investigate the universality and applicable conditions of the post-reduction strategy. Benefitting from the NIR-II emission of Cr4+, it has great application potential in optical temperature measurement and spectral analysis.

Osteosarcoma, as the most prevalent primary malignant bone tumor, has consistently shown suboptimal clinical treatment outcomes. In this manuscript, we proposed an efficacious and novel phototherapy strategy for osteosarcoma based on a photoresponsive near-infrared (NIR) material, CaCO 3 -ICG-DDP-PEG-PT (CIDP-PT), which contained calcium carbonate (CaCO 3 ) and Cisplatin (DDP) via the gas dispersion method, and indocyanine green (ICG) was encapsulated through a nanoprecipitation reaction by employing DSPE-PEG 2000 -COOH to enhance the stability and hydrophilicity. The targeting oligopeptide (PT) for osteosarcoma cells was inserted into the outer membrane of nanoparticles, thereby further enhancing the tumor-targeting capability through receptor-mediated binding. The prepared CIDP-PT nanoparticles exhibited high targeting efficiency and multifunctionality. In vitro and in vivo experiments demonstrated that CIDP-PT significantly enhanced the therapeutic efficacy for osteosarcoma via the NIR-II photoresponsive and tumor-targeting approach. It achieves efficient tumor cell eradication through multiple modalities while maintaining an exceptional level of biocompatibility. Hence, the CIDP-PT nanocarriers possess considerable practical value for future clinical applications in adjunct osteosarcoma phototherapy.

Background Fluorescence imaging as the beacon for optical navigation has wildly developed in preclinical studies due to its prominent advantages, including noninvasiveness and superior temporal resolution. However, the traditional optical methods based on ultraviolet (UV, 200–400 nm) and visible light (Vis, 400–650 nm) limited by their low penetration, signal-to-noise ratio, and high background auto-fluorescence interference. Therefore, the development of near-infrared-II (NIR-II 1000–1700 nm) nanoprobe attracted significant attentions toward in vivo imaging. Regrettably, most of the NIR-II fluorescence probes, especially for inorganic NPs, were hardly excreted from the reticuloendothelial system (RES), yielding the anonymous long-term circulatory safety issue. Results Here, we develop a facile strategy for the fabrication of Nd3+-doped rare-earth core–shell nanoparticles (Nd-RENPs), NaGdF4:5%Nd@NaLuF4, with strong emission in the NIR-II window. What’s more, the Nd-RENPs could be quickly eliminated from the hepatobiliary pathway, reducing the potential risk with the long-term retention in the RES. Further, the Nd-RENPs are successfully utilized for NIR-II in vivo imaging and magnetic resonance imaging (MRI) contrast agents, enabling the precise detection of breast cancer. Conclusion sThe rationally designed Nd-RENPs nanoprobes manifest rapid-clearance property revealing the potential application toward the noninvasive preoperative imaging of tumor lesions and real-time intra-operative supervision.Graphical abstract

Photothermal therapy (PTT) is a promising local therapy playing an increasingly important role in tumor treatment. To maximize PTT efficacy, various near-infrared photoabsorbers have been developed. Among them, metal sulfides have attracted considerable interest due to the advantages of good stability and high photothermal conversion efficiency. However, the existing synthesis methods of metal-sulfide-based photoabsorbers suffer from the drawbacks of complicated procedures, low raw material utilization, and poor universality. Herein, we proposed a flexible, adjustable strategy capable of transforming commercial metal sulfides into injectable hydrogels for local PTT. We took copper sulfide (CuS) as a typical example, which has intense second-window near-infrared absorption (1064 nm), to systematically investigate its in vitro and in vivo characteristics. CuS hydrogel with good syringeability was synthesized by simply dispersing commercial CuS powders as photoabsorbers in alginate-Ca2+hydrogel. This synthesis strategy exhibits the unique merits of an ultra-simple synthesizing process, 100% loading efficiency, good biocompatibility, low cost, outstanding photothermal capacity, and good universality. The in vitro experiments indicated that the hydrogel exhibits favorable photothermal heating ability, and it obviously destroyed tumor cells under 1064 nm laser irradiation. After intratumoral administration in vivo, large-sized CuS particles in the hydrogel highly efficiently accumulated in tumor tissues, and robust local PTT was realized under mild laser irradiation (0.3 W/cm2). The developed strategy for the synthesis of CuS hydrogel provides a novel way to utilize commercial metal sulfides for diverse biological applications. Keywords:commercial copper sulfide (CuS);alginate;hydrogel;photothermal therapy (PTT);near-infrared II windows

The development of a NIR-II (near-infrared-II: 1000–1700 nm) nanophotoswitch is urgently needed, due to its deeper tissue penetration and higher resolution imaging. In this work, a new type of NIR-II upconversion (UC) and downconversion (DC) nanophotoswitch (NaErF4:Ho@NaYF4@NaYbF4:Tm@NaYF4 core–multishell nanoparticles) is presented. When the switch is open, ultraviolet (UV)–blue emissions and intense red UC (Er3+:654 nm) and NIR-II DC (Er3+:1520 nm) emissions are simultaneously obtained under 980 nm NIR-I (near-infrared-I: 700–1000 nm) excitation, which shows potential application for real-time visible- and NIR-II-imaging in the photodynamic therapy (PDT) process. When the switch is closed, UV–blue emissions disappear while red UC (Er3+:654 nm) and NIR-II UC (Ho3+:1154 nm) emissions remain under 1532 nm NIR-II excitation, which may be used for real-time visible- and NIR-II-imaging before conducting PDT. This novel NIR-II nanophotoswitch displays special advantages in the biomedical field.

Nanotheranostic agents that integrate diagnosis and treatment are promising for precision medicine, but they encounter some obstacles such as penetration depth and efficiency. In this study, novel carbon nitride-rose bengal nanoparticles (CN-RB NPs) with a graphite carbon nitride skeleton were synthesized by one-step thermal copolymerization. The enhanced absorption in the near-infrared-II region (NIR-II) endows CN-RB NPs with an excellent photothermal effect under 1064 nm laser irradiation, as well as an obvious photoacoustic signal for imaging in vivo. Interestingly, due to the introduced iodine element, CN-RB NPs exhibit enhanced radiation therapy, indicating that CN-RB NPs can achieve ideal therapeutic outcome through collaborative photothermal/radiation therapy under the guidance of NIR-II photoacoustic imaging. Moreover, CN-RB NPs demonstrate minimal side effects and long-term biological stability after 14 days. Therefore, the proposed new multifunctional nano-platform CN-RB NPs hold great potential in the application of deep therapeutics.

Small interfering RNA (siRNA) targeting the NOD-like receptor family pyrin domain-containing 3 (NLRP3) inflammasome has emerged as a promising therapeutic strategy to mitigate infarct volume and brain injury following ischemic stroke. However, the clinical translation of siRNA-based therapies is significantly hampered by the formidable blood-brain barrier (BBB), which restricts drug penetration into the central nervous system. To address this challenge, we have developed an innovative long-circulating near-infrared II (NIR-II) nanoparticle platform YWFC NPs, which is meticulously engineered to enhance BBB transcytosis and enable efficient delivery of siRNA targeting NLRP3 (siNLRP3@YWFC NPs) in preclinical models of ischemic stroke. Furthermore, we integrated advanced deep learning neural network algorithms to optimize in vivo NIR-II imaging of the cerebral infarct penumbra, achieving an improved signal-to-background ratio at 72 h poststroke. In vivo studies employing middle cerebral artery occlusion (MCAO) mouse models demonstrated that image-guided therapy with siNLRP3@YWFC NPs, guided by prolonged NIR-II imaging, resulted in significant therapeutic benefits.

Lanthanide-doped near-infrared-II (NIR-II) luminescent nanoparticles have shown great potential in many fields. However, most previous studies were focused on the sensitizer-activator coupled systems in which the NIR-II luminescence intensity is seriously restricted by the low dopants concentration . Improving the NIR-II emission intensity in lanthanide-doped nanoparticles has remained a daunting challenge. Herein, we report a systematic research on the enhancement of the NIR-II luminescence of Er 3+ through lanthanide-mediated energy modulation. It is found that introducing proper content of Yb 3+ , Ce 3+ , Tm 3+ , and Ho 3+ in the erbium sublattice can indeed enhance the NIR-II emission under 980 and 808 nm excitations by mediating the energy transfer processes. Doping of other lanthanide ions (e.g., Pr 3+ , Tb 3+ , Dy 3+ , and Sm 3+ ) only led to a quick decline in luminescence intensity. The NIR-II emission was further enhanced by designing a core-shell-shell nanostructure which consists of an NaYbF 4 interlayer to maximize the absorption of 980 nm excitation. The NIR-II luminescence exhibits superior penetration depth in biological tissue, showing great promise in biological applications .

The rapid development of minimized near-infrared (NIR) phosphor-converted LEDs (pc-LEDs) has evoked a large demand for broadband NIR phosphors and will enable breakthrough NIR spectroscopy applications. Consequently, the search for NIR pc-LEDs which can emit in the NIR-II window (1000–1700 nm) has become increasingly urgent. Herein, an unprecedented long-wavelength NIR MgTi2O5:Ni2+ phosphor that exhibits broadband NIR emission covering 1200 to 1700 nm with a full width at half maximum (FWHM) of 245 nm is successfully synthesized. By combining the MgTi2O5:Ni2+ phosphor with an ultraviolet LED chip, a high-performance NIR pc-LED is fabricated, which has a NIR output power of 3.6 mW at 300 mA driving current. These results indicate that the fabricated NIR-II pc-LED device is a promising candidate as a broadband light source for NIR spectroscopy applications including night vision, high-penetrating imaging, and information restoration. For instance, the secret information covered by ink can be recovered with the aid of an NIR-II pc-LED. This work not only provides a new material that has great potential as a NIR light source, but also demonstrates a set of approaches to expand the application of NIR spectroscopy and imaging technology.
