Core/Shell-Structured Ag2Te/Ag2Se Quantum Dots for High-Resolution In Vivo Fluorescence Imaging in the Near Infrared IIb Region

Kai Wang,Ku-Han Deng,Yi-Shen Tian,Meng-Ying Sun,Zi-li Yu,Zhi-Quan Tian,Zhi-Ling Zhang

Fluorescence in the near infrared IIb window (NIR-IIb, 1500–1700 nm) shows higher signal-to-noise ratio, better space-time resolution, and deeper tissue penetration. However, materials with excellent fluorescence properties in the NIR-IIb region were rarely reported. Ag2Te quantum dots (QDs) could emit variable fluorescence in the NIR-II window due to their narrow band gap. Unfortunately, the fluorescence in the NIR-IIb region was not strong enough to image in vivo because the lattice defects on the surface of Ag2Te increased non-radiative transitions and decreased fluorescence performance. In this work, we coated a Ag2Te core with a Ag2Se shell to passivate the lattice defects and enhance its fluorescence efficiency. The fluorescence property of the as-prepared Ag2Te/Ag2Se core/shell QDs is more than 10 times stronger than that of Ag2Te. The brighter Ag2Te/Ag2Se QDs showed high-resolution in vivo imaging in the NIR-IIb region to distinguish thin blood vessels up to 66 μm. Overall, Ag2Te/Ag2Se QDs with excellent fluorescence in the NIR-IIb region were prepared and showed great potential for biomedical imaging.

Biodegradable Near-Infrared-IIb Rare-Earth Nanoprobe Enables Time-Programmable Neuroimaging

Xue Zheng,Yijing Du,Zetao Dang,Ke Lan,Guantong Liu,Jingyan Ren,Xin Wang,Shoujun Zhu

Neural imaging plays a crucial role in understanding the structure and function of the living body. While near-infrared-II (NIR-II) imaging techniques have emerged as potential tools for non-invasive and high-resolution imaging of neural activity, there have been rare relevant reports on in vivo neural imaging. This study presents the development of biodegradable NIR-II rare-earth nanoprobes as a novel contrast agent for enhanced neural imaging. The biodegradable NIR-II rare-earth nanoprobes exhibit excellent long wavelength emission, high quantum yield, and tunable biodegradation time in vivo. This work demonstrates the effectiveness of these nanoprobes in enhancing the imaging depth and resolution in a mouse model of neural activity, thereby facilitating a better understanding of the structure and function of the nervous system. Furthermore, their biodegradability ensures minimal long-term toxicity and allows for repeatedly monitoring of damaged nerves during the recovery or deterioration process. The results highlight the potential of biodegradable NIR-II rare-earth nanoprobes for advancing neural imaging techniques and facilitating deeper insights into neuroscience research and clinical diagnostics.

NIR-IIb-triggered photodynamic therapy combined with chemotherapy platform based on rare-earth-doped nanoparticles

Tu Zhuo,Wan Yong,Ge Juan,Li Chen-Chen,Liang Tao,Li Zhen

Abstract Photodynamic therapy (PDT), with high spatiotemporal selectivity, minimal side effects and less drug resistance, has emerged as a promising approach for the treatment of cancer. However, inadequate penetration depth and poor efficacy seriously restrict its practical application. To address these challenges, we developed a combined therapy platform (RENP@C/D-FA) based on rare-earth-doped nanoparticles (RENPs) with a structure of NaErF 4 :Tm(0.5%)@NaYF 4 , on which photosensitizer Ce6 (chlorin e6) and chemo-therapeutic drug (DOX) (doxorubicin) were loaded by encapsulating with an amphiphilic polymer distearoyl phosphatidylethanolamine-polyethylene glycol-folic acid (DSPE-PEG-FA). RENPs with NaErF 4 as the matrix possess multiple excitation/emission peaks, enabling activation of PDT under near-infrared-IIb (NIR-IIb, 1500–1700 nm) excitation and luminescence imaging in NIR-IIb region by absorbing 808 nm near-infrared photons. Furthermore, DOX can be selectively released in the acidic microenvironment of tumors. Benefiting from the tumor targeting capability of folic acid, this platform can successfully ablate tumor cells and inhibit tumor growth by combining PDT and chemotherapy. This innovative approach holds great promise as a powerful tool for cancer treatment, showing the potential to revolutionize PDT. Graphical abstract

Aggregation-induced emission dots assisted non-invasive fluorescence hysterography in near-infrared IIb window

Xiaoming Yu,Yanyun Ying,Zhe Feng,Ji Qi,Junyan Zheng,Yuhuang Zhang,Juan Liu,Jun Qian,Ben Zhong Tang,Dan Zhang

Uterine diseases seriously threaten the physical and mental health of women. The main principle, when clinicians adopt examinations, is to achieve efficient diagnosis without negative effect on the physical function including fertility. Hysterography in near-infrared (NIR) IIb window (1500–1700 nm) presents perceptibly enhanced signal to background ratio (SBR) and higher penetration capability compared with those beyond 1000 nm and 1300 nm, but lays down high requirements for the biosafety of fluorophores at the same time. Assisted by the biologically excretable aggregation-induced emission (AIE) dots, non-invasive NIR-IIb fluorescence hysterography visualized typical Y-shaped uteruses, real-time uterine peristalsis or the uterine lesions (mimetic disease statuses in clinic) in mouse models. Significantly, after intrauterine perfusion, the reproductive capacity was unimpaired via fertility assessment and histological analysis. This work could inspire some new ideas for non-invasive clinical diagnosis of uterine diseases and effectively promote the clinical translation of AIE dots.

Plasmonic-Fluorescent Janus Au-PbS Nanoparticles with Bright Near-Infrared-IIb Fluorescence and Photothermal Effect for Computed Tomography Imaging-Guided Combination Cancer Therapy

Xue-Hui Shi,Leiming Tao,Lei Wang,Xing Liu,Shu-Lin Liu,Zhi-Gang Wang

In recent years, the application of both fluorescence and photothermal systems in one system has received great attention in the biomedical field. Yet, synthesizing a nanomaterial with both fluorescence (radiative decay) and a photothermal effect (nonradiative decay) is rarely reported. Here, we proposed novel plasmonic-fluorescent Janus Au-PbS nanoparticles (NPs) with tunable near-infrared II (NIR-II) fluorescence and photothermal effect to be used as a highly effective nanotheranostic for dual-modal NIR-IIb fluorescence imaging (FI)/computed tomography imaging (CT)-guided photothermal therapy. In addition, we investigated the equilibrium mechanism between the fluorescence and photothermal effects to guide the synthesis of this type of nanomaterial and found that the fluorescent semiconductor PbS quantum dots (QDs) receive electrons from Au NPs in Janus Au-PbS NPs, which could maintain the good fluorescence properties of Janus Au-PbS NPs. In addition, the alteration of electron and hole recombination between PbS QDs and Au NPs in Janus Au-PbS NPs increased nonradiative decay, leading to enhanced photothermal effects of Janus Au-PbS NPs. The electron receiver and electron–hole interactions in Janus Au-PbS NPs offered a fundamentally new means of engineering materials with fluorescence and photothermal properties through tailoring electron redistribution in Janus NPs.

Ultrasound-Activatable Nanoprobes for Imaging-Guided Inhibition of Immunosuppressor Production in Tumor Microenvironment

Jian Guo,Wen-Ping Lin,Tao Tang,Fushou Liu,Fangtao Tan,Longshua Qin,Wendi Xu,Hao Li,Zhi-Jun Sun,Ran Cui,Mingxi Zhang

The immunosuppressive tumor microenvironment (TME), characterized by elevated adenosine levels, poses a significant barrier to effective cancer immunotherapy. Herein, we report an ultrasound-activatable nanoprobe based on piezoelectric quantum dots (QDs) emitting in the near-infrared-IIb (NIR-IIb) window. The nanoprobe integrates a fluorescent QDs core with tumor-targeting peptides and CD73 inhibitors conjugated via a reactive oxygen species (ROS)-responsive linker. Attributed to the optimized piezoelectricity of the QDs core, this nanoprobe could generate ROS under deep-penetrating ultrasound irradiation. The bright NIR-IIb fluorescence and tumor-targeting ability enable the noninvasive delineation of the tumor margin from normal tissue, guiding the optimal scope and timing of ultrasound treatment. The precise ultrasound irradiation triggers the cleavage of the ROS-responsive linker and the release of the CD73 inhibitor, which effectively inhibits the production of adenosine in TME. This reduction alleviates immunosuppression and robustly activates antitumor immunity in mice, achieving significant therapeutic efficacy. This work provides a promising strategy for overcoming adenosine-mediated immunosuppression and advancing precise ultrasound therapy for cancers.

Ti3C2 MXene Nanosheets Functionalized with NaErF4:0.5%Tm@NaLuF4 Nanoparticles for Dual-Modal Near-Infrared IIb/Magnetic Resonance Imaging-Guided Tumor Hyperthermia

Jialin Pan,Meiling Zhang,Gege Fu,Li Zhang,Huimei Yu,Xu Yan,Fangmeng Liu,Peng Sun,Xiaoteng Jia,Xiaomin Liu,Geyu Lu

Although the advent of two-dimensional (2D) MXenes has substantially increased the family of 2D materials and diverse applications, the rational design of MXene-based composites and their specialized applications in theranostic biomedicine remain a challenge. In this work, we developed, for the first time, Ti3C2 MXene nanosheets for near-infrared (NIR)-IIb (1530 nm) and magnetic resonance (MR) imaging-guided photothermal therapy. Surface engineering and functionalization of 2D Ti3C2 MXene nanosheets with NaErF4:0.5%Tm@NaLuF4 (NaErF4) nanoparticles result in NaErF4@Ti3C2 composites that emit in the NIR-IIb region at 1530 nm under 808 nm excitation, conferring NIR-IIb imaging guidance and monitoring capabilities during tumor hyperthermia. Due to the inherent magnetic properties of the Er3+ ions, the NaErF4@Ti3C2 composite can also be used for efficient T2-weighted MR imaging. In particular, based on the high photothermal conversion capability (43.62% at 808 nm irradiation) and photothermal stability, the designed composites were further explored for efficient tumor ablation with an inhibition ratio of 92.9%, which was systematically demonstrated both in vitro and in vivo. No significant nanocomposite toxicity was observed at the injected dose. This work not only expands the performance of MXene-based multifunctional nanostructures but also highlights the promise of their versatile applications in cancer therapy.

Near-Infrared-IIb Fluorescent Nanozymes with Surface Charge Flipping Ability for Imaging-Guided Therapy of Inflammatory Bowel Disease

Fushou Liu,Tao Tang,Jian Guo,Wendi Xu,Fangtao Tan,Linfeng Wang,Mingxi Zhang,Ran Cui

Inflammatory bowel disease (IBD) is a common chronic gastrointestinal inflammatory disease with an increasing global incidence. However, a significant challenge in IBD management lies in the coordination between high stability and targeting properties of diagnostic and therapeutic agents. Here, we engineer near-infrared-IIb (NIR-IIb) fluorescent nanozymes with pH-responsive surface charge flipping ability for imaging-guided therapy of IBD. The nanozymes exhibit protonation-induced positive charge (+2.6 mV) in gastric acid for enhanced stability, switching to negative charge (−7.2 mV) in intestinal fluid for targeted accumulation. Therefore, these NIR-IIb fluorescent nanozymes could markedly bypass gastric acid barriers and target damaged colon tissue, revealing a significant NIR-IIb signal in early-stage IBD models versus the control. Meanwhile, Mn doping confers a reactive oxygen species scavenging ability, alleviating inflammation in colitis mice. This dual-functional platform overcomes gastrointestinal barriers, enabling precise lesion targeting and therapy monitoring and offering a promising strategy for personalized IBD management.

Biodegradable near-infrared-IIb lanthanide-doped inorganic nanoparticles with red up-conversion luminescence for bioimaging and photodynamic therapy

Liu Lin,Shi Junpeng,Peng Shanshan,Zhong Hongyun,Lin Peng,Wang Jinyuan,Sun Xia,Song Liang,Yuan Quan,Zhang Yun

The near-infrared-II (NIR-II) lanthanide-doped inorganic nanoparticles (NPs) have received considerable attention because of their low photon scattering and autofluorescence for deep tissue penetration. These NIR-II NPs have a “rigid” and physiologically inert crystal lattice (such as NaYF 4 :Yb,Er) and cannot be degraded in vivo , limiting their biomedical applications. Changing the chemical structure to achieve the transition from “rigid” to “soft” crystals is a feasible method for realizing their biodegradable applications. Herein, we developed a type of biodegradable NIR-IIb lanthanide-doped inorganic NPs, Na 3 ZrF 7 :Yb,Er,Ce (NZF). By optimizing the regulation of Na and doping with Ce 3+ , the NIR-IIb down-conversion luminescence (DCL) intensity of NZF NPs increased 37 times compared with Na 3 ZrF 7 :Yb,Er. Additionally, because of their “soft” crystal lattice features, water can destroy their crystal structure, and the degradation products can be excreted from the organs and tissues. Furthermore, NZF NPs exhibit red-emitting up-conversion luminescence (UCL). After surface modifications, we performed the proof of concept in biodegradable bioimaging applications and photodynamic therapy based on their NIR-IIb DCL and red-emitting UCL. These findings will benefit the future biomedical applications of lanthanide-doped inorganic NPs.

Near-infrared-IIb emitting single-atom catalyst for imaging-guided therapy of blood-brain barrier breakdown after traumatic brain injury

Huang Biao,Tang Tao,Chen Shi-Hui,Li Hao,Sun Zhi-Jun,Zhang Zhi-Lin,Zhang Mingxi,Cui Ran

The blood-brain barrier breakdown, as a prominent feature after traumatic brain injury, always triggers a cascade of biochemical events like inflammatory response and free radical-mediated oxidative damage, leading to neurological dysfunction. The dynamic monitoring the status of blood-brain barrier will provide potent guidance for adopting appropriate clinical intervention. Here, we engineer a near-infrared-IIb Ag 2 Te quantum dot-based Mn single-atom catalyst for imaging-guided therapy of blood-brain barrier breakdown of mice after traumatic brain injury. The dynamic change of blood-brain barrier, including the transient cerebral hypoperfusion and cerebrovascular damage, could be resolved with high spatiotemporal resolution (150 ms and ~ 9.6 µm). Notably, the isolated single Mn atoms on the surface of Ag 2 Te exhibited excellent catalytic activity for scavenging reactive oxygen species to alleviate neuroinflammation in brains. The timely injection of Mn single-atom catalyst guided by imaging significantly promoted the reconstruction of blood-brain barrier and recovery of neurological function after traumatic brain injury.
