An Implant-Free Nanosystem Enabling Synergistic Oxidative Damage Mitigation and Deep Brain Stimulation for Alleviating Parkinsonian Symptoms

Chen Zhou,Feifei Wu,Liqing He,Huijie Yan,Zhaowei Zhang,Shuang Zhao,Wenyi Huang,Yuhao Luo,Wei Zhao,Bulong Gao,Zherui Zhang,Yaofeng Zhou,Baiheng Wu,Junqiu Liu,Dingcheng Zhu

Deep brain stimulation (DBS) effectively alleviates motor symptoms in Parkinson’s disease (PD) patients; however, it necessitates permanent invasive implantation of conduits, and its therapeutic effects diminish as PD progresses. Herein, an implant-free NIR-II laser-activated intervention nanosystem that combines wireless DBS with antioxidative neuroprotection is developed to overcome the above challenges. Enzyme-like 2-(phenylselanyl)ethan-1-amine (SePh) and manganese dioxide (MnO2) and NIR-II absorber IR-1048 (IR) are integrated onto the mesoporous polydopamine (mPDA) core to form mPDA-SeMn-IR. Upon NIR-II laser irradiation, intraventricularly injected mPDA-SeMn-IR nanoparticles effectively activate endogenously expressed inositol 1,4,5-trisphosphate receptors, leading to Ca2+ outflux from the endoplasmic reticulum. Ca2+ signaling enhances tyrosine hydroxylase expression and activity and triggers dopamine release to enhance dopaminergic function. Meanwhile, mPDA-SeMn-IR rapidly eliminates excessive reactive oxygen species, synergistically enhancing long-term DBS efficiency by increasing neuron survival and restoring neuronal function for improved dopamine secretion. In vivo, mPDA-SeMn-IR regulates midbrain neurons and coiling behaviors in zebrafish and considerably alleviates motor symptoms in the PD model, proposing a promising therapeutic strategy to address existing challenges in PD therapies.

Near Infrared II Laser-Boosted Peroxynitrite Generation Hollow Copper Single Atom Nanozyme for Effective Bacterial Infection Treatment

Wei Tang,Yuan Fang,Linhao Xie,Wanqin Cai,Guichao Liu,Yan Lu,Yongguang Guan,Wei Xue,Jianfu Zhao,Siming Yu

The peroxynitrite anion (ONOO − ) is the product of a specific reaction between nitric oxide (NO) and the superoxide anion (O 2 •− ), showing great potential for treating multidrug-resistant bacterial infections. However, antibacterial materials with highly efficient ONOO − controlled release properties are lacking. In this study, hollow copper single atom nanozyme (Cu HSAz) with oxidase (OXD)-mimicking property is rationally designed and used for the first time as effective delivery carrier for the NO donor N, N ′-di-sec-butyl- N, N ′-dinitroso-1, 4-phenylenediamine (BNN6), obtaining the nanozyme Cu HSAz@BNN6. Upon near infrared (NIR)-II laser irradiation, Cu HSAz@BNN6 simultaneously releases substantial amounts of NO and O 2 •− , which subsequently lead to the ONOO − boosting via a cascade reaction. Remarkably, ONOO − displays excellent antibacterial efficiency by considerably inhibiting the activity of methicillin-resistant Staphylococcus aureus ( MRSA ) activity. Moreover, ONOO − is effective in treating MRSA -associated wound infections and promoting wound healing. An antibacterial mechanism study reveals that ONOO − triggers MRSA death by firstly damaging the MRSA membrane, followed by destructing MRSA metabolism to cut the adenosine triphosphate energy supply and boosts intracellular reactive oxygen species generation, which subsequently depletes intracellular glutathione and caused DNA breakage.

Low-temperature photothermal irradiation triggers alkyl radicals burst for potentiating cancer immunotherapy

Bo Ning,Yao Liu,Boshu Ouyang,Xiaomin Su,Huishu Guo,Zhiqing Pang,Shun Shen

Although low-temperature photothermal therapy (PTT) can sensitize tumors to immune checkpoint inhibition, its efficacy is still restricted in the deep and internal tumors without enough oxygen and lymphocytic infiltration . Non-oxygen-dependent alkyl radicals have been demonstrated to synergistically enhance PTT through up-regulating lipid peroxidation and reactive oxygen species (ROS). Herein, an innovative strategy based on alkyl radicals to augment immunogenetic cell death (ICD) caused by mild PTT was proposed to improve poor efficacy of immunotherapy, which composed of a photothermal material of Chinse ink, an azo-initiator of 2,2-azobis[2-(2- imidazoline -2- acyl ) propane ]dihydrochloride (AIPH) and a PD-L1 inhibitor of HY19991 (HY). Upon near-infrared-II laser irradiation, low-temperature (<45℃) stimulation induced a high expression of immune checkpoint receptor (PD-L1) in tumors and triggered a large amount alkyl radicals generated by AIPH. Significantly, the alkyl radicals augmented the ICD and increased the recruitment of tumor-infiltrating lymphocytes against tumors after transformation of the immunologically cold tumor microenvironment into hot by mild PTT. The released HY further enhanced the immunotherapy effect by blocking the binding of activated T lymphocytes and PD-L1. In vivo studies exhibited that the all-in-one hydrogel with synergistic mechanisms had an extraordinary ability to reverse the immunosuppressive microenvironment, stimulate innate and adaptive immune responses to eliminate tumors and prevent metastasis.
