Cationic Carbon Dots for Modification-Free Detection of Hyaluronidase via an Electrostatic-Controlled Ratiometric Fluorescence Assay

Weiqiang Yang,Jiancong Ni,Fang Luo,Wen Weng,Qiaohua Wei,Zhenyu Lin,Guonan Chen

Carbon dots (CDs) emerge as excellent fluorescent nanomaterials, but the full exploitation and application of their exceptional properties in the development of fluorescence assay are still rare. In this work, cationic carbon dots (C-CDs) covered with plenty of positive charges on the surface were synthesized through a facile ultrasonic method. Negatively charged hyaluronic acid (HA) caused the aggregation of positively charged C-CDs and neutral red (NR) along its linear chain via electrostatic adsorption, leading to a remarkable Förster resonance energy transfer (FRET) from C-CDs to NR. However, the presence of hyaluronidase (HAase) resulted in the enzymolysis of HA, as well as the liberation of C-CDs and NR. The corresponding change of fluorescence color from red to green-yellow afforded a reliable ratiometric assay for HAase. Also the ratio of fluorescence intensity for C-CDs (I525) to that for NR (I630) was used for quantitative detection of HAase. The proposed sensing system was easily operated in aqueous media with a detection limit of 0.05 U/mL. This strategy provides a new approach for the wider application of some special CDs in detecting biomolecules.

Modulation of assembly and disassembly of a new tetraphenylethene based nanosensor for highly selective detection of hyaluronidase

Xiangqian Li,Zhan Zhou,Yiping Tang,Cheng Cheng Zhang,Yuhui Zheng,Jinwei Gao,Qianming Wang

The development of nanoprobes with high sensitivity and specificity for tumor marker detection has gained increasing attention in biological applications. Here, we have designed and synthesized a novel 4,4',4”,4”'-(ethene-1,1,2,2-tetrayltetrakis(benzene-4,1-diyl))tetrakis (1-(4-bromobenzyl)pyridin-1-ium) bromide (TPE-4N + ) based aggregation induced emission (AIE) fluorescent sensor and it gives rise to electrostatic adsorption towards hyaluronic acid (HA), resulting in an effective emission recovery in yellow-greenish region. In the presence of hyaluronidase (HAase), the enzymatic digestion between HA and HAase induces the fluorescence quenching and this “ on-off ” change has been analyzed by two consecutive linear equations. The low detection limit is determined to be 0.02 U/mL by quantitative evaluation and its practical application has been verified by detecting human urine samples. It is promising that this new approach can be utilized to study a wide variety of other depolymerization reactions.

New insights into the action of bacterial chondroitinase AC I and hyaluronidase on hyaluronic acid

Lei Tao,Fei Song,Naiyu Xu,Duxin Li,Robert J. Linhardt,Zhenqing Zhang

Hyaluronic acid (HA), a glycosaminoglycan, is a linear polysaccharide with negative charge, composed of a repeating disaccharide unit [→4)-β- d -glucopyranosyluronic acid (1   →   3)-β- d - N -acetyl- d -glucoaminopyranose (1   →   ] n ([→4) GlcA (1   →   3) GlcNAc 1   →   ] n ). It is widely used in different applications based on its physicochemical properties associated with its molecular weight. Enzymatic digestion by polysaccharides lyases is one of the most important ways to decrease the molecular weight of HA. Thus, it is important to understand the action patterns of lyases acting on HA. In this study, the action patterns of two common lyases, Flavobacterial chondroitinase AC I and Streptomyces hyaluronidase, were investigated by analyzing HA oligosaccharide digestion products. HA oligosaccharides having an odd-number of saccharide residues were observed in the products of both lyases, but their distributions were quite different. Chondroitinase AC acted more efficiently at the β 1–4 glycosidic bond linking GlcNAc and GlcA. Oligosaccharides, having an even number of saccharide residues, and with an unsaturated uronic acid (4-deoxy-α- l - threo -hex-4-enepyranosyluronic acid, △UA) residue at their non-reducing end represent the major product. A minor amount of oligosaccharides having an odd number of saccharide residues resulted from the irregular terminal residues of HA substrate chains. Hyaluronidase showed a more complicated product mixture. Its minimum recognition and digestion domain is HA heptasaccharide and it could cleave both β 1–4 and β 1–3 glycosidic linkages. The HA oligosaccharides, generated with a 2-acetamido-2,3-di-deoxy-β- d - erythro -hex-2-enopyranose (△HexNAc) at the non-reducing end, are believed to be unstable and undergo breakdown immediately after their generation, and the oligosaccharides with △UA residue at the non-reducing end are formed. Thus, oligosaccharides having both an even and odd-number saccharide residues with a △UA residue at their non-reducing ends, represent the major products of hyaluronidase acting on HA.

Au@Pt nanozyme-based smart hydrogel for visual detection of hyaluronic acid

Mengyao Qi,Yi Ke,Yifei Li,Peng Li,Huakuo Zhou,Xia Zhang,Jieshi Chen,Jun Meng

Hyaluronidase (HAase), is a naturally occurring glycosidase with the ability to break down glycosidic bonds within hyaluronic acid (HA) chains. In this paper, we propose a portable smartphone and Surface-enhanced Raman scattering (SERS) dual-mode sensor based on Au@Pt nanozyme coated with HA hydrogel to realize sensitive detection of HAase. Au@Pt nanozyme has excellent peroxidase-like activity, oxidase-like activity and catalase-like activity under acidic conditions. When HAase is present, the hydrogel structure of HA was hydrolyzed and gradually released Au@Pt nanozyme. The released Au@Pt nanozyme is used to catalyze the hydrogen peroxide system, so that 3,3′,5,5′-tetramethylbenzidine (TMB) can be oxidized into blue 3,3′,5,5′-tetramethylbenzidine (TMB ox ). Therefore, based on the above strategies, a dual-mode sensing platform for HAase detection by smartphone and SERS was established. The linear range of the dual-mode sensor platform are 2–30 U/mL and 2–80 U/mL, and the detection limits are 1.0386 U/mL and 0.004 U/mL, respectively. The dual-mode smartphone and SERS sensor platform, a combination of smartphone and Raman spectrometer, is successfully further applied to biological urine samples. This portable dual-mode sensor based on intelligent hydrogel coated nanozyme not only opens up a new way for instant detection, but also has potential applications in clinical diagnosis.

A label-free and fluorescence turn-on assay for sensitive detection of hyaluronidase based on hyaluronan-induced perylene self-assembly

Lin Ma,Wenjuan Gao,Xue Han,Fengli Qu,Lian Xia,Rong-Mei Kong

Hyaluronidase (HAase) has been reported as a diagnostic and prognostic urinary biomarker for bladder cancer. Simple and sensitive methods capable of quantitatively detecting HAase are desired for tumor diagnosis and therapy at its early stage. Herein, a label-free and fluorescence “turn-on” assay for sensitive detection of HAase was established based on the electrostatic assembly of a water-soluble cationic perylene diimide derivative (PDI) and hyaluronic acid (HA). The electrostatic assembly of HA–PDI between the positively charged PDI and the negatively charged HA resulted in an efficient aggregation-caused quenching (ACQ) of PDI. In the presence of HAase, the enzyme catalyzed hydrolysis of HA led to the failure of aggregation of PDI and thus the fluorescence of PDI was retained. HA was used to function not only as a scaffold to agglomerate PDI but also as a substrate for enzymatic reactions. The efficient ACQ and the specific enzymatic reaction induced failure of aggregation of PDI endowed the developed HA–PDI assembly with high sensitivity and specificity for the detection of HAase with a detection limit of 0.03 U mL−1. Moreover, the facile detection process and rapid response of the assay made it applicable for reliable detection of HAase in human serum and urine samples.

Bioresponsive Hyaluronic Acid-Capped Mesoporous Silica Nanoparticles for Targeted Drug Delivery

Zhaowei Chen,Zhenhua Li,Youhui Lin,Meili Yin,Jinsong Ren,Xiaogang Qu

Graphical Go nano! Mesoporous silica nanoparticle-based nanoreservoirs that are end-capped with hyaluronic acid have been constructed and demonstrate great potential for cell-specific targeting and controlled drug release in response to specific enzymes present at the tumor site (see figure). In this paper, we present a facile strategy to synthesize hyaluronic acid (HA) conjugated mesoporous silica nanoparticles (MSP) for targeted enzyme responsive drug delivery, in which the anchored HA polysaccharides not only act as capping agents but also as targeting ligands without the need of additional modification. The nanoconjugates possess many attractive features including chemical simplicity, high colloidal stability, good biocompatibility, cell-targeting ability, and precise cargo release, making them promising agents for biomedical applications. As a proof-of-concept demonstration, the nanoconjugates are shown to release cargoes from the interior pores of MSPs upon HA degradation in response to hyaluronidase-1 (Hyal-1). Moreover, after receptor-mediated endocytosis into cancer cells, the anchored HA was degraded into small fragments, facilitating the release of drugs to kill the cancer cells. Overall, we envision that this system might open the door to a new generation of carrier system for site-selective, controlled-release delivery of anticancer drugs.

A hyaluronic acid fluorescent hydrogel based on fluorescence resonance energy transfer for sensitive detection of hyaluronidase

Ge Minghao,Sun Jiaojiao,Chen Mingli,Tian Jingjing,Yin Huancai,Yin Jian

Due to its important role in tumor development and treatment, hyaluronidase (HAase) has been widely investigated in vitro and in vivo. However, such investigation was limited by the absence of sensitive and in situ detection methods. Herein, a hyaluronic acid (HA) hydrogel based on the fluorescence resonance energy transfer (FRET) effect was constructed for the detection of HAase. FITC and AuNPs were covalently coupled with two HA derivatives respectively to form a fluorescent donor-acceptor pair. In the presence of HAase, the hydrogel established by cross-linking of HA derivatives was hydrolyzed specifically. The FRET effect in the hydrogel disappeared and the fluorescence intensity increased proportionally with the changes in the concentration of the HAase. Experiments proved that the HAase sensing system had a wide response range (0.5–100 U/mL), good anti-interference, and excellent biocompatibility. When the hydrogel was used for 3D culture of lung cancer cells, in situ fluorescent response could be achieved. Graphical abstract

A comparative study of the ameliorative effects of hyaluronic acid oligosaccharides and hyaluronic acid on DSS-induced colitis in mice and research on relevant mechanisms

Wei Liu,Yuan-Yuan Liu,Meng-Qiu Zhang,Miao-Zhen Qin,Yuan-Yuan Yang,Bi-Wen Liu,Dong-Jian Zhang,Cui-Hua Jiang,Zhi-Qi Yin,Min Lu,Jian-ping Shen,Jian Zhang

As a dietary supplement, hyaluronic acid (HA) has exhibited appreciable immunomodulatory activity and an ameliorative effect on rodent colitis. However, its high viscosity is not only refractory to absorb through the gut, but also causes flatulence. In contrast to HA, hyaluronic acid oligosaccharides (o-HAs) can overcome the above-mentioned constraints, but their treatment effect still remains ill-defined contemporarily. Herein, the current study intends to compare the modulatory effects of HA and o-HA on colitis and assess the underlying molecular mechanism. We first showed that o-HA had a better preventive effect than HA in alleviating colitis symptoms, as evidenced by lower body weight loss, lower disease activity index scores, a lower inflammatory response (TNF-α, IL-6, IL-1β, p-NF-κB), and more intact colon epithelial integrity in vivo. The best efficiency was observed in the o-HA treated group with a dosage of 30 mg kg−1. In an in vitro barrier function assay, o-HA exerted a better protective effect on the transepithelial electrical resistance (TEER), FITC permeability, and wound healing and modulated the expression of tight junction (TJ) proteins (ZO-1, occludin) in lipopolysaccharide (LPS)-stimulated Caco-2 cells. In summary, both HA and o-HA showed the potential to reduce inflammation and ameliorate intestinal damage in DSS-induced colitis and LPS-induced inflammation, but o-HA had improved outcomes. The results also provided a glimpse of the latent mechanism by which HA and o-HA enhanced intestinal barrier function via MLCK/p-MLC signaling pathway suppression.

Hyaluronan promotes the regeneration of vascular smooth muscle with potent contractile function in rapidly biodegradable vascular grafts

Kang Qin,Fei Wang,Russell M.L. Simpson,Xueni Zheng,He Wang,Yanhua Hu,Zhixian Gao,Qingbo Xu,Qiang Zhao

The regeneration of smooth muscle with physiological functions has been a key challenge in vascular tissue engineering . Hyaluronan (HA), as a major component of the extracellular matrix, plays a vital role in regulating tissue injury and repair. In this study, a biomimetic vascular graft was prepared by co-electrospinning of synthetic degradable polymers and native ECM components including collagen type-I as well as low and high molecular weight HA (LMW HA and HMW HA). Upon implantation in the rat abdominal aorta, the grafts exhibited sustained HA release that effectively enhanced the regeneration of vascular smooth muscle. Besides, LMW HA loaded vascular grafts demonstrated rapid endothelialization compared to the other groups. More importantly, HA-loaded poly(L-lactide-co-caprolactone) grafts demonstrated an optimal vascular media layer accompanied by well-organized elastin fibers after long-term implantation (6 months), and they maintained potent physiological function up to 1/3 that of the native artery . In contrast, inadequate smooth muscle regeneration was observed in poly(ε-caprolactone) grafts due to slow degradation restricting the regeneration. The mechanism was further investigated and explained by the HA-induced migration of smooth muscle cell (SMC) via CD44-mediated signaling. Besides, low molecular weight HA can promote the migration of vascular progenitor cells that further differentiate into SMCs. These results highlight the importance of HA in the regeneration of functional vascular smooth muscle, and provide a new insight into the fabrication of tissue engineering vascular grafts (TEVGs) via combining rapidly degradable polymers and bioactive ECM components that hold great translational potential.

Biochemical Characterization and Mechanism of Thermostability of the Thermophilic Hyaluronate Lyase TcHly8D

Yuzhu Zhang,Hao Wu,Zheng Fu,Shilong Zhang,Meiling Zheng,Jiaxia Sun,Zhongxia Lu,Rilei Yu,Wengong Yu,Feng Han

Hyaluronate lyases are widely used in medicine and biochemical engineering and are also applied as a tool enzyme to prepare oligosaccharides with various biological activities. To date, only a few hyaluronate lyases are on sale with poor thermostability. In this study, a PL8 hyaluronate lyase, TcHly8D, was found from Thermasporomyces composti and expressed in Escherichia coli with a maximum yield of 1.77 × 109 U/L (3.14 g/L) in a 5-L bioreactor. The recombinant TcHly8D exhibited a high hyaluronate lyase activity of 5.64 × 105 U/mg and an excellent thermostability with half-lives of 184.9 h at 60 °C. Fifty micrograms of TcHly8D could catalyze 5 g of hyaluronic acid with an oligosaccharide yield of 84.8% in 4 h. The salt bridges, hydrogen bonds, and proline residues, but not disulfide bonds, played important roles in the thermostability of TcHly8D. These findings provide insights into the multifunctional application potential of TcHly8D in agriculture, medicine, and the food industry.

A fluorescent hyaluronic acid-gelatin hydrogel for traceable articular cartilage regeneration

Haofeng Qiu,Xufeng Mao,Gaoke Pan,Ximing Cai,Xin Zhao,Xiang Wu,Lei Shao,Haijiao Mao,Dangsheng Xiong,Rong Wang

Post-traumatic osteoarthritis remains a significant clinical challenge, with limited effective treatment options. Conventional hydrogels for articular cartilage repair primarily focus on the bioactivity of the materials but often overlook the importance of monitoring the hydrogel's behavior in situ . This study presents the development of a novel injectable, traceable fluorescent hydrogel designed to prevent cartilage degradation and promote cartilage regeneration. The hydrogel was synthesized by forming a dynamically crosslinked network of oxidized hyaluronic acid (OHA) and adipic dihydrazide-grafted HA (HA-ADH), followed by photo-crosslinking with methacrylated gelatin (GelMA). Furthermore, GelMA was covalently conjugated with Rhodamine B, enabling real-time tracking of hydrogel degradation in situ . Experimental results demonstrated that the fluorescence of the hydrogel could effectively penetrate both skin and cartilage tissues, allowing for real-time monitoring. Additionally, the hydrogel exhibited superior lubrication (with a coefficient of friction of 0.134, comparable to HA) and favorable rheological properties, along with excellent biocompatibility. It also promoted cell differentiation and migration. In vivo studies showed that the hydrogel facilitated cartilage repair and prevented cartilage degradation over a 56-day period. Given its mechanical properties, biocompatibility, and fluorescence-based monitoring capability, this hydrogel shows great promise for real-time tracking in cartilage repair for the treatment of post-traumatic osteoarthritis.

Preparation of hyaluronic acid-loaded Harmine polymeric micelles and in vitro effect anti-breast cancer

Xiaohui Tang,Munire Kurban,Ipargul Hafiz,Qi Shen,Mei Wang

Aims To prepare hyaluronic acid-loaded Harmine polymeric micelles with CD 44 targeting properties and to investigate their anti-breast cancer effects in vitro . Methods The carboxyl group on hyaluronic acid is coupled to the amino group on 3,5-bis(trifluoromethyl)benzylamine by an amidation reaction. And the polymeric micelles self-assemble to encapsulate the Harmine in a hydrophobic core, characterized the polymer micelles by IR, 19 F-NMR, Malvern particle sizing, release, hemolysis, and other experiments. Used CD 44 -positive MDA-MB-231 cells and CD 44 -negative MCF-7 cells as tumor models. The effect of polymer micelles on breast cancer cells in vitro by cytotoxicity assay, confocal, and flow cytometry. Results The prepared polymer micelles had a uniform particle size of about 200 nm, good dispersion, PDI < 0.3, encapsulation rate up to 87%, drug loading of 4.12±0.03%, and negative charge. Hyaluronidase has a good enzymatic effect on polymeric micelles, with a hemolysis rate of less than 1%. It showed some dose-dependent toxicity to both MDA-MB-231 and MCF-7, with increased uptake of polymer micelles by CD 44 -positive MDA-MB-231 compared to CD 44 -negative MCF-7 cells and significant effects of polymer micelles on apoptosis and cycling in both cell types. These results suggest that the hyaluronic acid-loaded Harmine polymer micelles designed in this study are effective in killing breast cancer cells while at the same time reducing the toxicity of Harmine and improving its slow-release targeting.
