Two series of multicomponent silicone hydrogels based on poly (ethylene glycol)- polydimethylsiloxane-poly (ethylene glycol) (PEG-PDMS-PEG) triblock oligomers were prepared by copolymerization of silicon-containing monomers methacrylate-terminated PEG-PDMS-PEG (MTSM), tris (trimethylsiloxy)-3-methacryloxpropylsilane (TRIS), and hydrophilic monomers such as N,N-dimethylacrylamide (DMA), N-vinylpyrrolidone (NVP), and hydroxypropyl-methacrylate (HPMA). The influences of PEG chain length on hydrophilicity, oxygen permeability, and mechanical properties of silicone hydrogels were explored. The hydrophilic properties of silicone hydrogels were characterized by water contact angle, equilibrium water content, and swelling-dehydration process. The results showed that the water contact angles of dry samples decreased while the swollen ones increased with the increase of PEG chain length. The equilibrium water content decreased first and then increased as the length of the PEG chain increased. The results of swelling and dehydration process showed that PEG chains improved the water-retaining capacity of silicone hydrogels. Moreover, the protein adsorption of samples with PEG chains decreased. The surface morphologies of silicone hydrogels were characterized by scanning electron microscope (SEM), and a reconstruction model was proposed. In addition, the oxygen permeability and mechanical properties of silicone hydrogels also varied with the length of the PEG chain. These results could provide a theoretical reference for the design and modification of new hydrogels.
The properties of silicone hydrogels obtained by copolymerization of MTSM with hydrophilic monomers vary with the chain length of PEG, when PEG chains are presented as a segment of MTSM.

To clarify the influence of molecular weight of siloxane macromonomer on phase separation morphology, oxygen permeability, and mechanical properties, silicone hydrogels were prepared by copolymerizing mixtures of methacrylate-terminated siloxane macromonomer (MTSM), silicon-containing monomers tris(trimethylsiloxy)-3-methacryloxpropylsilane, and three hydrophilic monomers N , N -dimethylacrylamide, N -vinylpyrrolidone, and hydroxypropyl methacrylate. The number of Si–O–Si repeating units was equal in every silicone hydrogel while the molecular weight of MTSM ranged from 1000 to 10000 g/mol. The oxygen permeability coefficient (Dk), equilibrium water content (EWC), light transmittance, mechanical properties, and internal morphologies of obtained silicone hydrogels were characterized, and their relationships were discussed in detail. The results showed that the Dk value increased first and then decreased with the chain length of MTSM increasing. The EWC presented an increasing trend and the light transmittance decreased with the increase of MTSM chain length. The elongation increased while the modulus and the tensile strengths of the silicone hydrogels decreased along with the MTSM molecular weight increasing. The internal morphologies of the silicone hydrogels were observed by transmission electron microscope. The results indicated that the silicone hydrogels presented different phase separation structures depending on the molecular chain length of MTSM. The continuous silicone phase formed and made significant contributions to high Dk value as the molecular chain length of MTSM was moderate. Besides, a model was proposed to explain the effect mechanism of the MTSM chain length on the oxygen permeability and the EWC. This work provided a facile method to preparing the silicone hydrogel materials with desirable water absorption and high oxygen permeability by adjusting the molecular weight of macromonomers.

Silicone hydrogels have emerged as promising candidates for ocular drug delivery due to their exceptional oxygen permeability. However, their inherent hydrophobicity often results in suboptimal drug-loading capacity and patient discomfort. To address these limitations, this study introduces an innovative dual-network silicone/β-CD-HA hydrogel. In this design, the primary silicone network preserves high oxygen permeability, while the secondary β-CD-HA network enhances hydrophilicity and introduces additional drug-loading sites via host-guest interactions. Levofloxacin (LVF), a broad-spectrum antibiotic, was incorporated into the hydrogel through non-covalent binding with β-CD, and the system’s drug-loading capacity, sustained release profile, in vitro bactericidal activity, and post-loading properties were systematically evaluated. The results demonstrated a total LVF loading of 959.90 ± 36.60 μg/lens in the silicone/β-CD-HA hydrogel, with sustained release extending up to 8 h. Bactericidal assays revealed markedly reduced E. coli survival rates in treated groups: 14.46 ± 0.76 % (p < 0.0001) for LVF-loaded silicone hydrogel and 10.42 ± 1.66 % (p < 0.0001) for the β-CD-HA/silicone hydrogel, underscoring the enhanced antimicrobial efficacy of the dual-network system. The integration of β-CD-HA significantly improved hydrogel hydrophilicity, increasing equilibrium water content (EWC) from 27.69 ± 0.18 % to 45.60 ± 0.19 % and reducing the surface contact angle from 99.41 ± 0.24° to 70.36 ± 0.14°. Crucially, the modified hydrogel retained functionality-critical properties, exhibiting 93.45 ± 0.42 % light transmittance and 110.45 ± 0.32 barrer oxygen permeability values well within practical application standards. Post-drug-loading characterization showed minimal changes in oxygen permeability (105.47 ± 0.18 barrer), transmittance, or mechanical integrity, confirming structural stability. These findings collectively highlight the dual-network silicone/β-CD-HA hydrogel as a robust platform for extended ocular drug delivery, effectively balancing enhanced drug loading, sustained release kinetics, without compromising essential material properties.
