Inflammatory bowel disease (IBD) management is often hindered by limited efficacy and significant systemic side effects of traditional oral medications, owing to their lack of targeted delivery and multifunctional capabilities. This study introduces a novel orally sequence-targeted delivery system, QT-CMOF@HAS nanocube, based on hydrophobic cross-linked CD-MOF (CMOF) for the precise delivery of quercetin (Qu) to the inflamed colonic lesions. To enhance the mitochondrial targeting capabilities of Qu, a mitochondrial-functionalized Qu-(5-carboxypentyl) (triphenyl) phosphonium bromide (TPP) precursor complex (QT) was initially synthesized and loaded into the CMOF. A chitosan/glutathione-responsive hyaluronic acid (HAS) shell was subsequently coated onto the CMOF, forming the QT-CMOF@HAS nanoplatform to further enhance the gastrointestinal tolerance. Our results demonstrated that QT-CMOF@HAS significantly alleviated colitis symptoms by modulating the repolarization of pro-inflammatory M1 macrophages to anti-inflammatory M2 phenotypes and deactivation of the TLR4/MyD88/NF-κB pathway, thus reducing the production of inflammatory cytokines. Furthermore, the integrity of the intestinal mucosal barrier and the gut microbiota were enhanced. This study highlights the potential of QT-CMOF@HAS nanocube as a promising therapeutic tool for IBD treatment, offering a multifaceted approach to tackle inflammation and support mucosal healing in a targeted and controlled manner.

The ever-growing development of advanced biomaterials has substantially influenced the diagnosis and treatment against various diseases. In this study, a novel type of bio-inspired cross-linked polymer has been constructed with prominent size-tunability from micro- to nano-dimension and versatile enhanced drug loading mechanisms. In the meanwhile, this functional polymer has been applied to sustained and ultrasound (US)-triggered/promoted stimuli-responsive drug release upon an external US irradiation. Proof-of-principle in vivo animal studies proved their utility as a useful tool for a precision assessment of drug deposition and targeting delivery as well as a state-of-the-art visualized therapeutic strategy by this means. These valuable merits validated the as-reported functional polymer a useful tool for the precision assessment of drug deposition and targeted delivery, and also opened up an era to the state-of-the-art visualized therapeutic translations.

Metal–organic framework (MOF)-based drug delivery nanosystems with both precise drug release and multidrug codelivery capabilities have emerged as promising candidates for cancer treatment. However, challenges are posed by the limited number of suitable payload types, uncontrollable drug leakage, and lack of chemical groups for postmodification. To overcome those challenges, we developed a core–shell nanocomposite composed of zeolitic imidazolate framework-90 (ZIF-90) coated with spermine-modified acetalated dextran (SAD) by a facile microfluidics-based nanoprecipitation method. This nanocomposite could serve as a multidrug storage reservoir for the loading of two drugs with distinct properties, where the hydrophilic doxorubicin (DOX) was coordinately attached to the ZIF-90 framework, and hydrophobic photosensitizer IR780 was loaded into the SAD shell, enabling the combination therapy of photodynamic treatment with chemotherapy. Meanwhile, equipping ZIF-90 with a SAD shell not only substantially improved the pH-responsive drug release of ZIF-90 but also enabled the postformation conjugation of ZIF-90 with hyaluronic acid for specific CD44 recognition, thereby facilitating precise drug delivery to CD44-overexpressed tumor. Such a simple microfluidics-based strategy can efficiently overcome the limitations of solely MOF-based DDSs and greatly extend the flexibility of MOF biomedical applications.

Accurate and effective drug delivery in tumor cells significantly improves the curative effect with high drug delivery efficiency, low toxicity and side effects and has become an urgent demand for anticancer therapy. In this paper, a novel traceable and targeted drug delivery nanosystem (i.e. AuNF-nanocarriers) with high drug encapsulation and pH-controlled release was prepared based on gold nanoflowers (AuNFs) for efficient intracellular SERS imaging-guided chemo-phototherapy. SERS-active flower-like gold nanoparticles with large surface area were synthesized first and then modified with Raman and RGD molecules in sequence to prepare bright, traceable and targeted SERS tags of A549 human lung cancer cells. Furthermore, thiolated-PAA (PAA-SH) was synthesized and utilized for the first time to modify the SERS tags with a layer of negative charges for efficient pH-dependent loading and release of the anticancer drug doxorubicin. Based on the A549 human lung cancer cell model, the availability of the proposed AuNF-nanocarriers for efficient intracellular SERS imaging-guided chemo-phototherapy was studied and the results indicate that the AuNF-based drug delivery system exhibited attractive characteristics such as good stability, efficiency and pH-controlled drug loading and release, traceable and targeted delivery, as well as SERS imaging and chemo-phototherapy functions, and shows great potential for powerful SERS-imaging and as a theranostic candidate for precision nanomedicine that could achieve sensitive and accurate tumor detection and therapy.

This study successfully constructed an amphiphilic universal nanodelivery system (Trp-CS-PC) by modifying chitosan (CS) with hydrophobic L-Trp and dynamically self-assembling it with pectin (PC). This system achieved efficient and universal encapsulation of hydrophilic bovine collagen peptides (BCP) and hydrophobic curcumin (CUR) (encapsulation efficiency: BCP, 92.2±3.1%; CUR, 73.4±4.5%). Mechanistic studies indicate that the hydrophilic backbone in Trp-CS-PC fixes BCP through hydrogen bonding networks, while the hydrophobic microdomain stabilizes CUR through π-π stacking interactions. Trp-CS and PC formed a core-shell structure via pH-responsive ionic crosslinking, which maintained a tight network in the gastric environment, significantly reducing the release rates of BCP and CUR in gastric fluid (BCP: 18.23±1.05%; CUR: 36.56±2.17%). Trp modification significantly improved the solubility (5.95±0.38 mg/mL) and mucosal adhesion of CS, making this delivery system possess excellent storage stability (size variation <5% within 28 days) and intestinal retention ability (cumulative release of BCP and CUR <59% after gastrointestinal digestion). Furthermore, in vitro activity evaluation results showed that the biological activity retention of BCP and CUR delivered by Trp-CS-PC was significantly higher than that of the control group (P<0.05) . This study provides new ideas and strategies for the research and application of universal oral delivery systems for hydrophilic and hydrophobic bioactive substances.

Targeted delivery of proteins into desired cell groups is crucial for disease therapy and cellular functionalization. However, current delivery methods face severe side effects and off-target problems, making it difficult to achieve cell-targeted protein delivery. Herein, we developed a user-friendly membrane fusion liposome for cancer cell-targeted protein delivery. Phosphorothioated DNA-mediated membrane fusion was employed as an efficient transmembrane delivery approach. The phosphorothioated DNA was capped with the AS1411 aptamer, which specifically recognizes nucleolar proteins on the cancer cell membrane, enabling a controllable delivery of proteins into targeted cancer cells. This delivery system exhibited commendable biocompatibility and targeted delivery ability, thereby realizing highly effective cancer cell inhibition in vitro. The in vivo results further suggested that the membrane protein-responsive membrane fusion delivery system offers a new avenue for highly biocompatible and targeted protein delivery.

Biologic drugs hold immense promise for medical treatments, but their oral delivery remains a daunting challenge due to the harsh digestive environment and restricted gastrointestinal absorption. Here, inspired by the porcupinefish’s ability to inflate itself and deploy its spines for defense, we proposed an intestinal microneedle robot designed to absorb intestinal fluids for rapid inflation and inject drug-loaded microneedles into the insensate intestinal wall for drug delivery. Upon reaching the equilibrium volume, the microneedle robot leverages rhythmic peristaltic contraction for mucosa penetration. The robot’s barbed microneedles can then detach from its body during peristaltic relaxation and retain in the mucosa for drug releasing. Extensive in vivo experiments involving 14 minipigs confirmed the effectiveness of the intestinal peristalsis for microrobot actuation and demonstrated comparable insulin delivery efficacy to subcutaneous injection. The ingestible peristalsis-actuated microneedle robots may transform the oral administration of biologic drugs that primary relies on parenteral injection currently.

The oral delivery of chemotherapeutics has the potential to revolutionize anticancer medicine but has had limited clinical success as a consequence of delivery challenges. The reconstruction of molecular structures will modulate the physicochemical properties of parent drugs and thereby
should facilitate the incorporation of prodrugs into appropriate delivery platforms. Herein we investigated the possibility of combining a drug reconstruction strategy with a self-emulsifying drug delivery system (SEDDS) for the simultaneous oral co-delivery of two antitumor agents. To demonstrate
this concept, we constructed a hydrophobic heterodimeric prodrug 1 comprised of 7-ethyl-10-hydroxy-camptothecin (SN38) and capsaicin via a short succinic acid linker with the intent of alleviating the polarity of both drugs. Compound 1 was miscible with polysorbate 80 (Tween
80) and formed a transparent solution (termed 1-SEDDS) upon blending with water. Interestingly, the resulting 1-SEDDS nanomedicine was stable in the acidic environment of the gastrointestinal (GI) tract but concurrently released dual therapeutically active drugs in the small
intestine that is neutral. Finally, in vivo studies showed that orally administered 1-SEDDS outperformed intravenously injected CPT-11, a clinically approved SN38 prodrug, in terms of its antitumor effect. Collectively, these results provided a novel heterodimeric prodrug design
strategy for the oral administration of dual anticancer therapeutics in the form of a single platform. Further extending this concept to other potent chemotherapeutics could significantly enhance the potential of SEDDS-based platforms for combination cancer therapies.

Allergic contact dermatitis (ACD) is an inflammatory dermatitis with a high morbidity and recurrence rate. Scientific attention is focused on the development of safe and comfortable therapeutics of ACD. Herein, we propose a natural matrine-integrated pollen delivery system for the ACD treatment. Sunflower pollens were collected and defatted to serve as adhesive drug carriers for matrine. Specifically, the exquisite porous and hollow structures of the pollen shells can absorb matrine and realize the sustained drug release. Besides, the prickly surface morphology can strongly adhere to the inflamed skin sites, which can prolong the duration of the drug. By utilizing them in an ACD model and an acute pruritus model of mice, we have demonstrated that these matrine-integrated pollen shells can decrease the swelling degree of mice ears and weight loss, down-regulate inflammatory response, and improve the scratching times. These results indicate that our matrine-integrated pollen delivery systems have great potential to serve as natural topical preparations for skin disease therapy.

Though siRNA-based therapy has achieved great progress, efficient siRNA delivery remains a challenge. Here, we synthesized a copolymer PAsp(-N=C-PEG)-PCys-PAsp(DETA) consisting of a poly(aspartate) block grafted with comb-like PEG side chains via a pH-sensitive imine bond (PAsp(-N=C-PEG) block), a poly(l-cysteine) block with a thiol group (PCys block), and a cationic poly(aspartate) block grafted with diethylenetriamine (PAsp(DETA) block). The cationic polymers efficiently complexed siRNA into polyplexes, showing a sandwich-like structure with a PAsp(-N=C-PEG) out-layer, a crosslinked PCys interlayer, and a complexing core of siRNA and PAsp(DETA). Low pH-triggered breakage of pH-sensitive imine bonds caused PEG shedding. The disulfide bond-crosslinking and pH-triggered PEG shedding synergistically decreased the polyplexes’ size from 75 nm to 26 nm. To neutralize excessive positive charges and introduce the targeting ligand, the polyplexes without a PEG layer were coated with an anionic copolymer modified with the targeting ligand lauric acid. The resulting polyplexes exhibited high transfection efficiency and lysosomal escape capacity. This study provides a promising strategy to engineer the size and surface of polyplexes, allowing long blood circulation and targeted delivery of siRNA.Keywords:small polyplex;pH-sensitive PEG shedding;disulfide bond-crosslinking;siRNA delivery;targeting delivery

An oral multi-unit delivery system was developed by incorporating the nanoparticle (NP) into the nanofiber mat and its efficiency for intestinal-specific delivery and controlled release of a peptide (insulin) was investigated. Initially, the influence of deacetylation degree (DD) of chitosan and ionic gelation methods on the properties of NPs was studied. High DD (95%) chitosan was attributed to higher encapsulation efficiency and stability when crosslinked with polyanion tripolyphosphate. Subsequently, the multi-unit system was fabricated using a pH-sensitive polymer (sodium alginate) as the coating layer to further encapsulate the NP. Fiber mat with an average diameter of 481 ± 47 nm could significantly decrease the burst release of insulin in acidic condition and release most amount of insulin (>60%) in the simulated intestinal medium. Furthermore, the encapsulated peptide remained in good integrity. This multi-unit carrier provides the better-designed vehicle for intestinal-specific delivery and controlled release of the peptide.

Optic neuropathy, characterized by the irreversible degeneration of RGCs, leads to vision loss for which treatment options remained limited. Nanoparticle-based therapeutic strategies have been developed to stimulate RGCs regeneration; however, insufficient nanoparticles delivery to RGCs and a lack of robust stimulation of RGCs intrinsic regenerative capacity, have limited their application. Here, we demonstrate that a RGC-targeting Tet1 conjugated to liposome delivery system (Lipo-T) enhances therapeutic agent delivery to RGCs while minimizing off-target effect for activating glial cells. Targeted delivery of a mTOR-specific activator MHY1485 to RGCs via Lipo-T (MHY@Lipo-T) significantly enhances in vivo RGCs survival, neurite outgrowth, and axonal regeneration without retinal inflammation from unwanted glial activation. By conferring sustained MHY1485 to degenerated retina through Lipo-T injection, and with its high target ability potential, the first use of MHY@Lipo-T expands new therapeutic option for optic nerve injury.
