Bone pain is the primary problem for patients with osteoporosis. Ketoprofen is clinically used to treat osteoporotic pain, while long-term oral administration of ketoprofen can cause some side effects. In addition, osteoporosis is also accompanied by bone mass loss and inflammation. In this study, we designed a multifunctional drug (Ket@Mg-MOF-74) adopted Mg-MOF-74 to load ketoprofen to treat osteoporotic pain, bone loss and inflammation comprehensively. Mg-MOF-74 was prepared, and the physicochemical characterization proved that it had excellent physical and chemical stability. Ket@Mg-MOF-74 was synthesized by post-synthetic modification method and a high loading rate of ketoprofen was confirmed. Drug release and ion release experiments indicated Ket@Mg-MOF-74 had a good controlled release of ketoprofen and Mg in solution. Cell experiments in vitro proved the compound drug could significantly reduce the expression of pain-related genes of cyclooxygenase 2 (COX2), obviously up-regulated the expression of osteogenic cytokines and remarkably down-regulated the secretion of pro-inflammatory factors. Therefore, Ket@Mg-MOF-74 is believed a promising painkiller for osteoporotic bone pain, with the function of anti-inflammatory and promoting bone formation.

The solubility remains critical in drugs development, manufacturing, and application. S-Ketoprofen (SKTP) solubility in methanol/ethanol/2-propanol/acetone + water (278.15–318.15 K) was measured by the isothermal saturation technique. The impacts of solvent effect, Hansen solubility parameters (HSPs), and preferential solvation on solubility and model correlation were analyzed. The SKTP solubility increased with decreasing water content in alcohols–water but exhibited cosolvency at w = 0.80 in acetone–water; the maximum mole fraction solubility is 0.1072. The HSPs explained the occurrence of cosolvency molecularly (minimum Δδt = 0.08 at w = 0.80, acetone composition). The solvent effect analysis showed that solute–solvent or solvent–solvent interactions account for 63.44 and 32.60% of solvent effect. Thus, hydrogen bonding and nonspecific dipolarity/polarizability interactions dominate SKTP dissolution. The preferential solvation investigation indicated that SKTP was preferentially dissolved by cosolvents within 0.32 < x1 < 1 for methanol–water, 0.23 < x1 < 1 for ethanol–water, 0.17 < x1 < 1 for 2-propanol–water, and 0.21 < x1 < 0.55 for acetone–water but preferentially dissolved by water in other region. The correlation results suggested that the maximum values of relative average deviation and root-mean-square deviation were 4.84% and 9.45 × 10–4, thus Apelbat–Jouyban–Acree, Jouyban–Acree, and van’t Hoff–Jouyban–Acree models can correlate the solubility data accurately.

A functional biopolymer was synthesized by introducing stearic acid (SA) to the backbone of xylan (X) obtained from viscose fiber mills. The SA–X derivatives were characterized by various tests for analysis of the functional properties. Effects on the esterification conditions including molar ratio, time and temperature were discussed, and the degree of substitution (DS) was in the range of 0.34–1.54. SA–X nanoparticles (SA–X NPs) were synthesized by dialysis, and the average particle size of the NPs was about 194 nm. Ketoprofen (KPF) was chosen as a hydrophobic model drug for the loading function evaluation, and the final encapsulation efficiency was about 64%. The KPF release profile in buffer standard solution with pH 11.0 and 2.0 were tested, and the final released amount were 43.6% and 53.8%, respectively. Thus, the fabrication of functional SA–X NPs provided a new way to improve the unreasonable application of factory biopolymer by-products.

An electrochemical sensor for detecting ketoprofen was constructed by in-situ grown copper cobaltate (CuCo 2 O 4 ) nanoneedle arrays on a carbon cloth (CC) substrate. The resulting porous nanoneedle arrays not only expose numerous electrochemically active sites but also significantly enhance the electrochemical apparent active area and current transmission efficiency. By leveraging its electrochemical properties, the sensor achieves an impressive detection limit for ketoprofen of 0.7 pM, with a linear range spanning from 2 pM ~ 2 µM. Furthermore, the sensor exhibits remarkable reproducibility, anti-interference capabilities, and stability. Notably, the developed sensor also performed ketoprofen detection on real samples (including drug formulations and wastewater) and demonstrated excellent recognition ability. These exceptional performances can be attributed to the direct growth of CuCo 2 O 4 nanoneedle arrays on the CC substrate, which facilitates a robust electrical connection, provides abundant electrocatalytic active sites, and expands the apparent active area. Consequently, these improvements contribute to the efficient trace detection capabilities of the ketoprofen sensor. Graphical Abstract
