Amide Modification of Glycolipid Biosurfactants as Promising Biocompatible Antibacterial Agents

Tianyou Cui,Lianpeng Ge,Mengqian Zhao,Li Luo,Xuwei Long

Discovering new antibacterial agents is crucial to addressing the increasing risk of bacterial infections induced by antimicrobial resistance in food and agricultural industries. Here, biocompatible acidic-type sophorolipids (ASLs) and glucolipids (GLs) prepared via chemical modification of natural sophorolipids from fermentation were functionalized via amide modification for use as potential antibacterial agents. It was found that the arginine methyl ester derivative of GLs (GLs-d-Arg-OMe) showed excellent antibacterial activity, killing more than 99.99% of Escherichia coli at 200 mg/L. The sterilization dosage of the GLs against Bacillus subtilis, Bacillus cereus, and Staphylococcus aureus was 16–64 mg/L, in contrast to 32–64 mg/L for the fungus Candida albicans. In particular, GLs-d-Arg-OMe showed the best biocompatibility with a therapeutic index of up to 18. It was shown that amide modification of glycolipids can effectively improve antibacterial activity while maintaining biocompatibility, which can be exploited for the development of novel antibiotics in food and agricultural fields.

The removal of aqueous uranium by SBA-15 modified with phosphoramide: a combined experimental and DFT study

Yiming Ren,Ruizhu Yang,Lang Shao,Hao Tang,Shaofei Wang,Jianlong Zhao,Jingrong Zhong,Chuipeng Kong

Phosphoramide-modified ordered mesoporous silica (SBA-DEPA) materials were prepared via a two-step process involving: (1) the synthesis of phosphoramide via amidation of phosphoryl chloride with a primary amine and (2) modification of the phosphoramide onto SBA-15. The successful preparation was confirmed by FT-IR and NMR spectroscopy. As indicated by the ICP-AES and XRF analysis results, the phosphoramide group has a grafting ratio as high as 12.5%. The morphological information and N2 adsorption–desorption technique proved the highly ordered structure and large specific area of the material, respectively. An excellent performance for uranium sorption was found with a loading maximum of 311.6 mg g−1, a marked promotion relative to unmodified SBA-15. A strong pH and CO2 dependence suggested that near neutral conditions favored the maximum uranium sorption. To our surprise, the sorption took only 5 min to reach a 90% capacity and 20 min for equilibrium, which is extraordinary when compared to many other sorbent materials. The ionic strength partially influences the sorption, which indicates outer and inner sphere sorption both play parts in the process. A sorption capacity of over 90% of the original indicates the excellent reproducibility of SBA-DEPA. DFT calculations based on cluster models suggested a ‘tri-dentate-like’ structure for the uranyl-surface binding site, which may explain the excellent sorption ability of this material.

Preparation of 6-Amino-N-hydroxyhexanamide-Modified Porous Chelating Resin for Adsorption of Heavy Metal Ions

Shaomin Liu,Zihan Wang,Mingyi He,Jinglin Zhu

The pollution of water bodies by heavy metal ions has recently become a global concern. In this experiment, a novel chelating resin, D851-6-AHHA, was synthesized by grafting 6-amino-N-hydroxyhexanamide (6-AHHA) onto the (-CH 2 N-(CH 2 COOH) 2 ) group of the D851 resin, which contained a hydroxamic acid group, amide group, and some carboxyl groups. This resin was developed for the purpose of removing heavy metal ions, such as Cr(III) and Pb(II), from water. The findings from static adsorption experiments demonstrated the remarkable adsorption effectiveness of D851-6-AHHA resin towards Cr(III) and Pb(II). Specifically, the maximum adsorption capacities for Cr(III) and Pb(II) were determined to be 91.50 mg/g and 611.92 mg/g, respectively. Furthermore, the adsorption kinetics of heavy metal ions by D851-6-AHHA resin followed the quasi-second-order kinetic model, while the adsorption isotherms followed the Langmuir model. These findings suggest that the adsorption process was characterized by monolayer chemisorption. The adsorption mechanism of D851-6-AHHA resin was comprehensively investigated through SEM, XRD, FT-IR, and XPS analyses, revealing a high efficiency of D851-6-AHHA resin in adsorbing Cr(III) and Pb(II). Specifically, the (-C(=O)NHOH) group exhibited a notable affinity for Cr(III) and Pb(II), forming stable multi-elemental ring structures with them. Additionally, dynamic adsorption experiments conducted using fixed-bed setups further validated the effectiveness of D851-6-AHHA resin in removing heavy metal ions from aqueous solutions. In conclusion, the experimental findings underscored the efficacy of D851-6-AHHA resin as a highly efficient adsorbent for remediating water bodies contaminated by heavy metal ions.

Synthesis of amide-incorporated silica monolith stationary phase for the separation of benzene derivatives with better chromatographic performance: Experimental and computational analysis

Faiz Ali,Nashrah Azad,Muhammad Ismail,Zeid A. ALOthman,Won Jo Cheong

A new stationary phase was developed, characterized, packed in stainless column, and evaluated for the separation of five benzene derivatives in HPLC. Separation behaviors were studied through computational simulations. The Sol-gel technique was used for the synthesis of particles originating from silica monolith while reversible addition-fragmentation chain transfer polymerization (RAFT) was used to chemically bind the amide-containing organic polymer at the surface of silica particles. The polymer bound particles originated from silica monolith were characterized by SEM, FTIR, EDX, and BET/BJH analysis. An HPLC column was packed with polymer bound silica particles which effectively separated a mixture of five benzene derivatives with very good chromatographic performance using methanol/H2O (62/38, V/V%) having 0.05% trifluoroacetic acid with a flow rate of 0.37 µL/min. The average N value/column for phenol, 4-methyl-2-nitroaniline, acetophenone, benzene, and toluene were 22,400, 22,566, 27,333, 24,566, and 24,616, respectively. Density functional theory (DFT) was carried out for analyzing the interaction between analytes and polymer bound silica particles which agreed with the elution order of analytes from the column with few exceptions. The molecular electrostatic potential (MEP) map, frontier molecular orbitals (FMOs), and interaction energy of analytes were computed. The interaction energies of Phenol, 4-methyl-2-nitroaniline, acetophenone, benzene, and toluene are 6.943174, −0.52581, −0.27069, −0.23434, and −0.36344 electron volt, respectively.

Effect of siloxane backbone length on the physicochemical properties of gluconamide-modified polysiloxane surfactants

Xiyang Zhou,Yanyun Bai,Lingxiao Guo,Guoyong Wang

Three isostructural gluconamide-modified polysiloxane (GL@Si) surfactants of varying siloxane backbone lengths (molecular weights) were designed and synthesized via an amidation reaction. Their physicochemical properties and potential applications are discussed herein. Each of the three GL@Sis exhibits good surface activity. The critical aggregation concentrations (CAC) of GL@Sis solutions decrease with increasing siloxane backbone length. Surface tension at the CAC (γ CAC ) increases with molecular weight. Dynamic surface tension results indicate that lower molecular weight GL@Sis exhibit improved diffusion ability compared to larger GL@Sis. Contact angle measurements indicate that lower molecular weights and higher concentrations of GL@Sis produce smaller equilibrium contact angles. Emulsifying measurements reveal that GL@Sis forms better emulsions with soybean oil than liquid paraffin, with lower molecular weight GL@Sis demonstrating superior emulsifying capabilities. Moreover, GL@Sis exhibit good foamability and foam stability. GL@Sis possess smoothness, softness, and anti-yellowing properties. The fiber surface is visually smooth after finishing with GL@Si4-8.

Temperature-Responsive Electrocatalysis Based on Poly(N-Isopropylacrylamide)-Modified Graphene Oxide (PNIPAm-GO)

Hairan Zhang,Qiuyue Zhang,Ling Zhang,Tingting Pei,Erni Li,Huiting Wang,Qian Zhang,Lixin Xia

Graphical Making the switch : A thermally responsive poly( N -isopropylacrylamide)-modified graphene oxide (PNIPAm-GO) nanocomposite was synthesized by using a covalent “grafting-from” strategy. It was further applied as an intelligent PNIPAm-GO/GC electrode to display switchable electrocatalysis toward 1,4-dihydro-β-nicotinamide adenine dinucleotide and dopamine (see scheme). Poly( N -isopropylacrylamide)-modified graphene oxide (PNIPAm-GO), which is a type of thermally responsive GO, was designed and synthesized through a covalent “grafting-from” strategy. The as-prepared modified nanosheets integrated the individual advantages of two components, such as the thermal sensitivity of the PNIPAm terminal as well as the conductivity and the open 2D structure of the GO substrate. PNIPAm-GO was able to perform the reversible regulation of hydrophilicity/hydrophobicity in aqueous solution upon variations in the temperature. Such a unique property might also lead to the utilization of PNIPAm-GO as an intelligent electrode material to achieve a switchable electrochemical response toward a [Fe(CN) 6 ] 3−/4− probe. The PNIPAm-GO modified glassy carbon electrode (PNIPAm-GO/GC electrode) was able to exhibit better electrochemical performance in an ON/OFF switching effect than the PNIPAm-modified glassy carbon electrode (PNIPAm/GC electrode) without GO owing to the intrinsic properties and large surface area of the introduced GO. Moreover, it was found that the PNIPAm-GO/GC electrode also displayed excellent thermally responsive electrocatalysis toward the detection of 1,4-dihydro-β-nicotinamide adenine dinucleotide (NADH) and dopamine (DA), which resulted in two different catalytic statuses on the same electrode. This kind of switchable catalytic performance of the PNIPAm-GO/GC electrode might greatly enhance the flexibility of its application, and thus it is expected to have wide potential for applications in the fields of biosensors and biocatalysis.

Spectroscopic studies on noncovalent binding of nicotinamide–modified BRCA1 (856–871) analogs to calf thymus DNA

Boyuan Pan,Mingxiu Lv,Heng Du,Dongxin Zhao,Kui Lu

Various peptide drugs have entered the market with the development of molecular biology. Peptide drugs are used for treat diseases such as diabetes, breast cancer, and HIV infection. In this study, three nicotinamide–modified peptides were synthesized by modifying the N–terminus of BRCA1 (856–871, Y856R, K862Y, R866W) peptide with three nicotinic acid derivatives using solid–phase peptide synthesis. The results of calf thymus DNA (ctDNA) binding activity indicated that binding constants of BRCA1 (856–871, Y856R, K862Y, R866W) (P0) and three nicotinamide–modified peptides (P1, P2, and P3) to ctDNA were 1.89 × 10 3 , 2.97 × 10 4 , 7.61 × 10 4 , and 8.09 × 10 4 L·mol −1 , respectively. The binding affinity of the modified peptides was superior to that of BRCA1 (856–871, Y856R, K862Y, R866W). ΔH θ  < 0 and ΔS θ  < 0 indicated that van der Waals force and hydrogen bond contributed most to peptide–ctDNA binding. Results obtained by Circular dichroism (CD) indicated that peptide binding interaction led to conformational changes in ctDNA. Ultraviolet–visible (UV) spectroscopy, ethidium bromide (EB) competition experiments, DNA melting experiments, and viscosity measurements verified that peptides interacted with ctDNA via groove binding. Ionic strength experiments manifested that electrostatic binding was also involved in peptide–ctDNA binding.

Revisiting matrix hydrogel composed of gelatin and hyaluronic acid and its application in cartilage regeneration

Huangrong Zhu,Fengling Liu,Xinrang Zhai,Zhicheng Tong,Huimin Li,Wei Dong,Wei Wei,Chong Teng

With the increasing incidence of knee osteoarthritis (KOA), the reparation of cartilage defects is gaining more attention. Given that tissue integration plays a critical role in repairing cartilage defects, tissue adhesive hydrogels are highly needed in clinics. We constructed a biomacromolecule-based bioadhesive matrix hydrogel and applied it to promote cartilage regeneration. The hydrogel was composed of methacrylate gelatin and N-(2-aminoethyl)-4-(4-(hydroxymethyl)-2-methoxy-5-nitroso) butyl amide modified hyaluronic acid (HANB). The methacrylate gelatin provided a stable hydrogel network as a scaffold, and the HANB served as a tissue-adhesive agent and could be favorable for the chondrogenesis of stem cells. Additionally, the chemically modified HA increased the swelling ratio and compressive modulus of the hydrogels. The results of our in vitro study revealed that the hydrogel was compatible with bone marrow stromal cells . In vivo, the hyaluronic-acid-containing hydrogels were found to promote articular cartilage regeneration in the defect site. Therefore, this biomaterial provides promising potential for cartilage repair.
