The first use of yeast as a support in the molecular imprinting field combined with atom transfer radical polymerization was described. Then, the as-prepared molecularly imprinted polymers were characterized by Fourier transmission infrared spectrometry, scanning electron microscope, thermogravimetric analysis, and elemental analysis. The obtained imprinted polymers demonstrated elliptical-shaped particles with the thickness of imprinting layer of 0.63 μm. The batch mode experiments were adopted to investigate the adsorption equilibrium, kinetics, and selectivity. The kinetic properties of imprinted polymers were well described by the pseudo-second-order kinetic equation, indicating the chemical process was the rate-limiting step for the adsorption of cefalexin (CFX). The equilibrium data were well fitted by the Freundlich isotherm, and the multimolecular layers adsorption capacity of imprinted polymers was 34.07 mg g −1 at 298 K. The selectivity analysis suggested that the imprinted polymers exhibited excellent selective recognition for CFX in the presence of other compounds with related structure. Finally, the analytical method based on the imprinted polymers extraction coupled with high-performance liquid chromatograph was successfully used for CFX analysis in spiked pork and water samples.

Temperature responsive molecularly imprinted polymers (T-MIPs) were prepared based on the surface of yeast by electron transfer atom transfer radical polymerization (AGET ATRP). The as-prepared T-MIPs were charcterized by FT-IR, SEM, TGA and elemental analysis, which indicated that T-MIPs exhibited thermal stability and composed of temperature responsive imprinted layer. Then T-MIPs were evaluated as sorbents to selectively recognise and release cefalexin (CFX) molecules. The results suggested binding properties of T-MIPs were related to the testing temperature. The maximum adsorption capacity of T-MIPs at 303 K was 59.4 mg g −1 , and the maximum release proportion for T-MIPs at 293 K in water for 24 h was 71.08%. The selective recognition experiments demonstrated high affinity and selectivity of T-MIPs towards CFX over competitive compounds, and the specific recognition of binding sites may be based on the distinct size, structure and functional group to the template molecules.
High levels of blood glucose are always associated with numerous complications including cholesterol abnormalities. Therefore, it is important to simultaneously monitor blood glucose and cholesterol levels in patients with diabetes during the management of chronic diseases. In this study, a glucose dehydrogenase fromAspergillus oryzaeTI and a cholesterol oxidase fromChromobacteriumsp. DS-1 were displayed on the surface ofSaccharomyces cerevisiae, respectively, using the yeast surface display system at a high copy number. In addition, two whole-cell biosensors were constructed through the immobilization of the above yeast cells on electrodes, for electrochemical detection of glucose and cholesterol. The assay time was 8.5 s for the glucose biosensors and 30 s for the cholesterol biosensors. Under optimal conditions, the cholesterol biosensor exhibited a linear range from 2 to 6 mmol·L−1. The glucose biosensor responded efficiently to the presence of glucose at a concentration range of 20–600 mg·dL−1(1.4–33.3 mmol·L−1) and showed excellent anti-xylose interference properties. Both biosensors exhibited good performance at room temperature and remained stable over a three-week storage period.Keywords:whole-cell biosensor;yeast surface display;cholesterol oxidase;glucose dehydrogenase;electrochemical detection
