Enhancement of the wheat straw pretreatment in pulping: The microbial intelligent response

Hongling Liu,Zhicheng Xue,Feng Liang,BoBo Miao,Tengfei Wang,Di Huang,Haibo Yuan,Yi Jiang

In this study, Bacillus licheniformis (BL) was employed for the pretreatment of wheat straw, utilizing an intelligent regulatory mechanism to modulate the secretion of various enzymes. As a result, the cuticle and sclerenchyma within the lignocellulosic array were gradually degraded. Following a 36 h treatment period, the cellulose content in the pulp reached 77.18 %, whereas the lignin and hemicellulose contents continued to decrease. Additionally, the pulp brightness increased by 66.3 % after 48 h of treatment, and the tensile and tear indexes increased by 25.2 % and 61.5 %, respectively. Furthermore, we elucidated how BL intelligently regulated the surface characteristics of wheat stalks to highlight its advantages over direct enzyme treatments. These findings demonstrate the feasibility of using bacteria for wheat straw pretreatment in the papermaking process.

Enhancing removal performance of ciprofloxacin hydrochloride by Chlorella vulgaris-bacteria consortium: Insights of microbial responses and stoichiometry

Yongrui Pi,Yan Li,Wenpeng Jia,Yongzheng Tang

The microalgae-bacteria consortia (MBC) system has demonstrated significant potential in enhancing the removal efficiency of antibiotics from wastewater. This study systematically evaluated the treatment of ciprofloxacin hydrochloride (CIP) wastewater using pure microalgae ( Chlorella vulgaris ) (PM), pure bacteria (PB), and the MBC formed by the combination of PM and PB. The MBC system achieved an impressive CIP removal rate of 94.6 ± 0.6 % at an initial concentration of 10 mg/L. As the initial concentration of CIP increased from 10 to 80 mg/L, the biodegradation rate declined from 0.42 to 0.13 d - ¹ , with the half-life of CIP significantly increasing from 1.66 to 5.23 days. The MBC notably enhanced CIP removal efficiency by promoting microbial metabolic activity and increasing the concentration of photosynthetic pigments. The secretion of CIP-degrading enzymes to mitigate intracellular oxidative stress indicates the superior tolerance and degradation capacity of the MBC system towards CIP. Acinetobacter johnsonii was identified as the primary contributor to the CIP biodegradation. The molar yield coefficient (MYC) for the bacteria is nearly 2-times fold of that for C. vulgaris based on the growth-process equations of cells on CIP. Collectively, these findings provide valuable insights into the mechanisms by which MBC outperforms PM or PB in the treatment of CIP-contaminated wastewater, thereby contributing to advancements in sustainable wastewater management practices.

Engineering a thermophilic Bacillus thermoamylovorans for degradation of polyethylene terephthalate waste

Yi-Mei Cai,Cui-Can Lu,Tian-Tian Wang,Li-Jun Song,Yu-Ming Deng,Han Xu,Sheng Chen,Wei Xia,Jing Wu,Zheng-Fei Yan

Biodegradation presents a sustainable alternative for recycling polyethylene terephthalate (PET) waste. Enzymatic PET degradation typically requires enzyme preparation, which limits its industrial applicability. Herein, we developed a novel thermophilic biocatalyst using Bacillus thermoamylovorans JQ3, engineered to express PET hydrolase, capable of simultaneous enzyme secretion and PET degradation. The engineered strain harboring PET hydrolase variant from the bacterium HR29 (TurboPETase), achieved up to 100% weight loss of PET at 60°C. It exceeded the sum of the degradation rates (17.7%) of the wild strain and TurboPETase treatment alone, representing the highest degradation rate among previously reported engineered strains. Moreover, this engineered strain demonstrated broad specificity for various polyester plastics, with biocatalytic activity 10- to 50-fold higher than that of the wild strain. Nearly 100% degradation of polycaprolactone (PCL) was observed at a solids loading of 10 g/L. This study establishes B. thermoamylovorans as a promising platform for developing environmentally friendly biocatalytic approaches to address plastic waste challenges, paving the way for more sustainable plastic degradation and recycling solutions.

The mechanism of short hypha formation and high protein production system mediated by cell wall integrity signaling pathway in Aspergillus niger

Leyi Yu,Tiantian Wang,Bin Wang,Li Pan

Aspergillus niger is a cell factory widely used in industries to produce proteases, organic acids, drugs, and other substances. The hyphal morphology of A. niger is a complex differentiated elongated tubular structure, which limits its basic research and application. In this study, the mpkA , bck1 , steC , and Tpk2 genes were successfully deleted using a quick way to knock out genes based on the RNP (Ribonucleoprotein) complex. The study showed that the knockout of mpkA and bck1 kinase gene strains resulted in smaller, denser colonies, short rod-shaped hypha, and a significant increase in glucoamylase secretion. The mechanism of short hypha formation and high protein production for A. niger is the cell wall integrity signaling (CWIS) pathway. The CWIS pathway passed through the bck1 - mkkA - mpkA tertiary kinase to deliver phosphorylation signals to the rlmA transcription factor, which regulated the expression of the cell wall synthesis gene agsA , thus regulating hyphal morphology. The mpkA kinase regulated the expression of the transcription factor amyR , which affected the expression of the genes glaA and amyA , thus enhancing the expression of proteins in A. niger . This study provides a strategy for the regulation of hyphal morphology and promotes the application of A. niger in industrial production.

Efficient extracellular laccase secretion via bio-designed secretory apparatuses to enhance bacterial utilization of recalcitrant lignin

Lanfang Cao,Lu Lin,Haiyan Sui,Heng Wang,Zhichao Zhang,Nianzhi Jiao,Jizhong Zhou

Microbial-driven lignin conversion has a significant impact on the biogeochemistry of global ecosystems and biomass utilization. During this process, secretion of extracellular ligninolytic enzymes is the first, essential step, yet there is a significant challenge to the secretion of these high-redox potential enzymes in bacteria, especially in Gram-negative bacteria. In this study, genome and proteome analyses enabled us to bio-design two types of extracellular secretory apparatus, both of which effectively secreted a heterologous laccase in Pseudomonas putida, up to ∼300 U mL−1. Importantly, a strong cooperation between P. putida A514 cells and the extracellular laccase, which was either released to the medium or displayed on the cell surface, was observed to significantly promote cell growth (9 × 1010 CFUs mL−1) and lignin consumption (12.5%). Chemical analyses of lignin further confirmed the effect on lignin utilization. Moreover, we demonstrated that a free laccase-A514 cell system exhibited greater effect on lignin utilization than that of a cell-immobilized laccase complex, which challenges the current view of bacterial polymer catabolism, and suggests the importance of secretory pathways and sufficient reaction surface for lignin biocatalysis. Our study advances the knowledge of secretion mechanisms in Gram-negative bacteria and provides novel insights into the lignin utilization by extracellular lignolytic enzyme-bacterial cell systems.
