Investigating enhancement of protease and lysozyme combination pretreatment on hydrolysis of sludge organics under humic acid inhibition

Xiang Zou,Junguo He,Xinlei Pan,Qiupeng Cai,Shengye Duan,Yijie Zhong,Xinxin Cui,Jie Zhang

This study investigated the impact of humic acid (HA) on enzymatic pretreatment efficiency, focusing on sludge properties and HA molecular structure. The results showed that enzymatic pretreatment alleviates HA inhibition, improving hydrolysis efficiency. In the presence of HA, soluble proteins and polysaccharides in the enzyme-cocktail group reached 27.7 mg/L and 23.9 mg/L, 1.4 and 1.3 times higher than the blank group, respectively. The enzyme-cocktail group also had the highest soluble DNA concentration (19.4 mg/L) and the lowest viable cell proportion (69.3 %), indicating effective cell lysis. Enzyme-cocktail pretreatment reduced electrostatic repulsion, enhancing the mobility of extracellular organics. Enzyme interactions with HA released internal hydrolases and decreased amide groups on the HA surface, increasing the availability of biodegradable substrates. Overall, enzymatic pretreatment proves effective in mitigating HA-induced inhibition, thereby improving sludge biodegradation and enhancing carbon recovery in anaerobic fermentation.

Assessment of enzyme addition strategies on the enhancement of lipid yield from microalgae

Yi Zhang,Xihui Kang,Feng Zhen,Zhongming Wang,Xiaoying Kong,Yongming Sun

Enzymatic hydrolysis is emerging as a promising unconventional pretreatment method for disrupting microalgae cells. However, the sustainability of biological enzyme needs to be improved due to low efficiency of enzymatic hydrolysis with single enzyme. Cellulase , xylanase , pectinase , snailase, lysozyme and trypsin were selected to pretreat microalgae Scenedesmus sp. for lipid extraction. The results shown that the pretreatment of a multi-enzyme combination of cellulase, xylanase and pectinase perform a better enzymatic hydrolysis effect than each single enzyme. Results shown that different sequences of enzymatic hydrolysis had a significant effect on the lipid extraction performance from microalgae, the lipid recovery yield of 100% (total lipids content) could be achieved with the sequence of stepwise enzymatic hydrolysis: pectinase (10 mg g -1 dry weight microalgae), cellulase (20 mg g -1 ) and xylanase (14 mg g -1 ). The SEM image, FTIR measurement, HPLC and XPS analysis indicated that the outer layer of trilaminar sheaths in the microaglal cell walls was first degraded by pectinase, and then cellulose and hemicellulose in the inner layer were destroyed by cellulase and xylanase. This study provides an efficient and environmental-friendly route for lipid extraction as a new strategy of enzymatic hydrolysis to valorize high-value added products in microalgae downstream processing .

Lysozyme coupling protease pretreatment to relieve the humic acid inhibition on excess sludge anaerobic fermentation

Xiang Zou,Junguo He,Xinlei Pan,Qiupeng Cai,Shengye Duan,Xinxin Cui,Yijie Zhong,Jie Zhang

The asynchronous dosed protease and lysozyme combination pretreatment was proved to be effective in enhancing the anaerobic fermentation of waste activated sludge (WAS). However, humic acid (HA) in the sludge could interact with hydrolase and restrain the hydrolysis efficiency, thus inhibiting short-chain fatty acids (SCFAs) production. This study investigated the effectiveness and mechanism of enzymatic pretreatment against HA. Results showed that the enzyme cocktail method increased the extracellular bioavailable contents by 34 %, which raised SCFAs production by 89.69 % (1269.65 mg COD /L). The balanced ratio of hydrolysis and fermentation communities suggested that the small molecular organics generated by the hydrolysis community could be sufficiently utilized by fermentation communities. The metabolism of amino acids and glucose was facilitated, and the activities of key enzymes were enhanced. These results clarified the effect of asynchronous enzyme cocktail pretreatment against HA inhibition and contributed to SCFAs production, which offered fresh perspectives on carbon recovery.

Effects of Low Temperature–Ultrasound–Papain (LTUP) Combined Treatments on Purine Removal from Pork Loin and Its Influence on Meat Quality and Nutritional Value

Jiaojiao Yuan,Cheng Yang,Jialing Cao,Lianfu Zhang

A combined pretreatment method of “low temperature-ultrasound-papain” (LTUP) was proposed to remove the purine of pork loins. Compared with untreated pork loin, under optimal conditions (temperature 58 °C, ultrasound density 100 W/cm2, and papain concentration 0.085%), the purine removal rate of treated pork loin could reach 59.29 ± 1.39%. The meat quality of pork loin treated with the LTUP method such as hardness and chewiness decreased by 58.37% and 64.38%, respectively, and the in vitro protein digestibility was increased by 19.64%; the cooking loss was decreased by 15.45%, compared with the simulated household blanching process (HT). In view of the high purine removal rate, the losses of free amino acids and soluble peptides were acceptable and reasonable. SEM and LF-NMR results showed that low temperature and ultrasound combined with papain treatment opened a channel for purine transfer and promoted purine dissolution by affecting the protein structure of pork loin. In addition, the migration of water within the muscle tissue was also related to purine removal. In summary, LTUP is recommended as an efficient and green way for the meat industry to remove purine.
