Mechanism of patulin biodegradation by a reductase from Saccharomyces cerevisiae and its potential application to apple juice

Chao Yang,Chen Hu,Lingxuan Huang,Bangzhu Peng

Bioenzymatic degradation exhibits great potential for mycotoxins removal. So far, little is known about patulin (PAT) degrading enzymes from Saccharomyces cerevisiae . Here, the degradation mechanism of PAT by a free methionine-R-sulfoxide reductase (FRMSR) from S. cerevisiae was investigated. The results showed that purified FRMSR had high degradability without cofactor and displayed strong substrate specificity. The optimal degradation conditions in aqueous solution were 37 °C and pH 7.0. Isothermal titration calorimetry and molecular docking suggested that the PAT degradation by FRMSR was related to the hydrogen bonds formed between amino acids with PAT. Site-specific mutagenesis indicated that the mutation of Asp151 had the most significant effect on the degradation rate. Furthermore, the addition of FRMSR successfully degraded 88.16 % of PAT in apple juice without affecting its soluble solids content, pH value, titrable acidity and total phenols. These findings could provide valuable insights into the development of PAT-degrading enzymes in apple products and their industrial applications.

Influence of monomer composition of poly(butylene succinate-co-hexamethylene succinate)s on their enzymatic hydrolysis by Fusarium solani cutinase

Ting Hu,Yujun Wang,Jingwei Lin,Zhanyong Wang

Poly(butylene succinate) (PBS), poly(hexamethylene succinate) (PHS) and three kinds of poly(butylene succinate-co-hexamethylene succinate)s (P(BS-co-HS)s) were enzymatically hydrolyzed by Fusarium solani cutinase. The results showed that the characteristics of the enzymatic hydrolysis of these polyesters were mainly affected by crystallinity, thermal properties and the BS/HS ratio. The enzymatic hydrolysis rates of the polyesters are as follows: P(BS-co-HS52) ≈ P(BS-co-HS71) > PHS > P(BS-co-HS32) > PBS. Furthermore, with increasing HS content, both the degree of crystallinity (Xc) and melting temperature (Tm) of the polyesters first decreased and then increased. P(BS-co-HS52) and P(BS-co-HS71) had the lowest Xc and the lowest Tm, thus had the highest hydrolysis rate; this shows that the hydrolysis rate is affected by Xc and Tm. The results also showed that BS/HS ratio could affect the physical properties and degradability of polyesters. Thus, it is possible to prepare polyesters with various physical properties and degradability for different applications by adjusting BS/HS ratio. The crystalline and amorphous regions of the polyesters were both hydrolyzed, during which parts of the crystalline regions were converted into the amorphous regions. Finally, we found that the crystal structure and thermal stability of the polyesters were not affected by the enzymatic hydrolysis.

Specific Ion Effects on the Enzymatic Degradation of Polyester Films

Zhang Jian,Zhu Jie,Hua Zan,Liu Guang-Ming

Soil environment on earth contains a variety of ions, which are expected to play a vital role in the biodegradation of plastics discarded in the environment. In this work, poly(butyleneadipate- co -terephthalate) (PBAT) is employed as a model biodegradable plastic to study the specific ion effects on the enzymatic degradation of polyester plastics. The results show that the specific ion effects on the enzymatic degradation rate of the PBAT films and on the catalytic rate constant for the enzymatic hydrolysis of the ester bonds are strongly dependent on temperature and ionic strength. Both the enzymatic degradation rate and catalytic rate constant decrease following the trends Na + > K + > Ca 2+ and Cl − > SO 4 2− > NO 3 − for cations and anions, respectively, indicating that the ion-specific enzymatic degradation of the PBAT films is closely correlated with the specific ion effects on enzymatic hydrolysis of the ester bonds. Our study shows that the specific ion effects on the enzyme activity can be understood by taking into account the ion-specific cation-anion interaction, ionic dispersion force, salting-out effect and salting-in effect. This study of specific ion effects on the enzymatic hydrolysis of the ester bonds and the resultant enzymatic degradation of the PBAT films would offer us a new clue to develop new biodegradable, environmentally friendly synthetic plastics.

Enzyme-Driven Biodegradation of Ti3C2 MXene: Unveiling Peroxidase-Mediated Pathways and Enhanced Bioaccumulation Risks in Aquatic Systems

Weibiao Ye,Wenhao Lao,Lan Wu,Dong-Xing Guan,Chao Zhang,Yiping Feng,Liang Mao

2D MXene nanosheets are increasingly attracting interest due to their promising applications in materials science and biomedicine. However, the environmental fate of MXenes, particularly their biotransformation, is poorly understood. Here, the biodegradability of Ti3C2 MXene nanosheets was investigated using a plant horseradish peroxidase (HRP)-mediated reaction. The degradation rates of Ti3C2 MXene nanosheets were determined to be first-order in both HRP and H2O2 dosage. Material characterizations and product identifications revealed that peroxidase-catalyzed the oxidation of Ti3C2 MXene in the presence of H2O2, resulting in the formation of holes on its basal plane, the generation of titanium dioxide (TiO2) nanoparticles, and the release of CO2. The assessment using Daphnia magna revealed visible toxicity and enrichment of Ti3C2 MXene to aquatic organisms, with LC50 values of pristine Ti3C2 MXene to Daphnia as 55.41, 7.24, and 2.97 mg L-1 at 48, 72, and 96 h, respectively. Substantial accumulation (74.30 μg Ti mg-1 of dry tissue) of Ti3C2 MXene by Daphnia was observed after 48 h of exposure. Furthermore, the biological effects of HRP-degraded Ti3C2 MXene products on Daphnia were examined. Although the toxicity to Daphnia was reduced, a substantial increase in the bioaccumulation of Ti3C2 MXene biodegradation products (137.36 μg Ti mg-1 of dry tissue) was observed. These findings reveal that enzymatic degradation alters the size and surface chemistry of Ti3C2 MXene, potentially changing its toxicity and altering its environmental compatibility.

Folic acid-coupled phosphorylated Fe2+ chelating peptide: The properties of Fe2+ chelating capacity and mechanisms of anti-gastrointestinal enzyme degradation

Shanting Lin,Xiao Hu,Shengjun Chen,Hui Huang,Xiaoshan Long,Zhenxing Li

The aims of this study were to investigate the effect of precise phosphorylation of Ser in GL-Hyp-GPSGEEGKR on its Fe 2+ chelating capacity, and the effects and mechanisms of folic acid coupling to GL-Hyp-GPS (PO4) GEEGKR on its resistance against degradation by gastrointestinal enzymes. The results demonstrated that GL-Hyp-GPS (PO4) GEEGKR exhibited significantly enhanced Fe 2+ chelating capability compared to GL-Hyp-GPSGEEGKR. This was attributed to the phosphorylation of Ser introducing more Fe 2+ chelating sites. Furthermore, the results of UV–Vis, FTIR, and 1 H NMR showed that folic acid has successfully coupled with GL-Hyp-GPS (PO4) GEEGKR to generate folic acid-GL-Hyp-GPS (PO4) GEEGKR. Folic acid-GL-Hyp-GPS (PO4) GEEGKR demonstrated superior resistance against degradation by gastrointestinal enzymes. This may be attributed to the coupling of folic acid altering the original zeta potential of the peptide and the steric hindrance effect of folic acid hindering the interaction between enzymes and peptide. These findings can provide valuable insights for the production of high Fe 2+ chelating peptides that resist gastrointestinal enzyme degradation.

Enhanced enzymatic degradation of viscous marine polysaccharide by the self-propulsion of urease-driven micromotor

Yihong Chen,Lili Yin,Hao Dong,Yang Hu,Xiangzhao Mao

Efficient enzymatic degradation of marine polysaccharides is hindered by the limited mass transfer induced from the high viscosity. In this study, taking viscous chitosan as an example, a urease-powered hollow mesoporous silica micromotor (UHMSMM) was prepared as the carrier to immobilize chitosanase (Chi), aiming at improving the degradation of chitosan by self-propulsion. At a urea concentration of 300 mM, Chi@UHMSMM exhibited the maximum motion behavior, confirmed by the doubled diffusion coefficient, even in viscous chitosan. The enhanced motion behavior improved the enzymatic performance. The catalytic efficiency was significantly increased, confirmed by the enhancement (3.8 times) of k c at / K m value. It is demonstrated that the autonomous movement could induce the micromixing of fluid in the microenvironment around Chi according to the simulated result, contributing to improved substrate affinity and catalytic efficiency. Also, the immobilization of Chi on UHMSMM induced a common improvement of enzyme immobilization, including enhanced thermo-stability and acid resistance. As a result, Chi@UHMSMM could work as an efficient platform for degrading chitosan, with a 125.75 % yield enhancement within 40 min. In this work, the reported strategy, which accelerates the enzymatic degradation of chitosan by the self-propulsion of enzyme-driven micro/nanomotors offers a novel approach for improving enzyme reactions in high-viscosity systems.

Physically crosslinked silk fibroin/hyaluronic acid scaffolds

Yupin Guan,Haining You,Junyi Cai,Qiang Zhang,Shuqin Yan,Renchuan You

Combining the properties of natural protein and polysaccharide is a promising strategy to generate bioactive biomaterials with controlled structure. Here, a new method of preparing water‐insoluble silk fibroin/hyaluronic acid (SF/HA) scaffolds with tunable performances using an all-aqueous process is reported. Freezing-induced assembly was used to form silk I crystallization in the SF/HA blends. Silk I crystallization enhanced the stability of SF/HA scaffolds in water by forming silk I crystal networks to entrap blended HA without chemical cross-linking. Increasing HA content significantly enhanced the flexibility and water binding capacity of porous scaffolds, but high amount of HA reduced the water-stability of porous scaffolds due to insufficient silk I crystal cross-links. The enzymatic degradation behavior of the SF/HA scaffolds was investigated, revealing that the regulation ability of HA in the SF scaffolds. This novel nonchemically cross-linked protein/polysaccharide scaffold may be useful for soft tissue engineering due to excellent biocompatibility and tunable performances.

Synthesis, physical properties and enzymatic degradation of bio-based poly(butylene adipate-co-butylene furandicarboxylate) copolyesters

Weidong Zhou,Xiaowei Wang,Bing Yang,Ying Xu,Wei Zhang,Yajie Zhang,Junhui Ji

A series of bio-based poly(butylene adipate-co-butylene furandicarboxylate) (PBAFs) copolyesters were synthesized from 2,5-furandicarboxylic acid (FDCA), adipic acid (AA), and 1,4-butanediol (BDO) through a two-step polycondensation reaction. The copolyesters were characterized by 1 H NMR, GPC , DSC , XRD and tensile tests, and their enzymatic degradation behaviors were also investigated. They were random copolymers whose composition was well controlled and the weight average molecular weight ( M w ) ranged from 54,100 to 76,800 g/mol. By combining the results of DSC and XRD, with increasing FDCA content, PBAFs changed from semi-crystalline polymers to nearly amorphous polymers, then to semi-crystalline polymers again. Specifically, the crystallizability and melting temperature ( T m ) decreased with FDCA content 0–50 mol%, but rose again at FDCA content 75–100 mol%. And, the glass transition temperature ( T g ) increased continuously with increasing FDCA content. Consequently, the tensile modulus and strength decreased but the ultimate elongation increased at lower FDCA content (0–50 mol%), which were converse at higher FDCA content (75–100 mol%). Especially, the P(BA-40 mol% BF) shows outstanding elasticity and rebound resilience. In addition, the influences of FDCA content on the enzymatic degradation by lipase from porcine pancreas were studied in terms of the weight loss and morphological change. At FDCA content of 0–50 mol%, the copolyesters showed biodegradability but only the degradation rate of P(BA-10 mol% BF) was faster than PBA. When the FDCA content were 75–100 mol%, they were actually un-degradable. Thus, depending on their composition, PBAFs might find applications from biodegradable elastomers to thermoplastics .

Facile Ag+ assisted bonding strategy built a defect-low hybrid layer with intrinsic antibacterial and enzymolysis-inhibitory property

Yajie Cheng,Yadong Chen,Chang Shu,Chenying Zhou,Zhenzhen Zhang,Yan Tu,Qiaojie Luo,Xiaodong Li

As the most widely used material for dental tissue repair, dental resin composites face durability challenges, whose longevity critically depends on the hybrid layer's integrity. Incomplete adhesive infiltration within the demineralized dentin matrix (DDM) creates structural defects in this layer, rendering it vulnerable to stress concentration, enzymatic degradation, and bacterial invasion. These factors contribute to secondary caries, the predominant complication of resin-based restorations. Enhancing both adhesive infiltration and the hybrid layer’s antibacterial capacity is thus pivotal to extending restoration lifespan. Previous studies revealed that strong metal ion chelation can mobilize confined water to facilitate hydrophobic monomer infiltration, significantly improving dentin bonding efficacy and durability. In this study, leveraging the dual advantages of Ag⁺—potent chelation and antibacterial activity—we treated DDM with Ag⁺. A brief 20-second application enabled confined water release, enhanced adhesive infiltration, and conferred durable antibacterial functionality to chemically modified DDM. Additionally, matrix metalloproteinases (MMPs) activated during bonding were effectively inhibited. Crucially, subsequent light irradiation reduced Ag⁺ to metallic silver, enhancing structural stability by orders of magnitude. This approach successfully established a low-defect hybrid layer with intrinsic, long-lasting antimicrobial properties. The strategy offers a clinically viable solution for achieving durable dentin restoration with integrated antibacterial defense.

Partially Bio-Based and Biodegradable Poly(Propylene Terephthalate-Co-Adipate) Copolymers: Synthesis, Thermal Properties, and Enzymatic Degradation Behavior

Ping Song,Mingjun Li,Haonan Wang,Yi Cheng,Zhiyong Wei

A series of partially bio-based and biodegradable poly(propylene terephthalate-co-adipate) (PPTA) random copolymers with different components were prepared by the melt polycondensation of petro-based adipic acid and terephthalic acid with bio-based 1,3-propanediol. The microstructure, crystallization behavior, thermal properties, and enzymatic degradation properties were further investigated. The thermal decomposition kinetics was deeply analyzed using Friedman’s method, with the thermal degradation activation energy ranging from 297.8 to 302.1 kJ/mol. The crystallinity and wettability of the copolymers decreased with the increase in the content of the third unit, but they were lower than those of the homopolymer. The thermal degradation activation energyE, carbon residue, and reaction levelnall showed a decreasing trend. Meanwhile, the initial thermal decomposition temperature (Td) was higher than 350 °C, which can meet the requirements for processing and use. The PPTA copolymer material still showed excellent thermal stability. Adding PA units could regulate the crystallinity, wettability, and degradation rate of PPTA copolymers. The composition of PPTA copolymers in different degradation cycles was characterized by1H NMR analysis. Further, the copolymers’ surface morphology during the process of enzymatic degradation also was observed by scanning electron microscopy (SEM). The copolymers’ enzymatic degradation accorded with the surface degradation mechanism. The copolymers showed significant degradation behavior within 30 days, and the rate increased with increasing PA content when the PA content exceeded 45.36%.

Pepsin-inspired polyurethanes containing a tyrosine–fumaric acid–tyrosine segment

Haoxiang Lu,Wei Wang,Zhen Zheng,Peiyu Sun,Xinling Wang,Feng-Chih Chang

Two kinds of novel peptide mimetic pepsin-inspired degradable polyurethanes were synthesized and characterized. A synthetic pseudo tripeptide acted as chain extender and provided active sites for pepsin biodegradation. In vitrodegradation behavior was investigated in simulated gastric fluid containing pepsin, and assessed by 1H NMR, mass loss and SEM. The results indicated that the pseudo peptide bonds in polyurethanes were effectively digested by the action of pepsin. Furthermore, the rate and degree of pepsin-inspired polyurethanes in simulated gastric fluid containing pepsin could be controlled by alteration of the soft segments and activity of pepsin. The influence of the polymers on human umbilical vein endothelial cells was evaluated by WST-1 assay. The results indicated that the polymers sustained much higher cell viability than the controls.

Enzymatically degradable oxidized dextran–chitosan hydrogels with an anisotropic aligned porous structure

Jingjun Wu,Qian Zhao,Chengzhen Liang,Tao Xie

Enzyme-degradable hydrogels with aligned pores were prepared via unidirectional freezing of a mixed solution of oxidized dextran (oxDex) and chitosan (CHI) followed by cryo-gelation. At the frozen state, imine bonds as the crosslinking points of the hydrogels were formed as a result of the reaction between the aldehyde groups on the oxDex and the amino groups on the CHI. The morphologies of the obtained hydrogels were studied by confocal laser scanning microscopy (CLSM). Microtubular pores aligned along the freezing direction were obtained in the swollen state of the hydrogels. The pore size was adjustable ranging from 20 to 100 μm by controlling the freezing temperature in the unidirectional freezing step. Upon exposure of the hydrogels to chitosanase solution, chitosan can be hydrolyzed into short oligosaccharide fragments leading to collapse and degradation of the gel network. The degradation rate can be tuned by adjusting the gel composition and enzyme concentration.
