Btn2p influences arginine metabolism and ethanol tolerance in Saccharomyces cerevisiae during Huangjiu fermentation

Ruosi Fang,Ting Xia,Shuhui Wu,Jiamin He,Gongnian Xiao

Ethanol produced during the Huangjiu fermentation can adversely affect the growth and metabolism of Saccharomyces cerevisiae . It has been established that arginine, one of the major amino acids found in Huangjiu , can protect yeast cells against ethanol stress. Previous studies have indicated that BTN2 , which influences arginine metabolism, is associated with ethanol tolerance in yeast. In this study, we evaluated the response of BTN2 to ethanol stress based on analyses of cell growth, morphology, membrane permeability, membrane integrity, ROS and how BTN2 influenced intracellular metabolites, enzymes, and transcriptional genes in the arginine metabolic pathway. Our findings revealed that BTN2 promoted significantly increases in the growth and ethanol tolerance of S. cerevisiae g. Under 10% ethanol stress, RT-qPCR showed that BTN2 Δ ( BTN2 knock out strain) up-regulated CAR1 , CAR2 , GAP1 , DUR1,2 and DUR3 to increase arginine and urea degradation, BTN2 ↑ ( BTN2 over-expressing strain) up-regulated CAR1 , CAN1 and DUR3 to enhance arginine uptake, catabolism and urea reabsorption, while TPS1 was up-regulated to improve ethanol tolerance ability. Yeast up-regulated both CAR1 (Arginase) and DUR3 (urea transporter) genes in BTN2 knockout and over-expression strains to counteract the ethanol effects. Our findings revealed a novel arginine metabolism under ethanol tolerance of a core functional yeast during Huangjiu fermentation. These findings could contribute to the development of an approach for inhibiting toxic ethyl carbamate (EC) production during Huangjiu fermentation.

Promoting Proton Conductivity and Methanol-Resistance of PVdF-Based PEM Membrane by Incorporating Dispersive CNT Dotted With α-Zr(HPO4)2∙H2O

Pengtao Lei,Xiangyang Xu,Chenyu Huang,Dongsheng Chen,Zhenkun Jiang

To optimize the proton transport performance of polyvinylidene fluoride (PVdF), a proton exchange membrane (PEM) material, carbon nanotube (CNT), and zirconium hydrogen phosphate (ZrP) have been blended into the PVdF matrix. Adopting a hydrothermal synthesis route, the acidic ZrP can be better conjugated with the positively charged CNT functionalized by cetyltrimethylammonium bromide (CTAB). As visualized using electroscopes, ZrP nanocrystallines are intimately dotted and homogeneously distributed on CNT. The ZrP–PVdF interaction can be attributed to hydrogen bonding between hydroxyl and F-terminal groups. By incorporating well CNT/ZrP into PVdF via solution casting, the pore architecture can be regulated; namely, the macro-voids in PVdF are greatly reduced. The composite membranes exhibit tensile stress of 48–52 MPa while sustaining the thermal stability of PVdF. The incorporated CNT/ZrP can suppress void porosity and enhance film hydrophilicity. With improved wettability, the methanol permeability can be reduced to less than 2 × 10 −9 cm 2 ∙s −1 , ca. one-sixth that of pure PVdF. The proton conductivity for PVdF-CNT 0.03 -ZrP 0.06 , with CNT and ZrP mass content respectively 3 and 6 wt% that of PVdF, attains 0.089 S·cm −1 at 100°C, showing upgraded conducting capability. The membrane selectivity can be remarkably enhanced, verifying that CNT/ZrP blending is a facile and profitable approach for PEM optimization.

Solid–Solid Separation Approach for Preparation of Carbon-Supported Cobalt Carbide Nanoparticle Catalysts for Oxygen Reduction

Sefiu Abolaji Rasaki,Hangjia Shen,Tiju Thomas,Minghui Yang

Transition metal carbides have received significant attention for oxygen reduction reaction (ORR) applications primarily owing to their good conductivity. However, cobalt carbide based catalysts (CoCx) are difficult to prepare, owing to the preferential formation of metallic cobalt and cobalt oxide instead. Also, cobalt carbide nanoparticles rapidly coarsen and undergo grain growth. This is a problem as it results in low current density and poor active site exposures. In this study, carbon-supported CoCx (C@CoCx) is successfully synthesized using the solid–solid separation synthesis method. The limitations identified to be associated with cobalt carbide nanoparticle based electrocatalysts are overcome. Here, the synthetic approach ensures good distribution and protection of CoCx on in situ generated carbon nanotubes. This results in robust activities with an appreciable onset potential of ∼0.92 V, a half-wave potential of ∼0.8 V, and a durability of ∼100% (tested on ∼22 h continuous operation runs). In alkaline medium, C@CoCx compares favorably with commercial platinum dispersed on carbon (20 wt % Pt/C) and shows a higher current density of ∼300 μA/cm2 at an electrode potential of 0.6 V. All of these indicate that there is a synergistic charge transfer between CoCx and an in situ generated carbon nanotube. This is due to intimate contact that exists between the two phases; it is in fact responsible for superior methanol tolerance observed, making it a good candidate for direct methanol fuel cells.

High catalytic activity of Ti4O7/CNTs oxygen reduction reaction (ORR) electrocatalysts with excellent circulation and methanol resistance

Lei Yimin,Wei Yuxiang,Wu Dekai,Zhu Qing,Sun Yanan,Zhou Xuejiao,Xi He,Zhong Peng,Sun Jie

Electro catalysts play a vital role in facilitating the reaction process in the oxygen reduction reaction (ORR) of fuel cells. In methanol fuel cells, methanol easily penetrates the diaphragm rendering the ORR catalyst inactive. Although commercial platinum exhibits excellent ORR catalytic activity, its poor methanol resistance and long-time durability severely restrict its sustainable development and application. This work synthesized Ti 4 O 7 /CNT nanocomposite electrocatalysts by using the sol-gel method followed by high-temperature carbothermal reduction. The obtained catalysts exhibit a large electrochemically active surface area (347.89 cm 2 ) and low interfacial charge transfer resistance (64.5 Ω). The reduction potential (0.73 V), onset potential (0.93 V), half-wave potential (0.73 V), and electron transfer number (3.7) of this electrocatalyst are all close to those of the Pt/C electrocatalysts in alkaline medium. Simultaneously, it also possesses remarkable methanol tolerance and long-term durability, whose relative current density can maintain above 74% after cycling for 24 h.

Efficient asymmetric synthesis of chiral alcohols using high 2-propanol tolerance alcohol dehydrogenase SmADH2 via an environmentally friendly TBCR system

Zeyu Yang,Hengwei Fu,Wenjie Ye,Youyu Xie,Qinghai Liu,Hualei Wang,Dongzhi Wei

Alcohol dehydrogenases (ADHs) together with the economical substrate-coupled cofactor regeneration system play a pivotal role in the asymmetric synthesis of chiral alcohols; however, severe challenges concerning the poor tolerance of enzymes to 2-propanol and the adverse effects of the by-product, acetone, limit its applications, causing this strategy to lapse. Herein, a novel ADH gene smadh2 was identified from Stenotrophomonas maltophilia by traditional genome mining technology. The gene was cloned into Escherichia coli cells and then expressed to yield SmADH2. SmADH2 has a broad substrate spectrum and exhibits excellent tolerance and superb activity to 2-propanol even at 10.5 M (80%, v/v) concentration. Moreover, a new thermostatic bubble column reactor (TBCR) system is successfully designed to alleviate the inhibition of the by-product acetone by gas flow and continuously supplement 2-propanol. The organic waste can be simultaneously recovered for the purpose of green synthesis. In the sustainable system, structurally diverse chiral alcohols are synthesised at a high substrate loading (>150 g L−1) without adding external coenzymes. Among these, about 780 g L−1 (6 M) ethyl acetoacetate is completely converted into ethyl (R)-3-hydroxybutyrate in only 2.5 h with 99.9% ee and 7488 g L−1 d−1 space–time yield. Molecular dynamics simulation results shed light on the high catalytic activity toward the substrate. Therefore, the high 2-propanol tolerance SmADH2 with the TBCR system proves to be a potent biocatalytic strategy for the synthesis of chiral alcohols on an industrial scale.

PtCoN supported on TiN-modified carbon nanotubes (PtCoN/TiN–CNT) as efficient oxygen reduction reaction catalysts in acidic medium

Shengzhou Chen,Qiuchan Huang,Wei Yang,Hanbo Zou,Huangwang Mai,JiaHai Wang

Herein, new catalysts for the oxygen reduction reaction (ORR) (Pt CoN nanoparticles supported on TiN–carbon nanotubes (CNTs)) were successfully prepared via the simple polymerized complex method and heat treatment in NH 3 . Through this facile two-step synthesis, metal chlorides were first transformed to the corresponding oxides, then reduced to metal particles or nitrides . The synthesized Pt CoN/TiN–CNT electrocatalysts showed excellent electrochemical activity for oxygen reduction and high tolerance against methanol in acidic medium. The ORR onset potential of Pt 0.3 CoN/TiN–CNT (0.812 V vs. RHE) is higher than that of 40 wt% commercial Pt/C (0.789 V vs. RHE) due to the synergistic effect between nano-platinum particles and metal nitrides. The TiN-coated CNT support provided high electron transfer rate and catalytic tolerance in methanol solution. The number of transferred electrons was examined by rotating ring-disk electrode measurements, which concurred with the result of the Koutecky–Levich equation for a four-electron mechanism.

Au Nanowires@Pd-Polyethylenimine Nanohybrids as Highly Active and Methanol-Tolerant Electrocatalysts toward Oxygen Reduction Reaction in Alkaline Media

Qi Xue,Juan Bai,Congcong Han,Pei Chen,Jia-Xing Jiang,Yu Chen

Optimizing the shape, structure, and interface of noble metal bimetallic nanostructures significantly improves their catalytic properties. Recently, Au@Pd core–shell bimetallic nanostructures have attracted considerable interest for fuel cell applications. Regrettably, most of the research mainly focuses on the morphological and structural control whereas interfacial control is neglected. In this work, polyethylenimine (PEI) functionalized Au nanowires@Pd core–shell bimetallic nanohybrids (Au-NWs@Pd@PEI) are synthesized by a chemical reduction method using Au nanowires (Au-NWs) as seeds in the presence of PEI and demonstrate their application toward oxygen reduction reaction (ORR) in an alkaline solution. Electrochemical results display that Au-NWs@Pd@PEI with optimal component shows enhanced ORR activity, selectivity, and durability compared to Pt/C electrocatalyst. Physical characterizations demonstrate that the electronic property of Pd shell mainly is affected by PEI polymer rather than Au-NWs core. Relative to PEI functionalized Pd nanowires without Au core, ORR activity enhancement of Au-NWs@Pd@PEI is exclusively ascribed to the surface strain effect. Meanwhile, loose-packed PEI polymer on the Pd surface can work as barrier nanosieves to exclusively prevent the access of methanol to Pd sites, which imparts Au-NWs@Pd@PEI with enhanced methanol tolerance toward ORR in an alkaline solution.

Salt-Resistant and Ethanol-Resistant Monoamine Oxidases: New Sight for yobN Mining from Bacillus and Biogenic Amine Degradation Mechanism in Fermented Food

Qilin Yang,Shuangping Liu,Aibao Sun,Xin Li,Tiantian Liu,Xiao Han,Jian Mao

Amine oxidases have strong capabilities for the degradation of biogenic amines (BAs) in fermented foods. However, their application is limited by substrate specificity, and high concentrations of ethanol and salt can hinder their effectiveness. This study presents a novel approach utilizing comparative genomics, protein clustering analysis, and full homology modeling to identify three amine oxidases: KCYOBN from the salt-resistant Bacillus subtilis, and LYYOBN1 and LYYOBN2 from the ethanol-resistant Bacillus cereus. These enzymes are highly similar in structure and exhibit broad substrate specificities. KCYOBN maintains over 84% relative activity at 20% (w/v) NaCl, while LYYOBN1 and LYYOBN2 retain over 32 and 21% relative activity at 25% vol ethanol. Variations in key residues are one of the reasons for the differences in tolerance. In fermented foods, KCYOBN degraded 45.97% of the total BAs in cooking wine and 38.33% in fish sauce. LYYOBN1 achieved the highest degradation rate of 32.93% in huangjiu. LYYOBN2 exhibited degradation rates of 30.00% in soy sauce and 35.14% in wine with no significant impact on flavor compounds. The significance of this work lies in the identification of the novel salt-resistant KCYOBN and ethanol-resistant LYYOBN1 and LYYOBN2 through this new method, which can simultaneously degrade multiple BAs and have a broad application potential.

Simple one-pot synthesis of platinum-palladium nanoflowers with enhanced catalytic activity and methanol-tolerance for oxygen reduction in acid media

Jie-Ning Zheng,Li-Li He,Fang-Yi Chen,Ai-Jun Wang,Meng-Wei Xue,Jiu-Ju Feng

In this work, bimetallic alloyed platinum-palladium (PtPd) nanoflowers are fabricated by one-pot solvothermal co-reduction of Pt (II) acetylacetonate and Pd (II) acetylacetonate in oleylamine system. The as-prepared nanostructures show the enhanced electrocatalytic activity for oxygen reduction reaction (ORR), dominated by a four-electron pathway based on the Koutecky-Levich plots, mainly owing to the inhibition of the formation of Pt–OH ad via the downshift of d-band center for Pt. Meanwhile, PtPd nanoflowers display good methanol tolerance and improved stability for ORR. The chronoamperometry test reveals that the current of PtPd nanoflowers remains 45.9% of its original value within 6000 s, much higher than those of commercial Pt (36.7%) and Pd (32.2%) black catalysts. Therefore, PtPd nanoflowers with unique interconnected structures can be used as a promising cathode catalyst in direct methanol fuel cells.

Establishment and application of a modified membrane-blot assay for Rhizomucor miehei lipases aimed at improving their methanol tolerance and thermostability

Dong He,Wen Luo,Zhiyuan Wang,Pengmei Lv,Zhenhong Yuan,Shaowei Huang,Jingliang Xv

Directed evolution has been proved an effective way to improve the stability of proteins, but high throughput screening assays for directed evolution with simultaneous improvement of two or more properties are still rare. In this study, we aimed to establish a membrane-blot assay for use in the high-throughput screening of Rhizomucor miehei lipases (RMLs). With the assistance of the membrane-blot screening assay, a mutant E47K named G10 that showed improved thermal stability was detected in the first round of error-prone PCR. Using G10 as the parent, two variants G10-11 and G10-20 that showed improved thermal stability and methanol tolerance without loss of activity compared to the wild type RML were obtained. The T 50 60 -value of G10-11 and G10-20 increased by 12   °C and 6.5   °C, respectively. After incubation for 1   h, the remaining residual activity of G10-11 and G10-20 was 63.45% and 74.33%, respectively, in 50% methanol, and 15.98% and 30.22%, respectively, in 80% methanol. Thus, we successfully developed a membrane-blot assay that could be used for the high-throughput screening of RMLs with improved thermostability and methanol tolerance. Based on our findings, we believe that our newly developed membrane-blot assay will have potential applications in directed evolution in the future.

Tannic acid decorated AuPd lavender-like nanochains for enhanced oxygen reduction electrocatalysis

Shuli Yin,Songliang Liu,Shiqian Jiao,Hugang Zhang,You Xu,Ziqiang Wang,Xiaonian Li,Liang Wang,Hongjing Wang

Benefiting from the interaction of inorganic and organic building blocks, the design of metal–organic nanohybrids is of great significance for the oxygen reduction reaction (ORR). Herein, a universal strategy is presented to controllably synthesize tannic acid (TA) functionalized AuPd lavender-like nanochains (AuPd@TA LCs) by a chemical reduction method at low temperature. Due to the unique structural characteristics and polyphenolic modification, the AuPd@TA LCs show superior ORR performance in an alkaline electrolyte. Moreover, the surface decorated TA polymers can act as a molecular screen window to prevent methanol molecules from accessing the active site of the catalyst, thus resulting in high methanol tolerance and catalytic stability. The presented molecular screen window strategy is highly valuable for designing ORR electrocatalysts with high methanol tolerance.

Copper-Based Catalysts Confined in Carbon Nanocage Reactors for Condensed Ester Hydrogenation: Tuning Copper Species by Confined SiO2 and Methanol Resistance

Yaqi Yao,Xiaoqian Wu,Bo Chen,Zhiyu Tu,Oliver Y. Gutiérrez,Ying Cui,Jie Wang,Jiacheng Huang,Yan Xu,Han Sun,Haijun Chen,Zhenxin Yan,Donghai Mei,Yujun Zhao,Johannes A. Lercher

Hydrogenation of aliphatic esters to natural alcohols is an important strategy for the efficient utilization of biomass-derived oils. The synthesis of highly active copper (Cu)-based catalysts is a challenge for condensed-phase ester hydrogenation due to the difficulties in controlling active sites and catalyst deactivation. Here, a copper-based catalyst confined in a carbon nanocage reactor was successfully designed and prepared. The copper catalyst with 56 wt % SiO2 exhibited the best performance because of the optimum proportion of Cu+ and Cu0 sites and high dispersion. The interaction between SiO2 and Cu particles contributes to the formation of Cu+ species, which is the key site for the adsorption of carbonyl groups. Meanwhile, the confinement effect of the carbon nanocages effectively inhibited the agglomeration of the copper particles. The catalysts exhibited not only excellent thermal stability but also superior methanol resistance in comparison with the Cu/SiO2 catalyst. On the basis of the density functional theory (DFT) calculations results, methanol resistance should be attributed to the fewer hydroxyl groups on the catalyst surface, which increase the activation barrier for the dissociation of silica, allowing the stable holding of the copper species in the methanol solvent.
