In this study, a pH-responsive amphiphilic alginate (Ugi-Alg) was synthesized via Ugi reaction without using a catalyst. The structure of Ugi-Alg was confirmed by FT-IR and 1H NMR spectroscopy. Amphiphilic alginate can form micelles in an aqueous medium due to it's amphiphilic nature.. The impacts of Na+ concentration and pH on the micelle size were characterized by dynamic light scattering (DLS) and transmission electron microscopy (TEM). The dynamic light scattering observations showed that micelle size increases with the decrease in Na+ concentration in aqueous solution. However, the micelle size decreases first and then increases as the pH value decreases from 5.3 to 2.0. Transmission electron microscopy confirmed that the mean size of micelles is 30–200 nm. In addition, a model hydrophobic pesticide (acetamiprid) was loaded in the micelles. The encapsulation efficiency and release behavior of micelles were studied, which could be controlled by Na+ concentration and pH. The results indicated that encapsulation efficiency of acetamiprid increases from 55% to 96% due to the increase in Na+ concentration from 0.01 M to 0.3 M. Moreover, with the decrease in pH from 5.3 to 2.0, encapsulation efficiency increases from 55% to 80%. Furthermore, the data of acetamiprid release kinetics could be well-fitted by the Weibull model.

The extensive use of pesticides in modern agriculture has raised concerns about environmental contamination and adverse human health effects. Therefore, developing highly sensitive detection methods to identify pesticide residues is crucial for food safety and ecosystem protection. In this study, the Fe 2 AlB 2 MAX phase was synthesized and characterized. After evaluating its peroxidase-mimetic performance using a colorimetric method, a new, sensitive, and simple dual fluorescence-colorimetric sensor was developed for the quantification of two common pesticides, acetamiprid (ACP) and imidacloprid (IMP), using fluorescein (FL). The developed method is based on the inhibitory impact of ACP and IMP on the enzymatic performance of the Fe 2 AlB 2 MAX phase, specifically the inhibition of hydroxyl radical ( OH) generation, which enhances the absorption and emission intensities of FL. The results confirmed that OH generated through the breakdown of H 2 O 2 via the catalytic activity of the MAX phase can decrease the intrinsic absorption and emission intensities of FL. However, ACP and IMP inhibit the peroxidase-like activity of the MAX phase, leading to increased absorption and emission intensities of FL. The limit of detection values calculated for spectrophotometric and spectrofluorimetric quantifications were 2.8 μM and 0.051 μM for ACP and 1.72 μM and 0.013 μM for IMP, respectively. Furthermore, the proposed method was successfully utilized to accurately and reliably determine ACP and IMP in spiked real samples with satisfactory accuracy and precision. These developed methods offer several advantages, making them promising candidates for the direct, rapid screening of pesticide residues.

Time-resolved fluorescent lateral immunoassay strip (TRFLIS) is a reliable and rapid method for detecting acetamiprid. However, its sensitivity is often affected by the structural patterns and stability of the fluorescent probe. Researchers have shown significant interests in using goat anti-mouse IgG (GaMIgG) which is indirectly bound to time-resolved fluorescent microsphere (TRFM) and antibody. This allowed for oriented modification of the antibody. However, the stability of fluorescent probe in this binding mode remained unexplored. Herein, 1-ethyl-(3-dimethylaminopropyl) carbodiimide hydrochloride was innovatively used as a cross-linking agent to enhance the binding of antibody to GaMIgG, which improved the stability of the fluorescent probe. Under optimal working conditions, this strategy exhibited a wide linear response range of 5–700 ng/mL. Its limit of detection (LOD) was 0.62 ng/mL, the visual LOD was 5 ng/mL, and the limit of quantification (LOQ) of 2.06 ng/mL. Additionally, under tomato matrix, leek matrix and Chinese cabbage matrix, the linear response ranges were 5–400, 5–300, and 5–700 ng/mL, with LODs of 0.16, 0.60, and 0.41 ng/mL, with LOQs of 0.53, 2.01 and 1.37 ng/mL, respectively. In conclusion, this strategy effectively reduced the dosage of acetamiprid antibody compared with TRFM directly linking acetamiprid antibody, and greatly increased the sensitivity of TRFLIS. Meanwhile, it demonstrated outstanding specificity and accuracy in acetamiprid detection and had been successfully applied to vegetable samples. This method enables rapid and accurate detection of large-volume samples by combining qualitative and quantitative methods. As such, it has great potential in the development of low-cost and high-performance immunochromatographic platforms.
