Citrus fruits are susceptible to microbial infection during storage and transportation, leading to weight loss, quality deterioration and even physiological decay. In this study, a layer-by-layer (LbL) self-assembly edible coating based on chitosan and Sodium carboxymethyl cellulose (CMC) was prepared using electrostatic deposition. Postharvest citrus fruits (Citrus reticulatecv. ‘Chunxiang’) were coated either with the LbL coating, which exhibited an increased transmission rate of water vapor, or with single chitosan coating. The data of some physiological indexes of citrus coated with LbL coating and single coating were compared after being stored at 13 ± 2 °C and at relative humidity (RH) at 80–85% (shelf-life condition) for 10 days. Results demonstrated that the LbL deposited coating was effective at maintaining morphological and microstructural attributes, increasing the surface brightness, firmness and the content of titratable acid and ascorbic acid, as well as reducing the weight loss after storage by about 0.8%. Results in the present study indicated that the LbL edible coating could have the potential to maintain postharvest citrus quality during storage. Keywords:chitosan;CMC;FTIR;SEM;postharvest citrus;quality

Citrus is susceptible to Penicillium digitatum ( P. digitatum ) infection in post-harvest storage, resulting in enormous economic losses. This study aimed to investigate the antifungal activity and potential mechanism of the combination of Polymethoxylated flavones (PMFs) and citral (two natural antifungal components derived from citrus) against P. digitatum in vitro and citrus fruit. The results show that PMFs can enhance the antifungal activity of citral nanoemulsion, and PMFs-loaded citral nanoemulsion (PCT) has significant antifungal activity in a concentration-dependent manner. PCT can evidently inhibit spore germination and mycelial growth in vitro , and effectively control the growth of green mold on postharvest citrus fruit. Furthermore, PCT treatment resulted in the alteration of mycelia morphology, accumulation of reactive oxygen species, and membrane lipid peroxidation. These changes can disrupt the normal structure and function of the cell membrane, as evidenced by the reduction of total lipid and ergosterol content in the mycelia and the stronger red fluorescence of the cells emitted after PI staining. Based on the above results, we infer that PCT has a strong inhibitory effect on P. digitatum , and its potential mechanism is related to the destruction of the cell membrane. Therefore, PCT can be considered as a botanical fungicide for citrus preservation.

Citrus fruit post-harvest decay has caused economic loss for fruit-dependent industries worldwide in recent decades. However, bio-preservatives offer a promising and sustainable solution. In this study, culture experiments were carried out to assess the ability of Bacillus velezensis HY19, which we previously identified, to inhibit common pathogenic fungi in postharvest citrus fruit. HY19 was capable of producing extracellular siderophores and hydrolases (protease, phosphatase, and cellulase). The cells and metabolites (fermentation broth and volatiles) of the bacterium exhibited strong suppression against Penicillium italicum , P. digitatum , and Colletotrichum gloeosporioides . LC-MS analysis of B. velezensis HY19 fermentation broth (BFB) revealed 13 antimicrobial substances (simvastatin, cinnamic acid, cyclopentanone, etc.), 3 antifungal pesticide intermediates (M-cresol, 4-hydroxybenzaldehyde, and L-menthyl acetoacetate), and P-salicylic acid. GC-MS detected 18 antimicrobial substances (2-nonanol, 2-nonanone, benzoic acid, etc.) in HY19's volatile organic compounds (VOCs). Transcriptome analysis of P. digitatum exposed to BFB stress revealed that numerous genes involved in diverse biological processes, such as DNA replication and cell wall biosynthesis, were differentially expressed. Moreover, BFB inhibited the expression of multiple genes responsible for the biosynthesis of N-glycan and proteins in P. digitatum . BFB enhanced antioxidant enzyme activities in citrus peels, reduced electrolyte leakage and reactive oxygen species (ROS) accumulation, and induced the disease resistance in citrus fruit, leading to the decrease in the infection of pathogenic fungi on citrus fruit. During storage at low-temperature (4–8 °C), BFB inhibited the disease incidence in citrus fruit for up to 90 days and achieved over 60% control efficacy. Additionally, BFB had no impact on fruit quality, making it a lower risk option. Considering the benefits of sustainability and environmentally friendly options, it could be concluded that BFB was an effective alternative for controlling citrus diseases and had the potential to be used as a fruit preservative.

This study investigated the efficacy of trans-2-hexenal in preventing green mold on postharvest citrus fruit and its mode of action against imazalil-resistant Penicillium digitatum Pdw03. Trans-2-hexenal inhibited Pdw03 at a minimum inhibitory concentration (MIC) and a minimum fungicidal concentration (MFC) of 0.25 mL L −1 . Trans-2-hexenal reduced the incidence of green mold, which is a plant volatile compound with potent antifungal properties . Ultrastructural analysis revealed that trans-2-hexenal altered the mitochondrial structure of Pdw03. Trans-2-hexenal also increased the amount of reactive oxygen species, which hindered mitochondrial energy metabolism of Pdw03, resulting in mitochondrial structural damage and malfunction, and a decrease in mitochondrial membrane potential. In addition, RNA-seq analysis revealed that trans-2-hexenal altered the expression of some energy metabolism genes associated with the tricarboxylic acid (TCA) cycle, glycolysis, and oxidative phosphorylation of Pdw03. Superoxide dismutase , catalase , glutathione S-transferase, and glutathione peroxidase activities were altered, and expression of the relevant gene was also quantified by real-time fluorescence quantitative PCR . Trans-2-hexenal damaged ribosomes, primarily by downregulation of the ribosomal structural component-associated genes RPL3 , RPS18 , RPL12 , RPL26 , RPL13A , RPL9 , RPL23 , and RPL4 , and then by reducing protein synthesis to achieve the inhibitory effect.

This study aimed to investigate the effect of Pichia galeiformis on disease resistance and elucidate the changes in phenylpropane biosynthesis treated by P. galeiformis in postharvest citrus. The results showed that P. galeiformis reduced the disease incidence and lesion diameters without direct contact with the pathogen Penicillium digitatum. Transcriptome analysis revealed that phenylpropanoid biosynthesis was triggered by P. galeiformis. Genes encoding phenylpropanoid biosynthesis were upregulated, including phenylalanine ammonia-lyase (PAL), 4-coumaroyl-CoA ligase (4CL), cinnamate-4-hydroxylase (C4H), peroxidase (POD), cinnamyl alcohol dehydrogenase (CAD), O-methyltransferase, and hydroxyl cinnamoyl transferase. Moreover, P. galeiformis increased the activity of PAL, 4CL, C4H, POD, polyphenol oxidase, and CAD in citrus pericarp. In addition, P. galeiformis treated citrus displayed higher levels of total phenolic compounds, flavonoid, and lignin and higher amounts of ferulic and sinapic acid. In conclusion, these results suggested that P. galeiformis could induce resistance through modulating the pathway of phenylpropanoid biosynthesis in postharvest citrus.
