Copper is often used as a growth promoter, at the same time copper is one of the most important essential trace elements for fur animals, especially Rex rabbits. However, too much copper added to the diet may harm animal health, and copper excreted in feces can pollute the environment. In this study, 3-month-old Rex rabbits were randomly divided into four groups and fed a basal diet containing 0, 30, 60, or 120 mg/kg Cu for 5 weeks. The diet supplemented with 30 mg/kg Cu significantly increased ( P < 0.05) the average daily feed intake (ADFI) and the average daily gain (ADG) and also the activity of serum Cu–Zn (zinc) superoxide dismutase and the digestibility of ether extract. Supplemental Cu up to 120 mg/kg did not significantly adversely affect the Zn metabolism of growing Rex rabbits. Overall, the data in this study indicate that 30 mg/kg is the optimal level of Cu supplementation in the diet of growing Rex rabbits. The results will provide a reference to improve the breeding of Rex rabbits and possibly other animals. In follow-up studies, the amount of copper in the diet should be reduced as much as possible from the baseline of 30 mg/kg copper.

Amino acids in the brain modulate eating behavior and energy balance. The aim of the present study was to investigate the effects of DL-methionine (DL-Met), L-lysine monohydrochloride (L-Lys-HCl), and taurine (Tau) on feed intake and the mRNA expression levels of appetite-related hypothalamic neuropeptides in chicks. DL-Met, L-Lys-HCl, or Tau was intracerebroventricularly (ICV) administered to 5-day-old layer-type chicks, and the feed intake was recorded until 1 h post-injection. Quantitative PCR was performed to determine the hypothalamic mRNA expression levels of neuropeptide Y ( NPY ) and its receptors, agouti-related protein ( AgRP ), pro-opiomelanocortin ( POMC ), cocaine- and amphetamine-regulated transcript ( CART ), and melanocortin receptors. Our results indicated that injection time and treatment have a significant effect on food intake. Food intake decreased significantly at 30 and 60 minutes after ICV administration of DL-Met (1.678 μmol), L-Lys-HCl (6.856 μmol), and Tau (5.006 μmol) compared to the control group ( P > 0.05). On the contrary, a high dose of each amino acid did not alter feed intake ( P > 0.05). The result showed that ICV injection of DL-Met (1.678 μmol) significantly decreased the mRNA expressions of NPY , NPY4R , and NPY6R ( P < 0.05), but had no significant effect on the mRNA expressions of NPY1R, NPY5R, and AgRP in the hypothalamus ( P > 0.05). Similarly, ICV injection of L-Lys-HCl (6.856 μmol) and Tau (5.006 μmol) significantly decreased mRNA levels of NPY, NPY4R, and NPY6R ( P < 0.05), without affecting the mRNA expressions of NPY1R, NPY5R, and AgRP ( P > 0.05). DL-Met had no effect on anorexigenic gene expression ( P > 0.05). L-Lys-HCl upregulated MC1R ( P < 0.05), and Tau upregulated POMC and MC3R ( P < 0.05), and both had no influence on CART, MC4R , or MC5R ( P > 0.05). The result suggests that both orexigenic and anorexigenic neuropeptide genes are involved in the effects of DL-Met, L-Lys-HCl, and Tau. The effect of DL-Met, L-Lys-HCl, and Tau acts in a dose-dependent manner in the hypothalamus to influence feed intake.

Piglet enteritis is a major problem that needs to be solved urgently in modern pig production. Paeonol (Pae) has been used as a novel treatment option due to its good medicinal value. This study purported to elucidate the regulatory mechanism of Pae on dextran sodium sulfate (DSS)-induced ulcerative colitis (UC) in weaned piglets. A total of 36 crossbred (Duroc × Landrace × Yorkshire) weaned piglets were stochastically split into six groups: the control group, DSS group, 0.2% Pae group, 0.4% Pae group, 0.8% Pae group, and mesalazine group. The control and DSS groups were fed with a basic diet, the three Pae and mesalazine groups were fed with 0.2, 0.4, 0.8%, and 2 g mesalazine per kilogram of basic diet throughout the study. On the 15th day of the test period, the control group was gavaged with 10 ml of normal saline, while the remaining five groups were gavaged with 10 ml 5% DSS solution for 13 days. The study lasted for 27 days. The results showed that the 0.8% Pae group significantly increased the average daily feed intake (ADFI) and Occludin mRNA expression in the colon of piglets ( P < 0.05). The 0.2% Pae group markedly increased the average daily gain (ADG) and zonula occludens-1 (ZO-1) mRNA expression ( P < 0.05). In the 0.2% and 0.4% Pae groups, the feed-to-gain ratio (F/G) was significantly reduced and the mRNA expression levels of Caspase-8, respectively, markedly enhanced the mRNA expression levels of transforming growth factor-β (TGF-β) and interleukins-4 (IL-4) ( P < 0.05). In the 0.8% Pae group, the relative abundance of Campilobacterota was significantly reduced ( P < 0.05). In the 0.4% Pae group, the relative abundance of Firmicutes was notably increased ( P < 0.05). In the 0.2 and 0.8% Pae groups, the relative abundance of Prevotella was markedly increased ( P < 0.05). In the 0.2% Pae group, the contents of propionic acid, butyric acid, and valerate acid were markedly higher ( P < 0.05). Thus, it is speculated that Pae may regulate the balance of anti-inflammatory/pro-inflammatory factors, improve intestinal tight junction expression, reduce apoptosis, and improve intestinal microflora structure and growth performance of piglets, thereby restoring intestinal barrier function and alleviating DSS-induced UC in piglets.
