Nanodiamonds (NDs) with nitrogen-vacancy (NV) centers are emerging as powerful quantum nanosensors (QNs) in biomedical applications due to their exceptional sensitivity. However, achieving optimal diagnostics performance necessitates both high sensitivity and selectivity; especially in practical biomedical settings, it remains challenging for QNs to provide quantitative analyses when multiple analytes are present. Here, we present a biosensing platform that integrates DNA logic gates (DLGs) with spin-based quantum sensing, termed DLG-QN for ultrasensitive and ultraselective diagnostics. Utilizing an AND DLG, both NDs and magnetic beads (MBs) are functionalized with hairpin DNA strands. In the presence of both miRNA-21 and miRNA-155─key biomarkers overexpressed in cancer─the hairpin DNAs undergo conformational changes that facilitate DNA-guided self-assembly of NDs and MBs, enriching the target signal. Resonant microwave modulation of ND fluorescence emission allows for high signal-to-noise ratio (SNR) detection by separating the signal from background fluorescence via spin-enhanced analysis. This platform demonstrated ultrasensitive and ultraselective detection of miRNA-21 and miRNA-155 with a limit of detection of 19.8 fM, highlighting its potential as a general biosensing strategy for precision diagnostics involving multiple biomarkers.

Sensitive and accurate miRNA detection is important in cancer diagnosis but remains challenging owing to the essential features of miRNAs, such as their small size, high homology, and low abundance. This work proposes a novel electrochemical (EC)-enhanced quantum sensor achieving quantitative detection of miRNA-155 with simultaneous EC sensing. Specifically, fluorescent nanodiamonds/MXene nanocomposites were synthesized and modified with dual-mode signal labels, enabling miRNA-155 concentration measurement via T1 relaxation time of nitrogen-vacancy (NV) centers and EC signals. Quantum sensing was enhanced via external voltage during the EC process, which modulated the negatively charged state of the NV centers, thereby improving the sensitivity and accuracy of miRNA-155 detection. EC sensing improved the accuracy and reliability of miRNA-155 detection while enhancing quantum sensing. The limit of detection (LOD) of the EC-enhanced quantum biosensor reached 10.0 aM, nearly 106 and 10 times lower than the reported LODs of a quantum sensor using bulk diamond and fluorescent sensors, respectively. The LOD of EC sensing was 2.6 aM, aligning with previous reports. The findings of the study indicated that quantum sensing combined with EC sensing can achieve ultrasensitive miRNA-155 detection with high accuracy and reliability, providing an advanced approach for early cancer diagnosis.

Conventional point-of-care testing (POCT) has limitations in sensitivity with high risks of missed detection or false positive, which restrains its applications for routine outpatient care analysis and early clinical diagnosis. By merits of the cutting-edge quantum precision metrology, this study devised a mini quantum sensor via magnetofluorescent qubit tagging and tunning on core–shelled fluorescent nanodiamond FND@SiO2. Comprehensive characterizations confirmed the formation of FND biolabels, while spectroscopies secured no degradation in spin-state transition after surface modification. A methodical parametrization was deliberated and decided, accomplishing a wide-field modulation depth ≥15% in ∼ zero field, which laid foundation for supersensitive sensing at single-FND resolution. Using viral nucleocapsid protein as a model marker, an ultralow limit of detection (LOD) was obtained by lock-in analysis, outperforming conventional colorimetry and immunofluorescence by > 2000 fold. Multianalyte and affinity assays were also enabled on this platform. Further by resort to artificial-intelligence (AI) augmentation in the Unet-ConvLSTM-Attention architecture, authentic qubit dots were identified by pixelwise survey through pristine qubit queues. Such processing not just improved pronouncedly the probing precision but also achieved deterministic detections down to a single protein in human saliva with an ultimate LOD as much as 7800-times lower than that of colloidal Au approach, which competed with the RT-qPCR threshold and the certified critical value of SIMOA, the gold standard. Hence, by AI-aided digitization on optic qubits, this REASSURED-compliant contraption may promise a next-generation POCT solution with unparalleled sensitivity, speed, and cost-effectiveness, which in whole confers a conclusive proof of the prowess of the burgeoning quantum metrics in biosensing.

Optically levitated nanodumbbells in vacuum are excellent candidates for thermodynamics, macroscopic quantum mechanics, precision measurements and quantum sensing. Silica (SiO 2 ) material, with extremely low absorption of near-infrared light and super mechanical strength, has been the most potential material for optically levitated systems. Here we synthesize high-purity solid SiO 2 nanodumbbells via Stöber method by introducing acetone for the induced aggregation of SiO 2 nanospheres. The nanodumbbells show high uniformity and their sizes are tunable. Previous experimental results demonstrated that the synthetic nanodumbbells can be applied in GHz nanomechanical rotors and can withstand the tensile strength of over 13 GPa. This work supports batch production and high yield of SiO 2 nanodumbbells, which engineers a new material platform to advance levitated optomechanics.

The NV center in diamonds has been widely employed in quantum sensing, quantum computing, and bioimaging due to its controllable ground-state spin, detectable magnetic resonance, excellent photostability, favorable biocompatibility, and chemical inertness. However, conventional excitation using 532 nm green light still exhibits certain limitations in practical applications. To address this, we propose a novel NV center excitation method based on the upconversion of near-infrared light to green emission. Through the synthesis of molybdenum-doped NaYF4: 20% Yb3+, 1.5% Er3+upconversion materials, efficient excitation of NV centers has been achieved. Both UC-LED luminescence spectroscopy and ODMR measurements confirm that the green light generated via the upconversion process exhibits sufficient intensity to effectively excite NV centers. Meanwhile, the characteristic sharp emission peaks of rare-earth upconversion materials eliminate the need for optical filters, facilitating device miniaturization, and a miniaturized UC-LED sensor has been developed.

Diamond nitrogen-vacancy (NV) center-based magnetometry provides a unique opportunity for quantum bio-sensing. However, NV centers are not sensitive to parameters such as temperature and pressure, and immune to many biochemical parameters such as pH and non-magnetic biomolecules. Here, we propose a scheme that can potentially enable the measurement of various biochemical parameters using diamond quantum sensing, by employing stimulus-responsive hydrogels as a spacing transducer in-between a nanodiamond (ND, with NV centers) and magnetic nanoparticles (MNPs). The volume phase transition of hydrogel upon stimulation leads to sharp variation in the separation distance between the MNPs and the ND. This in turn changes the magnetic field that the NV centers can detect sensitively. We construct a temperature sensor under this hybrid scheme and show the proof-of-the-principle demonstration of reversible temperature sensing. Applications in the detection of other bio-relevant parameters are envisioned if appropriate types of hydrogels can be engineered.

Nitrogen-vacancy (NV) centers in diamond demonstrate advantages in biosensing due to their exceptional photostability and long spin coherence time. However, clinical applications of NV centers are significantly limited because their spin states lack responsiveness to nonmagnetic biomolecules. This work presents a nanocatalytic-amplified quantum sensing platform targeting tyrosinase (TYR), a key biomarker for melanoma. The platform achieves ultrasensitive detection of nonmagnetic biomarkers through nanocatalytic signal transduction. Specifically, we develop plasmonic nanodiamond (FND@Au–Ag) functionalized hydrogel microneedles. The captured TYR triggers a cascade reaction that induces the nanocatalytic generation of hydroxyl radicals (•OH), thereby establishing an AND logic gate based on the dynamic response mechanism of spin relaxation time (T1). By integrating three-modal signals (fluorescence, colorimetry, and T1), the platform achieves a wide dynamic detection range of 0.01–200 U/mL and an ultralow detection limit (0.003 U/mL) for TYR. Ultimately, a genetic algorithm-optimized backpropagation neural network is developed to enhance the reliability of concentration prediction in complex biological matrices. This work proposes an approach to minimally invasive melanoma screening, overcoming the inherent limitations of traditional quantum sensing systems in detecting nonmagnetic biomarkers.

Real-time temperature monitoring inside living organisms provides a direct measure of their biological activities. However, it is challenging to reduce the size of biocompatible thermometers down to submicrometers, despite their potential applications for the thermal imaging of subtissue structures with single-cell resolution. Here, using quantum nanothermometers based on optically accessible electron spins in nanodiamonds, we demonstrate in vivo real-time temperature monitoring inside Caenorhabditis elegans worms. We developed a microscope system that integrates a quick-docking sample chamber, particle tracking, and an error correction filter for temperature monitoring of mobile nanodiamonds inside live adult worms with a precision of ±0.22°C. With this system, we determined temperature increases based on the worms’ thermogenic responses during the chemical stimuli of mitochondrial uncouplers. Our technique demonstrates the submicrometer localization of temperature information in living animals and direct identification of their pharmacological thermogenesis, which may allow for quantification of their biological activities based on temperature.

Accurate detection of microRNA concentrations is essential for early cancer diagnosis and monitoring. Existing methods typically provide a linear response within a limited concentration range, which hinders their precise clinical application. This study presents a novel sensing approach using the fluorescence properties of nitrogen vacancy (NV) centers. Magnetic particles capture and enrich fluorescent nanodiamonds (FNDs), and effectively address signal inconsistencies due to variations in FND particles by applying a voltage that regulates the NV center fluorescence. Our method achieves a linear response to microRNAs across a broad concentration range (10 fM to 1 μM), with the determination coefficient (R²) of the fluorescence signal, enhanced from 0.8711 to 0.9951 through voltage modulation. Additionally, optimizing experimental conditions and enhancing FND fluorescence increased the signal intensity, achieving a minimum detection limit of 7.17 fM. The combination of sensitive detection and an extensive linear range suggests the potential of this strategy as a universal detection method for the early diagnosis of various diseases.
