We exposed zebrafish ( Danio rerio ) to different concentrations of lead and cadmium, and monitored them for survival at 24, 48, 72, and 96 h. Metal toxicity was predicted and compared using the dose–response and general threshold survival models in terms of required data sets, fit performance, and applicability. It is necessary to study the mechanism of resistance to heavy metals in microbiological processes. In this study, Ochrobactrum MT180101 was used as the microbial source of an membrane bioreactor to investigate its degradation efficiency for electroplating wastewater and the copper-resistant mechanism. Meanwhile, excitation emission matrix-parallel factor, scanning electron microscope , atomic force microscope, fourier transform infrared spectroscopy , X-ray photoelectron spectroscopy and proteome analyses were applied to explain the comprehensive mechanism of the Ochrobactrum MT180101 resisting heavy metal toxicity. The results indicated that the Ochrobactrum MT180101 resisted heavy metal toxicity with the following pathways: i) binding metal cations on cell wall surfaces, ii) generating microbial products such as protein to chelate and stabilize the metal cations, iii) bio-transporting heavy metals from the intramembrane to the outer membrane by means of intracellular transport, and iv) reducing heavy metals through enzyme-mediated biotransformation . The results ensure that Ochrobactrum MT180101 was a copper-resistant bacterium that can be used in the pretreatment or deep treatment of electroplating wastewater.

Heavy metals released from municipal solid waste incinerators have become a major environmental concern. A comprehensive knowledge of metal vapor condensation in fly ash particles during incineration is essential for alleviating heavy metal toxicity, and for optimizing incineration process parameters and flue-gas cleaning systems. In this paper, the condensation of zinc vapor during flue-gas cooling in a 200 t/d fluidized bed incinerator and a 150 t/d moving grate incinerator was characterized and comparatively studied using high resolution synchrotron X-ray absorption spectroscopy (XAS). Principal component analysis, target transformation, and linear combination fitting were employed to identify zinc species directly from size fractionated fly ash particles. The chemical reaction behaviors of different zinc species were described by thermodynamic equilibrium simulations. Consistent with previous theoretical analysis and laboratory scale tests, the condensation behavior of zinc in an industrial incineration system is mainly affected by the sulfur/chlorine ratio and the inorganic particulates. It is found that zinc chloride is the major zinc species in a moving grate incinerator but willemite dominates in the fluidized bed incinerator. The high sulfur and silica/alumina particle concentration in the fluidized bed system changes the condensation propensity of vapors of Zn compounds. Adjusting the concentrations of SO 2 in flue-gas can inhibit the formation of zinc chlorides. Silica, alumina, aluminosilicates, and calcium-based compounds are potential sorbents for transforming zinc to less harmful species. To prevent toxic zinc species contained in fine particles from escaping into the atmosphere, wet scrubbers are more suitable for cleaning flue-gases in moving grate incineration systems, while improving the efficiency of dust removal is more important for fluidized bed incineration systems.

Key message Cytosolic G6PDH contributes to soybean tolerance to heavy metal exposure by alleviating oxidative damage, and in this process, nitric oxide may act upstream of cytosolic G6PDH. Heavy metal (HM) pollution in soil significantly impairs agricultural production and represents a substantial risk to food security. However, the underlying mechanism by which glucose-6-phosphate dehydrogenase (G6PDH or G6PD, EC 1.1.1.49) alleviates HM toxicity needs further clarification. In this study, the role of G6PDH in mitigating the toxicity of HMs (chromium, cadmium, copper, lead, and mercury) was examined in soybean ( Glycine max L.) using pharmacological and transgenic approaches. The findings indicate that the enhanced G6PDH activity observed during HM exposure was attributable to cytosolic G6PDH induction. HM stress induced the expression of cytosolic G6PDH genes ( GmG6PD6 , GmG6PD7 , and GmG6PD8 ), particularly GmG6PD7 . Overexpression of GmG6PD7 in soybean hairy roots enhanced G6PDH activity and HM tolerance. The addition of a G6PDH inhibitor during HM exposure markedly reduced the levels of NADPH, ascorbic acid (ASA), and reduced glutathione (GSH) in soybean roots, thereby exacerbating oxidative damage. In contrast, overexpression of GmG6PD7 significantly increased the contents of NADPH, ASA, and GSH in transgenic soybean roots under HM exposure, thereby alleviating oxidative damage. Furthermore, nitric oxide (NO) stimulated an elevation in cytosolic G6PDH activity and GmG6PD7 expression under HM exposure. Notably, G6PDH demonstrated comparable functionality in response to all five HMs examined. In summary, the NO-mediated induction of cytosolic G6PDH enhances soybean resistance to HM exposure through strengthening the antioxidant defense system.

The presence of dissolved organic matter (DOM) strongly influences the bioavailability of metals in aquatic environments; however, the association between the binding activities and the concentrations of DOM compositions is not well documented, leading to uncertainties in metal toxicity assessment. We creatively quantify the mitigation and acceleration effects of DOM compositions on cadmium (Cd) bioaccumulation and toxicity in zebrafish larvae using abiotic ligand (ABLs) and biotic ligand (BLs) in a toxicokinetic–toxicodynamic (TK–TD) model. The BL–TK–TD model could accurately predict the protective effect of fulvic acid while overestimating the complexing capacity of citric acid . The model also could successfully simulate the protective effects of native DOM in most cases from 32 natural water bodies in China. The observed LC50 values of Cd showed a peak effect for the native DOM fraction comprising hydrophilic acidic contents (3.55 ± 0.44 mg L − 1 ) in natural water from 32 sites. The BL–TK–TD model provides practically useful information to identify the effect of different DOM compositions on metal bioavailability and toxicity in aquatic environments and guides future water management policies aimed at controlling aquatic heavy metal pollution.

The discovery of cuproptosis provides a new perspective for copper overload treatment of cancer. However, the existing copper ion carriers only show oversimplified copper consignment and limited anti-tumor effect, which limits their further clinical application. Herein, we first design and synthesize two copper (II) complexes Py-Cu and TPE-Cu for synergistic cuproptosis, ferroptosis, and apoptosis anticancer therapy. In vitro , Py-Cu and TPE-Cu could induce dihydrolipoamide S -acetyltransferase (DLAT) aggregation, leading to cellular cuproptosis in PC-3 and CT-26 cells. Meanwhile, Py-Cu and TPE-Cu can amplify cellular oxidative stress by simultaneously boosting reactive oxygen species (ROS) production and depleting reduced glutathione (GSH), causing down-regulation of glutathione peroxides 4 (GPX4) expression and inducing ferroptosis. Moreover, Py-Cu and TPE-Cu could induce efficient apoptosis in vitro . To specifically release drugs at the tumor site, Py-Cu and TPE-Cu were co -assembled with hypoxia-responsive polymer PAL into nanoparticles, named NanoPy-Cu and NanoTPE-Cu, respectively. In vivo , NanoPy-Cu and NanoTPE-Cu showed superior tumor suppression effect both in the CT-26 tumor-bearing BALB/c mice model and ovarian (OV) cancer PDX-bearing BALB/c nude mice model. Here, we first reported two copper (II) complexes with unexpected broad anti-tumor activity, which synergized with cuproptosis, ferroptosis, and apoptosis to kill tumor cells.
