A Hydrogen Peroxide Sensor Based on Pt@Au Nanoparticles Loading to Polyethyleneimine Functionalized Carbon Nanotubes

Wei-Wei WANG,Yu QIU,Shao-Peng ZHANG,Jia-Wei LI,Xiao-Quan LU,Xiu-Hui LIU

A novel hydrogen peroxide sensor was fabricated by the seed-mediated growth method. Firstly, polyethyleneimine (PEI) functionalized multiwalled carbon nanotubes (MWNTs) were used as growth scaffold on a glass carbon electrode (GCE). Then, Au nanoparticles were uniformly electrodeposited as seeds. Finally, Pt nanoparticles (PtNPs) grew on Au PtNPs to form Pt@Au core-shell structure nanocomposite. A new type of electrochemical sensor based on Pt@Au/PEI-MWNTs nanocomposites for detection of hydrogen peroxide was then developed, and the designed Pt@Au/PEI-MWNTs/GCE was characterized by electrochemical methods and field emission scanning electron microscopy (FESEM). The differential pulse experiment results showed that the modified electrode exhibited excellent electrocatalytic activity towards the reduction of H 2 O 2 with the wide linear range from 9.2×10 −8 to 1.3×10 −3 M. The correlation coefficient was 0.9994 and the detection limit was down to 3.1×10 −8 M at the signal-to-noise ratio of 3.

Development of a Gold Nanoparticle-Linked Immunosorbent Assay of Staphylococcal Enterotoxin B Detection with Extremely High Sensitivity by Determination of Gold Atom Content Using Graphite Furnace Atomic Absorption Spectrometry

Chaojun Song,Yutao Liu,Jinwei Hu,Yupu Zhu,Zhengjun Ma,Jiayue Xi,Minxuan Cui,Leiqi Ren,Li Fan

Highly sensitive staphylococcal enterotoxin B (SEB) assay is of great importance for the prevention of toxic diseases caused by SEB. In this study, we present a gold nanoparticle (AuNP)-linked immunosorbent assay (ALISA) for detecting SEB in a sandwich format using a pair of SEB specific monoclonal antibodies (mAbs) performed in microplates. First, the detection mAb was labeled with AuNPs of different particle sizes (15, 40 and 60 nm). Then the sandwich immunosorbent assay for SEB detection was performed routinely in a microplate except for using AuNPs-labeled detection mAb. Next, the AuNPs adsorbed on the microplate were dissolved with aqua regia and the content of gold atoms was determined by graphite furnace atomic absorption spectrometry (GFAAS). Finally, a standard curve was drawn of the gold atomic content against the corresponding SEB concentration. The detection time of ALISA was about 2.5 h. AuNPs at 60 nm showed the highest sensitivity with an actual measured limit of detection (LOD) of 0.125 pg/mL and a dynamic range of 0.125–32 pg/mL. AuNPs at 40 nm had an actual measured LOD of 0.5 pg/mL and a dynamic range of 0.5 to 128 pg/mL. AuNPs at 15 nm had an actual measured LOD of 5 pg/mL, with a dynamic range of 5–1280 pg/mL. With detection mAb labeled with AuNPs at 60 nm, ALISA’s intra- and interassay coefficient variations (CV) at three concentrations (2, 8, and 20 pg/mL) were all lower than 12% and the average recovery level was ranged from 92.7% to 95.0%, indicating a high precision and accuracy of the ALISA method. Moreover, the ALISA method could be successfully applied to the detection of various food, environmental, and biological samples. Therefore, the successful establishment of the ALISA method for SEB detection might provide a powerful tool for food hygiene supervision, environmental management, and anti-terrorism procedures and this method might achieve detection and high-throughput analysis automatically in the near future, even though GFAAS testing remains costly at present. Keywords:gold nanoparticles;gold nanoparticle-linked immunosorbent assay;staphylococcal enterotoxin B

One-pot synthesis of highly dispersed PtAu nanoparticles–CTAB–graphene nanocomposites for nonenzyme hydrogen peroxide sensor

Panpan Liu,Jiawei Li,Xiuhui Liu,Ming Li,Xiaoquan Lu

A simple and efficient approach was developed to fabricate an amperometric sensor for hydrogen peroxide (H 2 O 2 ) determination based on one-pot synthesis of highly dispersed PtAu nanoparticles–CTAB–graphene nanocomposites. In this study, graphene oxide was first functionalized with a cationic surfactant, cetyl trimethyl ammonium bromide (CTAB); then PtAu nanoparticles (NPs) were deposited on graphene (GR) via a simple chemical co-reduction method. A large amounts of highly dispersed bimetallic PtAu nanoparticles could be well loaded on the surface of the graphene, as revealed by transmission electron microscopy (TEM). In addition, PtAuNPs–CTAB–GR nanocomposites were also characterized by electrochemical methods including cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). The experimental results demonstrated that constructed sensor exhibited good catalytic activity toward H 2 O 2 , and obtained a wide linear range from 5.0   ×   10 −9 to 4.8   ×   10 −6   mol/L with a limit of detection (LOD) of 1.7   ×   10 −9   mol/L (S/N   =   3). Moreover, it could also be applied to real samples analysis. The excellent performance of this H 2 O 2 sensor could be ascribed to graphene being used as effective load matrix for the deposition of PtAuNPs and synergistic effect of bimetallic PtAuNPs and graphene nanomaterials.

SERS detection of urea and ammonium sulfate adulterants in milk with coffee ring effect

Abid Hussain,Da-Wen Sun,Hongbin Pu

In the current work, surface-enhanced Raman scattering (SERS) based on gold nanoparticles (AuNPs) and silver-coated gold nanoparticles (Au@AgNPs) with coffee ring effect was employed to simultaneously ascertain urea and ammonium sulfate (AmS) in milk. A small drop (2 µL) of milk with adulterants was dried on a gold-coated slide to examine concentrations ranging from 5, 10, 20, 40 and 80 mg/dL based on spectra ranging from 400 to 1500 cm.−1 A uniform distribution of analytes, with enhanced Raman signals was detected in a small region (maximum 1.9 mm) of coffee ring across the centre of coffee ring pattern. Nanoparticles with core (Au) diameter of 26 nm and shell thickness (Ag) of 6.5 nm were confirmed using transmission electron microscopy (TEM) images. A strong Raman peak at 980 cm−1 was assigned to AmS, while that at 1001 cm−1 was ascribed to urea. With AuNPs, coefficients of determination (R2) of 0.9873 and 0.9859 were achieved for urea and AmS, respectively, while for Au@AgNPs values of 0.9827 and 0.9855 were obtained for urea and AmS, respectively. This study revealed that SERS based on coffee ring effect has the potential to be further exploited for detecting other banned and hazardous adulterants in milk and milk products.

Detection of ADAMTS-4 Activity Using a Fluorogenic Peptide-Conjugated Au Nanoparticle Probe in Human Knee Synovial Fluid

Shi Peng,Qiang Zheng,Xin Zhang,Linghui Dai,Jingxian Zhu,Yanbin Pi,Xiaoqing Hu,Wenqing Cheng,Chunyan Zhou,Yinlin Sha,Yingfang Ao

A disintegrin and metalloproteinase with thrombospondin motif-4 (ADAMTS-4) plays a pivotal role in degrading aggrecan, which is an early event in cartilage degrading joint diseases such as osteoarthritis (OA). Detection of ADAMTS-4 activity could provide useful clinical information for early diagnosis of such diseases and disease-modifying therapy. Therefore, we developed a ADAMTS-4 detective fluorescent turn-on AuNP probe (ADAMTS-4-D-Au probe) by conjugating gold nanoparticles with a FITC-modified ADAMTS-4-specific peptide (DVQEFRGVTAVIR). When the ADAMTS-4-D-Au probe was incubated with ADAMTS-4, the fluorescence recovered and fluorescence intensity markedly increased in proportion to concentrations of ADAMTS-4 and the probe. A nearly 3-fold increase in fluorescent intensity in response to only 3.9 pM of ADAMTS-4 was detected, whereas almost no fluorescence recovery was observed when the probe was incubated with matrix metalloproteinase (MMP)-1, -3, and -13. These results indicate a relative high sensitivity and specificity of the probe. Moreover, ADAMTS-4-D-Au probe was used to detect ADAMTS-4 activity in synovial fluid from 11 knee surgery patients. A substantial increase in fluorescent intensity was observed in the acute joint injury group as compared to the chronic joint injury and end-stage OA groups, indicating that this simple and low-cost sensing system might serve as a new detection method for ADAMTS-4 activity in biological samples and in screens for inhibitors for ADAMTS-4-related joint diseases. Additionally, this probe could be a potential biomarker for early diagnosis of cartilage-degrading joint diseases.

Colloids containing gadolinium-capped gold nanoparticles as high relaxivity dual-modality contrast agents for CT and MRI

ChengCai Zeng,XiaoNi Shi,Bo Wu,Dong Zhang,WeiLu Zhang

A type of novel macromolecular colloid was prepared from gadolinium-based poly( l -succinimide) and gold nanoparticles (GNPs) with Au S covalent bonds. The colloid displayed improved stability in aqueous media and regular arrays in partial region. Moreover, these materials enhanced the contrast 9 times more than that of the corresponding uncoated Au compound when the Au mass content was only 0.15%. Therefore, these materials might have potential as dual-modality contrast agents for magnetic resonance imaging (MRI) and computed tomography (CT).

Sub-10-nm multicolored gold nanoparticles for colorimetric determination of antibiotics via formation of interlocking rings

Leng Yumin,Fu Yu,Lu Zhiwen,Sang Zhipei,Liu Kecheng,Du Chenxi,Ma Linfeng

A general approach is presented for synthesis of multicolored gold nanoparticles (GNPs) by Au(I)-mediated generation of interlocking rings in proteins and antibiotics. The Au(I) ions are shuttled from proteins to antibiotics, and this causes the formation of interlocking rings. The multicolored GNPs of different sizes were synthesized in the rings by using the rapid nucleation method. To take the unique colors of GNPs, a functional array was designed for the colorimetric determination and discrimination of antibiotics, specifically of amoxicillin, chlortetracycline, erythromycin, spiramycin, neomycin, thiamphenicol, gentamycin and lincomycin. The method is based on the “three color” (RGB) principle. The color response patterns are characteristic for each antibiotic and can be quantitatively differentiated by statistical techniques. The limits of detection (LOD, at S/ N  = 3) for spiramycin (Sp) have been calculated to be 0.18 μM and 0.10 μM in water and milk, respectively. The good linear range (from 0.3 to 3.5 μM) has been used for the quantitative assay of Sp in a certified reference material. Schematic presentation of gold nanoparticles (GNPs) synthesis via formation of interlocking rings in protein and antibiotics. The Au(I) ions mediate protein and antibiotics to be interlocking rings, which are quickly fixed via microwave reaction. The GNPs are synthesized and assembled in the rings.

(Pd, Au, Ag) nanoparticles decorated well-ordered macroporous carbon for electrochemical sensing applications

Shuang Dong,Zhou Yang,Baoliang Liu,Jintao Zhang,Peng Xu,Meng Xiang,Tong Lu

The research and develpoment of biomass waste derived carbon materials have attracted tremendous research interests in fabrication of high performance electrochemical sensors. In this work, noble metal NPs (i.e., Pd, Au and Ag) loading on Loofah sponge-derived well-ordered macroporous carbon (OMC)/ N -doped carbon thin layer (N-C) was designed by a general approach. This polydopamine (PDA)-based method has the advantages of simplicity, high efficiency and versatility. In virtue of the synergistic contribution from 3D macroporous carbon scaffold and highly active nobel metal NPs, these as-prepared Pd@OMC/N-C, Au@OMC/N-C and Ag@OMC/N-C electrode exhibit excellent electrochemical sensing performances for H 2 O 2 reduction, glucose oxidation and nitrite oxidation, respectively. Thus, 3D OMC/N-C loaded noble metal nanoparticles are promising materials for a wide spectrum of electrochemical sensor and biosensor applications.

A simple, fast, and sensitive colorimetric assay for visual detection of berberine in human plasma by NaHSO4-optimized gold nanoparticles

Ziwei Hu,Mingshan Xie,Danting Yang,Dong Chen,Jingyi Jian,Haibin Li,Kaisong Yuan,Zhengjin Jiang,Haibo Zhou

Berberine is an important anti-inflammatory drug widely used for the treatment of heart and intestinal disorders. The quantification of berberine is usually required in clinic medical assays. Herein, we report simple, fast and sensitive colorimetric visualization of berberine in human plasma using citrate-stabilized gold nanoparticles (AuNPs). Upon exposure to μg mL−1 levels of berberine, the AuNP solution exhibits a highly sensitive color change from red to blue and rapid aggregation kinetics within the initial 10 min, which can be directly seen with the naked eye and monitored by UV-vis absorbance spectroscopy. As confirmed by comparison with potential coexisting foreign substances, the presence of positively charged berberine induces the aggregation of the AuNPs owing to the electrostatic attraction and coordination interactions between the ring nitrogen atoms of berberine and the AuNPs, causing a color change of the AuNP suspension. Further evidence is that the sensitivity is significantly improved when NaHSO4 is added to promote the ligand exchange between citrate and berberine at the surface of the AuNPs. The NaHSO4-optimized AuNP system provides a simple colorimetric assay for the rapid detection of berberine in human plasma. The limit of detection (LOD) is 0.24 μg mL−1 by the naked eye and 0.06 μg mL−1 by UV-vis spectroscopy.

A novel sensing platform for sensitive cholesterol detection by using positively charged gold nanoparticles

Xiangmin Miao,Zhiyuan Cheng,Zongbing Li,Po Wang

This work described a label-free platform for sensitive cholesterol detection upon the precipitation of cetyltrimethyl ammonium bromide (CTAB) stabilized gold nanoparticles (AuNPs). Modifying with CTAB, the stability of AuNPs was greatly improved compared to conventional used AuNPs, and the surface of them was positively charged, which could effectively adsorb with negatively charged DNA detection probe (DP) based on electrostatic adsorption, and inducing the precipitation of them. Tremendous H 2 O 2 would produce based on the reaction of cholesterol with oxygen by using cholesterol oxidase (ChO x ) as the catalyzer, and followed the happen of Fenton reaction in the presence of Fe 2+ to generate OH , inducing the cleavage of DP into DNA fragments. As a result, the adsorption ability of AuNPs decreased, accompanying with a decrease of AuNPs precipitation. Then, based on detecting the absorption spectra change of AuNPs in supernatant along with the increase of cholesterol concentration, sensitive and simple visual-based cholesterol detection was realized with a detection limit of 335   pM. Such method showed high selectivity for the detection of cholesterol. Moreover, the high stability of such AuNPs in solution that contained salt, protein or other metal ions, making our sensing method have a potential application in bioanalysis.

Visual detection of Fusarium temperatum by using CRISPR-Cas12a empowered LAMP assay coupled with AuNPs-based colorimetric reaction

Yingchun Li,Yaqin Zhang,Fange Kong,Chunxia Wang,Shanshan Chen,Jiasi Wang,Di Wang

Fusarium temperatum ( F. temperatum ) causes maize stalk rot disease, reducing grain yield and producing multiple harmful mycotoxins that threaten food safety and quality. However, current detection methods are limited by the requirement of well-trained personnel, complicated operations and expensive instruments. To address this challenge, gold nanoparticles (AuNPs)-based colorimetric assay using loop-mediated isothermal amplification (LAMP) coupled with CRISPR-Cas12a (Au-CRISPR) was tested for F. temperatum detection. The assay utilizes DNA-modified AuNPs as a colorimetric probe and the results can be readily observed by the naked eye. After optimization, the method can specifically detect as low as 100 copies/μL of target DNA or 10 −8  ng/μL of extracted DNA from F. temperatum . The detection platform allows high-throughput testing by integrating microtiter plates and microplate reader. Furthermore, a portable smartphone-based device was developed to realize point of care (POC) detection of F. temperatum in resource-limited settings. This simple, inexpensive and sensitive detection platform has great potential for the applications in field detection.

Hydration-responsive keratin fibers with ultra-high shape recovery via reversible protein conformational change and Au enhancement

Xiaoyun Xu,Zhuang Wang,Qi Zhang,Ke Zhang,Min Li,Jieqiong Yang,Yi Zhao,Qinfei Ke,Qingbao Guan,Jinlian Hu

Fabricating shape-memory materials from natural macromolecules has gained popularity to facilitate the development of bio-friendly smart materials and devices. However, current biobased shape-memory materials have shortcomings, including insufficient mechanical properties and low shape recovery rate in their real applications. Herein, this work aims to develop hierarchically structured keratin-based fibers with high strength and shape-memory properties in response to hydration. The gold nanoparticle is adopted for enhancement in the keratin system, and a well-aligned anisotropic structure with α-helix to the fiber axis is established in the regenerated keratin fibers by wet spinning. The strong crosslinking effect from Au S bonding endows the keratin fibers with outstanding shape-memory performance, exhibiting a shape fixity of ~92 % and a near-full shape recovery rate of ~96 %. In this system, the transition between α and β conformation under external force and water stimulation is assigned as the fundamental mechanism in response to hydration. In hydration and dehydration, the hydrogen bonds act as ‘switches’ that enable deformation, while the strong Au S bonds are ‘net points’ to maintain the original and temporary shapes. This strategy can promote metal nanomaterial anchoring as a methodology for fabricating shape-memory protein-based fibers with excellent mechanical properties and shape-memory performance.
