Effects of phenolic acid grafted-chitosan hydrocolloids on the aldehyde contents from lipid oxidation in golden pompano (Trachinotus blochii) fillets during pan-frying

Mantong Zhao,Ying Sun,Yikai Huang,Wei Yang,Haohao Shi,Jiamei Wang,Zhongyuan Liu,Xueying Zhang,Chuan Li,Guanghua Xia,Haohao Wu,Xuanri Shen,Dayong Zhou

The effects of phenolic acid grafted-chitosan hydrocolloids (CS-g-GA/FA) on aldehyde contents from lipid oxidation in golden pompano fillets during pan-frying was investigated with an established high-performance liquid chromatography-mass spectrum method. Results indicated that pan-frying induced profound lipid oxidation and aldehydes generation with propanal, hexanal, nonanal, trans , trans -2,4-decadienal, and 4-hydroxy-2-nonenal as the abundant species. CS-g-FA and CS-g-GA effectively decreased their contents by 23.74–27.42 %, 61.69–67.42 %, 41.83–53.91 %, 29.91–48.79 %, and 61.57–65.39 % after 3 min. Most aldehyde contents decreased with the extension of pan-frying time due to the volatilization and reaction. In terms of substrate depletion, CS-g-phenolic acids effectively inhibited unsaturated fatty acids oxidation due to their decent antioxidant activity than CS. The significant lower retention rates of aldehydes in the CS-g-phenolic acids groups compared with control in chemical mode confirmed the carbonyl ammonia condensation. These results suggested that CS-g-phenolic acids serve as novel coating to reduce hazardous compounds during aquatic products thermal processing.

Impacts of zein-fucoidan nanoparticles with and without curcumin on gel properties of golden threadfin bream (Nemipterus virgatus) surimi

Shaoxuan Yu,Geyu Pang,Shiyang Li,Shengjuan Lv,Zhengpeng Wei,Jinmei Wang,Haifang Xiao,Lanlan Zhu

Zein-fucoidan nanoparticles (ZFNPs) as an excellent colloidal delivery system for bioactive compounds, such as curcumin (CUR), show a great application potential in the food industry. Herein, various amount of ZFNPs, ZFNPs loaded with CUR (CZFNPs) and free CUR were added into golden threadfin bream ( Nemipterus virgatus ) surimi to explore their impacts on the gel properties. Dynamic rheological tests showed that storage modulus (G') and loss modulus (G") of surimi gels with the additive amount of ZFNPs and CZFNPs under 0.8 % significantly increased and their loss tangent (tan δ) slightly reduced during thermal gelation, indicating stronger networks and better structural configuration. Puncture test and textural profile analysis confirmed the improvements in breaking force, hardness, chewiness and gumminess of surimi gels. Furthermore, 0.8 % of ZFNPs and CZFNPs induced conformational changes of surimi proteins from α-helix to β-sheet and random coil, and then facilitating the cross-link between proteins. Low-field nuclear magnetic resonance revealed that ZFNPs and CZFNPs promoted the redistribution of water in surimi gel networks, converting more free water to tightly bounded water, and thus increasing the water holding capacity. However, the addition of equivalent amount of CUR caused detrimental to surimi gel properties. In conclusion, ZFNPs and CZFNPs conferred better gel properties and nutritional characteristics to surimi gels, and this study provided a theoretical basis for the development of high-quality surimi products.

Insulin-like growth factor binding protein 5b of Trachinotus ovatus and its heparin-binding motif play a critical role in host antibacterial immune responses via NF-κB pathway

Du Hehe,Zhou Yongcan,Du Xiangyu,Zhang Panpan,Cao Zhenjie,Sun Yun

Introduction: Insulin-like growth factor binding protein 5 (IGFBP5) exerts an essential biological role in many processes, including apoptosis, cellular differentiation, growth, and immune responses. However, compared to mammalians, our knowledge of IGFBP5 in teleosts remains limited. Methods: In this study, TroIGFBP5b, an IGFBP5 homologue from golden pompano (Trachinotus ovatus) was identified. Quantitative real-time PCR (qRT-PCR) was used to check its mRNA expression level in healthy condition and after stimulation. In vivo overexpression and RNAi knockdown method were performed to evaluate the antibacterial profile. We constructed a mutant in which HBM was deleted to better understand the mechanism of its role in antibacterial immunity. Subcellular localization and nuclear translocation were verified by immunoblotting. Further, proliferation of head kidney lymphocytes (HKLs) and phagocytic activity of head kidney macrophages (HKMs) were detected through CCK-8 assay and flow cytometry. Immunofluorescence microscopy assay (IFA) and dual luciferase reporter (DLR) assay were used to evaluate the activity in nuclear factor-κB (NF-κβ) pathway. Results: The TroIGFBP5b mRNA expression level was upregulated after bacterial stimulation. In vivo, TroIGFBP5b overexpression significantly improved the antibacterial immunity of fish. In contrast, TroIGFBP5b knockdown significantly decreased this ability. Subcellular localization results showed that TroIGFBP5b and TroIGFBP5b-δHBM were both present in the cytoplasm of GPS cells. After stimulation, TroIGFBP5b-δHBM lost the ability to transfer from the cytoplasm to the nucleus. In addition, rTroIGFBP5b promoted the proliferation of HKLs and phagocytosis of HKMs, whereas rTroIGFBP5b-δHBM, suppressed these facilitation effects. Moreover, the in vivo antibacterial ability of TroIGFBP5b was suppressed and the effects of promoting expression of proinflammatory cytokines in immune tissues were nearly lost after HBM deletion. Furthermore, TroIGFBP5b induced NF-κβ promoter activity and promoted nuclear translocation of p65, while these effects were inhibited when the HBM was deleted.Discussion: Taken together, our results suggest that TroIGFBP5b plays an important role in golden pompano antibacterial immunity and activation of the NF-κβ signalling pathway, providing the first evidence that the HBM of TroIGFBP5b plays a critical role in these processes in teleosts.

Metabolomic profiles and potential biomarkers identification among targeted muscles of fresh hybrid grouper (Epinephelus lanceolatus × Epinephelus fuscoguttatus) and golden pompano (Trachinotus ovatus)

Ahtisham Ali,Jinfang Wang,Wei Jiang,Shuai Wei,Qinxiu Sun,Qiuyu Xia,Zefu Wang,Zongyuan Han,Yang Liu,Shucheng Liu

Differential metabolites and potential biomarkers of hybrid grouper and golden pompano muscles were identified in this study. Principal component analysis (PCA) and partial least squares discriminant analysis (PLS-DA) were performed to evaluate liquid chromatography-mass spectrometry (LC-MS) metabolomic analysis. Among the metabolites, 1678 co-contained differential metabolites in positive modes and 1445 co-contained differential metabolites in negative modes were investigated. Sixteen most differential metabolites were determined as potential biomarkers in different muscle groups. The top 10 KEGG-enriched pathways were identified. The sensory characteristics of groups were highest in aroma and sulfur-organic compounds in both fishes. Most organic acid contents in hybrid grouper were significantly higher than those of golden pompano. This present study provides a baseline for a better understanding of potential metabolites involved in muscle quality and taste as well as could provide a piece of further knowledge to identify the significant role of key biomarkers in fish muscles.

Effects of ferulic acid grafted-chitosan hydrocolloid on the quality preservation of golden pompano (Trachinotus blochii) fillets during refrigerated storage with emphasis on lable-free proteomics

Mantong Zhao,Zhongyuan Liu,Ying Sun,Wei Yang,Haohao Shi,Xueying Zhang,Jiamei Wang,Guanghua Xia,Haohao Wu

The effect of ferulic acid grafted-chitosan hydrocolloid (CS-g-FA) on the quality preservation of golden pompano fillets during refrigerated storage was investigated with emphasis on label-free proteomics analysis. It was indicated that CS-g-FA significantly inhibited spoilage bacteria growth, decreased total volatile basic nitrogen (TVB-N) content, retarded lipid and protein oxidation, and delayed water migration in fillets, thus maintaining the texture properties of fillets. In detail, CS-g-FA treatment caused the least total viable counts of 6.55 logCFU/g, TVB-N content of 21.56 mg N/100g, thiobarbituric acid reactive substances value of 1.50 mg MDA/kg, and carbonyl value of 1.26 μmol/g protein, respectively, after 15 day. Furthermore, 27 shared differentially expressed proteins (DEPs) closely related with freshness were identified compared to the fresh sample. CS-g-FA inhibited structural proteins degradation, while reduced the expression of metabolic enzymes to maintain fillets quality and extend the shelf-life. Several DEPs could be identified as potential proteomics markers for golden pompano freshness assessment. Thus, CS-g-FA serve as promising coating for quality preservation of aquatic products.
