A novel and sensitive fluorescence analysis platform was constructed for the detection of chlorogenic acid (CGA) using carbon dots (C-dots) with prominent sensitivity and selectivity. Excitation-dependent emission fluorescence C-dots were fabricated using citric acid and l -histidine as precursors through an efficient one-step hydrothermal treatment. The maximum excitation and emission wavelength of the as-synthesized C-dots were 340 nm and 414 nm, respectively. Moreover, the as-prepared C-dots displayed excellent water solubility and good photostability. The fluorescence quantum yield of the as-prepared C-dots was measured to be about 22% using quinine sulfate as the reference. Furthermore, the obtained C-dots were applied to the detection of CGA accompanied with a wide linear range from 1.53 μmol L −1 to 80.0 μmol L −1 as well as a limit detection of 0.46 μmol L −1 . More importantly, the proposed fluorescence method was successfully used to analyse CGA in coffee and honeysuckle.

Using honeysuckle as raw material, chlorogenic acid (CGA) was extracted with different alcohols. Based on the single-factor experiment design, the relationship between each parameter and the response value was explored by Box–Behnken method to optimize the process conditions. Best extraction results were obtained under the conditions of solid-to-liquid ratio of 1:20, the ultrasonic time of 40 min, the ultrasonic vibrator power of 240 w, and the CGA extraction rate of 2.98%. The experimental data show that the extraction rate of CGA is related to the length of the alcohol carbon chain and the number of hydroxyl groups in the extractant. The results from this work can provide technical basis for the safe and efficient production of CGA from honeysuckle.

To investigate the dietary exposure risk of flonicamid application on Lonicerae Japonicae Flos and the effect of flonicamid on constituents of Lonicerae Japonicae Flos, field experiments were conducted in Fengqiu, Henan province, and flonicamid residue in samples collected was detected by gas chromatography equipped with electron capture detector (GC-ECD). And chlorogenic acid and luteoloside were determined by HPLC. Dietary exposure risk assessment was conducted through comparing the estimated daily intake (EDI) which was calculated by using the consumed residual level along with the acceptable daily intake (ADI). The effect of flonicamid on chlorogenic acid and luteoloside were obtained by ANOVA statistical analysis and least significant difference (LSD)-t test. The results showed that the terminal-residue contents of flonicamid were under 1.6 mg kg−1. And risk quotient ranged from 0.0011 to 0.0028, indicating the long-term exposure to flonicamid residual through consumption of Lonicerae Japonicae Flos in consumers was relatively low. Flonicamid could suppress the generation of luteoloside, so it was not advised to be used in L. japonica flowering phase. The study aims at providing the useful suggestion on the reasonable flonicamid usage and the reference for the establishment of maximum residue limits (MRLs) of flonicamid in Lonicerae Japonicae Flos.

In order to fulfill people's requirements for food quality and safety, it is a promising strategy to develop intelligent biodegradable food packaging materials. Herein, honeysuckle extracts/attapulgite/chitosan composite films containing natural carbon dots were fabricated for intelligent food packaging. Different characterization techniques were employed to study the obtained composite films, while the physicochemical properties, optical properties, antibacterial and antioxidant activities of composite films were determined. The obtained composite films presented good mechanical, antibacterial and antioxidant properties, and the antibacterial ratios of composite films against Escherichia coli and Staphylococcus aureus were 99.27 ± 0.18 % and 98.85 ± 0.65 %, respectively. When the added amount of honeysuckle extracts/attapulgite nanocomposites was 4.76 %, the tensile strength and elongation at break of composite films reached 24.9 ± 2.35 MPa and 64.8 ± 2.11 %, respectively, which were obviously higher than that of pure chitosan film. Furthermore, the composite films exhibited excellent UV shielding and blue fluorescence properties, as well as pH-sensitivity due to the presence of caffeoquinic acid-based natural carbon dots derived from honeysuckle extract. Therefore, the composite films indicated a potential application for intelligent food packaging.

Flos Lonicerae was an important Chinese medicine. In this research, a microwave assisted extraction method was applied for the extraction of chlorogenic acid from Flos Lonicerae . The operating conditions were optimized using a Box–Behnken design test. Under the optimal conditions, the extraction yield of chlorogenic acid reached 32.52 ± 1.31 mg/g. Next, a direct analysis in real time mass spectrometry (DART-MS) method was utilized to quantify of chlorogenic acid in Flos Lonicerae extracts. The primary parameters were optimized to obtain maximum signal intensity. In the detection process of the actual samples, the results obtained by DART–MS are consistent with those obtained by HPLC method with short detection time and acceptable repeatability and precision (<15%). In addition, the DART–MS/MS method has several advantages, such as speed, low cost and simplicity. Therefore, the DART-MS method is an efficient method that can be applied in the quantification of chlorogenic acid in Flos Lonicerae.

This study aims to observe the protective action of Flos Lonicerae (FL) aqueous extract against acetaminophen (AP)-induced liver injury and its mechanism. Results show that FL decreases AP-increased serum alanine/aspartate transaminases (ALT/AST) activity, as well as total bilirubin (TB) amount, in mice. Histological evaluation of the liver further confirms the protection of FL against AP-induced hepatotoxicity. TdT-mediated biotin-dUTP nick-end labeling (TUNEL) assay shows that FL reduces AP-increased apoptotic cells. Furthermore, AP-decreased liver glutamate-cysteine ligase (GCL) enzymatic activity and glutathione (GSH) amount are both reversed by FL because of the increased expression of the catalytic subunit of GCL (GCLC) protein. The amount of chlorogenic acid (CGA), caffeic acid, and luteolin, the main active compounds in FL, is detected by high-performance liquid chromatography (HPLC). In addition, cell viability assay demonstrates that polyphenols in FL, such as CGA, caffeic acid, as well as isochlorogenic acids A, B, and C, can reverse AP-induced cytotoxicity. In conclusion, FL can prevent AP-induced liver injury by inhibiting apoptosis. The cellular antioxidant enzyme GCL is also involved in such protection. Polyphenols may be the main active hepato-protective ingredients in FL.

The intestinal absorption of active herbal constituents plays an important role in the biomedical efficacy of Traditional Chinese Medicine (TCM) formulations after oral administration. TCM compounds with low oral bioavailability can reach the distal intestine and then interact with intestinal flora, influencing the botanical pharmacological effects. In this study, in vitro digestion and an ex vivo Ussing chamber model were utilized to evaluate the intestinal absorption behavior of Forsythiae Fructus - Lonicerae Japonicae Flos -containing Yinqiao Jiedu Granule (YQJDG). It was found that the jejunum exhibited active absorption effects for some components of the formula, while the oral bioavailability of other herbal ingredients was low. Through further research using a combined UPLC-MS/MS and 16S rDNA sequencing technique, we studied the existence of the reciprocal interactions between YQJDG and gut microbiome. The in vitro fecal fermentation results showed that YQJDG significantly impacted the microbial community composition. The YQJDG markedly increased the abundance of beneficial microorganisms, such as Parabacteroides distasonis and Streptococcus gallolyticus subsp. Macedonicus, and suppressed the abundance of conditional pathogens including Prevotella steorerea, Haemophilus parainfluenzae , and Bacteroides . These effects may potentially contribute to the body’s immune functions and anti-inflammatory capacities. UPLC-MS/MS analysis suggested that the fecal microbiota chemically transformed constituents with low bioavailability to more readily absorbed potentially active metabolites. These findings provided valuable insights into the absorption characteristics of YQJDG and its interaction with the gut microbiome, further facilitating our understanding of precise pharmacological mechanisms of action of this Chinese herbal formulation.

Introduction: The quality of traditional Chinese medicine is based on the content of their secondary metabolites, which vary with habitat adaptation and ecological factors. This study focuses on Lonicera japonica Flos (LJF), a key traditional herbal medicine, and aims to evaluate how ecological factors impact its quality. Methods: We developed a new evaluation method combining high-performance liquid chromatography (HPLC) fingerprinting technology and MaxEnt models to assess the effects of ecological factors on LJF quality. The MaxEnt model was used to predict suitable habitats for current and future scenarios, while HPLC was employed to analyze the contents of key compounds. We also used ArcGIS for spatial analysis to create a quality zoning map. Results: The analysis identified 21 common chromatographic peaks, with significant variations in the contents of Hyperoside, Rutin, Chlorogenic acid, Cynaroside, and Isochlorogenic acid A across different habitats. Key environmental variables influencing LJF distribution were identified, including temperature, precipitation, and elevation. The current suitable habitats primarily include regions south of the Yangtze River. Under future climate scenarios, suitable areas are expected to shift, with notable expansions in southern Gansu, southeastern Tibet, and southern Liaoning. The spatial distribution maps revealed that high-quality LJF is predominantly found in central and southern Hebei, northern Henan, central Shandong, central Sichuan, southern Guangdong, and Taiwan.Discussion: The study indicates that suitable growth areas can promote the accumulation of certain secondary metabolites in plants, as the accumulation of these metabolites varies. The results underscore the necessity of optimizing quality based on cultivation practices. The integration of HPLC fingerprinting technology and the MaxEnt model provides valuable insights for the conservation and cultivation of herbal resources, offering a new perspective on evaluating the impact of ecological factors on the quality of traditional Chinese medicines.

Thiamethoxam is one of the top three neonicotinoids found in the environment and in food. The widespread use of thiamethoxam in medicinal and edible herbs threatens their safe use. Therefore, it is particularly important to develop a rapid and simple approach to detect thiamethoxam residues in herbal medicines. In this study, colloidal carbon-based immunochromatographic strip (CNP-ICS) and colloidal gold-based immunochromatographic test strip (GNP-ICS) methods were respectively developed for the determination of thiamethoxam . The cutoff value of CNP-ICS was 0.1 ng mL −1 , which is six times more sensitive than that of the GNP-ICS method (0.6 ng mL −1 ). The CNP-ICS method was applied to detect thiamethoxam in medicinal and edible Lonicerae Japonicae Flos (LJF) for the first time, with a limit of detection of 10 ng g −1 . The detection results of thiamethoxam in 26 batches of LJF samples by the CNP-ICS were consistent with those of the LC–MS/MS method. High residue levels (10.16–530.40 ng g −1 ) and high detection rates (69.23%) of thiamethoxam in LJF were observed. The CNP-ICS prepared in this study has the merits of low cost, simple preparation, non-toxicity, and high sensitivity, which can be utilized for rapid field screening of thiamethoxam residues in complex matrices such as herbal medicines. Globally, honeysuckle is consumed as a food and administered as a medicinal agent. However, pesticide residues in honeysuckle limit its application and development of the honeysuckle industry, affecting food safety and endangering human health. Here, the degradation kinetics of 11 typical pesticides, including insecticides, fungicides , and an acaricide , in honeysuckle were investigated. The half-lives of pesticides in Henan and Liaoning fields were 1.90–4.33 and 2.05–4.62 d, respectively. The processing factors (PFs) of these pesticides after oven, sun, and shade drying ranged from 3.52 to 11.2. After decocting, the PFs of the pesticides were <1. Twenty degradation products were identified using ultra high performance liquid chromatography-quadrupole time-of-flight mass spectrometry , and pathways were proposed based on drying and decoction. The ecotoxicities of the degradation products were evaluated using the Toxicity Estimation Software Tool . Finally, the acute hazard indices of these pesticides, as determined via dietary exposure assessment combined with the PFs, were 0.227 and 0.911 for adults and children, respectively. Thus, special populations, such as children, require particularly careful risk control in terms of dietary exposure.

Chemotherapy is considered a most effective way to treat cancer. However, it is very common that chemotherapy causes unbearable mental and physical side effects to cancer patients, which ultimately reduces the patients’ confidence of overcoming diseases and compromises the treatment of chemotherapy. Cisplatin (DDP), a widely used anticancer agent for various types of cancers, also damages nontumor cells and tissues, which are mostly related to the activation of the inflammation pathway. Previously, we have discovered a few rational formulas of food as medicine materials that reduced systemic inflammation in in vitro and in vivo models. Hence, this study reports the ability of an optimized traditional Chinese anti-inflammatory formulation capable of synergizing the antitumor effect of DDP in vitro and in vivo. More significantly, by formulation of two anti-inflammatory herbal medicine, the Chrysanthemum × morifolium (Ramat.) Hemsl [Asteraceae] and Lonicera japonica Thunb [Caprifoliaceae] with a mediator Glycyrrhiza uralensis Fisch. ex DC [Fabaceae], a best formula relieved the kidney damage imposed by DDP. Treatments of various combinations of major chemical components of the three herbs also exhibited a similar trend for lowering the DDP-induced nephrotoxicity; however, contrary to that of the formula of herbal extracts, all chemical formulas could not recover the body weight and food intake of the tumor-bearing mice treated by DDP. Our findings suggested that the therapeutic index of DDP-based chemotherapy was able to be improved by minimizing toxicities from the two-herb formula to inhibit the inflammation in mouse tumor models and DDP-induced acute kidney injury mouse models.

Lonicera japonica Thunb. leaves were used to biosynthesize silver nanoparticles (AgNPs), while its antibacterial and antifungal properties were investigated. Reaction conditions of AgNPs which characterized were optimized. The maximum absorption wavelength of the AgNPs was located about 400 nm, with a spherical shape and a particle size of 14.87 ± 4.07 nm. Produced AgNPs showed effective antibacterial activities against tested bacterial strains. Minimal inhibitory concentrations and minimum bactericidal concentrations of the AgNPs were 0.25 and 0.5 mg/ml for Escherichia coli ( E. coli ), and 0.5 and 1.0 mg/ml for Staphylococcus aureus , respectively. According to the mechanism study, AgNPs generated morphological changes in E. coli and disrupted membrane integrity. Furthermore, AgNPs which showed effective dose-dependent inhibitory effects against Penicillium citrinum could suppress the growth of fungal mycelium on citrus peels. Conclusively, AgNPs generated by this facile and green method showed effective antimicrobial activity, which is promising in food processing and preservation. Practical applications Silver nanoparticles have attracted enormous attention because of their intrinsic properties which are advantageous in many applications, especially in agriculture and food industry. Silver nanoparticles were synthesized by medicine food homology plants Lonicera japonica Thunb. leaves for the first time, which was a green synthesis method. Synthesized AgNPs showed good antibacterial and antifungal properties, which has the potential use in citrus preservations.
