The effect of imidazole on the enhancement of gadolinium-porphyrin phosphorescence at room temperature

Lixin Zang,Huimin Zhao,Jianyu Hua,Feng Qin,Yangdong Zheng,Zhiguo Zhang,Wenwu Cao

A mechanism for the enhanced room-temperature phosphorescence (RTP) of gadolinium-coordinated hematoporphyrin monomethyl ether (Gd-HMME) in the presence of imidazole and free gadolinium ions (Gd3+) is revealed. Imidazole, the molten solvent used in the synthesis of Gd-HMME, was effective in enhancing the Gd-HMME RTP. In the presence of imidazole, further enhancement of the Gd-HMME RTP was observed upon adding Gd3+. Overall, a 40-fold enhancement of Gd-HMME RTP intensity was achieved by adding both imidazole and Gd3+. In addition, there was an increase in the RTP lifetime. Through spectroscopic analysis, we deduced that a protective medium is formed by the imidazole and the degree of this protection is further increased by Gd3+. The protective medium enhances the Gd-HMME RTP by partially inhibiting energy transfer from the lowest triplet state of Gd-HMME to oxygen. This was demonstrated by the presence of lower levels of singlet oxygen in the Gd-HMME solution after the addition of imidazole. These results indicate that imidazole could have potential application as an RTP enhancer or triplet state protector.

Gadolinium–porphyrin based polymer nanotheranostics for fluorescence/magnetic resonance imaging guided photodynamic therapy

Wandi Chen,Junkai Zhao,Mengfei Hou,Mo Yang,Changqing Yi

Nanotheranostics for fluorescence/magnetic resonance (FL/MR) dual-modal imaging guided photodynamic therapy (PDT) are highly desirable in precision and personalized medicine. In this study, a facile non-covalent electrostatic interaction induced self-assembly strategy is developed to effectively encapsulate gadolinium porphyrin (Gd-TCPP) into homogeneous supramolecular nanoparticles (referred to as Gd-PNPs). Gd-PNPs exhibit the following advantages: (1) excellent FL imaging property, high longitudinal relaxivity (16.157 mM−1 s−1), and good singlet oxygen (1O2) production property; (2) excellent long-term colloidal stability, dispersity and biocompatibility; and (3) enhanced in vivo FL/MR imaging guided tumor growth inhibition efficiency for CT 26 tumor-bearing mice. This study provides a new strategy to design and synthesize metalloporphyrin-based nanotheranostics for imaging-guided cancer therapy with enhanced theranostic properties.

Folic acid-nanoscale gadolinium-porphyrin metal-organic frameworks: fluorescence and magnetic resonance dual-modality imaging and photodynamic therapy in hepatocellular carcinoma

Yang Chen,Wei Liu,Yue Shang,Peipei Cao,Jianlin Cui,Zongjin Li,Xuebo Yin,Yuhao Li

Background Hepatocellular carcinoma (HCC) is the most common primary liver cancer and severely threatens human health. Since the prognosis of advanced HCC remains poor, there is an urgent need to develop new therapeutic approaches. Porphyrin metal-organic frameworks are a class of porous organic-inorganic hybrid functional materials with good biocompatibility. Methods Gadolinium-porphyrin metal-organic frameworks were used as a skeleton for folic acid (FA) conjugation to synthesize a novel type of nanoparticle, denoted as folic acid-nanoscale gadolinium-porphyrin metal-organic frameworks (FA-NPMOFs). The FA-NPMOFs were characterized using transmission electron microscopy, Fourier transform infrared spectroscopy and thermogravimetric-differential thermal analysis. The biotoxicity and imaging capability of the FA-NPMOFs were determined using HepG2 cells and embryonic and larval zebrafish. The delivery and photodynamic therapeutic effect of FA-NPMOFs were explored in transgenic zebrafish with doxycycline-induced HCC. Results FA-NPMOFs were spherical in structure with good dispersion and water solubility. They showed low biotoxicity, emitted bright red fluorescence, and exhibited an excellent magnetic resonance imaging capability, both in vitro and in vivo . Meanwhile, the FA-NPMOFs exhibited a strong affinity for folate receptor (FR)-expressing cells and were delivered to the tumor site in a targeted manner. Moreover, HCC tumor cells were eliminated following laser irradiation. Conclusion FA-NPMOFs can be used for dual-modality imaging and photodynamic therapy in HCC and show promise for use as a carrier in new therapies for HCC and other FR-positive tumors.

Imaging-Guided Synergistic Photo-Chemotherapy Using Doxorubicin-Loaded Gadolinium Porphyrin-Based Metal–Organic Framework Nanosheets

Chen Yuan,Yang Li,Ziqing Xu,Chuanbing Wang,Jie Zhang,Yingying Jin,Zhihui Chen,Hanyao Sun,Feiyun Wu,Qingling Zhang,Yuxia Tang,Shouju Wang

Porphyrin-based metal–organic framework (MOF) nanosheets have promising potential for drug loading and imaging-guided photodynamic therapy. However, the feasibility of performing imaging-guided three-channel photodynamic-photothermal-chemotherapy to treat cancer was seldom reported. The study showed that the gadolinium-porphyrin-based MOF nanosystem (DOX-Gd-TCPP) was constructed by loading doxorubicin (DOX) on the surface of tetrakis (4-carboxyphenyl) porphyrin (TCPP) with chelated gadolinium(III) ions. DOX-Gd-TCPP displayed superior drug loading capacity (up to 60%). Compared to separate photothermal, photodynamic, or chemotherapy, DOX-Gd-TCPP nanosheets showed significantly higher efficacy against cancer through the combination effect of three-channel therapeutic modalities. Meanwhile, DOX-Gd-TCPP nanosheets exhibited excellent fluorescence and magnetic resonance imaging capability. It is predicted that the imaging-guided synergistic photo-chemotherapy strategy suggests that DOX-Gd-TCPP MOF nanosheets could potentially be used in cancer theranostics technology.

Central metal-derived co-assembly of biomimetic GdTPP/ZnTPP porphyrin nanocomposites for enhanced dual-modal imaging-guided photodynamic therapy

Jiefei Wang,Zhongjie Wang,Yong Zhong,Yan Zou,Chong Wang,Haigang Wu,Albert Lee,Weitao Yang,Xiao Wang,Yanjie Liu,Dongya Zhang,Jiliang Yan,Mingcong Hao,Meng Zheng,Roger Chung,Feng Bai,Bingyang Shi

Dual-modal imaging guided photodynamic therapy (PDT) of multifunctional nanocomposites holds great promise for precision tumor theranostics. However, poor heterogeneous interfacial compatibility between functional components, low hydrophilicity and complicated preparation of nanocomposites remain major obstacles for further bioapplication . Herein, a facile central metal-derived co-assembly strategy is developed to effectively integrate gadolinium porphyrin (GdTPP) contrast agent and Zinc porphyrin (ZnTPP) photosensitizer into a homogeneous GdTPP/ZnTPP nanocomposites (GZNs). GZNs possesses the following advantages: (1) Greatly improved interfacial compatibility facilitated by incorporating two metalporphyrins with same group (phenyl-) and different central metal atoms (Zn and Gd) leading to higher yield (4.7–5 fold) than either monocomponent nanoparticles . (2) Poor dispersity of GdTPP nanoparticles is greatly improved after integrating with ZnTPP blocks. (3) The GZNs inherit excellent fluorescence imaging, high relaxation rate (8.18 mM −1  s −1 ) and singlet oxygen production from two raw metalporphyrins. After camouflaging with homotypic cancer cell membrane for immunologic escape, the HeLa membrane coated GZNs (mGZNs) show enhanced in vivo MR/FL imaging guided anti-tumor targeting efficiency of 80.6% for HeLa cells . Our new strategy using central metal-derived co-assembly of homogeneous building blocks greatly improves interfacial compatibility to achieve combined functions for visualized cancer theranostics.
