The use of low-cost working electrodes is of great importance for the development of sensor industrialization. Acupuncture needles electrode (ANE) was selected as working microelectrode, a sensor was designed based on poly(3,4-ethylenedioxythiophene)-polystyrene sulfonate (PEDOT: PSS) and chitosan (CS) modified covalent organic frameworks (COFs) (CS/COFs) for detection of cadmium (Cd(II)). First, a mixture of PEDOT and PSS was electrodeposited on the surface of the acupuncture needles to synthesize a PEDOT: PSS as conductive polymeric film to support the electron transfer capability and enhance the conductive efficiency of the sensor. Then, CS/COFs was self-assembled and attached to the PEDOT: PSS/ANE surface by π-π stacking intermolecular interaction forces and the viscosity of CS, thus increasing the specific surface area of the electrode. Differential pulse anodic stripping voltammetry (DPASV) was used as the electrochemical method for detection of Cd(II). And the results indicated that the CS/COFs/PEDOT: PSS/ANE was linearly responsive in the range of 400 μg/L to 2400 μg/L, and the LOD was 13.8 ng/L for Cd(II). The constructed sensor also showed great selectivity, reproducibility, and stability. Additionally, the sensor demonstrated outstanding recovery in the detection of Cd(II) in real sample scallops. This sensing construction method based on acupuncture needles as working electrodes is of great significance to the development of sensor industrialization and aquatic product safety detection.

Promethazine (PMZ) is an effective antihistamine that is used as a nerve tranquilizer to treat mental disorders. However, drug abuse causes harm to the human body and also pollutes the environment to a certain extent. Therefore, it is crucial to develop a highly selective and sensitive biosensor for PMZ determination. An acupuncture needle (AN) was used as an electrode in 2015, and further research on the electrode's essence in electrochemistry is needed. In this work, a sensor based on a surface imprinted film coordinated Au/Sn biometal was first fabricated on AN via electrochemistry. The obtained cavities showed complementary and suitable sites for “N atom” electron transfer through the phenyl ring structure in promethazine, which is rigorous for the configuration near the interface. Under the optimal conditions, MIP/Au/Sn/ANE exhibits a good linear relationship in the range of 0.5 μM–500 μM, and the detection limit (LOD) is 0.14 μM (S/N = 3). The sensor exhibits good repeatability, stability, and selectivity and can be successfully used to analyze and detect PMZ in human serum and environmental water. The findings are scientifically significant for AN electrochemistry and the sensors have potential for in vivo medicamentosus monitoring in the future.

Epinephrine (EP) is an important neurotransmitter that plays a vital role in mammalian central nervous and hormone systems. So, the monitoring of EP is significant to evaluate human health. Until now, developing new methods for detecting EP has attracted increasing interest to avoid the previous disadvantages, such as high cost, long time-consuming, and professional operation. Acupuncture is a kind of traditional Chinese treatment. Herein, stainless steel needle, an acupuncture tool, was used as an electrochemical matrix and functionalized to be a simple, effective sensor for detecting EP. The acupuncture needle showed a typical electrochemical behavior. Gold nanoparticles coordinated polydopamine-modified acupuncture needle (AuNPs/PDA/AN) was fabricated via the green route of pure electron-transfer induced reaction of electrodepositing. The covered interface exhibited synergistic response performance to EP. The electron transfer of the phenolic hydroxyl group in EP molecular was enhanced via the superficial active site, showing the notable current peak. The generated current intensity can be used as an index to reflect the corresponding concentration. Under the optimized conditions, the fabricated biosensor expressed a wide and good linear relationship from 1 μM to 1000 μM, and a detection limit of 0.26 μM, which are competitive with former relative reports. In practical application, the fabricated sensor has been effectively used to analyze EP in real human serum, and also realized the real-time detection of EP secreted by PC12 cells irritated by salbutamol. The sensor was found to perform high selectivity , stability, and reproducibility, which is promising in the future given the advantages of electrochemical sensors , including low price, fast response, and simple operation.

To obtain depth profiles of surface-enhanced Raman scattering (SERS) information in living systems, a SERS-active needle was structured by acupuncture needles, gold nanoshells (GNSs), and polystyrene, which were used as carriers, SERS-active elements to be absorbed on the carriers, and coatings to protect the absorbed GNSs from being erased during insertion, respectively. The SERS-active needle is minimally invasive for entering and exiting the body. The interspaces between the GNSs became vessels to collect diffused fluids at different depths after a SERS-active needle was inserted into an agarose gel, and the SERS intensity profile on the SERS-active needle coincided with the concentration profile of Nile Blue A (NBA) in the gel. SERS detection in vitro avoided the signal attenuation in gels, and the SERS detection at different spots of the SERS-active needle provided a depth profile of the NBA molecule in the gel. In vivo experiments of NBA and 6-mercaptopurine confirmed that the SERS-active needle could collect fluids in living systems easily with minimal invasion and provide information about depth profiles of target molecules in tissues.

Accurate determination of potassium ion (K + ) concentration in fingertip blood, soil pore water, pipette solution, and sweat is crucial for performing biological analysis, evaluating soil nutrients levels, ensuring experimental precision, and monitoring electrolyte balance. However, current electrochemical K + sensors often require large sample volumes and oversized reference electrodes, which limits their applicability for the aforementioned small-volume samples. In this paper, a K + sensor integrated with a glass capillary and a spiral reference electrode was proposed for detecting K + concentrations in small-volume samples. A K + -selective membrane (K + -ISM)/reduced graphene oxide-coated acupuncture needle (working electrode) was spirally wrapped with a chitosan-graphene/AgCl-modified Ag wire (reference electrode). This assembly was then inserted into a glass capillary, forming an anisotropic diffusion region of an annular cylindrical gap with width 410 µm and height 20 mm. It was found that the capillary action of the glass capillary results in a raised liquid level of the sample inside it compared to that in the container, which promotes efficient contact between the small-volume sample and the K + sensor. Besides, the formed anisotropic diffusion region limits the K + diffusion from the bulk solution to the K + -ISM, which leads to a larger potentiometric response of the K + -ISM. The glass capillary-assembled K + sensor displays high performance, including a sensitivity 58.3 mV/dec, a linear range 10 −5 –10 −1 M, and a detection limit 1.26 × 10 −6 M. Moreover, it reliably determines K + concentrations in artificial sweat of microliter volume. These results facilitate accurate detection of K + concentration in fingertip blood, soil pore water, and pipette solution.

The development of sensitive and rapid methods for analysis and detection of small molecules is highly desirable for medical diagnostics and therapeutics. We report an acupuncture needle functionalized with gold nanoparticles (Au NPs) and a macrocyclic amine (MA) Raman tag as the platform to realize the sensitive detection of adenosine triphosphate (ATP) by surface-enhanced Raman spectroscopy (SERS). The assembled Au NPs with abundant hot spots on the surface of the needle avoids the aggregation of Au NPs and results in a good signal response. Moreover, there is strong combination between ATP and MA through electrostatic adsorption, hydrogen-bonding interactions, and π–π stacking, and as a consequence, this functionalized needle can be used as a SERS platform for detection of ATP (25 nM) through a decrease of the Raman signal of MA resulting from the high chemical affinity of ATP for MA. Specially, the Au NP/MA-functionalized needle is conveniently used to monitor ATP (100 nM) added to serum, and demonstrates great promise in the study and detection of ATP in a complex sample, laying the foundation for SERS applications in complex acupuncture specimens with fast response and simple operation.

Chlorpromazine (CPZ) is a medicine for nervous system disorders. Measuring CPZ in vivo can assist doctors in evaluating patients’ blood drug concentration and monitoring drug metabolism. Therefore, an accurate in vivo detection of CPZ is crucial. In recent years, the acupuncture needle, traditionally used in Chinese medicine, has emerged as a potential electrode in the field of electrochemistry, with promising applications for in vivo detection. In this study, Au/Cu nanoparticles were electrodeposited onto an acupuncture needle electrode (ANE) to improve electrical conductivity and provide an electro-catalytic surface. Subsequently, 3-aminophenylboronic acid and CPZ were attracted to each other through intermolecular forces; at the same time, the interaction force of Au–S between CPZ and the AuNPs made the polymer layer grow around the CPZ molecules on the modified electrode surface. The imprinted nanocavities showed highly selective and sensitive detection performance for CPZ after elution. Inside the recognizable site and microenvironment of the cavities, the captured CPZ molecule provided a suitable configuration for the fluent electron transfer of the electroactive group within a short range from the Au/Cu bimetal. Under ideal conditions, the MIP/Au/Cu/ANE exhibited two good linear ranges of 0.1–100 μM and 100–1000 μM with a detection limit of 0.07 μM. Moreover, the sensors showed great selectivity, good stability and excellent repeatability, making them suitable for CPZ detection in human serum. This provides a novel idea for real-time and in vivo CPZ detection.

We report a unique nanosensing platform by combining modern nanotechnology with traditional acupuncture needle to prepare graphene-modified acupuncture needle (G-AN) and using it for sensitive detection of neurotransmitters via electrochemistry. An electrochemical deposition method was employed to deposit Au nanoparticles (AuNPs) on the tip surface of the traditional acupuncture needle, while the other part of the needle was coated with insulation paste. Subsequently, the G-AN was obtained by cyclic voltammetry reduction of a graphene oxide solution on the surface of the AuNPs. To investigate the sensing property of the G-AN, pH dependence was measured by recording the open circuit potential in the various pH buffer solutions ranging from 2.0 to 10.0. What’s more, the G-AN was further used for detection of dopamine (DA) with a limit of detection of 0.24 μM. This novel G-AN exhibited a good sensitivity and selectivity and could realize direct detection of DA in human serum.

This study reports an acupuncture needle modified with an iron-porphyrin functionalized graphene composite (FGPC) for real-time monitoring of nitric oxide (NO) release in acupoints of rats. A gold film was first deposited to the needle surface to enhance the conductivity. The FGPC was prepared via hydrothermal synthesis, and subsequently applied to the tip surface of acupuncture needle by electrochemical deposition method. The functionalized needle enabled a specific and sensitive detection of NO based on the favorably catalytic properties of iron-porphyrin and the excellent conductivity of graphene. Amperometric data showed that the needle achieved not only a low detection limit down to 3.2 nM in PBS solution, but also a satisfactory selectivity. Interestingly, the functionalized needle could be inserted into the acupoints of rats for real-time monitoring of NO in vivo . It was found that a remarkable response to NO was respectively obtained in different acupoints when stimulated by L -arginine ( L -Arg), revealing that the release of NO was detectable in acupoints. We expect this work would showcase the applications of acupuncture needle in detecting some important signaling molecules in vivo , and exploring the mechanism of acupuncture treatment.

A novel electrochemical microsensor is constructed on a traditional acupuncture needle (AN) and used to monitor a biomarker of the SARS-CoV-2-N protein. The reversible interaction of the borate bond between the cis-diol in this glycoprotein and the phenylboronic acid in 4-mercaptophenylboronic acid (4-MPBA) is effectively exerted. This interaction was applied to anchor the SARS-CoV-2-N protein onto 4-MPBA, which is covalently self-assemblied onto electrodeposited AuNPs by the S-Au bond. Meldola blue was then electropolymerized around the protein template. After the template was eluting, three-dimensional nanocavities complementary with the protein were generated within the polymelola blue (pMB) layer. Interestingly, nanocavities could play a channel role for the electron-transfer of outer [Fe(CN)6]3−/4−, and the signal of electrochemical probe could be hindered after recombining the SARS-CoV-2-N protein, which laid a platform for the detection of this biomarker. After optimizing the influencing factors, the prepared microsensor exhibits a linear range of 0.1 ~ 1000 ng mL−1 with a low detection limit of 0.01 ng mL−1 (S/N = 3). In particular, the sensing ability was dramatically affected by the thickness correlative factors for the polymer matrix. The suitable thickness is effective for the sensing signals, which corresponds to the behavior of surface-imprinted polymer. The microsensor showed comparatively high sensitivity and selectivity and practically detected the SARS-CoV-2-N protein in the serum sample, which is of scientific significance for the development of the electrochemical microsensor and acupuncture.

Osteopontin (OPN) is an important biomarker for reflecting osteoarthritic inflammation and endochondral ossification. In the field of electroanalysis, OPN belongs to non-electroactive protein, which is usually detected by means of an outer probe or biolabel. Here, a novel microsensor can directly electroanalyze OPN, which was constructed by coordinating surface molecularly imprinted polymer (SMIP) with a built-in electroactive probe of poly (methylene blue) (pMB) on an acupuncture needle microelectrode (ANME). The OPN template can be reversibly anchored using 4-mercaptophenylboric acid (4-MBPA) via a borate bond between phenylboronic acid and the external cis-diol of the glycoprotein. Methylene blue (MB) and dopamine (DA) were sequentially electropolymerized and grown around templates, which played pivotal roles in the detection signal from the built-in pMB through the imprinted nanocavities. After the recombination of OPN molecules with imprinted nanocavities, the current strength of built-in pMB could be impeded with a highly sensitive response. This microsensor shows a linear relationship to the concentration of OPN from 0.01 to 1000 ng mL-1 with a detection limit of 3 pg mL-1. The microsensor also exhibits high selectivity and stability, which is attributed to the recognizing ability of the imprinted nanocavities and the hindrance and anti-interference function of coated polydopamine (pDA). This strategy of preparing a sensor shows practical and scientific significance for functionalizing microelectrodes and constructing microsensors for non-electroactive glycoproteins. In the future, it will be fascinating to integrate this microsensor with the acupuncture technique.

Glucose concentration in the body is a very important index for health evaluation. Nonenzymatic glucose biosensors attract persistent interest and research. The acupuncture technique has been developed for treating the disease for thousands of years. Here, a stainless-steel acupuncture needle (AN) was first used to sense glucose after functionalizing with bimetal of gold nanoparticles (AuNPs) and nickel nanoparticles (NiNPs). AuNPs were electrodeposited as particle clusters, and the NiNPs could be further electrodeposited around AuNPs to form a nano-pinecone shape with a size of about 600 nm. Au/AN exhibited the best electroconductivity compared to the weakest Ni/AN and medium Ni/Au/AN interfaces. Three nanotechnically processed interfaces showed obvious electrochemical response behavior for glucose compared with the bare AN. Surprisingly, the matrix of AuNPs could drastically boost the electrochemical response of NiNPs for glucose, which might be due to the abundance of active sites in the bimetallic hierarchy. The Ni/Au/AN electrode possessed an additive signal compared with Au/AN and Ni/AN. Under the optimized conditions, the prepared nonenzymatic biosensor exhibited two linear relationships in the wide concentration range of 0.5 μM to 1.5 mM and 1.5–5 mM with sensitivities of 766.02 and 368.77 (μA/mM/cm2), and the limit of detection was 0.14 μM. Moreover, the Ni/Au/AN showed superior performance over reported electrodes with great selectivity, stability, and reproducibility. The apparent Km of 0.161 mM was lower than that of glucose oxidase, implying that the Ni/Au-modified AN exhibited a strong affinity for glucose. The fabricated biosensor can be successfully used for the detection of glucose concentration in human serum, which provided a leading direction and potential for clinical point-of-care monitoring.
