Determination of bergenin based on the electrochemiluminescence quenching of tris(2,2′-bipyridine)-ruthenium(II)

Yinggui Zhu,Lili Zou,Qin Dong,Derong Jiang

Quenching effects of bergenin, based on the electrochemiluminescence (ECL) of the tris(2,2′-bipyridyl)-ruthenium(II) (Ru(bpy) 3 2+ )/tri-n-propylamine (TPrA) system in aqueous solution, is been described. The quenching behavior can be observed with a 100-fold excess of bergenin over Ru(bpy) 3 2+ . In the presence of 0.1 m TPrA, the Stern–Volmer constant ( K SV ) of the ECL quenching is as high as 1.16 × 10 4 M −1 for bergenin. The logarithmic plot of the inhibited ECL versus logarithmic plot of the concentration of bergenin was linear over the range 3.0 × 10 −6 –1.0 × 10 −4 mol/L. The corresponding limit of detection was 6.0 × 10 −7 mol/L for bergenin (S/N = 3). In the mechanism of quenching it is believed that the competition of the active free radicals between Ru(bpy) 3 2+ /TPrA and bergenin was the key factor for the ECL inhibition of the system. Photoluminescence, cyclic voltammetry, coupled with bulk electrolysis, supports the supposition mechanism of the Ru(bpy) 3 2+ /TPrA–bergenin system. Copyright © 2015 John Wiley & Sons, Ltd.

Ruthenium(II) Complexes Coupled by Erianin via a Flexible Carbon Chain as a Potential Stabilizer of c-myc G-Quadruplex DNA

Zhixiang Wang,Wentao Liu,Guohu Li,Jiacheng Wang,Bin Zhao,Peishan Huang,Wenjie Mei

Herein, two novel ruthenium(II) complexes coupled by erianinviaa flexible carbon chain, [Ru(phen)2(L1-(CH2)4-erianin)](ClO4)2(L1= 2-(2-(tri-fluoromethyphenyl))-imidazo [4,5f][1–10]phenanthroline (1) and [Ru(phen)2(L2-(CH2)4-eria)](ClO4)2(L2= 2-(4-(tri-fluoromethyphenyl))-imidazo [4,5f][1,10]phenanthroline (2), have been synthesized and investigated as a potential G-quadruplex(G4) DNA stabilizer. Both complexes, especially 2, can bind toc-mycG4 DNA with high affinity by electronic spectra, and the binding constant calculated for 1 and 2 is about 15.1 and 2.05 × 107M−1, respectively. This was further confirmed by the increase in fluorescence intensity for both complexes. Moreover, the positive band at 265 nm in the CD spectra ofc-mycG4 DNA decreased treated with 2, indicating that 2 may bind toc-mycG4 DNA through extern groove binding mode. Furthermore, fluorescence resonance energy transfer (FRET) assay indicated that the melting point ofc-mycG4 DNA treated with 1 and 2 increased 15.5 and 16.5 °C, respectively. Finally, molecular docking showed that 1 can bind toc-mycG4 DNA in the extern groove formed by base pairs G7–G9 and G22–A24, and 2 inserts into the small groove ofc-mycG4 DNA formed by base pairs T19–A24. In summary, these ruthenium(II) complexes, especially 2, can be developed as potentialc-mycG4 DNA stabilizers and will be exploited as potential anticancer agents in the future. Keywords:ruthenium(II) complexes;erianin;c-mycG-quadruplex DNA;stabilizer

Ruthenium(II) containing supramolecular polymers with cyclopentadithiophene–benzothiazole conjugated bridges for photovoltaic applications

Kui Feng,Xiaoyan Shen,Ying Li,Yujiang He,Dong Huang,Qiang Peng

Two novel supramolecular polymers (PHBTRU and PFBTRU) containing ruthenium(II) metal ions and bis-terpyridyl ligands with cyclopentadithiophene–benzothiazole conjugated bridges, have been successfully synthesized for application in polymer solar cells by the supramolecular self-assembly process. Both polymers exhibit appropriate solubility in organic solvents and excellent thermal stability with decomposition temperatures over 400 °C. The UV-vis and cyclic voltammetry results show that the resulting polymers have good light-harvesting abilities, low bandgaps as well as low-lying HOMO levels. Compared to polymer PHBTRU, the fluorine atom effect significantly improves the above optical and electrochemical properties of PFBTRU. The photovoltaic devices based on PHBTRU and PFBTRU were fabricated and evaluated with a conventional structure of ITO/PEDOT:PSS/polymer:PC71BM/Ca/Al. The PFBTRU device exhibited the highest power conversion efficiency (PCE) of 2.66% with a Voc of 0.73 V, a Jsc of 7.12 mA cm−2 and a FF of 0.51, which was obtained without any additives and/or posttreatment of the active films. The photovoltaic performance achieved in this work is the best result of the reported supramolecular polymers being used in conventional BHJ polymer solar cells.

Photocytoxicity in vitro studies of ruthenium (II) complexes inducing cell death through targeting DNA

Xue-Wen Liu,Liang Ma,Geng-Jun He,Xia Hu,Yuan-Dao Chen,Song-Bai Zhang

Photodynamic therapy (PDT) has been proven to be an important and effective therapy method with good spatial and temporal accuracy in clinical cancer treatment. Among many PDT candidates, Ru(II)-based compounds have received more attention due to their excellent photochemical and photophysical properties, high singlet oxygen ( 1 O 2 ) photogeneration, and DNA affinities. In this study, three ruthenium(II) complexes (RC1, RC2, RC3) have been designed by varying the aromatic plane area of the main ligand to adjust their 1 O 2 photogeneration, DNA affinities, DNA photodamage, and photocytoxicity. All complexes display no cytotoxicity in the dark against Hela, A549, HepG2, and BEL-7402 cells. RC1 displays no photocytotoxicity against all tumor cells. RC2 and RC3 showed high photocytotoxicity against BEL-7402 cells. The cellular uptake, cell migration, colonies assay, apoptosis analysis, and cell cycle analysis of RC2 and RC3 confirmed that the ruthenium(II) complexes might effectively enter the cancer cells, and lead to BEL-7402 cells apoptosis through DNA damage pathway. DNA photocleavage, DNA binding, and 1 O 2 measurement experiments demonstrated the DNA damage caused by RC2 and RC3 under light, compared to RC1, might be due to their high DNA affinities, large 1 O 2 quantum yields, and large lipophilicity by varying the aromatic plane area of the main ligand. Time dependent density functional theory (TDDFT) calculated results further show that two complexes exhibit the photocytotoxicity possibly via type II photoreaction mechanism because of the larger Δ S-T than O 2 and strong SOC between S n and T m . Hence, rational design of ruthenium(II) complexes by increasing DNA affinities, 1 O 2 quantum yields and lipophilicity can be regarded as an effective approach for enhanced photodynamic therapy (PDT).

Ruthenium(II) complexes synthesized as potential inducers of conformation inversion of AS1411 G4 DNA

Peishan Huang,Wenhui Deng,Qiong Wu,Wanwan Hong,Jing Liu,Jiangbo Liang,Bin Zhao,Wenjie Mei

Herein, four ruthenium(II) complexes, [Ru(L) 2 (PEIP)(CH 2 ) 4 Br)](ClO 4 ) 2 (L = phen, 1 ; bpy, 2 , PEIP = 2-(2-phenyl-ethenyl)-1 H -imidazole-[4,5 f ][1,10]phenanthroline) and [Ru(L) 2 (PEIP)(CH 2 ) 4 Erianin)](ClO 4 ) 2 (L = phen, 1E ; bpy, 2E ) have been synthesized and investigated as promising inducer to promote conformation inversion of AS1411 G4 DNA. All of these complexes can bind and stabilize AS1411 G4 DNA. Subsequent investigations reveal that in the presence of 1 and 1E, the described circular dichroism signal of AS1411 G4 DNA was reversed. Furthermore, it is discovered that in the presence of 1 and 1E, AS1411 G4 DNA can condense to a somewhat loose nano-network. Moreover, the analysis using molecular docking demonstrates that the conformation inversion of AS1411 G4 DNA is largely dependent on the binding sites of 1 and 1E. In conclusion, these complexes will be used for tumor diagnostics in the clinical context because of their capacity to stably bind AS1411 G4 DNA and dramatically boost its fluorescence emission. Besides, these complexes show promise as inducers of AS1411 G4 DNA conformational inversion.

Microwave-assisted synthesis of ruthenium(II) complexes containing levofloxacin-induced G2/M phase arrest by triggering DNA damage

Ruotong Liu,Chanling Yuan,Yin Feng,Jiayi Qian,Xiaoting Huang,Qiutong Chen,Shuyuan Zhou,Yin Ding,Bingbing Zhai,Wenjie Mei,Liangzhong Yao

Ru(II) complexes have attracted increasing attention as promising antitumor agents for their relatively low toxicity, high affinity to DNA molecules, and correlation with multiple targets. Meanwhile, quinolones are synthetic antibacterial agents widely used in the clinical practice. In this paper, two novel Ru(II) complexes coordinated by levofloxacin (LOFLX), [Ru(bpy)2(LOFLX)]·2ClO4 (1), and [Ru(dmbpy)2(LOFLX)]·2ClO4 (2) (bpy = 2,2′-bipyridine, dmbpy = 4,4′-dimethyl-2,2′-bipyridine) were synthesized with high efficiency under microwave irradiation and characterized by ESI-MS, 1H NMR, and 13C NMR. The binding behavior of these complexes with double-strand calf thymus DNA(CT-DNA) was investigated using spectroscopy, molecular docking, and density functional theory calculations. Results showed that 2 exhibited higher binding affinity than 1 and LOFLX. Further studies showed that 2 could induce the G2/M phase arrest of A549 cells via DNA damage. In summary, these results indicated that 2 could be developed as a potential anticancer agent in treatment of lung cancer through the induction of cell cycle arrest at G2/M phase by triggering DNA damage.

Highly Luminescent and Self-Enhanced Electrochemiluminescence of Tris(bipyridine) Ruthenium(II) Nanohybrid and Its Sensing Application for Label-Free Detection of MicroRNA

Jing Ye,Guoyong Liu,Mengxia Yan,Qiuju Zhu,Liping Zhu,Jianshe Huang,Xiurong Yang

Inspired by the coreactive activity of carbon nanodots (CDs) and branched polyethylenimine (BPEI) toward electrochemiluminescence (ECL) of Ru(bpy)32+, a highly luminescent and self-enhanced ECL nanohybrid (Ru-BCDs) was synthesized through covalently linking BPEI-coated carbon dots (BCDs) with Tris (4,4′-dicarboxylic acid-2,2′-bipyridyl) ruthenium(II) dichloride (Ru(dcbpy)32+). The composition and morphological characterization demonstrated that the spherical Ru-BCDs particles with 12.1 ± 1.4 nm diameter were obtained. The enhanced ECL property of Ru-BCDs was proved to originate from the dual coreactive contribution of BPEI and CDs as coreactants as well as the intramolecular electron transfer process, which could shorten the electron transfer path and minimize energy loss. A carbon nitride nanosheet (CNN) was utilized to stabilize the Ru-BCDs-modified glassy carbon electrode, which greatly improved the stability of solid-state ECL. By utilizing the affinity discrepancy of the CNN to single-stranded and double-stranded nucleic acids, a label-free and signal-on ECL biosensor was constructed for the determination of microRNA-133a (miR-133a), a potential biomarker of acute myocardial infarction. The designed biosensor exhibited good performance of miR-133a detection with a detection limit of 60 fM and could be used for the detection of real human serum with satisfactory results. The self-enhanced ECL nanohybrid with distinguished ECL efficiency holds a promising prospect in biosensing and bioimaging applications.

Imaging of the nuclei of living tumor cells by novel ruthenium(II) complexes coordinated with 6-chloro-5-hydroxylpyrido[3,2-a]phenazine

Yang Ding,Qiong Wu,Kangdi Zheng,Linkun An,Xiaoying Hu,Wenjie Mei

Two novel ruthenium(II) complexes coordinated with 6-chloro-5-hydroxylpyrido[3,2-a]phenazine (CQM), [Ru(L)2(CQM)]ClO4 [L = 1,10-phenanthroline, (1) and 2,2′-bipyridine, (2)], were investigated as potential fluorescent probes to track dynamic changes in the nuclei of living cells. Confocal laser technology was used to observe their co-location inside the cells. The results showed that both complexes were taken up by HepG2 cells, especially 1, which was localized in the cell nuclei, whereas 2 was distributed in the cell nuclei and mitochondria. Further studies by real-time fluorescence observations revealed that 1 rapidly entered the living cells, namely, HepG2, HeLa and MCF-7 cells, imaged the dynamic changes of the nuclei of living tumor cells, and exhibited low toxicity toward cells. The results demonstrated that 1 may be developed into a novel fluorescent probe for living cell nuclei. This study facilitates the development of fluorescent chemosensors with metal complexes.

Self-enhanced electrogenerated chemiluminescence of ruthenium(II) complexes conjugated with Schiff bases

Panpan Li,Zhaoyu Jin,Meilian Zhao,Yanxue Xu,Yong Guo,Dan Xiao

In this work, we obtain two ruthenium(II) complexes with Schiff base cavities that exhibit significantly enhanced electrochemiluminescence (ECL) intensity and quantum efficiency due to Ru(bpy)32+ in aqueous solution, without the addition of tri-n-propylamine (TPrA). The great increase in ECL intensity is confirmed to be due to the electrochemical oxidation of phenolic hydroxyl groups and the resonant structure of imino radicals. Thus, the electrons are transferred intramolecularly to the Ru(III) center, leading to efficient generation of the excited state of Ru(II)*. Subsequently, we find that Co2+ bonding with the salen cavity can selectively and quickly quench the ECL signal. Furthermore, we study the mechanism of the process by which Co2+ hinders the oxidation of phenolic groups and blocks the electron transfer from imino radicals to the Ru center. Thus, a highly sensitive and selective ECL probe for the recognition of Co2+ was developed with a stable response over a concentration range of 0.9 μM to 6.3 μM and a detection limit as low as 21 nM.

New luminescent probe for the selective detection of dopamine based on in situ prepared Ru(II) complex-sodium dodecyl benzyl sulfonate assembly

Sujitraj Sheth,Mengyuan Li,Qijun Song

Due to clinical importance, detection of dopamine by using easy and rapid method is still ongoing challenge. Here we present a simple and quite efficient method for dopamine (DA) detection in alkalescent medium using in situ prepared Ru(II) complex and sodium dodecyl benzyl sulfonate (SDBS) as highly luminescent luminophore. The luminescence enhancement in the Ru(II) complex (Ru-CIP) has been observed in the miceller medium formed by SDBS. The capability to successively quench the luminescence intensity has been tested for variety of molecules and only dopamine as analyte found to be able to quench luminescence effectively. Hence selective quenching of luminescence by dopamine was used as a tool to detect dopamine and two linear concentration ranges has been established from 0.1 μM to 1μM and from 2 μM to 10 μM with limit of detection (LOD) is 6.6 nM (S/N = 3). Spectral evidence showed that luminescence quenching mechanism arose via Forster resonance energy transfer (FRET) among oxidized DA (i.e. DA quinone) and in situ generated Ru-CIP and SDBS assembly. Due to ultra sensitivity and high selectivity of the prescribed (Ru-CIP-SDBS) luminescent probe has a strong potential for practical analytical application in clinical diagnosis.

Phosphonate-Substituted Ruthenium(II) Bipyridyl Derivative as a Photoelectrochemical Probe for Sensitive and Selective Detection of Mercury(II) in Biofluids

Shuo Wu,Wenjuan Tu,Yanqiu Zhao,Xiuyun Wang,Jie Song,Xinlan Yang

A ruthenium(II) bipyridyl derivative photoelectrochemical probe, Ru-1, is synthesized and coupled with TiO2 nanoparticles (Ru-1/TiO2) for the specific recognition and highly sensitive photoelectrochemical (PEC) detection of Hg2+ in a series of biofluids. The probe is designed with a chromophore, a thiocyanate recognition unit, a π-conjugated photoelectron-transfer pathway, and a phosphonate anchor. TiO2 nanoparticles with strong affinity to phosphonate and suitable conduction band energy are used as intermediate layers to increase the Ru-1 adsorption amount and amplify the photocurrent response. Under irradiation, the Ru-1/TiO2/fluorine-doped tin oxide (FTO), with strong visible light-harvesting capacity, aqueous stability, and efficient photoelectron transfer, shows a high and stable photocurrent response. In the presence of Hg2+, however, the specific Hg2+ and NCS coordination changes the photophysical properties of Ru-1, imposing the probe with a wider band gap, a weaker absorbance, and a poorer photoelectron and hole separation efficiency, thus resulting in a significant photocurrent decrease. On the basis of the Hg2+-induced photocurrent change, the Ru-1/TiO2/FTO shows good selectivity and high sensitivity toward the PEC detection of Hg2+, with wide linear ranges from 10–12 to 10–7 and 10–7 to 10–3 g/mL, and a low limit of detection of 0.63 pg/mL. The PEC probe is recyclable and accurate for selective detection of Hg2+ in urine, serum, and cell extracts. The whole analysis can be completed within 15 min. These good analytical performances indicate that the PEC method might have great potential for the onsite detection of small molecules in biosystems.
