Tanshinones, the major bioactive components inSalvia miltiorrhizaBunge (Danshen), are synthesized via the mevalonic acid (MVA) pathway or the 2-C-methyl-D-erythritol-4-phosphate (MEP) pathway and the downstream biosynthesis pathway. In this study, the bacterial component lipopolysaccharide (LPS) was utilized as a novel elicitor to induce the wild type hairy roots ofS. miltiorrhiza.HPLC analysis revealed that LPS treatment resulted in a significant accumulation of cryptotanshinone (CT) and dihydrotanshinone I (DTI). qRT-PCR analysis confirmed that biosynthesis genes such asSmAACTandSmHMGSfrom the MVA pathway,SmDXSandSmHDRfrom the MEP pathway, andSmCPS,SmKSLandSmCYP76AH1from the downstream pathway were markedly upregulated by LPS in a time-dependent manner. Furthermore, transcription factorsSmWRKY1 andSmWRKY2, which can activate the expression ofSmDXR,SmDXSandSmCPS, were also increased by LPS. Since Ca2+signaling is essential for the LPS-triggered immune response, Ca2+channel blocker LaCl3and CaM antagonist W-7 were used to investigate the role of Ca2+signaling in tanshinone biosynthesis. HPLC analysis demonstrated that both LaCl3and W-7 diminished LPS-induced tanshinone accumulation. The downstream biosynthesis genes includingSmCPSandSmCYP76AH1were especially regulated by Ca2+signaling. To summarize, LPS enhances tanshinone biosynthesis throughSmWRKY1- andSmWRKY2-regulated pathways relying on Ca2+signaling. Ca2+signal transduction plays a key role in regulating tanshinone biosynthesis inS. miltiorrhiza.Keywords:lipopolysaccharide;tanshinones;Ca2+signaling;SmWRKY

Background The filamentous fungus Trichoderma reesei Rut-C30 is one of the most vital fungi for the production of cellulases, which can be used for biofuel production from lignocellulose. Nevertheless, the mechanism of transmission of external stimuli and signals in modulating cellulase production in T. reesei Rut-C30 remains unclear. Calcium is a known second messenger regulating cellulase gene expression in T. reesei. Results In this study, we found that a biologically relevant extracellular Mn2+ concentration markedly stimulates cellulase production, total protein secretion, and the intracellular Mn2+ concentration of Rut-C30, a cellulase hyper-producing strain of T. reesei. Furthermore, we identified two Mn2+ transport proteins, designated as TPHO84-1 and TPHO84-2, indicating that they are upstream in the signaling pathway that leads to cellulase upregulation. We also found that Mn2+ induced a significant increase in cytosolic Ca2+ concentration, and that this increased cytosolic Ca2+ might be a key step in the Mn2+-mediated regulation of cellulase gene transcription and production. The utilization of LaCl3 to block plasma membrane Ca2+ channels, and deletion of crz1 (calcineurin-responsive zinc finger transcription factor 1) to interrupt calcium signaling, showed that Mn2+ exerts the induction of cellulase genes via calcium channels and calcium signaling. To substantiate this, we identified a Ca2+/Mn2+ P-type ATPase, TPMR1, which could play a pivotal role in Ca2+/Mn2+ homeostasis and Mn2+ induction of cellulase genes in T. reesei Rut-C30.Conclusion sTaken together, our results revealed for the first time that Mn2+ stimulates cellulase production, and demonstrates that Mn2+ upregulates cellulase genes via calcium channels and calcium signaling. Our research also provides a direction to facilitate enhanced cellulase production by T. reesei.

The effect of sodium chloride (NaCl) on cadmium (Cd) uptake, translocation, and oxidative stress was investigated using 2 tobacco cultivars differing in Cd tolerance. The growth inhibition of the tobacco plants exposed to Cd toxicity was in part alleviated by moderate addition of NaCl in the culture solution. Cadmium concentration of shoots and roots in the 2 cultivars increased with increasing Cd levels in the solution and decreased with the addition of NaCl. The addition of NaCl could alleviate the oxidative stress caused by Cd toxicity, as reflected by reduced production of malondialdehyde and recovered or enhanced activities of antioxidative enzymes catalase and glutathione peroxidase. The results also showed that the enhancement of antioxidative enzyme activity by NaCl for the tobacco plants exposed to Cd stress is related to induced Ca signaling. When severe defects bone occur, limited mobility in patients is accompanied by delayed signal transmission and osteogenesis. A sonodynamic therapy based on ultrasound-driven piezoelectric scaffolds is designed to restore cellular connections for regeneration. A series of 3D-printed scaffolds are fabricated using different polyurethane-based composites (hydroxyapatite/BaTiO 3 ) and further polarized to achieve a high piezoelectric constant. Driven by low-intensity pulsed ultrasound (LIPUS), the electrical signals from polarized scaffolds are optimized, achieving an increased open-circuit voltage (197 mV) due to the interconnected piezoelectric phase. The ultrasound-driven piezoelectric cues elevated the cytosolic and mitochondrial calcium levels in bone marrow mesenchymal stem cells (BMSCs), where calcium ions acted as a key second messenger, fortifying mitochondrial energy metabolism and exciting cellular activity. After applying the sonodynamic therapy to rat skull defects, the calcium signaling pathway, oxidative phosphorylation, and citrate cycle are actuated in the energy metabolism through functioning networks, while the HIF-1 signaling pathway on angiogenesis, and the AMPK and MAPK signaling pathways on osteogenesis collectively improved. The optimized sonodynamic therapy facilitated ≈27.85% osteogenesis (8 W) with early neovascularization (4 W) in the defects. Given the high efficiency of the piezoelectric-transducing network, the sonodynamic therapy offers a sustained mitochondria-excited treatment to maintain bone viability.

In this study, the effect of polyethylene glycol 8000 (PEG8000) stress on cellulase biosynthesis in Trichoderma reesei CICC2626 via calcium signaling was investigated, and a plausible mechanism by which intracellular Ca 2+ regulates the transcription of cellulase genes was proposed. The results indicated that the total cellulase (filter paper-hydrolyzing activity [FPase]), endoglucanase (carboxymethyl cellulase activity [CMCase]), and β-glucosidase activities of the strain were 1.3-, 1.2-, and 1.3-fold higher than those of the control (no PEG8000 addition) at a final concentration of 1.5% (w/v) PEG8000. Moreover, the transcriptional levels of cellulase genes, protein concentrations, and biomass increased. With the synergistic use of commercial cellulase and T. reesei CICC2626 cellulase to hydrolyze alkali-pretreated rice straw, the released reducing sugar concentration reached 372.7 mg/g, and the cellulose content (22.7%, 0.32 g) was significantly lower than the initial content (62.5%, 1.88 g). Transcriptome data showed that 12 lignocellulose degradation–related genes were significantly upregulated in the presence of 1.5% PEG8000. Furthermore, the addition of Ca 2+ inhibitors and deletion of crz1 (calcineurin-responsive zinc finger 1-encoding gene, which is related to the calcium signaling pathway) demonstrated that calcium signaling plays a dominant role in PEG8000-induced cellulase genes overexpression. These results revealed a link between PEG8000 induction and calcium signaling transduction in T. reesei CICC2626. Moreover, this study also provides a novel inducer for enhanced cellulase production. Key points • Cellulase biosynthesis in Trichoderma reesei could be enhanced by PEG8000 • PEG8000 could induce a cytosolic Ca 2+ burst in Trichoderma reesei • The activated calcium signaling was involved in cellulase biosynthesis Graphical abstract

Gray mold , caused by Botrytis cinerea is an intractable fungal disease that causes extensive damage to agricultural products. In the search for novel antifungal active ingredients, we discovered a linear pyranocoumarin Pd-D-V was effective against B. cinerea in both in vitro and in vivo assays. Furthermore, this study investigated the effects of Ca 2+ and the Ca 2+ -calcineurin signaling pathway on its antifungal activity against B. cinerea . The results indicated that Pd-D-V reduced the concentration of Ca 2+ in the mycelia of B. cinerea ; CaCl 2 , the Ca 2+ channel blocker verapamil , or the calcineurin inhibitor cyclosporin A could affect the sensitivity of Pd-D-V against B. cinerea ; the expression of genes ( Bccch1, Bcmid1, BccnA, Bccnb1, Bcpmc1, and Bcpmr1 ) of the Ca 2+ -calcineurin signaling pathway decreased after Pd-D-V treatment. In summary, Pd-D-V is compound for developing fungicides against B. cinerea . Pd-D-V can reduce intracellular Ca 2+ concentration and disturb Ca 2+ homeostasis . The Ca 2+ -calcineurin signaling pathway is important in the antifungal activity of Pd-D-V against B. cinerea .

Heat stress is one of the most remarkable abiotic factors limiting the growth and productivity of Ulva prolifera . The calcium signalling pathway plays an important role in the plant heat resistance response. However, the physiology and molecular mechanisms by which the calcium signalling pathway regulates the U. prolifera response to high temperature remain largely unknown. Changes in the physiology, transcriptome , and proteome of U. prolifera in response to high temperature were investigated with exogenous application of CaCl 2 (denoted UpCa) and the calmodulin (CaM) inhibitor W-7 (N-(6-aminohexyl)-5-chloro-1-naphthalenesulfonamide hydrochloride, denoted UpW7) to fill this gap. In total, 33,475 differentially expressed genes (DEGs) and 869 differentially expressed proteins (DEPs) were identified between the control and UpCa treatments. At the mRNA level, the upregulated DEGs were enriched in the RNA transport , mRNA surveillance, carotenoid biosynthesis, basal transcription factor, porphyrin and chlorophyll metabolism, diterpenoid biosynthesis, and flavonoid biosynthesis pathways. The downregulated DEGs were enriched in photosynthesis, plant hormone signal transduction , photosynthesis-antenna proteins, and oxidative phosphorylation . At the protein level, upregulated proteins were enriched in alpha-linolenic acid metabolism, photosynthesis-antenna proteins, and plant–pathogen interactions. The downregulated proteins were enriched in endocytosis , mismatch repair, ribosomes, RNA degradation, and spliceosomes . The addition of exogenous CaCl 2 induced the expression of peroxidase , heat shock 70, heat shock 90, nuclear pore complex protein Nup62, MFS transporter, calcium-dependent protein kinase, calcium/calmodulin-dependent protein kinase kinase 1, calcium-binding protein calmodulin-like (CML), Ca 2+ -transporting ATPase, and serine/threonine-protein kinase at both the mRNA and protein levels. In addition, the same downregulated DEPs and DEGs were correlated and included the ApaG protein, chaperonin GroEL , proteasome inhibitor subunit 1 (PI31), serine/arginine repetitive matrix protein 1, glutamine synthetase, and F-type H + -transporting ATPase subunit alpha. The addition of W7 inhibited Ca 2+ -related gene expression, and the expression of genes and proteins involved in phagosome , photosynthesis-antenna proteins, RNA transport, plant–pathogen interactions, protein processing in the endoplasmic reticulum , and protein export were downregulated. However, exogenous CaCl 2 and W7 increased the expression of TFs, such as Myb and G2-like. In a subsequent study, the addition of CaCl 2 significantly increased and W7 significantly inhibited catalase and superoxide dismutase activity in U. prolifera under high-temperature stress. The expression of the CaM gene was significantly increased at 24 h in U. prolifera under high-temperature stress; CaCl 2 and W7 significantly inhibited the expression of the CaM gene. In summary, Ca 2+ activated Ca 2+ -related channels to regulate downstream transcript and response proteins; however, the addition of Ca 2+ and W7 inhibited photosynthesis. These results provide insights into the mechanism by which calcium increases the mechanical strength of U. prolifera in response to high-temperature stress.

Background Trichoderma reesei is renowned for its cellulase-producing ability and is used for biofuel production from lignocellulose. In plants and fungi, cellulase production is induced by cellulose and suppressed by glucose; however, whether metformin can enhance cellulase production and mitochondrial function in T. reesei remains unclear. Metformin reduces blood glucose levels by inhibiting hepatic gluconeogenesis; therefore, it is worth investigating whether metformin transmission modulates cellulase biosynthesis in T. reesei . Results Metformin increased cellulase production and the transcription of cellulase-related genes. It also enhanced the concentrations of Ca 2+ in the cytosol and mitochondria and regulated the transcription levels of cellulase-related genes by modulating calcium homeostasis in T. reesei QM6a. In addition, metformin was identified as an antioxidant that can enhance cellulase activity by reducing reactive oxygen species (ROS). Our results demonstrated that metformin influences the state of mitochondria by enhancing mitochondrial activity and membrane potential to promote cellulase production. Conclusion Collectively, these results indicate that metformin enhances cellulase production, scavenges ROS, and protects mitochondrial activity in T. reesei . Graphical Abstract

An increase in exoenzyme production can be enhanced by environmental stresses such as graphene oxide (GO) stress, but the link between the two events is still unclear. In this work, the effect of GO as an environmental stress factor on exoenzyme (lignocellulolytic enzyme, amylase, peptidase, and protease) biosynthesis was investigated in Bacillus subtilis Z2, and a plausible mechanism by which cytosolic Ca2+ regulates lignocellulolytic enzyme production in B. subtilis Z2 subjected to GO stress was proposed. The filter paper-hydrolyzing (FPase [representing total cellulase]), carboxymethylcellulase (CMCase [representing endoglucanase]), and β-glucosidase activities and extracellular protein concentration of the wild-type strain under 10 μg/mL GO stress were 1.37-, 1.64-, 1.24-, and 1.16-fold those of the control (without GO stress), respectively. Correspondingly, the transcription levels of lignocellulolytic enzyme genes, cytosolic Ca2+ level, and biomass concentration of B. subtilis were all increased. With lignocellulolytic enzyme from B. subtilis used to hydrolyze alkali-pretreated rice straw, the released reducing sugar concentration reached 265.53 mg/g, and the removal rates of cellulose, hemicellulose, and lignin were 52.4%, 30.1%, and 7.5%, respectively. Furthermore, transcriptome data revealed that intracellular Ca2+ homeostasis played a key role in regulating the levels of gene transcription related to the synthesis of lignocellulolytic enzymes and exoenzymes. Finally, the use of Ca2+ inhibitors (LaCl3 and EDTA) and deletion of spcF (a calmodulin-like protein gene) further demonstrated that the overexpression of those genes was regulated via calcium signaling in B. subtilis subjected to GO stress.IMPORTANCE To effectively convert lignocellulose into fermentable sugars, high lignocellulolytic enzyme loading is needed. Graphene oxide (GO) has been shown to promote exoenzyme (lignocellulolytic enzyme, amylase, peptidase, and protease) production in some microorganisms; however, the regulatory mechanism of the biosynthesis of lignocellulolytic enzymes under GO stress remains unclear. In this work, the lignocellulolytic enzyme production of B. subtilis under GO stress was investigated, and the potential mechanism by which B. subtilis enhanced lignocellulolytic enzyme production through the calcium signaling pathway under GO stress was proposed. This work revealed the role of calcium signaling in the production of enzymes under external environmental stress and provided a direction to facilitate lignocellulolytic enzyme production by B. subtilis.

Key message
Three annexin genes may be involved in the ripening progress of strawberry fruit. Phytohormones and calcium regulate the expressions of three annexin genes during strawberry fruit ripening.
Abstract Plant annexins are multi-functional membrane- and Ca 2+ -binding proteins that are involved in various developmental progresses and stress responses. Three annexins FaAnn5a , FaAnn5b and FaAnn8 cDNA obtained from strawberry fruit encode amino acid sequences of approximately 35 kDa containing four annexin repeats, Ca 2+ -binding site, GTP-binding motif, peroxidase residue, and conserved amino acid residues of tryptophan, arginine and cysteine. During fruit development, the transcript levels of FaAnn5a and FaAnn5b increased while FaAnn5b declined after 3/4R stage. The expression patterns of annexins suggested their potential roles in strawberry fruit development and ripening. Expressions of annexin genes were also highly correlated with hormone levels. In addition, exogenous abscisic acid (ABA) enhanced the expressions of FaAnn5a and FaAnn8 while exogenous auxin (IAA) retarded it. However, both ABA and IAA promoted the transcript levels of FaAnn5b , indicating the independent regulation of annexins in fruit likely due to multi-functions of their large family. The responses of annexin genes to exogenous ABA and IAA inhibitors verified the involvement of annexins in plant hormone signaling. Besides, calcium restrained the expressions of FaAnn5s ( FaAnn5a and FaAnn5b ) but promoted the expression of FaAnn8 . Effects of calcium and ethylene glycol tetraacetic acid (EGTA) on the transcript levels of annexins confirmed that calcium likely mediated hormone signal transduction pathways, which helped to elucidate the mechanism of calcium in fruit ripening. Therefore, FaAnn5s and FaAnn8 might be involved in plant hormones’ regulation in the development and ripening of strawberry fruit through calcium signaling in the downstream.

Activation of NADPH oxidase 2 (NOX2) triggers reactive oxygen species (ROS) generation, both of which are essential for robust microbial clearance by phagocytes. However, it is unknown whether inhibition of NOX2 activation or ROS generation affects cellular phagocytosis. Here, we found that the classic NOX2 inhibitor diphenyleneiodonium (DPI) induced uptake of E. coli by murine peritoneal macrophages through enhancing phagocytosis, and this effect was temperature-sensitive and attenuated by cytochalasin D as well as chemical inhibition of Syk and PLCγ, two downstream kinases involved in actin polymerization during phagocytosis. DPI also decreased the production of TNF-α and IL-6 resulting from E. coli stimulation. The DPI-induced enhancement of phagocytosis was independent of NOX2 inhibition or ROS generation but depended on increased intracellular calcium and activation of the p38 MAPK signaling pathway. Furthermore, DPI enhanced bacterial elimination and ameliorated inflammation in E. coli -infected mice, leading to improved survival. Our results demonstrate that DPI facilitates ROS-independent bacterial phagocytosis by macrophages through activation of calcium and p38 MAPK signaling pathways.

Summary cADPR is a well-recognized signaling molecule by modulating the RyRs, but considerable debate exists regarding whether cADPR can bind to and gate the TRPM2 channel, which mediates oxidative stress signaling in diverse physiological and pathological processes. Here, we show that purified cADPR evoked TRPM2 channel currents in both whole-cell and cell-free single-channel recordings and specific binding of cADPR to the purified NUDT9-H domain of TRPM2 by surface plasmon resonance. Furthermore, by combining computational modeling with electrophysiological recordings, we show that the TRPM2 channels carrying point mutations at H1346, T1347, L1379, S1391, E1409, and L1484 possess distinct sensitivity profiles for ADPR and cADPR. These results clearly indicate cADPR is a bona fide activator at the TRPM2 channel and clearly delineate the structural basis for cADPR binding, which not only lead to a better understanding in the gating mechanism of TRPM2 channel but also shed light on a cADPR-induced RyRs-independent Ca 2+ signaling mechanism.
