Sonochemical fabrication and characterization of β-Lactoglobulin-Galactooligosaccharide conjugates: Enhancement of calcium absorption

Jiayuan Liu,Xinyu Hou,Ziyuan Wang,Fei Gao,Gaoqi Ye,Jie Liu

Solid-in-oil-in-water (S/O/W) emulsions containing calcium carbonate (CaCO 3 ) are a potential new type of calcium-fortified emulsion system. This study developed ultrasonication-pretreated β-lactoglobulin (ULG)-galactooligosaccharide (GOS) conjugates using a novel combined method involving ultrasonication and Maillard reaction. This synergistic approach significantly increased the grafting degree and induced structural modifications of protein. The ULG-GOS conjugates exhibited superior emulsifying properties and interfacial stabilization capacity through exposed hydrophobic domains and strengthened electrostatic repulsion. When applied as stabilizers for S/O/W emulsions, ULG-GOS conjugates form a homogeneous system with significantly improved environmental stability. Notably, in vitro digestion studies revealed a 2-fold increase in calcium bioaccessibility (58.77 ± 1.75 %) of ULG-GOS stabilized system compared to native LG-stabilized systems. This study underscored the critical role of ultrasonication in enhancing the functionality of LG-GOS conjugates, which were essential to nutritional formulations designed to facilitate calcium absorption.

The threshold effect of Ce-Zn compound pollution on maize roots: implications for plant tolerance to combined metal pollution

Lei Wang,Zhe Li,Dongdong Tuo,Mingze Li,Mengtong Guo,Wenqian Zhang,Xiaodong Wang,Fuxin Hang,Ying Zhang

Heavy metal pollution is becoming increasingly severe, and rare earth elements (REEs) play a crucial role in influencing the environmental impact of heavy metals. Mixed heavy metal pollution presents a significant challenge due to its complex effects. While many studies have focused on single heavy metal pollution, relatively few have examined the pollution caused by rare earth-heavy metal composites. This study investigated the toxic effects of Ce-Zn compound pollution on maize roots and used the integrated biomarker response (IBR) to assess the toxicity of different concentrations of Ce-Zn compound pollution on maize roots. The results showed that Ce-Zn compound pollution had a threshold effect: promoting maize growth at low concentrations but inhibiting growth at a threshold concentration of Ce 50 mg/kg-Zn 50 mg/kg. Ce-Zn compound pollution-induced oxidative stress in maize. When the pollution concentration exceeded the threshold, the H₂O₂ content in maize roots increased by 3.30 and 3.56 times, respectively, compared to the CK group, while the O₂⁻ content increased by 1.45 and 1.48 times, respectively. Cell structure damage was observed, and the levels of antioxidant enzymes—peroxidase (POD), catalase (CAT), and superoxide dismutase (SOD)—in maize roots gradually increased. At low concentrations, the Ca²⁺ absorption rate in maize roots increased by 82.99 % compared to CK. By regulating the Ca²⁺ concentration, the absorption of H⁺ and other ions was affected, enhancing intracellular enzyme activity and improving the tolerance of maize roots to compound pollution. These findings enhance our understanding of the harmful effects of combined pollution from rare earth metals and heavy metals on plants.

Preparation of Calcium-Chelating Peptides from Squid Skin and Evaluation of Calcium Absorption Capacity in Caco-2 Cell Monolayer Model

Jihao Zeng,Xue Bai,Yongli Zhang,Qianyu Le,Jinhong Wu,Huiyun Chen

To develop a highly bioavailable calcium supplement, this study utilized Peruvian squid ( Dosidicus gigas ) skin as a raw material. Through alkaline protease hydrolysis and enzymatic membrane reactor separation, three molecular weight fractions of squid skin peptides were obtained, followed by calcium ion chelation to synthesize calcium-chelating peptides (CCPs-SS). Systematic characterization revealed that the less than 1 kDa fraction of CCPs-SS exhibited superior antioxidant capacity (82.18%) and calcium chelation efficiency (77.14%) in cellular models compared to higher molecular weight counterparts. Optimal synthesis conditions were identified as 60 °C, pH 9, and 12 mg/mL calcium chloride concentration. Post-chelation analyses demonstrated significant physicochemical alterations for CCPs-SS: ζ-potential shifted from −18.4 mV to −10.47 mV, while particle size increased from 476.75 nm to 664.4 nm. Notably, membrane separation enhanced phenylalanine and leucine molar concentrations by 25.5% and 57.6%, respectively, suggesting structural modifications that potentiate bioactivity. These findings demonstrate an innovative strategy for converting squid processing byproducts into functional nutraceuticals, which not only addresses calcium deficiency challenges but also promotes resource sustainability by utilizing waste materials.

Determination of phosphoryl-oligosaccharides obtained from Canna edulis Ker starch

Jianhua Liu,Hu Li,Jinhong Wu,Fan Xie,Juan Zhang,Zhengwu Wang

Phosphoryl oligosaccharides (POs) are a new type of functional oligosaccharide that promote the absorption of calcium. Canna edulis Ker (edible canna) starch contains a small amount of phosphorus and may be a source in the production of POs. Herein, the POs fraction was separated from Canna edulis Ker starch for the first time using enzymatic reactions that produce isomalto-oligosaccharides (IMOs). Fourier transform infrared (FT-IR) spectroscopy, nuclear magnetic resonance (NMR) spectroscopy, and high-performance anion exchange chromatography (HPAEC) were used to determine the presence and composition of the isolated POs. The POs that were obtained from Canna edulis Ker starch were mainly composed of phosphoryl maltotriose, maltotetraose, maltopentaose, maltohexaose, and maltoheptaose. These findings provide new insight into the simultaneous production of IMOs and POs from Canna edulis Ker starch by enzymatic reaction.

Bioactive peptide isolated from casein phosphopeptides promotes calcium uptake in vitro and in vivo

Guo Liu,Shengwei Sun,Baoyan Guo,Benchun Miao,Zhen Luo,Zumeng Xia,Danyang Ying,Fei Liu,Bin Guo,Jian Tang,Yong Cao,Jianyin Miao

Casein phosphopeptides (CPPs) have been demonstrated to be calcium chelators. Unfortunately, few studies have been reported on the effects of CPPs on the mechanism of the uptake and absorption of Ca2+ and bone metabolism. In this study, a monomeric peptide fraction isolated by RP-HPLC (F6-1) that possessed high calcium transport capacity in Caco-2 cell monolayers was separated and characterized. The effects of F6-1 on the absorption mechanisms of Ca2+ in a Caco-2 monolayer model and bone metabolism in rats were investigated. F6-1 was isolated by preparative and analytical RP-HPLC. Results for calcium transport suggested that the rates of Ca2+ transportation by F6-1 were approximately 2.57, 2.87 and 2.38 times higher than those in the control group at 30, 60 and 120 min, respectively. Results of ultraviolet (UV) spectroscopy indicated that the intensity of UV absorption changed because of the binding of Ca2+ to F6-1. Analysis of transepithelial electrical resistance (TEER) and the expression of TRPV6 in Caco-2 cells showed that F6-1 was likely to influence the transcellular pathway of intestinal absorption of Ca2+ rather than the paracellular pathway. Furthermore, the F6-1 group (1% Ca, 0.03% F6-1) exhibited increases in serum Ca2+ levels, femur length and femur Ca and decreases in serum alkaline phosphatase (ALP) levels and urinary pyridinoline content in a Sprague-Dawley rat model, which implied that F6-1 was beneficial for bone calcification. Overall, our results suggested that F6-1 enhanced the transport of Ca2+ in Caco-2 cells by affecting the transcellular pathway by upregulating the expression of TRPV6. F6-1 also improved bone formation and prevented bone resorption to benefit bone health in rats, which provided a basis for using F6-1 in calcium supplements or functional foods.

Effect of the phosphorylation structure in casein phosphopeptides on the proliferation, differentiation, and mineralization of osteoblasts and its mechanism

Wanying Zhong,Jian He,Wen Huang,Guangling Yin,Guo Liu,Yong Cao,Jianyin Miao

Our previous studies have shown that highly phosphorylated casein phosphopeptides (residues 1–25) P5 could efficiently bind calcium and promote intestinal calcium absorption, and enhanced bone development in rats. The purpose of this study was to investigate the effect of the phosphorylation structure in P5 on the proliferation, differentiation, and mineralization of osteoblasts (MC3T3-E1) and its mechanism. P5 was obtained by high-performance liquid chromatography (HPLC) and non-phosphorylated peptide P5-0 was obtained by chemical synthesis. Compared with the control group, the proliferation rate of MC3T3-E1 cells treated by P5 was 1.10 times that of P5-0 at 200 μg mL−1. P5 caused the cell cycle retention of MC3T3-E1 cells in the G2/M phase, while P5-0 had no significant difference in the G2/M phase. MC3T3-E1 cells incubated with P5 showed stronger alkaline phosphatase (ALP) activity than with P5-0, suggesting a tendency to promote cellular differentiation. Compared to the P5-0 treatment group, the P5 treatment group at concentrations of 10 μg mL−1 showed significant differences in the mineralization rates (p < 0.05). P5 significantly upregulated the expressions of Runx2, ALP, ColIα1, and OCN compared with the control group (p < 0.05). In addition, in silico molecular docking showed that the binding force of the P5-EGFR complex was stronger than that of the P5-0-EGFR complex, which was significantly related to the phosphorylation structure in P5 and might be an important reason for osteoblast proliferation. In conclusion, the phosphorylation structure and amino acid composition in P5 stimulated the osteogenic activity of MC3T3-E1 cells, and could be expected to be a functional food for the prevention of osteoporosis.

Structural characterization and calcium absorption-promoting effect of sucrose-calcium chelate in Caco-2 monolayer cells and mice

Qian Du,Ruiyan Wang,Zeyuan Deng,Jianqun Zhou,Nan Li,Wenwen Li,Liufeng Zheng

Sucrose emerges as a chelating agent to form a stable sucrose-metal-ion chelate that can potentially improve metal-ion absorption. This study aimed to analyze the structure of sucrose-calcium chelate and its potential to promote calcium absorption in both Caco-2 monolayer cells and mice. The characterization results showed that calcium ions mainly chelated with hydroxyl groups in sucrose to produce sucrose-calcium chelate, altering the crystal structure of sucrose (forming polymer particles) and improving its thermal stability. Sucrose-calcium chelate dose dependently increased the amount of calcium uptake, retention, and transport in the Caco-2 monolayer cell model. Compared to CaCl 2 , there was a significant improvement in the proportion of absorbed calcium utilized for transport but not retention (93.13 ± 1.75% vs. 67.67 ± 7.55%). Further treatment of calcium channel inhibitors demonstrated the active transport of sucrose-calcium chelate through Cav1.3. Cellular thermal shift assay and quantitative reverse transcription-polymerase chain reaction (qRT-PCR) assays indicated that the ability of sucrose-calcium chelate to promote calcium transport was attributed to its superior ability to bind with PMCA1b, a calcium transporter located on the basement membrane, and stimulate its gene expression compared to CaCl 2 . Pharmacokinetic analysis of mice confirmed the calcium absorption-promoting effect of sucrose-calcium chelate, as evident by the higher serum calcium level (44.12 ± 1.90 mg/L vs. 37.42 ± 1.88 mmol/L) and intestinal PMCA1b gene expression than CaCl 2 . These findings offer a new understanding of how sucrose-calcium chelate enhances intestinal calcium absorption and could be used as an ingredient in functional foods to treat calcium deficiency. Practical Application The development of high-quality calcium supplements is crucial for addressing the various adverse symptoms associated with calcium deficiency. This study aimed to prepare a sucrose-calcium chelate and analyze its structure, as well as its potential to enhance calcium absorption in Caco-2 monolayer cells and mice. The results demonstrated that the sucrose-calcium chelate effectively promoted calcium absorption. Notably, its ability to enhance calcium transport was linked to its strong binding with PMCA1b, a calcium transporter located on the basement membrane, and its capacity to stimulate PMCA1b gene expression. These findings contribute to a deeper understanding of how the sucrose-calcium chelate enhances intestinal calcium absorption and suggest its potential use as an ingredient in functional foods for treating calcium deficiency.

Application of a Mytilus edulis-derived promoting calcium absorption peptide in calcium phosphate cements for bone

Zhe Xu,Zhixuan Zhu,Hui Chen,Lingyu Han,Pujie Shi,Xiufang Dong,Di Wu,Ming Du,Tingting Li

The IEELEEELEAER peptide (PIE) identified from the protein hydrolysate of Mytilus edulis is reported to enhance osteoblast growth and differentiation, which also possesses a superior bone formation ability both in vitro and in vivo . Moreover, PIE bound to calcium spontaneously at the stoichiometry of 1:1, and there were amino nitrogen and carboxyl oxygen atoms in 2 glutamic acid residues at the calcium-binding sites in the PIE. The PIE–calcium complex facilitated calcium uptake through the Caco-2 cell monolayers. Incorporation of PIE into calcium phosphate cements enhanced calcium ion uptake and proliferation of osteoblasts and inhibit bacteria. This study suggest that calcium phosphate cements supplemented with PIE can serve as a potentially efficient material for bone graft used during spinal surgery.

The triacylglycerol structure and composition of a human milk fat substitute affect the absorption of fatty acids and calcium, lipid metabolism and bile acid metabolism in newly-weaned Sprague-Dawley rats

Lin Zhu,Shuaizhen Fang,Wenwen Liu,Hong Zhang,Yaqiong Zhang,Zhuohong Xie,Puyu Yang,Jianchun Wan,Boyan Gao,Liangli (Lucy) Yu

In this study, the effect of sn-2 palmitic triacylglycerols (sn-2 palmitic TAGs) and the ratio between the two major sn-2 palmitic TAGs (OPL to OPO ratio) in a human milk fat substitute (HMFS) on growth, fatty acid and calcium absorptions, and lipid and bile acid metabolic alterations was investigated in Sprague-Dawley rats. After 4 weeks of high-fat feeding, rats fed with the HMFS containing a sn-2 palmitic acid content of 57.87% and an OPL to OPO ratio of 1.4 showed the lowest TAG accumulation in their livers and hypertrophy of perirenal adipocytes, compared to the groups fed with fats containing a lower sn-2 palmitic acid content or a lower OPL to OPO ratio. Meanwhile, synergistically improved absorption of fatty acids and calcium and increased levels of total bile acids (BAs), especially for the tauro-conjugated BAs (TCDCA, TUDCA, TαMCA, TβMCA, TDCA and TωMCA), were observed in rats by both increasing the sn-2 palmitic acid content and the OPL to OPO ratio in HMFS. In addition, the levels of total BAs and tauro-conjugated BAs were negatively correlated with serum TAG, TC, and LDL-c levels and positively correlated with HDL-c levels according to Spearman's correlation analysis (P < 0.05). Collectively, these findings present new nutritional evidence for the potential effects of the TAG structure and composition of a human milk fat substitute on the growth and lipid and bile acid metabolism of the host in infancy.

Enhancement of Calcium Chelating Activity in Peptides from Sea Cucumber Ovum through Phosphorylation Modification

Lingyu Han,Yaoyao Li,Bing Hu,Wei Wang,Jianming Guo,Jixin Yang,Nuo Dong,Yingmei Li,Tingting Li

Recently, phosphorylation has been applied to peptides to enhance their physiological activity, taking advantage of its modification benefits and the extensive study of functional peptides. In this study, water-soluble peptides (WSPs) of sea cucumber ovum were phosphorylated in order to improve the latter’s calcium binding capacity and calcium absorption. Enzymatic hydrolysis methods were screened via ultraviolet-visible absorption spectroscopy (UV–Vis), the fluorescence spectrum, and calcium chelating ability. Phosphorylated water-soluble peptides (P-WSPs) were characterized via high-performance liquid chromatography, the circular dichroism spectrum, Fourier transform infrared spectroscopy (FTIR), UV–Vis spectroscopy, surface hydrophobicity, and fluorescence spectroscopy. The phosphorus content, calcium chelation rate and absorption rate were investigated. The results demonstrated that phosphorylation enhanced the calcium chelating capacity of WSPs, with the highest capacity reaching 0.96 mmol/L. Phosphate ions caused esterification events, and the carboxyl, amino, and phosphate groups of WSPs and P-WSPs interacted with calcium ions to form these bonds. Calcium-chelated phosphorylated water-soluble peptides (P-WSPs-Ca) demonstrated outstanding stability (calcium retention rates > 80%) in gastrointestinal processes. Our study indicates that these chelates have significant potential to develop into calcium supplements with superior efficacy, bioactivity, and stability.
