Preparation and optimization of calcium pectate beads for cell encapsulation

Shan Zhao,Ying Zhang,Yang Liu,Fan Yang,Zhilong Xiu,Xiaojun Ma,Guangwei Sun

A variety of natural polymers are considered suitable for cell entrapment. Pectin, a natural polysaccharide found in the cell walls of all higher terrestrial plants, presents great potential for cell encapsulation because of its ability to form gels. In this study, we investigated the influences of intrinsic and extrinsic factors on the formation and properties of calcium pectate (CP). Beads of CP prepared with 30–35% methyl esterification presented improved gel properties. The molecular weight ( M w ), calcium content, and gel time did not significantly affect the gel properties. The gelling properties, sphericity factor (SF), and swelling ratio were significantly influenced by the degree of methyl esterification (DE) and pectin concentration. The DE (<40%), M w , and calcium concentration had negative effects on the SFs of CP beads. Moreover, increases in the DE (<40%) and pectin concentration increased the swelling ratio. The C3A cells encapsulated in optimized CP beads maintained good viability and proliferation for up to 2 weeks. In conclusion, CP beads are a potential encapsulation material for cell immobilization and application in related fields. © 2017 Wiley Periodicals, Inc. J. Appl. Polym. Sci. 2018 , 135 , 45685.

Optimization of preparation conditions for calcium pectinate with response surface methodology and its application for cell encapsulation

Shan Zhao,Ying Zhang,Yang Liu,Fan Yang,Wenguang Yu,Shixin Zhang,Xiaojun Ma,Guangwei Sun

The most notable and unique property of pectin is its ability to form gels, which is the foundation of its many functions and applications. To obtain the desired pectin hydrogel beads for tissue engineering or biological applications, the combined effect of the key factors of gel properties of calcium pectinate (CP) beads were investigated by response surface methodology (RSM). The results derived from RSM indicated that the model equation of average size and mechanical stability were significant and could be used to describe the process under a wide range of preparation conditions. The optimum condition for preparing variables were gained graphically. Moreover, the degree of methyl-esterification (DE) of pectin and pectin - calcium concentrates showed significant and combined effort on sphericity factor (SF). The mechanical stability of CP beads was significantly affected by the quadratic contributions of the pectin concentration, the interaction effects between pectin and calcium concentration, and the linear contribution of pectin concentration. The viability and proliferation of cells encapsulated in optimal CP beads demonstrated that the optimal formula results in better gel properties and are more suitable to cell encapsulation.

Advanced targeted curcumin delivery using biodegradable hierarchical microspheres with calcium pectinate matrix and hyaluronic acid moieties for enhancing colitis amelioration

Jin Feng,Zhen Wang,Xingyu Zhao,Lujing Xu,Cong Teng,Songbai Liu,Wuyang Huang,Ying Li

Oral targeted delivery systems for food bioactive compounds have attracted considerable attention for improving the efficiency of nutritional interventions. In this study, a hierarchical curcumin carrier with sequence-targeting capability was fabricated to mitigate colitis through a convenient and environmentally friendly approach. Initially, curcumin-loaded nanostructured lipid carriers (NLC) with hyaluronic acid (HA) moieties (HA-NLC) were prepared using ovalbumin-HA conjugates as emulsifiers. Subsequently, HA-NLC was immobilized within a calcium pectinate matrix using electrospray, leading to the formation of supramolecular microspheres (HA-NLC@MPs) approximately 140 μm in size. Results suggested that the pectin matrix preserved the integrity of the carrier and prevented curcumin leakage during transit through the upper gastrointestinal tract, while selectively degrading in response to colon bacteria. Moreover, the exposed HA moieties on the released HA-NLC facilitated the transcellular absorption of curcumin by inflamed colonic enterocytes through cluster of differentiation-44 receptor-mediated endocytosis. In vitro and in vivo studies demonstrated predominant curcumin accumulation in the colorectal tissues of colitis mice using HA-NLC@MPs as carrier. Curcumin delivered via HA-NLC@MPs demonstrated superior effects in alleviating colitis compared with that in the nanocarriers, through the modulation of macrophage polarization, TLR4/MyD88/NF-κB signaling cascade, and gut microbiota homeostasis.

A New Method for Determination of Pectin Content Using Spectrophotometry

Furong Wang,Chao Du,Junjun Chen,Lisheng Shi,Hailong Li

The study aimed at developing a new spectrophotometric method for determining the pectin content. Take commercial pectin as an example, and the method is based on the reaction of copper ions with pectin to produce copper pectate. The spectrophotometer was used to measure the remaining content of copper ions so as to calculate the pectin content. This method eliminated the weight step and avoided the error associated with it. Effects of reaction time, temperature, and pH on absorbance were also studied. Additionally, the accuracy of this method was verified. It indicated excellent repeatability and accuracy with the relative standard deviation of 2.09%. In addition, three different plant types were used to demonstrate the reliability of the method. To summarize, this method can be widely used for the determination of pectin content in many materials. Keywords:copper pectate;calcium pectate;ultraviolet and visible spectrophotometer

Load mechanism and release behaviour of synephrine-loaded calcium pectinate beads: Experiments characterizations, theoretical calculations and mathematical modeling

Peng Zhou,Min Zheng,Xiangzhou Li,Jun Zhou,Wensheng Li,Yanhong Yang

It is an appropriate strategy to construct the carrier material with polysaccharide pectin, which is the characteristics of good bio-compatible, safe and non-toxic, avoiding the functional loss of bioactive ingredients and achieve sustained release . However, the loading mechanism of the active ingredient and the release behaviour of the active ingredient from the carrier material is still at the stage of conjecture. In this study, a kind of synephrine-loaded calcium pectinate beads (SCPB) with high encapsulation efficiency (95.6 %), loading capacity (11.5 %) and excellent controlled release behaviour was constructed. The interaction between synephrine (SYN) and quaternary ammonium fructus aurantii immaturus pectin (QFAIP) was revealed by FTIR, NMR and density functional theory (DFT) calculation. An inter-molecular hydrogen bond and Van der Waals forces between 7-OH, 11-OH and 10-NH of SYN and -OH, -C=O and N + (CH 3 ) 3 of QFAIP were formed. The release experiment in vitro showed that the QFAIP could effectively avoid the release of SYN in gastric fluid, and also realized the slow and full release of SYN in intestinal tract. Moreover, the release mechanism of SCPB in simulated gastric fluid (SGF) was Fickian diffusion, while in simulated intestinal fluid (SIF) was a non-Fickian diffusion controlled by both diffusion and skeleton dissolution.
