Preparation, Characterization and Stability of Calcium-Binding Peptides Derived from Chicken Blood

Jing Yang,Jing Shi,Ying Zhou,Ye Zou,Weimin Xu,Xiudong Xia,Daoying Wang

Calcium-binding peptides have gained significant attention due to their potential applications in various fields. In this study, we aimed to prepare, characterize, and evaluate the stability of calcium-binding peptides derived from chicken blood. Chicken hemoglobin peptides (CPs) were obtained by protease hydrolysis and were applied to prepare chicken hemoglobin peptide–calcium chelate (CP-Ca). The preparation conditions were optimized, and the characteristics and stability of CP-Ca were analyzed. The optimal chelating conditions were determined by single-factor and response surface tests, and the maximum calcium ion chelating rate was 77.54%. Amino acid analysis indicated that glutamic acid and aspartic acid motifs played an important role in the chelation of the calcium ions and CP. According to the characterization analysis, CP-Ca was a different substance compared with CP; calcium ions chelated CPs via the sites of carbonyl oxygen, carboxyl oxygen, and amino nitrogen groups; and after the chelation, the structure changed from a smooth homogeneous plate to compact granular. The stability analysis showed that CP-Ca was stable at different temperatures, pH, and gastrointestinal conditions. The study indicates that chicken blood is a promising source of peptide–calcium chelates, providing a theoretical basis for application in functional foods and improving the utilization value of chicken blood.

Label-free quantification proteomics reveals novel calcium binding proteins in matrix vesicles isolated from mineralizing Saos-2 cells

Xiaoying Zhou,Yazhou Cui,Jing Luan,Xiaoyan Zhou,Genglin Zhang,Xiumei Zhang,Jinxiang Han

Matrix vesicles (MVs) involved in the initiation of mineralization by deposition of hydroxyapatite (HA) in their lumen are released by the budding of mineralization-competent cells during skeletogenesis and bone development. To identify additional mineralization-related proteins, MVs were isolated from non-stimulated and stimulated Saos-2 cells in culture via an ExoquickTM approach and the corresponding proteomes were identified and quantified with label-free quantitative proteome technology. The isolated MVs were confirmed by electron microscopy, alkaline phosphatase activity (ALP), biomarkers, and mineral formation analyses. Label-free quantitative proteome analysis revealed that 19 calcium binding proteins (CaBPs), including Grp94, calnexin, calreticulin, calmodulin, and S100A4/A10, were up-regulated in MVs of Saos-2 cells upon stimulation of mineralization. This result provides new clues to study the mechanism of the initiation of MV-mediated mineralization.

Deciphering the calcium-binding mechanism of a novel cod bone-derived peptide: A synergistic experimental and computational analysis

Qiaoji Tian,Li Hao,Qian Zhang,Huilin Huang,Chaoxin Tu,Chaozhong Fan,Hu Hou

A novel cod bone peptide GETGPA with high calcium chelating capacity (4.60 ± 1.06 μg/mg) was prepared and synthesized to elucidate the binding mechanism of peptide with Ca 2+ . The chelation sites, binding modes, and coordination bond formation mechanism were systematically investigated by experimental methods combined with molecular dynamics simulation and density functional theory calculation. FTIR and Raman spectra showed that the oxygen atoms of carboxyl groups were likely involved in the binding of GETGPA with Ca 2+ . Furthermore, mass spectrometry identified Glu-2 as the primary calcium binding site of GETGPA. NMR indicated that the free carboxyl group of Ala-6 and Glu-2 might be participated in Ca 2+ chelation. There were multiple chelating modes in the system where multiple GETGPA molecules coexisted with Ca 2+ . The coordination bond formed between GETGPA and Ca 2+ exhibited excellent chemical and thermodynamic stability. This study provided new insights into the binding mechanisms of peptides and calcium.

The S100 calcium-binding protein A6 plays a crucial role in hepatic steatosis by mediating lipophagy

Qian Du,Tingting Zhu,Guorong Wen,Hai Jin,Jiaxing An,Jingyu Xu,Rui Xie,Jiaxing Zhu,Xiaoxu Yang,Ting Zhang,Qi Liu,Shun Yao,Xingyue Yang,Biguang Tuo,Xiong Ma

Background: S100 calcium-binding protein A6 (S100A6) is a calcium-binding protein that is involved in a variety of cellular processes, such as proliferation, apoptosis, and the cellular response to various stress stimuli. However, its role in NAFLD and associated metabolic diseases remains uncertain. Methods and Results: In this study, we revealed a new function and mechanism of S100A6 in NAFLD. S100A6 expression was upregulated in human and mouse livers with hepatic steatosis, and the depletion of hepatic S100A6 remarkably inhibited lipid accumulation, insulin resistance, inflammation, and obesity in a high-fat, high-cholesterol (HFHC) diet-induced murine hepatic steatosis model. In vitro mechanistic investigations showed that the depletion of S100A6 in hepatocytes restored lipophagy, suggesting S100A6 inhibition could alleviate HFHC-induced NAFLD. Moreover, S100A6 liver-specific ablation mediated by AAV9 alleviated NAFLD in obese mice. Conclusions: Our study demonstrates that S100A6 functions as a positive regulator of NAFLD, targeting the S100A6-lipophagy axis may be a promising treatment option for NAFLD and associated metabolic diseases.

Simultaneous Unlocking Optoelectronic and Interfacial Properties of C60 for Ultrasensitive Immunosensing by Coupling to Metal–Organic Framework

Qing Zhou,Guanghui Li,Kaiyang Chen,Hong Yang,Mengran Yang,Yuye Zhang,Yakun Wan,Yanfei Shen,Yuanjian Zhang

Due to exceptional electron-accepting ability, light-absorption, and a delocalized conjugated structure, buckminsterfullerene (C60) has attracted fascinating interest in the field of organic solar cells. However, poor delocalization and accumulation of electrons for pristine C60 in physiological aqueous solution and difficulties in conjugation with biomolecules limit its extended photovoltaic applications in bioassay. Herein, we reported the noncovalent coupling of C60 to an electronically complementary porphyrin-derived metal–organic framework (PCN-224) with carboxyl-group terminals. Such assembly not only offered a friendly interface for bioconjugation but also resulted in a long-range ordering C60@PCN-224 donor–acceptor system that demonstrated an unprecedented photocurrent enhancement up to 10 times with respect to each component. As an example, by further cooperating with Nanobodies, the as-prepared C60@PCN-224 was applied to a photoelectrochemical (PEC) immunosensor for S100 calcium-binding protein B with by far the most promising detection activities. This work may open a new venue to unlock the great potential of C60 in PEC biosensing with excellent performances.

Preparation and Characterization of a Sea Cucumber Collagen-Derived Peptide and the Inhibition of Pancreatic Lipase

Zhe Xu,Jiale Chang,Zijin Qin,Hanxiong Liu,Xu Yan,Shuang Guan,Xiufang Dong,Jianping Wu,Tingting Li

The effective modulation of pancreatic lipase activities is critical in maintaining blood triglycerides and cholesterol levels within normal physiological ranges. This study investigated the inhibitory effect of peptides from sea cucumber collagen on pancreatic lipase activity through interaction with the Ca-binding sites on the enzyme. Among the five sea cucumber collagen digests (SCCD), the pepsin-digested 2h+trypsin-digested 3h digest (SDP2h+T3h) showed the strongest inhibitory effect on pancreatic lipase. A peptide RDDPEPSYK (RDD), with a sequence of Arg-Asp-Asp-Pro-Glu-Pro-Ser-Tyr-Lys from SDP2h+T3h, was found to target the Ca-binding site of pancreatic lipase, interacting with certain residues including Asp184, Glu187, Lys163, etc. Additionally, a cell study on 3T3-L1 preadipocytes indicated RDD's capability to inhibit adipocyte differentiation, as well as decrease intracellular triglycerides, total cholesterol, and lipid content. The results from this study suggest that RDD is a targeted pancreatic lipase inhibitor with potential applications for obesity control.

CALCOCO2 prevents AngII-induced atrial remodeling by regulating the interaction between mitophagy and mitochondrial stress

Wanyue Sang,Xiaoji Yan,Lu Wang,Huaxin Sun,Yi Jian,Feifei Wang,Baopeng Tang,Yaodong Li

Background The biological functions of mitochondrial complexes are closely related to the development of atrial fibrillation (AF). Calcium binding and coiled-coil domain 2 (CALCOCO2) is a novel and specific receptor for mitophagy; however, its function in AF remains unknown. Therefore, this study aimed to investigate the role and molecular mechanisms of CALCOCO2 in AF, especially its regulatory mechanism in mitophagy and mitochondrial stress. Methods Mice and HL-1 cells were treated with AngII to establish in vitro and in vivo AF models. Additionally, we examined the effect of CALCOCO2 or DAP3 Binding Cell Death Enhancer 1 (DELE1) overexpression on mitophagy and mitochondrial stress in AF models. To investigate the role of mitophagy in the regulatory effects of CALCOCO2 in AF, HL-1 cells were treated with chloroquine, a mitophagy inhibitor. Moreover, mitochondrial parameters were examined using specific fluorescent probes, transmission electron microscopy, western blotting, immunohistochemistry, and confocal microscopy. Results AngII severely impaired the normal morphology and function of mitochondria; inhibited mitophagy; promoted atrial mitochondrial stress, fibrosis, and oxidative stress; and accelerated the progression of atrial remodeling in atrial myocytes. However, CALCOCO2 overexpression reversed/ameliorated these AF-induced changes. Additionally, CALCOCO2 overexpression restored mitochondrial homeostasis in atrial muscle by activating mitophagy and ameliorating mitochondrial stress. Mechanistically, DELE1 overexpression increased mitochondrial reactive oxygen species level and the expression of mitochondrial stress proteins (HRI, eIF2α, and ATF4) even in CALCOCO2-expressing in vitro AF models.. Conclusions CALCOCO2 may serve as a potential target for AF therapy to prevent or reverse the progression of atrial remodeling by regulating mitophagy and DELE1-mediated mitochondrial stress.

Oligodendroglia-to-pericyte conversion after lipopolysaccharide exposure is gender-dependent

Qingting Yu,Linyuan Zhang,Ting Xu,Jiapeng Shao,Falei Yuan,Zuisu Yang,Yuncheng Wu,Haiyan Lyu

To investigate the sex-dependent differentiation of Sox10 cells and their response to pathological conditions such as lipopolysaccharide (LPS) exposure or ischemia, we utilized Sox10 Cre-ER T2 , tdTomato mice. Tamoxifen administration induced the expression of red fluorescent protein (RFP) in these cells, facilitating their subsequent tracking and analysis after LPS injection and ischemia via immunofluorescence staining. Propidium iodide (PI) was injected to label necrotic cells following LPS administration. We found that the conversion of Sox10 cells to pericytes in female mice was significantly higher than in male mice, especially in those exposed to LPS. After LPS injection, the number of PI + necrotic cells were significantly greater in females than in males. Moreover, RFP + cells did not co-localize with glial fibrillary acidic protein (GFAP) or cluster of differentiation 11b (CD11b). Similarly, after brain ischemia, RFP + cells did not express cluster of differentiation 13 (CD13), neuronal nuclei (NeuN), GFAP, or ionised calcium binding adaptor molecule 1 (Iba-1). These findings indicate that the conversion of Sox10 cells to pericytes following LPS exposure is sex-dependent, with neither male nor female groups showing differentiation into other cell types after LPS exposure or under ischemic conditions. The differences in LPS-induced necrosis of pericytes between sexes may explain the variations in the conversion of Sox10 cells to pericytes in both sexes.

Dihydroartemisinin Modulates Enteric Glial Cell Heterogeneity to Alleviate Colitis

Peishan Qiu,Ying Chang,Xiaoyu Chen,Shaoqi Wang,Haihang Nie,Yuntian Hong,Meng Zhang,Haizhou Wang,Cong Xiao,Yuhua Chen,Lan Liu,Qiu Zhao

The precise mechanism underlying the therapeutic effects of dihydroartemisinin (DHA) in alleviating colitis remains incompletely understood. A strong correlation existed between the elevation of glial fibrillary acidic protein (GFAP) + /S100 calcium binding protein B (S100β) + enteric glial cells (EGCs) in inflamed colonic tissues and the disruption of the intestinal epithelial barrier (IEB) and gut vascular barrier (GVB) observed in chronic colitis. DHA demonstrated efficacy in restoring the functionality of the dual gut barrier while concurrently attenuating intestinal inflammation. Mechanistically, DHA inhibited the transformation of GFAP + EGCs into GFAP + /S100β + EGCs while promoting the differentiation of GFAP + /S100β + EGCs back into GFAP + EGCs. Furthermore, DHA induced apoptosis in GFAP + /S100β + EGCs by inducing cell cycle arrest at the G0/G1 phase. The initial mechanism is further validated that DHA regulates EGC heterogeneity by improving dysbiosis in colitis. These findings underscore the multifaceted therapeutic potential of DHA in ameliorating colitis by improving dysbiosis, modulating EGC heterogeneity, and preserving gut barrier integrity, thus offering promising avenues for novel therapeutic strategies for inflammatory bowel diseases.

Enhancement of Calcium Chelating Activity in Peptides from Sea Cucumber Ovum through Phosphorylation Modification

Lingyu Han,Yaoyao Li,Bing Hu,Wei Wang,Jianming Guo,Jixin Yang,Nuo Dong,Yingmei Li,Tingting Li

Recently, phosphorylation has been applied to peptides to enhance their physiological activity, taking advantage of its modification benefits and the extensive study of functional peptides. In this study, water-soluble peptides (WSPs) of sea cucumber ovum were phosphorylated in order to improve the latter’s calcium binding capacity and calcium absorption. Enzymatic hydrolysis methods were screened via ultraviolet-visible absorption spectroscopy (UV–Vis), the fluorescence spectrum, and calcium chelating ability. Phosphorylated water-soluble peptides (P-WSPs) were characterized via high-performance liquid chromatography, the circular dichroism spectrum, Fourier transform infrared spectroscopy (FTIR), UV–Vis spectroscopy, surface hydrophobicity, and fluorescence spectroscopy. The phosphorus content, calcium chelation rate and absorption rate were investigated. The results demonstrated that phosphorylation enhanced the calcium chelating capacity of WSPs, with the highest capacity reaching 0.96 mmol/L. Phosphate ions caused esterification events, and the carboxyl, amino, and phosphate groups of WSPs and P-WSPs interacted with calcium ions to form these bonds. Calcium-chelated phosphorylated water-soluble peptides (P-WSPs-Ca) demonstrated outstanding stability (calcium retention rates > 80%) in gastrointestinal processes. Our study indicates that these chelates have significant potential to develop into calcium supplements with superior efficacy, bioactivity, and stability.
