Enhanced Hydroxyapatite Growth and Osteogenic Activity on Polydopamine Coated Ti Implants

Xiao Xiang,Xu Yao,Fu Jijiang,Gao Biao,Huo Kaifu,Chu Paul K.

Titanium (Ti) and its alloy are widely used in biomedical implant but the poor bioactivity of Ti restricts some direct applications. Polydopamine (PDA), a functional polymeric substance mimicking the mussel adhesive protein, is deposited on Ti surface to improve the bioactivity by means of a mild and facile technique employing an alkaline dopamine solution. The PDA coated Ti implant accelerates nucleation and growth of hydroxyapatite (HAP), enhances osteoblast cell adhesion and proliferation, and improves the ALP activity thus having promising biomedical applications.

A novel dual-functional coating based on curcumin/APEG polymer with antibacterial and antifouling properties

Limin Qu,Xiangzhou Li,Jun Zhou,Kairui Cao,Qiuen Xie,Peng Zhou,Wei Qian,Yanhong Yang

Infection associated with implants and devices is a problem in medicine, especially when bacterial colonization leads to the formation of biofilms on the implant surface. Here, we report an effective strategy to develop dual-functional coatings on titanium implants with antibacterial and antifouling properties to combat biofilm infections. Coating monomers with antibacterial and antifouling blocks were prepared by conjugating curcumin-derived curcumin carboxylic acid (Cur-COOH) with different chain lengths of allyl polyethylene glycol (APEG; MW 1200 or 2400). The coating monomer (APEG 1200/2400 -Cur) was grafted onto the surface of the titanium implant through azobisisobutyronitrile (ABIN)-induced radical polymerization to form dual-functional coatings with antibacterial and antifouling properties. Scanning electron microscopy & energy dispersive spectroscopy (SEM & EDS), contact angle (CA), and X-ray spectroscopy (XPS) confirmed successful grafting of coupling compounds. The coatings exhibited excellent antibacterial activity against Gram-negative bacteria ( Escherichia coli ) and Gram-positive bacteria ( Staphylococcus aureus ) and prevented biofilm formation. The coatings also reduced the adsorption of proteins and the adhesion of platelets, indicating their excellent antifouling properties. Notably, the coatings also exhibited favorable biocompatibility with mammalian erythrocytes and pre-osteoblasts. In summary, this dual-functional coating showed remarkable potential for application in preventing and treating infections caused by biofilms.

Nitric Oxide-Mediated Dual-Functional Smart Titanium Implant Coating for Antibacterial and Osseointegration Promotion in Implant-Associated Infections

Zishuo Hou,Kun Wang,Guming Liu,Zhang Yuan,Haowei Peng,Yue Yuan,Hongbo Wei,Tengjiao Wang,Peng Li

The balance of antibacterial and osseointegration-promoting properties on titanium (Ti) implants in a simple and efficient manner is crucial for the management of implant-associated infections, a condition that has become a significant global health concern. Herein, a nitric oxide (NO)-mediated dual-function smart coating with antibacterial and osseointegration-promoting properties is developed for Ti implants. The coating leverages the distinct properties of NO at high and low concentrations to enable an on-demand functional switch. Specifically, antibacterial function is achieved through a rapid release of high-dose NO in response to the infection microenvironment and near-infrared stimulation. Once the infection is resolved and normal physiological conditions are restored, the coating gradually releases low-dose NO to promote osseointegration. In vitro tests confirm that the coating exhibits antibacterial ratio of 97.84% and 97.18% against methicillin-resistant Staphylococcus aureus and its biofilms, respectively, and demonstrates the ability to activate osteoblasts. The rat femoral implant-associated infection model further certifies that the responsive NO release mechanism of the coating efficiently facilitates the on-demand functional switch between antibacterial and osseointegration-promoting properties. Notably, the use of the dual-functional nonantibiotic agent, NO, significantly mitigates the risk of bacterial resistance.

Straddle-type heterostructure films endow titanium implant with NIR photocatalysis property for rapid sterilization

Shiwei Guan,Zhenhao Hou,Ji Tan,Xianming Zhang,Junyu Liu,Huihui Du,Hongqin Zhu,Yuqin Qiao,Zixiao Liu,Xuanyong Liu

Near-infrared (NIR) light-triggered photocatalysis antibacterial therapy has attracted significant attention. Herein, we designed a straddle-type heterostructure film of ZnS/Ag 2 S nanorod on the titanium implant, which showed excellent NIR photocatalytic ability. Under NIR, the electrons of Ag 2 S cascaded jump to ZnS conduction band (CB), and the electron of ZnS was half-jumped to Ag 2 S valence band (VB). The VB of Ag 2 S acted as a consumption platform for the holes of Ag 2 S and the electrons of ZnS, improving photocatalytic efficiency. To prove the feasibility of this mechanism, another straddle-type heterostructure of ZnS/Bi 2 S 3 with similar energy band structure was fabricated, and its NIR photocatalytic ability was also distinctly enhanced. Furthermore, the ZnS/Ag 2 S heterostructure films rapidly removed bacteria in vitro and in vivo under the NIR irradiation. This work innovatively discovered NIR photocatalytic effect of the straddle-type heterostructure, which complemented the design of heterostructures in photocatalysis.

Fungal Component Coating Enhances Titanium Implant-Bone Integration

Yuchen Shi,Lintao Wang,Yiming Niu,Na Yu,Panfei Xing,Lei Dong,Chunming Wang

Bioactive coating on bone and dental implants is widely used to increase their integration with the host tissue, with millions of implantations performed globally each year. Numerous coating strategies focus on creating a cell-adhesive surface by employing bone matrix components, but insufficient integration and its associated complications remain a substantial clinical challenge. Designing an implant coating to more comprehensively promote healing while appropriately modulating host immune response is still an unfulfilled promise. This study reports the development of a polysaccharide coating that can improve osseointegration by targeting and regulating inflammatory activity at the implant surface. Zymosan—a fungal component—is screened and covalently grafted onto titanium (Ti) substrates. The zymosan coating specifically stimulates macrophages through toll-like receptors into a proregenerative phenotype, which produces abundant osteogenic/angiogenic cytokines to enhance osteogenic activities locally. Further assessment in a rat femur condyle defect indicates that zymosan coated at a medium dose (0.1 µg mm −2 ) remarkably increases the integration of Ti column with bone tissue, causing no abnormality to the host. This is the first demonstration that modulating inflammation with an immunoactive coating material can promote bone-implant integration, which may inspire new strategies for the design of bone and dental implants.

Biomimetic Self-Assembling Metal–Organic Architectures with Non-Iridescent Structural Coloration for Synergetic Antibacterial and Osteogenic Activity of Implants

Chunlei Zhang,Guangyu Chu,Zesong Ruan,Ning Tang,Cunfeng Song,Qichao Li,Wenjie Zhou,Jiale Jin,Hossam Haick,Yunfeng Chen,Daxiang Cui

Materials in nature feature versatile and programmable interactions to render macroscopic architectures with multiscale structural arrangements. By rationally combining metal–carboxylate and metal–organophosphate coordination interactions, Au25(MHA)18 (MHA, 6-mercaptohexanoic acid) nanocluster self-assembled structural color coating films and phytic acid (PA)–metal coordination complexes are sequentially constructed on the surface of titanium implants. The Lewis acid–base coordination principle applies for these metal–organic coordination networks. The isotropic arrangement of nanoclusters with a short-range order is investigated via grazing incidence wide-angle X-ray scattering. The integration of robust M–O (M = Ti, Zr, Hf) and labile Cu–O coordination bonds with high connectivity of Au25(MHA)18 nanoclusters enables these artificial photonic structures to achieve a combination of mechanical stability and bacteriostatic activity. Moreover, the colorless and transparent PA–metal complex layer allows the viewing of the structural color and surface wettability switching to hydrophilic and makes feasible the interfacial biomineralization of hydroxyapatite. Collectively, these modular metal–organic coordination-driven assemblies are predictive and rational material design strategies with tunable hierarchy and diversity. The complete metal–organic architectures will not only help improve the physicochemical properties of the bone–implant interface with synergistic antibacterial and osseointegration activities but also can boost surface engineering of medical metal implants

In situ fabrication of NIR-II responsive TiO2 bio-metasurface for photothermal antibacterial and enhanced osseointegration

Fan Xiao,Chen-xiao Huang,Jun-hao Dai,Jun-hui Ye,Kang-jie Cheng,Xu Xu,Li-quan Deng,Jia You,Wei Wang,Wan-bing Jin

Titanium implants, widely used in orthopedic and dental applications, often face challenges due to inadequate osseointegration and the risk of postoperative infections , which can potentially lead to implant failure. To address this issue, we developed a titanium implant with an in-situ fabricated anatase TiO 2 metasurface , characterized by a subwavelength thickness and a preferential orientation of the (001) facets. This metasurface, which consists of uniformly distributed nano-crystallites of approximately 30 nm in size, aims to enhance both osseointegration and antibacterial properties. The addition of isopropanol (IPA) to the precursor solution used in the fabrication process played a crucial role in controlling the grain size and coating thickness of the metasurface. This process led to a redshift of the light absorption peak into the near-infrared (NIR)-II region, thereby increasing the penetration depth of photothermal therapy . The implants with TiO 2 metasurface exhibited increased hydrophilicity, rapid apatite formation in simulated body fluid, and an effective photothermal response under NIR-II light, showing significant antibacterial activity against E. coli and S. aureus . In vivo experiments in rabbits indicated superior bone regeneration and osseointegration around the implants with TiO 2 metasurface compared to the control titanium implants. At 4 weeks postoperatively, the bone volume to tissue volume (BV/TV) ratio within the implant was 53.94 %, and by 8 weeks, the implants were nearly entirely filled with new bone, with a BV/TV ratio of approximately 70 %. The findings suggest that the TiO 2 metasurface, which combines antibacterial and bioactive features, has the potential to reduce the risk of postoperative infections and improve clinical outcomes.

Construction of Ti implant-based responsive OPG release platform for enhanced osteoporotic bone regeneration via programmed regulation of osteogenesis and bone resorption

Kaiwen Yin,Zheng Liu,Chenyu Liao,Jiawei Li,Hao Wang,Kunhua Yang,Li Zhang,Yunfeng Li

The potential for persistent and controllable delivery of osteoprotegerin (OPG) under osteoporotic conditions has become an essential property of bone implants to promote osseointegration and osteogenesis. Herein, a microenvironment-responsive surface coating for titanium (Ti) implants by immobilizing metal-organic frameworks (MOFs) loaded with OPG onto alkali heat-treated Ti surfaces via gelatin (Gel) (AHT-MOF@OPG-GEL) was successfully fabricated. In weakly acidic environment, MOF@OPG can achieve responsive degradation and slow release of zinc ions (Zn2+) and OPG, which not only inhibits osteoclast activity and osteoclastic biomarker expressions, but also potentiates osteoblast proliferation and differentiation in vitro. Moreover, alkali heat treatment was favorable for the hydrophilicity and cell adhesion of the Ti surface. In addition, in vivo experiments in the osteoporotic bone defect model confirmed that the MOF-based functional coating could promote the generation of new bone around the implant, thereby enhancing the bonding strength between the implant and the surrounding bone tissue, and effectively facilitating the osseointegration of the implant with the bone tissue. In conclusion, this study is expected to provide new ideas for improving the osseointegration of implants in osteoporotic conditions.

Microarc Oxidation Coatings Doped with a Low Proportion of Yttrium Enhance the Osseointegration of Titanium Implants through the BMP/Smad Pathway

Chenyang Zhang,Chenghui Qian,Guang Yang,Yiying Zhu,Binbin Kang,Xiaohong Chen,Si Chen

Adding metal ions is a promising strategy to enhance the biological performance of titanium implants. In this study, we aimed to explore the effects of yttrium on the osseointegration of titanium implants. First, a series of yttrium-doped titanium surfaces were fabricated via microarc oxidation (MAO) by incorporating yttrium acetate into the electrolyte, and then the surface characteristics of different substrates were evaluated. Subsequently, the cellular behaviors of different coatings were assessed, and the osteointegration effects were examined using a rat model. Finally, high-throughput sequencing was employed to elucidate the underlying mechanisms of the yttrium-doped MAO coatings. As the results indicated, the proportion of yttrium in the coatings increased as the concentration of yttrium acetate improved. Surface characterization revealed that the yttrium-doped MAO coatings exhibited a homogeneous porous morphology, with comparable roughness and wettability to those of the undoped MAO coating, while the morphology became inconsistent when the yttrium acetate concentration reached 30 mM. The in vitro assays demonstrated that the addition of yttrium notably improved the cell adhesion, spreading, proliferation, and osteogenic differentiation of MAO coatings when doped with a low proportion, accompanied by enhanced osseointegration according to the in vivo experiments. Further exploration revealed a significant enrichment of osseointegration-related signaling factors and the activation of BMP/Smad signaling in the effects of yttrium-doped titanium coatings, which was attributed to the excessive accumulation of phosphorylated Smad1/5/9 in the nucleus. In summary, our work demonstrates that the use of MAO coatings doped with a low proportion of yttrium can enhance the osseointegration of titanium implants, providing an efficient strategy to optimize titanium implant performance.

Electro-deposition of Nd3+-doped metal-organic frameworks on titanium dioxide nanotube array coated by hydroxyapatite for anti-microbial and anticorrosive implant

Zhang Ziqi,Zhang Yan,Liu Yang,Zhang Siqi,Yao Kaida,Sun Yueqiu,Wang Xiangzhi,Huang Weimin,Liu Xiaobo

Metal-organic frameworks are well-aligned nanoporous crystals which can be applied to biomedical field, thanks to the large superficial area, ordered pores array, and biocompatible metallic ion composition. In this work, we fabricated a novel Nd 3+ ion-doped ZIF-8 since there are some reports showing that Nd 3+ has anti-carcinogenic properties and can be used for therapeutic application in aberrant angiogenesis recently. Therefore, we attempted to use electrochemical anodic oxidation and electrochemical in situ deposition techniques to produce coating which contains Nd 3+ on Ti implant. And then we detected the biotoxicity, antibacterial function, and corrosion resistance of this new coating so as to facilitate further research on Nd 3+ ion implantation in vivo. SEM and TEM were applied to observe the surface morphology. N 2 adsorption–desorption was used to investigate the specific surface area and pore structure. The XRD and XPS were carried out to investigate the compound surface including the chemical composition and element-bonding configurations. The corrosion resistance properties of the samples were tested by electrochemical method. ICP was conducted to investigate the release behavior.

Synthesis of rare earth doped MOF base coating on TiO2 nanotubes arrays by electrochemical method using as antibacterial implant material

Ziqi Zhang,Yan Zhang,Siqi Zhang,Kaida Yao,Yueqiu Sun,Yang Liu,Xiangzhi Wang,Weimin Huang

Although titanium (Ti) and its alloys are the most widely used biomedical implant materials in the world today due to their satisfactory mechanical strength, high corrosion resistance and neglecting cellular toxicity, implant failures still occur because of poor osseointegration and bacterial infection. Thus in this paper we synthesized an new Ce doped MIL metal-orgainc framework derivant and used it as an anti-bacterial and anti-corrosion additive coating on titanium with hydroxyapatite by electrochemical in situ deposition technique. The experimental results demonstrated that the MOF-hydroxyapatite composite coating can increase the corrosion resistance and antibacterial property of titanium without destroying its original biocompatibility.

Enhanced mechanical properties of porous titanium implants via in-situ synthesized titanium carbide in lamellar pore walls

Yufei Tang,Xuan Zhou,Qi Zhang,Lei Chen,Kang Zhao,Zixiang Wu

Directional lamellar porous titanium scaffolds are widely used as bone implant bearing materials because of their anisotropic pore structure . Their mechanical properties can be effectively improved by enhancing the strength of pore walls through the introduction of ceramics. In this work, porous titanium implants were prepared by freeze casting combined with TiH 2 decomposition. The graphene was introduced into the pore walls of porous titanium, which could transform into titanium carbide (TiC) in situ upon sintering. TiC was evenly distributed in the lamellar pore walls, and the interface was well bonded. The compression strength of the fabricated implants was up to 389.94 MPa when the graphene content was 3 wt%, which was 377.8% times as high as the porous titanium. The crack propagation was resisted by TiC because of the “pinning” effect on the pore wall. Some of TiC were pulled out from the matrix, and others were fractured. The strength of the fabricated implants was improved significantly by the large consumption of fracture energy. Also, fabricated porous titanium implants with TiC are suitable for bone implantation.
