Cadmium absorption and translocation of amaranth (Amaranthus mangostanus L.) affected by iron deficiency

Rong Zou,Li Wang,Yuncong C. Li,Zhaohui Tong,Wenmin Huo,Keyu Chi,Hongli Fan

Amaranth ( Amaranthus mangostanus L.) has superior capability for accumulating cadmium (Cd) and has the potential to be used for phytoremediation of Cd contaminated soils. Iron (Fe) is chemically similar to Cd and may mediate Cd-induced physiological or metabolic impacts in plants. The purpose was to investigate the model of time-dependent and concentration-dependent kinetics of Cd absorption under Fe deficiency, understanding the physiological mechanism of Cd absorption in amaranth roots. The kinetic characteristics of Cd uptake by amaranth grown in Cd enriched nutritional solution with or without Fe addition and with methanol-chloroform, carbonyl cyanide 3-chlorophenylhydrazone (CCCP), and lanthanum chloride (LaCl 3 ) were compared using 109 Cd 2+ isotope labeling technique. The results showed that Cd uptake was time-dependent and about 90–93% of uptake occurred during the first 150 min. The kinetics of Cd uptake showed that two stages were involved. The saturation stage fitted the Michaelis-Menten model when concentrations of Cd were lower than 12.71 μmol/L and then the absorption of Cd by roots was increased linearly during the second stage. Only linear absorption was observed with methanol-chloroform treatment while the metabolic inhibitor CCCP inhibited only the saturation absorption process, and the Ca channel inhibitor LaCl 3 partially inhibited the two stages of absorption. These results indicated that the root absorption of 109 Cd 2+ was enhanced under Fe deficiency which induced more Fe transporters in the root cell membrane, and the Ca channel, apoplastic and symplastic pathways enhanced the Cd absorption in roots.

Elevation of NO production increases Fe immobilization in the Fe-deficiency roots apoplast by decreasing pectin methylation of cell wall

Ye Yi Quan,Jin Chong Wei,Fan Shi Kai,Mao Qian Qian,Sun Cheng Liang,Yu Yan,Lin Xian Yong

Cell wall is the major component of root apoplast which is the main reservoir for iron in roots, while nitric oxide (NO) is involved in regulating the synthesis of cell wall. However, whether such regulation could influence the reutilization of iron stored in root apoplast remains unclear. In this study, we observed that iron deficiency elevated NO level in tomato ( Solanum lycopersicum ) roots. However, application of S-nitrosoglutathione, a NO donor, significantly enhanced iron retention in root apoplast of iron-deficient plants, accompanied with a decrease of iron level in xylem sap. Consequently, S-nitrosoglutathione treatment increased iron concentration in roots, but decreased it in shoots. The opposite was true for the NO scavenging treatment with 2-(4-carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide (cPTIO). Interestingly, S-nitrosoglutathione treatment increased pectin methylesterase activity and decreased degree of pectin methylation in root cell wall of both iron-deficient and iron-sufficient plants, which led to an increased iron retention in pectin fraction, thus increasing the binding capacity of iron to the extracted cell wall. Altogether, these results suggested that iron-deficiency-induced elevation of NO increases iron immobilization in root apoplast by decreasing pectin methylation in cell wall.

Low nitrate alleviates iron deficiency by regulating iron homeostasis in apple

Wei-Jian Sun,Jiu-Cheng Zhang,Xing-Long Ji,Zi-Quan Feng,Xun Wang,Wen-Jing Huang,Chun-Xiang You,Xiao-Fei Wang,Yu-Jin Hao

Iron (Fe) is an essential element for plant growth, development and metabolism. Due to its lack of solubility and low bioavailability in soil, Fe levels are usually far below the optimum amount for most plants' growth and development. In apple production, excessive use of nitrogen fertilizer may cause iron chlorosis symptoms in the newly growing leaves, but the regulatory mechanisms underlying this phenomenon are unclear. In this study, low nitrate (NO 3 − , LN) application alleviated the symptoms of Fe deficiency and promoted lower rhizosphere pH, which was beneficial for root Fe acquisition. At the same time, LN treatment increased citrate and abscisic acid accumulation in roots, which promoted Fe transport from root to shoot and maintained Fe homeostasis. Moreover, qRT-PCR analysis showed that nitrate application caused differential expression of genes related to Fe uptake and transport, as well as transcriptional regulators. In summary, our data reveal that low nitrate alleviated Fe deficiency through multiple pathways, demonstrating a new option for minimizing Fe deficiency by regulating the balance between nutrients.

Effect of iron deficiency on microstructure and magnetic properties of high dielectric constant YIG

Bixian Zhang,Kai Yang,Sihao Jian,Bei Ren,Ziqian Meng,Yinghui Tang,Yang Miao

With the rapid advancement of communication technologies toward higher frequencies and miniaturization, microwave devices are imposing increasingly stringent performance requirements on fundamental magnetic materials. Optimizing the saturation magnetization and dielectric constant of ferrite materials can enhance the transmission efficiency and frequency response of microwave devices, thereby meeting the evolving technical requirements. In this study, we synthesized a series of yttrium iron garnet ferrites with the chemical composition Bi 1.4 Ca 0.3 Y 1.3 Zr 0.15 Sn 0.15 In 0.2 Fe 4.5-x O 12-1.5x (0 ≤ x ≤ 0.3) using the conventional solid-state reaction method. The effects of iron deficiency on the structure and electromagnetic properties of these ferrites were systematically investigated. The results demonstrate that controlled iron deficiency effectively enhances saturation magnetization (4πMs) while reducing dielectric loss (tanδ), without altering the garnet phase structure or dielectric constant (ε'). However, excessive iron deficiency induces the formation of a secondary YFeO 3 phase, severely degrading material properties. The sample with x = 0.2 exhibits outstanding comprehensive performance, with a high saturation magnetization (1903 Gs), an elevated dielectric constant (25.2), and a low dielectric loss (4.67 × 10 -3 ). These high-performance ferrite materials show great potential for applications in next-generation microwave devices.

Novel Flaxseed Gum Nanocomposites Are Slow Release Iron Supplements

Shan Liang,Yu Huang,Youn Young Shim,Xiang Ma,Martin J. T. Reaney,Yong Wang

Nanocomposites, based on iron salts and soluble flaxseed gum (FG), were prepared as potential treatments of iron deficiency anemia (IDA). FG was extracted, characterized, and formulated into iron-loading nanocomposites via ion-exchange against FeCl3, Fe2(SO4)3, FeCl2, and FeSO4·7H2O. FG-iron nanocomposites preparation condition was optimized, and physicochemical properties of the nanocomposites were investigated. In vitro release kinetics of iron in simulated gastric fluid (SGF) was also evaluated. FG heteropolysaccharide, consisting of rhamnose (33.73%), arabinose (24.35%), xylose (14.23%), glucose (4.54%), and galactose (23.15%) monosaccharides, linked together via varieties of glycosidic bonds, was a good recipient for both ferric and ferrous irons under screened conditions (i.e., 80 °C, 2 h, I/G = 1:2). Iron loaded contents in the nanocomposites prepared from FG-FeCl3, FG-Fe2(SO4)3, FG-FeCl2, and FG-FeSO4·7H2O were 25.51%, 10.36%, 5.83%, and 22.83%, respectively. Iron in these nanocomposites was mostly in a bound state, especially in FG-FeCl3, due to chelation forming bonds between iron and polysaccharide hydroxyl or carboxyl groups and formed stable polysaccharide-iron crystal network structures. Free iron ions were effectively removed by ethanol treatments. Because of chelation, the nanocomposites delayed iron release in SGF and the release kinetics were consistent with Korsmeyer-Peppas model. This indicates that such complexes might reduce side effects of free iron in human stomach. Altogether, this study indicates that these synthetic FG-iron nanocomposites might be developed as novel iron supplements for iron deficiency, in which FG-FeCl3 is considered as the best option.

Enhanced Bioavailability of Ferric Pyrophosphate Delivery System Constructed with β-Glucan and Casein Phosphopeptide

Yiqiao Pei,Ye Zhang,Fang Tian,Yanrong Zhao,Xuguang Zhang,Steve W. Cui,Huali Wang,Jianbo Zhang,Hao Wang

Iron supplementation has been an important and complicated health topic. Available iron preparations generally have issues such as instability, inadequate absorption, and low consumer acceptability. This study provided a novel ferric pyrophosphate (FePP) delivery system, the FePP-casein-phosphopeptide-dextran (FCD) complex, with sodium pyrophosphate (NaPP) as a solvent promoter and casein-phosphopeptide (CPP) and β-glucan as wall materials. Compared to FePP, FCD showed better properties, including good water solubility (95% resolubilization), color stability (pH 3.5–6.5), compatibility with sensitive nutrients like vitamin C (VC), and lipid oxidation stability (delayed by approximately 50%). In vitro, FCD demonstrated gradual iron release in the gut with a release rate exceeding 80%. In vivo, FCD effectively alleviated iron deficiency anemia (IDA) symptoms in mice, with superior overall efficacy to FePP. Therefore, this novel FCD iron delivery system had considerable potential to be applied in nutritional food systems or iron supplements.

Mammalian SLC39A13 promotes ER/Golgi iron transport and iron homeostasis in multiple compartments

Li Huihui,Cui Yanmei,Hu Yule,Zhao Mengran,Li Kuanyu,Pang Xiaoyun,Sun Fei,Zhou Bing

Iron is a potent biochemical, and accurate homeostatic control is orchestrated by a network of interacting players at multiple levels. Although our understanding of organismal iron homeostasis has advanced, intracellular iron homeostasis is poorly understood, including coordination between organelles and iron export into the ER/Golgi. Here, we show that SLC39A13 (ZIP13), previously identified as a zinc transporter, promotes intracellular iron transport and reduces intracellular iron levels. ZIP13 loss causes an iron deficiency in the ER/Golgi and other intracellular compartments, such as lysosomes and mitochondria, as well as elevating iron in the cytosol. ZIP13 overexpression has the opposite effect, increasing iron in organellar compartments. We suggest that ZIP13 gatekeeps an iron trafficking route that shunts iron from the cytosol to the ER/Golgi hub. Zip13- knockout male mice have iron deposition in several tissues. These data demonstrate that mammalian ZIP13 is crucial for iron homeostasis and suggest a potential iron transport function.

Multifunctional Iron–Humic Acid Fertilizer from Ball Milling Double-Shelled Fe–N-doped Hollow Mesoporous Carbon Microspheres with Lignite

Yafu Tang,Yuechao Yang,Shanmin Hou,Dongdong Cheng,Yuanyuan Yao,Shugang Zhang,Jiazhuo Xie,Xiaoqi Wang,Xiaoxiao Ma,Zhen Yu,Shan Li

Iron (Fe) chelated by water-soluble humic substances can effectively overcome plant micronutrient deficiencies. However, the traditional synthesis methods are often complex, expensive, and thus not applicable to large-scale industrial productions. In this work, an ecofriendly and multifunctional Fe–humic acid (HA) complex was successfully prepared through simply ball milling lignite with double-shelled Fe–N hollow mesoporous carbon microspheres (DS–Fe–N–HC). The novel DS–Fe–N–HC was synthesized using a simple strategy and was first applied in activating HA from lignite by solid-phase ball milling to attain the water-soluble HA–iron fertilizer (WHIF). The physicochemical properties of the DS–Fe–N–HC nanocomposites and WHIF were systematically characterized by a series of techniques. A traditional Fe–HA complex is mainly obtained by mixing HA and iron ions, which has poor water solubility because HA is not activated. In comparison to samples prepared with other methods, WHIF contained more stable water-soluble HA. Furthermore, the as-obtained WHIF had higher amounts of proteins and amino sugar tannin as evidenced in FT-ICR MS analysis. Moreover, WHIF contributed to higher activities in chlorophyll content, height, steam diameter, and biomass of peanuts than other samples. The results demonstrate that WHIF is effective in reducing symptoms of iron deficiency in peanuts, which can be attributed to its iron-based compounds and water-soluble HA. The findings of this study provide a promising, cost-effective, and environmentally friendly solution to mitigate iron deficiency and promote the growth of peanuts for large-scale agriculture production.

Preparation of iron dextran complex and its effect on iron deficiency anemia in rats

Yuying He,Biyang Zhu,Huamai Qiu,Lijun You,Lianzhu Lin,Sandra Pedisić

Iron dextran complex is commonly used to treat iron deficiency anemia (IDA). In this work, dextran was degraded and oxidized by UV/H 2 O 2 treatment, and the product Dex-90 was chelated with FeCl 3 to prepare the iron dextran complex (Dex-90-Fe). Results showed that UV/H 2 O 2 treatment could significantly decrease the molecular weight of dextran and increase its carboxyl content, making it easier to chelate with iron (III). Moreover, Dex-90-Fe had a high iron content (30.77 % ± 0.87 %) and good thermal stability, which might be formed by Dex-90 with iron (III) via Fe O bond. Furthermore, the therapeutic effects of Dex-90-Fe on IDA were investigated in rats. After oral administration, Dex-90-Fe significantly improved hematological parameters, increased serum iron and serum ferritin, facilitated iron accumulation in the liver and spleen, and regulated the gene expressions of iron homeostasis. In summary, the iron dextran complex Dex-90-Fe prepared from UV/H 2 O 2 -degraded dextran could be developed as an iron supplement.
