Iron deficiency is the utmost critical problem of nutrition worldwide. Polysaccharide-iron chelate is a promising iron supplementation that exhibits good stability, high absorption rate, and fewer adverse reactions, but it still owns drawbacks of extra energy and lower iron content with respect to inorganic ones. Carboxylated polysaccharide-iron has been proven with enhanced iron content, however with poor aqueous solubility. Here, Fe 3+ concentrations controlled highly soluble carboxylated inulin-iron chelates were synthesized to promote the sustained release and efficient supplement of iron with no extra energy. Inulin, soluble dietary fiber, was initially reacted with succinic anhydride (SA) to get carboxylated inulin (IN-SA) consequently enhancing the complexing ability of Fe 3+ and its bioavailability. Interestingly, the aqueous solubility of the IN-SA-iron chelate (IN-SA-Fe) directly correlated with the Fe 3+ concentrations. Relatively higher or lower Fe 3+ concentrations resulted in highly soluble IN-SA-Fe, otherwise, leading to insolubility precipitates. The possible mechanism was proposed as the polymerization reaction of IN-SA in different degrees with Fe 3+ acting as the cross-linker. Moreover, IN-SA-Fe was demonstrated with properties of sustained slow-release of iron in artificial gastric juice and good in vivo biosafety. This work provides a simple, efficient, no extra energy soluble iron supplement for the treatment of iron deficiency.

Nanocomposites, based on iron salts and soluble flaxseed gum (FG), were prepared as potential treatments of iron deficiency anemia (IDA). FG was extracted, characterized, and formulated into iron-loading nanocomposites via ion-exchange against FeCl3, Fe2(SO4)3, FeCl2, and FeSO4·7H2O. FG-iron nanocomposites preparation condition was optimized, and physicochemical properties of the nanocomposites were investigated. In vitro release kinetics of iron in simulated gastric fluid (SGF) was also evaluated. FG heteropolysaccharide, consisting of rhamnose (33.73%), arabinose (24.35%), xylose (14.23%), glucose (4.54%), and galactose (23.15%) monosaccharides, linked together via varieties of glycosidic bonds, was a good recipient for both ferric and ferrous irons under screened conditions (i.e., 80 °C, 2 h, I/G = 1:2). Iron loaded contents in the nanocomposites prepared from FG-FeCl3, FG-Fe2(SO4)3, FG-FeCl2, and FG-FeSO4·7H2O were 25.51%, 10.36%, 5.83%, and 22.83%, respectively. Iron in these nanocomposites was mostly in a bound state, especially in FG-FeCl3, due to chelation forming bonds between iron and polysaccharide hydroxyl or carboxyl groups and formed stable polysaccharide-iron crystal network structures. Free iron ions were effectively removed by ethanol treatments. Because of chelation, the nanocomposites delayed iron release in SGF and the release kinetics were consistent with Korsmeyer-Peppas model. This indicates that such complexes might reduce side effects of free iron in human stomach. Altogether, this study indicates that these synthetic FG-iron nanocomposites might be developed as novel iron supplements for iron deficiency, in which FG-FeCl3 is considered as the best option.

Nanozyme has been proven to be an attractive and promising candidate to alleviate the current pressing medical problems. However, the unknown clinical safety and limited function beyond the catalysis of the most reported nanozymes cannot promise an ideal therapeutic outcome in further clinical application. Herein, we find that ferric maltol (FM), a clinically approved iron supplement synthesized through a facile scalable method, exhibits excellent peroxidase-like activity than natural horseradish peroxidase-like (HRP) and commonly reported Fe-based nanozymes, and also shows high antibacterial performance for methicillin-resistant Staphylococcus aureus (MRSA) elimination (100%) and wound disinfection. In addition, with added effects inherited from contained maltol, FM can accelerate skin barrier recovery. Therefore, the exploration of FM as a safe and desired nanozyme provides a timely alternative to current antibiotic therapy against drug-resistant bacteria.

Grifola frondosa protein was hydrolysed using Alcalase, and the resulting hydrolysates (CGFP) were filtered using 5 and 1 kDa nominal cut-off ultrafiltration membranes and separated using Sephadex gel chromatography . Two main fractions, named GFP-1 and GFP-2 were obtained. CGFP-Fe and GFP-Fe were synthesized using ferrous chelation. Results showed that the GFP-2 fraction with molecular weight of 963 Da had the highest Fe(II) chelating activity (2680 ± 40 μg/g). UV spectrum showed that the Fe(II) shifted the maximum absorption to shorter wavelengths. The peptides measured using FTIR showed that the GFP-Fe peptide and the un-chelated peptide had different C–O and C= O absorption peaks. The effects of CGFP, CGFP-Fe, GFP2-2 and GFP-Fe on splenocytes proliferation and cytokines secretion were studied in vitro . Both the GFP-2 and GFP-Fe had good immune-enhancing activity. Furthermore, GFP-Fe showed an ability to promote the proliferation of splenocytes and peritoneal macrophages. The iron-chelating peptide of Grifola frondosa still maintained its immune enhancing activity after in vitro gastrointestinal digestion. The results suggested that GFP-Fe might be beneficial as a new iron supplement and as an immunological enhancement .
