Coordination-responsive selenium-containing polymer micelles for controlled drug release

Wei Cao,Yang Li,Yu Yi,Shaobo Ji,Lingwu Zeng,Zhiwei Sun,Huaping Xu

A novel coordination-responsive system for the controlled release of doxorubicin was fabricated by complexing platinum cations with selenium-containing polymers. Doxorubicin loaded in the platinum-coordinating micelles can be released in a controlled manner through the competitive coordination of the platinum cations with glutathione. The coordination micelles are quite biocompatible as vehicles of drug delivery, thus opening a new avenue in multidrug systems for cooperative chemotherapy.

Highly selective fluorescence detection of Pt4+ over Pd2+ and Pt2+ using a polyethyleneimine-based nanosensor prepared via facile three-component reaction

Mingyue Chang,Moru Zhang,Haihong Hu,Shucai Liang

A novel polyethyleneimine (PEI)-based polymeric nanosensor (named PEIMP) was developed for specific fluorescence enhanced sensing of Pt 4+ ion in aqueous media. The sensor was fabricated via “one-pot” three-component reaction using ortho -phthalaldehyde (OPA), PEI and mercaptopurine as raw materials, by which the formation of isoindole fluorophore and its chemical grafting onto PEI chain were achieved simultaneously. The morphology, size and structure of PEIMP have been characterized by various techniques. In buffered aqueous solution (pH 7.0), PEIMP had the ability to specifically bind with Pt 4+ producing notable increase in fluorescence emission at 463 nm (excited at 395 nm). Based on investigations on the sensing mechanism, the fluorescence turn-on response towards Pt 4+ was attributed to the binding of Pt 4+ with purine group in PEIMP resulting in the inhibition of photoinduced electron transfer from purine to isoindole fluorophore. Under the optimal conditions (pH 7.0, incubated at 37 ℃ for 20 min) the detection of Pt 4+ could be achieved with the linear range of 0.1–10 μM and the detection limit of 80 nM. The sensor had the advantages of low-cost raw materials, simple and environmental-friendly synthesis and analytical detection procedures. What's more, it could selectively and sensitively detect Pt 4+ without the effects from common transition metal ions (Pb 2+ , Fe 3+ , Cr 3+ , Al 3+ , Ag + , Co 2+ , Hg 2+ , Cd 2+ , Cu 2+ , Mg 2+ , Ni 2+ , Mn 2+ , Zn 2+ ), especially precious metal   ions of Pt 2+ and Pd 2+ . The proposed method had been successfully applied to quantify Pt 4+ in wastewater and urine samples, and also proved to be potential for monitoring Pt 4+ in biological systems.

An aptamer-based colorimetric Pt(II) assay based on the use of gold nanoparticles and a cationic polymer

Sang Fuming,Liu Jia,Zhang Xue,Pan Jianxin

A colorimetric method is described for the determination of Pt(II). It is based on the use of gold nanoparticles (AuNPs) which are known to aggregate in the presence of a cationic polymer such as poly(diallyldimethylammonium chloride) (PDDA). If, however, a mismatched aptamer (AA) electrostatically binds to PDDA, aggregation is prevented. Upon the addition of Pt(II), it will bind to the aptamer and induce the formation of a hairpin structure. Hence, interaction between aptamer and PDDA is suppressed and PDDA will induce the aggregation of the AuNPs. This is accompanied by a color change from red to blue. The effect can be observed with bare eyes and quantified by colorimetry via measurement of the ratio of absorbances at 610 nm and 520 nm. Response is linear in the 0.24–2 μM Pt(II) concentration range, and the detection limit is 58 nM. The assay is completed within 15 min and selective for Pt(II) even in the presence of other metal ions. It was successfully applied to the rapid determination of Pt(II) in spiked soil samples. Schematic representation of the method for detection of Pt(II) based on the use of a cationic polymer and gold nanoparticles. In the presence of Pt(II), aptamer interacts with the Pt(II) and prevents the interaction between aptamer and cationic polymer. Hence, cationic polymer induce the aggregation of the AuNPs and lead to the color change from red to blue.

A novel ion-imprinted membrane induced by amphiphilic block copolymer for selective separation of Pt(IV) from aqueous solutions

Jianxian Zeng,Chaoqiang Lv,Guoqing Liu,Zhe Zhang,Zhihui Dong,JinYan Liu,Ying Wang

A novel ion-imprinted blend membrane was fabricated to separate selectively platinum(IV) from aqueous media. An amphiphilic copolymer poly(methyl methacrylate)-b-poly(4-vinylpyridine) (PMMA- b -P4VP) was firstly synthesized via the reversible addition fragmentation chain transfer polymerization , and characterized by 1 H NMR, FT-IR and GPC. Subsequently, poly(vinylidene fluoride) (PVDF) blend membranes were prepared using PMMA- b -P4VP as a blending additive. Effects of the following factors on performance of blend membranes were systematically examined: P4VP content of PMMA- b -P4VP, solvent type, and PMMA- b -P4VP/PVDF ratio. Further, platinum(IV) ion-imprinted membrane (Pt(IV)-IIM) was fabricated by blending polymer-Pt(IV) complexes with PVDF. SEM, FTIR-ATR, contact angle, water flux and adsorption capacity were employed to characterize all membranes. Results indicated that the membrane, which was prepared using the copolymer with high P4VP content (polymerization time 10 h) as a functional polymer , N,N-dimethylacetamide as a solvent and PMMA- b -P4VP/PVDF ratio of 30 wt%, exhibited an intriguing morphology and much higher performance. Pt(IV)-IIM showed higher adsorption and selectivity for Pt(IV) (selectivity coefficients of 27.66 ± 1.38 and 77.16 ± 3.86 for Pt(IV)/Cu(II) and Pt(IV)/Ni(II), respectively) compared with a non-imprinted membrane. During membrane filtration, Pt(IV)-IIM was effective and selective for separation of Pt(IV) in the presence of Cu(II) and Ni(II) ions. Also, Pt(IV)-IIM was chemical stable, and had a good regeneration performance.

Effects of selected two noble metal ions in medicine on the structure and activity of bovine serum albumin: A multi-spectral studies

Hongmei Zhang,Mengyue Zhang,Ying Wang,Yanqing Wang,Shixin Sun,Zhenghao Fei,Jian Cao

In the present study, the toxic influence of two metallic ions on serum albumin was studied by using fluorescence, UV/vis, circular dichroism spectroscopy, and molecular docking , with the aim of delineating the effects of Pt(II) and Au(III) ions on the secondary and tertiary structure , the transportation function, the thermal stability, and the esterase-like activity of bovine serum albumin (BSA). The conformational change occurring in protein structure was monitored by CD and fluorescence spectroscopy . Pt(II) and Au(III) could react with some functional groups of BSA, making the secondary structure change and increasing the microenvironment around Tyr residues of BSA. The content of α-helix decreased remarkably and the contents of β-sheet, and random coil increased with increasing concentration of Pt(II) or Au(III) ion. Compared with Pt(II) ion, Au(III) ion is more likely to damage the tertiary structure of BSA and a higher number of Tyr residues are affected in Au(III)-BSA system. The fluorescence and UV/vis spectral results indicated the formation of Pt(II)-BSA and Au(III)-BSA complexes. In addition, metal ion binding decreased the ANS binding ability, the reversibility of the thermal unfolding process and the esterase-like activity of BSA. Therefore, this paper is expected to improve the toxicity evaluation mechanism of platinum and gold ions.

Bioinspired Mechanically Responsive Hydrogel upon Redox Mediated by Dynamic Coordination between Telluroether and Platinum Ions

Feng Li,Xue Zhang,Shuxian Hu,Zhaoyue Lv,Jigang Lv,Wenting Yu,Xihan Xu,Dayong Yang

Inspired by natural lives, materials with switchable mechanical properties have been constructed by virtue of the development of dynamic chemistry. However, the limited dynamic reactions are hampering their further development. The progress in organotellurium chemistry brings opportunity to open new frontiers of dynamic reactions for fabrication of mechanically responsive materials. Herein, a bioinspired mechanically responsive hydrogel upon redox mediated by dynamic coordination between telluroether and platinum ions is reported. The low electronegativity and superior σ electron-donating capability of telluroether facilitate its coordination with Pt ions. Remarkably, the coordination interaction is highly redox-active. Telluroether can dissociate from Pt ions upon oxidation (e.g., H2O2) because the effect of the polar solvent dramatically reduces the thermodynamic activity of oxidized telluroether. In turn, the coordination interaction is easily regenerated via reduction (e.g., vitamin C) of oxidized telluroether. When the coordination system is introduced into the polymer network of a hydrogel via copolymerization of telluroether containing a monomer and N-hydroxyethylacrylamide, the hydrogel exhibits a switchable and regulatable microporous structure and mechanical strength upon redox stimuli by virtue of the dynamic coordination interaction between telluroether and Pt ions. This work provides a new set of dynamic coordination chemistry and a new strategy to prepare mechanically responsive hydrogel.
