Platinum (II)-coordinated Portulaca oleracea polysaccharides as metal-drug based polymers for anticancer study

Qianqian Han,Lirong Huang,Ying Wang,Shixin Sun,Hao Huang,Fei Li,Fangtian Wang,Ligen Chen,Hongmei Zhang,Yanqing Wang

Novel polysaccharide-platinum conjugated polymers bearing alendronate on Portulaca oleracea polysaccharides (PPS) were designed and synthesized. Their chemical structures and properties were characterized by Fourier transform infrared spectroscopy (FT-IR), 1 H NMR and 31 P NMR spectroscopy, Thermogravimetric analysis (TGA), X-ray powder diffraction (XRD), UV–vis spectrophotometer (UV–vis) and other analysis methods. The results demonstrated that alendronate can be used as the linker of Portulaca oleracea polysaccharides and platinum compounds. Portulaca oleracea polysaccharides-alendronate (PPS-ALN) conjugates exhibited stronger antioxidant ability than PPS. The cytotoxicity assay to cancer cells was tested in vitro, and the Portulaca oleracea polysaccharides-alendronate-platinum (PPS-ALN-Pt) conjugates strongly inhibited the proliferation of cancer cells than PPS and PPS-ALN. The evaluation of complexes affinity toward supercoiled plasmid DNA, displayed a high DNA interaction. Interestingly, the platinum conjugates displayed immunological competence in HeLa cells by cellular immunofluorescence assay. Besides, the cellular platinum accumulation of PPS-ALN-Pt conjugates was higher than that of cisplatin in HeLa cells, implying that the polysaccharide-platinum conjugated polymers might have a synergistically therapeutic application in metal anticancer drug delivery.

Synergistic roles of Pt0 and Pt2+ species in propane combustion over high-performance Pt/AlF3 catalysts

Xue Li,Yan-Rong Liu,Wen-Min Liao,Ai-Ping Jia,Yue-Juan Wang,Ji-Qing Lu,Meng-Fei Luo

Catalytic propane combustion was performed on a series of supported Pt/AlF 3 catalysts. These catalysts are very active and stable in the reaction, and the 0.2Pt/AlF 3 catalyst gives the highest specific reaction rate of 207.3 μmol g Pt −1  s −1 at 220 °C. The 1Pt/AlF 3 is much more active than the reference Pt/Al 2 O 3 catalyst, due to its higher content of metallic Pt species than that in the Pt/Al 2 O 3 . Furthermore, turnover frequency (TOF) based on Pt dispersion increases with the surface concentration of metallic Pt 0 in the catalyst, suggesting these species might be the active sites. Furthermore, synergistic roles of metallic and oxidized Pt species are found, as the TOFs calculated based on surface Pt 0 concentration reaches a maximum at the Pt 0 /Pt 2+ molar ratio of about 1. Such optimal Pt 0 /Pt 2+ ratio implies the requirement of Pt 0 Pt 2+ pair in the reaction, probably due to the activation and cleavage of C H bond on metallic Pt atom and the sequential oxidation on the oxidized Pt atoms.

Hollow porous nanoparticles with Pt skin on a Ag–Pt alloy structure as a highly active electrocatalyst for the oxygen reduction reaction

Tao Fu,Jun Fang,Chunsheng Wang,Jinbao Zhao

Aiming to reduce the dosage of the noble metal Pt and improve the catalytic activity of the catalyst in fuel cells, hollow porous Ag–Pt alloy nanoparticles with Pt coating are prepared via a facile controlled galvanic replacement reaction. Ag is used as the substrate to build a hollow porous structure and alloyed with Pt to minimize the tensile effect of the Ag on the deposited Pt skin which would significantly lower the catalytic performance of the Ag–Pt bimetallic catalyst. This hollow porous Ag/Pt bimetallic catalyst exhibits a long catalytic durability and a mass activity of 0.438 A mgPt−1 at 0.9 V (vs. RHE) towards the oxygen reduction reaction (ORR), which is ca. 3 times higher than that of the commercial Pt/C catalyst. The significant enhancement over the state-of-the-art Pt catalysts can be attributed to (1) the high surface area of the nanoparticles, (2) the more suitable d-band center of the Pt skin deposited on the Ag–Pt alloy substrate, and (3) the high thermal stability of the Ag–Pt alloy. Therefore, this work provides a new strategy for designing high-performance catalysts with low cost. In addition, the synthetic chemistry involved can possibly be extended for fabricating versatile catalysts with a similar structure.

Templated-Assisted Synthesis of Structurally Ordered Intermetallic Pt3Co with Ultralow Loading Supported on 3D Porous Carbon for Oxygen Reduction Reaction

Xiao-Feng Han,Nadia Batool,Wen-Tao Wang,Hao-Tian Teng,Li Zhang,Ruizhi Yang,Jing-Hua Tian

Simple and reliable mass production of platinum-based alloy catalysts with excellent activity and stability is an enormous challenge for the wide commercialization of proton-exchange membrane fuel cells (PEMFC), especially those with ultralow loading of Pt. Herein, an economical, highly durable, and efficient catalyst consisting of structurally ordered intermetallic Pt3Co alloy nanoparticles with ultralow Pt loading (1.4 wt %) supported on hierarchically porous carbon structure (three-dimensional, 3D Pt3Co/C) were synthesized with large-scale production by the NaCl-template-assisted approach. The obtained best sample, 3D Pt3Co/C#1, exhibited mass activities of 11.56 and 0.70 A mgPt–1 for oxygen reduction reactions (ORRs) in alkaline and acidic electrolytes, which are 60.8 and 6.4 times those of commercial Pt/C, respectively. Furthermore, the 3D Pt3Co/C#1 exhibited excellent stability both in acidic and alkaline electrolytes, with almost no decay of the half-wave potential after 5000 potential cycles. This work proposes a new high-yielding, simple, and environmentally friendly method to fabricate excellent Pt-based alloy electrocatalysts with ultralow loading of Pt, which opens up new hopes for the development of PEMFC.

Inhibition of thioredoxin reductase 1 correlates with platinum-based chemotherapeutic induced tissue injury

Ping Cheng,Huan Liu,Yinchuan Li,Peiling Pi,Yu Jiang,Shaozhen Zang,Xiaorong Li,Ailing Fu,Xiaoyuan Ren,Jianqiang Xu,Arne Holmgren,Jun Lu

Platinum-containing drugs (PtDs; e.g. cisplatin , carboplatin , and oxaliplatin) have been widely used as anticancer reagents against various cancers. However, treatment with these drugs results in undesirable adverse effects with unknown mechanisms. Herein, we found a strong correlation between the inhibitory effects of PtDs on cytosolic thioredoxin reductase (TXNRD1) and tissue injury. Of the PtDs tested, cisplatin was found to be the most effective inhibitory PtD against TXRND1, causing the severest kidney injury . The initial inhibition of TXNRD1 in the kidney resulted from cisplatin-induced transcriptional activation of Nrf2-regulated genes including Txnrd1 . However, the antioxidant responses in the kidney did not reverse the cisplatin-induced oxidation process. Nephrotoxicity was accompanied with an increase of protein glutathionylation and a cellular thiol redox environment oxidation. These results suggest that the changes of the cellular thiol-dependent redox environment regulated by TXNRD1 is a major event in the adverse effects of cisplatin in kidney.

Construction of Ti3C2T x MXene Supported Low-Platinum Electrocatalyst for Hydrogen Evolution Reaction by Direct Electrochemical Strategy

Xuan Jian,Tong-Tong Wei,Mi-Mi Zhang,Rui Li,Jian-Xin Liu,Zhen-Hai Liang

The design of high-performance catalysts for hydrogen evolution reaction (HER) is urgently needed in the development of renewable energy. Herein, we report a rapid strategy for synthesizing an efficient catalyst with ultralow-Pt nanoclusters on a two-dimensional titanium carbide (Ti3C2Tx) MXene support (denoted as Pt NCs-MXene, 5 μg cm−2) via electrophoretic deposition and cyclic voltammetry. The MXene support not only provides more active sites to anchor Pt nanoclusters (NCs) but also increases the electrochemically active surface area. The electrochemical HER performance of the as-synthesized Pt NCs-MXene catalyst was investigated in an acid electrolyte. The results showed that at a current density of 10 mA cm−2, the overpotential and Tafel slope of the Pt NCs-MXene were 40 mV and 50.8 mV dec−1, respectively, lower than those of the commercial 20 wt% Pt/C catalyst. The turnover frequency and mass activity of the Pt NCs-MXene were 7.09 H2 s−1 and 7.02 A mgPt−1, respectively, which were approximately 70 times those of the Pt/C catalyst. Moreover, the developed catalyst also exhibited a stable hydrogen evolution current for 18 h in a long-term cycling test. This work can provide novel and nontoxic methods for the preparation of low-Pt loading electrocatalysts for high-efficiency HER.

Harnessing Phosphato-Platinum Bonding Induced Supramolecular Assembly for Systemic Cisplatin Delivery

Yingqin Hou,Yaoyi Wang,Ruijue Wang,Weier Bao,Xiaobo Xi,Yunlong Sun,Shengtao Yang,Wei Wei,Hua Lu

To improve the therapeutic index of cisplatin (CDDP), we present here a new paradigm of drug-induced self-assembly by harnessing phosphato-platinum complexation. Specifically, we show that a phosphato-platinum cross-linked micelle (PpY/Pt) can be generated by using a block copolymer methoxy-poly(ethylene glycol)-block-poly(l-phosphotyrosine) (mPEG-b-PpY). Coating of PpY/Pt with a R9-iRGD peptide by simple mixing affords a targeting micelle with near neutral-charged surface (iPpY/Pt). The micelles feature in well-controlled sizes below 50 nm and high stability under physiological conditions, and can withstand various environmental stresses. Importantly, the micelles demonstrate on-demand drug release profiles in response to pathological cues such as high ATP concentration and acidic pH. In vitro, the micelles are efficiently internalized and almost equally potent compared to CDDP. Moreover, iPpY/Pt induce greater cytotoxicity than PpY/Pt in a 3D tumor spheroid model likely due to its deeper tumor penetration. In vivo, the micelles exhibit prolonged circulation half-lives, enhanced tumor accumulation, excellent tumor growth inhibition in a xenograft HeLa model and an orthotropic mammary 4T1 model, and improved safety profiles evidenced by the reduced nephrotoxicity. Together, this work demonstrates for the first time that phosphato-platinum complexation can be exploited for effective delivery of CDDP, and suggests a paradigm shift of constructing nanosystems for other anticancer metallodrugs.

High-value utilisation of PGM-containing residual oil: Recovery of inorganic acids, potassium, and PGMs using a zero-waste approach

Minghui Liu,Yutong Zhao,Quanzhong Cheng,Bingyang Tian,Ming Tian,Jian Zhang,Hui Zhang,Tianyan Xue,Tao Qi

Residual oil containing platinum group metals (PGMs), which is under-researched, can easily pose resource waste and environmental risks. PGMs feature as scarce strategic metals, and inorganic acids and potassium salts are also considered valuable. An integrated process for the harmless treatment and recovery of useful resources from residual oil is proposed herein. This work developed a zero-waste process based on the study of the main components and characteristics of the PGM-containing residual oil. The process consists of three modules: pre-treatment for phase separation, liquid-phase resource utilisation, and solid-phase resource utilisation. Separating the residual oil into liquid and solid phases allows for the maximum recovery of valuable components. However, concerns about the accurate determination of valued components emerged. Findings revealed that Fe and Ni are highly susceptible to spectral interference in the PGMs test when using the inductively coupled plasma method. After studying 26 PGM emission lines, Ir 212.681 nm, Pd 342.124 nm, Pt 299.797 nm, and Rh 343.489 nm were reliably identified. Finally, formic acid (81.5 g/t), acetic acid (117.2 kg/t), propionic acid (291.9 kg/t), butyric acid (3.6 kg/t), potassium salt (553.3 kg/t), Ir (27.8 g/t), Pd (10960.0 g/t), Pt (193.1 g/t), and Rh (109.8 g/t) were successfully obtained from the PGM-containing residual oil. This study provides a helpful reference for the determination of PGM concentrations and high-value utilisation of PGM-containing residual oil.

Development of nanotubes coated with platinum nanodendrites using a virus as a template

Jiawang Guo,Yuan Lin,Qian Wang

We developed a facile method to fabricate platinum (Pt) porous nanotubes coated with interconnected Pt dendrites using the tobacco mosaic virus (TMV) as a template. The surface-exposed arginine residues of the TMV induced the selective deposition of Pt seeds on the TMV outside surface, and poly(sodium-p-styrenesulfonate) (PSS) was chosen to stabilize the dispersity of TMV coated with Pt seeds (TMV/SPt). The limited space between the Pt seeds and their uniform distribution on the TMV exterior confined the growth of Pt dendrites, resulting in continuous dendritic platinum nanotubes (TMV/DPtNT). The synergistic effects of porous dendrites and anisotropic structures of the TMV/DPtNTs provided an increase in the active sites, the enhancement of transport efficiency and long-distance electron transfer, which greatly improved the catalytic activity. We also demonstrated that such nanotubes could be used in the detection of H2O2 with good sensitivity.

Anchoring ultrafine metallic and oxidized Pt nanoclusters on yolk-shell TiO2 for unprecedentedly high photocatalytic hydrogen production

Jun Jin,Chao Wang,Xiao-Ning Ren,Shao-Zhuan Huang,Min Wu,Li-Hua Chen,Tawfique Hasan,Bin-Jie Wang,Yu Li,Bao-Lian Su

We demonstrate an alkali modification process to produce highly dispersed ultrafine Pt nanoclusters with metallic Pt 0 and oxidized Pt 2+ species as co-catalyst anchored on nanosheet-constructed yolk-shell TiO 2 (NYTiO 2 -Pt) acting as light harvesting reactor for highly efficient photocatalytic H 2 production. Benefiting from the high surface area, highly dispersed ultrafine Pt nanoclusters (~0.6   nm) with Pt 0 and Pt 2+ species and special nanosheet-constructed yolk-shell structure, this novel light harvesting reactor exhibits excellent performance for photocatalytic H 2 production. The NYTiO 2 -Pt-0.5 (0.188   wt% Pt) demonstrates an unprecedentedly high H 2 evolution rate of 20.88   mmol   h −1   g −1 with excellent photocatalytic stability, which is 87 times than that of NYTiO 2 -Pt-3.0 (0.24   mmol   h −1   g −1 , 1.88   wt% Pt), and also much higher than those of other TiO 2 nanostructures with the same Pt content. Such H 2 evolution rate is the highest reported for photocatalytic H 2 production with such a low Pt content under simulated solar light. Our strategy here suggests that via alkali modifying the photocatalysts , we can not only enhance the H 2 production for solar energy conversion but also significantly decrease the noble metal content for cost saving.

THE ADSORPTION OF BiIII/Pt NANOCOMPOSITES AT PLATINUM ELECTRODE WITH HIGHLY ENHANCED ELECTROCATALYSIS TOWARD GLUCOSE

ZHUOYUAN YANG,YUQING MIAO,TIANRUI WANG,MINGSHU XIAO,XIAOCAI LIANG,YANG YANG,WEIWEI LI

In this paper, we first fabricated a nanoPt modified platinum electrode. Then through a simple method, the electrode surface was introduced with a submonolayer of bismuth that acted as an effective promoter. Cyclic voltammetry and other characterizations were employed. The obtained Bi III / nanoPt / Pt electrode exhibited two greatly increased oxidation peaks at negative and positive potential areas, respectively. The signals were far larger than that of platinum electrode because of the large true surface area of nanoparticles and the catalysis of bismuth adsorbed on platinum. In the presence of bismuth, the platinum active sites could combine with more OH - from bismuth hydroxyl to form a new active site for the oxidation of glucose. The prepared Bi III / nanoPt / Pt electrode given high sensitivity and excellent linearship to glucose detection and showed the potential application in the areas of electrocatalysis or electroanalysis.

Single-atom platinum or ruthenium on C4N as 2D high-performance electrocatalysts for oxygen reduction reaction

Zhou Yang,Meng Xiang,Yunfeng Zhu,Jia Hui,Yong Jiang,Shuang Dong,Chengbin Yu,Junfei Ou,Hengfei Qin

Pt/C is a practical electrocatalyst for oxygen reduction reaction (ORR) in metal-air batteries and fuel cells, however, also suffers the drawbacks of high cost and instability, which have propelled people to search for new substitutes, such as single-atom metal electrocatalysts. Herein, C 4 N was firstly synthesized from 2,3-diyldiamine phenazine via one-step hydrothermal method rather than the traditional way that two monomers react in organic solvent, and then loaded with single-atom metal (Pt or Ru) to form new electrocatalysts. The so-formed M@C 4 N (M = Pt or Ru) electrocatalysts show more excellent ORR performances with higher half-wave potential and better stability and methanol tolerance as compared with the commercial Pt/C. Specifically, the half-wave potential of 1%Pt@C 4 N and 0.5%Ru@C 4 N is 0.861 V and 0.828 V, respectively. Density functional theory calculations reveal that 1%Pt@C 4 N has stronger adsorption of reactant species and faster kinetics than 0.5%Ru@C 4 N in ORR pathway.
