Objective. To investigate the effects of mung bean peptide and its’ complexes on promoting lead excretion and neuroprotection of zebrafish. Methods. The lead poisoning models of zebrafish were established by lead acetate solution; the models were treated with high and low concentrations (58.3 and 175 μg/mL) of mung bean peptides, with high, medium, and low concentrations (27.8, 83.3, and 250 μg/mL) of mung bean peptide complexes, separately. The effects of the mung bean peptide complexes on the lead content, axonal fluorescence intensity, and peripheral motor nerve length changes were identified in the zebrafish model, and the effects of mung bean peptide and its’ complexes on zebrafish's lead excretion, axonal protection rate, and peripheral movement promotion rate of nerve regeneration were calculated. Results. The effects of high concentration of mung bean peptide (175 μg/mL) in promoting lead excretion was 29% (), and the effect of high concentration of mung bean peptide complexes (250 μg/mL) in promoting lead excretion was 30% (). The other concentrations of mung bean peptide and its’ complex groups did not show a noticeable lead excretion effect. The protective effects of mung bean peptide at concentrations of 58.3 and 175 μg/mL against zebrafish axonal injury were 98% and 101% (), and the peripheral nerve regeneration promotion effects were 29% () and 42% (), respectively. The protective effects of mung bean peptide complexes at concentrations of 27.8, 83.3, and 250 μg/mL against zebrafish axonal injury were 85%, 78%, and 93% (); peripheral nerve regeneration promotion rates were 46%, 50%, and 50% (). Conclusion. The mung bean peptide and its’ complexes can effectively promote the discharge of lead in the zebrafish lead poisoning and have protective and regeneration effects on zebrafish nerves.

Lead (Pb) contamination is a worldwide public health threaten. Besides close restraint of lead exposure, it's emergency to discover compounds that could help to cue toxicities caused by lead. Zebrafish embryos and early larvae can serve as valuable screening tools in early pre-clinical phase of drug screening and research. In order to establish a zebrafish lead poisoning model that could be used for drug screening and research, zebrafish embryos at 6 h post-fertilization (hpf) were treated with lead at different concentrations by soaking intermittently, raised in lead work solution at night while in fish water during the day. After treated for 90 h, death and severe trunk curvature were observed on zebrafish in 640 μM group, obvious dysplasia, blood toxicity, excessive reactive oxygen species (ROS), severe neurotoxicity, such as shorter length of peripheral motor neurons, neuronal apoptosis, and axonal injury, and neurobehavior impairment were induced by lead at 80, 160 and 320 μM, similar to phenotypes reported in rodent. Moreover, the mRNA level of genes related to neurodevelopment, memory, and antioxidation were significantly down regulated, and apoptosis-related genes were up regulated, consistent to zebrafish phenotypic change. Finally, zebrafish were intermittently exposed to 80 μM lead solution to establish the lead poisoning model, and the efficacy of a safe chelating agent Meso-2,3-Dimercaptosuccinic acid (DMSA) was tested at a series of concentrations to validate the zebrafish model. The result showed concentration-dependent decrease of lead content in zebrafish in DMSA treated groups compared with model group. The above data fully demonstrated a zebrafish model of lead poisoning suitable for drug screening was successfully developed, which was expected to provide a rapid and economic tools for discovering antidotes of lead and drugs of neuroprotection.

δ-Aminolevulinic acid (δ-ALA) plays a vital role in the diagnosis and clinical screening of diseases like lead poisoning, hepatic porphyria, and hereditary tyrosinemia type-1. However, because of the drawbacks of the existing methods, such as tedious pre-separations and analytical interference, it poses a serious challenge for an efficient, specific, and simple δ-ALA detection strategy. In this work, matrix-isopotential synchronous fluorescence spectrometry (MISFS) and the first derivative technique are combined for the direct determination of δ-ALA in urine samples. The proposed approach promises the detection of δ-ALA in urine samples with unknown background fluorescence, alleviating complicated and tedious pre-separation processes. Here, the fluorescent derivatization of δ-ALA was based on the Hantzsch reaction. MISF spectra were scanned along a matrix-isopotential trajectory, which combines the points of equal intensity in theoretical 3D contours. The contours were constructed from the excitation and emission spectra of δ-ALA and urine. The unknown background signal of the urine was suppressed by the first order derivative technique. Maximum sensitivity and appropriate selectivity were ensured by the study of the experimental variables in the δ-ALA bands, centered at its excitation and emission maxima (392.6 nm and 464.5 nm). The limits of detection and quantification, calculated according to IUPAC definitions, were 5.4 ppb and 18 ppb, respectively. The average recoveries were 97.2% ± 2.6%. For within-run precisions, the inter-day and intra-day CVs were 5.3% and 3.6%, respectively. The results achieved by the proposed method were consistent with those obtained by the conventional HPLC-FLD method.

The presence of lead as an environmental pollutant is widespread. However, safe and effective treatments for the resulting intestinal and liver damage from high levels of lead exposure remain limited. The study aimed to investigate the protective effects of dietary fiber and polyphenols in whole grain wheat flour on lead-induced mice. The results indicated that the daily intake of 12 mg of polyphenols, 0.5 g of dietary fiber, and their combination effectively reduced blood and liver lead accumulation by approximately 50 % in mice exposed to lead, while also mitigating lead-induced oxidative stress though a reduction in malondialdehyde levels and an enhancement in antioxidant enzyme activities including superoxide dismutase, catalase, and glutathione peroxidase. Furthermore, all three treatments enhanced cytokine secretion with the combined treatment exhibiting the highest efficacy. Specifically, the combination treatment decreased tumor necrosis factor-α and interleukin 1β by 56.78 %, 47.86 % in intestinal tissue while increasing increased interleukin 4 and interleukin 10 by 81.84 %, 145.14 %. Additionally, it promoted the expression of tight junction proteins like Zonula occludens-1, Occludin and Claudin-1. The study presented a potential strategy for alleviating liver and intestinal tract damage from high-dose lead exposure.

Heavy metal Pb is an inevitable ingredient of coal-fired flue gas and has been proven to be a source of inhibiting effect on the selective catalytic reduction (SCR) catalyst. In this paper, two types of Pb-poisoned catalyst, prepared by the incipient wetness impregnation and solid diffusion methods, are studied to unveil the poisoning effects of lead species on the V 2 O 5 -WO 3 /TiO 2 catalyst. The activity test results showed that lead species loaded by the incipient wetness impregnation method exhibited a significant inhibiting effect on the denitrification efficiency of the catalyst although a negligible activity loss was observed on the samples from the solid diffusion method. Characterizations of fresh and Pb-poisoned catalysts from incipient wetness impregnation method were applied using comprehensive techniques such as X-Ray diffraction (XRD), field emission scanning electron microscopy (FE-SEM), X-ray fluorescence (XRF), NH 3 -temperature programmed desorption (NH 3 -TPD), X-Ray photoelectron spectroscopy (XPS), and so forth. The analytical results indicated that a phase transformation occurred in tungsten species and the amorphous WO 3 converted into PbWO 4 , causing the accumulation and valence imbalance of active vanadium phase, which further decreased the surface acidity and activity of the V 2 O 5 -WO 3 /TiO 2 catalyst.

Blood lead poisoning is a universal and severe health problem that greatly threatens human health in various industries. Elimination of blood lead relying on chelating agents and combination with hemoperfusion adsorbents has achieved considerable progress, but it is still suffering from the compromised selectivity of adsorbents as well as in absence of real time monitoring during treatment. Herein, we proposed a selective blood lead adsorbent integrated with real-time visualized/electrochemical bi-modal monitoring based on TMPyP-bound guanosine-borate (GB) supramolecular hydrogel as potential smart hemoperfusion device. The GB hydrogel possessed stability in physiological environment, self-healing ability resistant to fluid shear, blood compatibility, selective adsorption of lead ions superior to conventional adsorbents, anti-fouling performance to blood components and renewability. Benefiting from binding with TMPyP and the intrinsic conductivity, GB hydrogel was endowed with the ability to qualitatively diagnose the presence of blood lead via simple color change and quantitatively reflect the amount of adsorbed lead from blood accurately through electrochemical technique . This work puts forward an integrated treatment/monitoring hemoperfusion device with high selectivity, simple fabrication and low-cost, providing a paradigm for next generation design of intelligent, monitorable theranostic hemopurification system, which is also an extensible platform for the other research fields such as environmental monitoring and remediation.

Ferritin is expected to alleviate lead-induced oxidative damage. Despite being a relatively stable macromolecular protein that can withstand high temperatures and extreme pH conditions without its structure and activity being compromised, oral administration of ferritin may still result in its degradation within the gastrointestinal tract, reducing its bioavailability. In recent years, microcapsules have emerged as an effective approach in oral protein delivery to enhance bioavailability. In this study, carboxymethyl-β-cyclodextrin grafted chitosan embedded oyster ( Crassostrea gigas ) ferritin (CMCD-CS@GF1) was fabricated with favorable encapsulation efficiency (57.73 ± 4.84 %) and loading capacity (5.78 ± 0.48 %), as well as a certain degree of resistance to digestion in the gastric environment. In the simulated gastric fluid with an acidic pH, the cumulative release of GF1 over 4 h was 24.18 %. In the simulated intestinal fluid with a neutral pH, the cumulative release of GF1 over 8 h was 70.56 %. Animal experiments showed that oyster ferritin could reduce the accumulation of lead in the body, enhance the activity of antioxidant enzymes, activate the Nrf2 signaling pathway, and down-regulate the NLRP3 signaling pathway to reduce oxidative stress and inflammation levels. The findings demonstrate that oyster ferritin may mitigate the toxic effects of lead in the body, providing a significant theoretical foundation for its application in functional foods.

Pb poisoning affects infant growth and development. However, dimercaptosuccinic acid (DMSA) as the current therapy for Pb poisoning exerts relatively significant toxic side effects in infants. Therefore, identifying a non-toxic treatment in this regard is particularly important. In this study, we aimed to investigate the therapeutic effect of an infant feces-derived probiotic strain, Lactobacillus casei SYF-08 (SYF-08), on Pb poisoning in young mice. The Pb levels in the organisms were detected via inductively coupled plasma mass spectrometry, while the therapeutic effect of SYF-08 on Pb-induced neural system damage was explored via the Morris water maze test, hematoxylin-eosin staining, and immunohistochemistry. Additionally, the molecular mechanisms underlying the protective effects of SYF-08 against Pb-induced intestinal damage were also explored via histological staining, 16S rRNA sequencing, untargeted metabolomics, qRT-PCR, and western blotting. In vivo experiments revealed that SYF-08 reduced blood and bone Pb levels and increased urinary Pb excretion. Additionally, SYF-08 alleviated Pb-induced pathological damage to the brain and ultimately improved the learning and cognitive abilities of the young mice. This treatment also restored intestinal microflora dysbiosis, regulated bile acid metabolism, and inhibited the FXR-NLRP3 signaling pathway. It also resulted in fewer adverse events than the DMSA treatment. In conclusion, our results provided valuable insights into the therapeutic role of SYF-08 in Pb poisoning and also suggested that its administration can significantly alleviate the Pb-induced damage.

Lead (Pb) poisoning is estimated to account for 1 % of the global disease burden. The gold standard for diagnosing lead poisoning in human body relies on blood lead level (BLL), which is always performed in hospitals using expensive instruments. However, there are still many countries and regions with a lack of medical resources (without enough professional medical staff and analytical instruments). To achieve a facile diagnosis of lead poisoning by ordinary residents (without any expertise), this study conducted a research study on 810 participants to discover and validate a new lead poisoning indicator (creatinine-corrected urinary lead level, cULL) beyond BLL in non-invasive samples. A point-of-care testing (POCT) device to measure cULL was developed, equipped with liquid-phase microextraction and electromembrane extraction on a paper-based analytical device for on-site separation of lead and creatinine in the urine, using a smartphone for the quantification of analytes. The cULL as a novel indicator and the POCT device developed could be effective in reducing the risk of damage from lead contamination.
