Ion-Imprinted Polymer Modified with Carbon Quantum Dots as a Highly Sensitive Copper(II) Ion Probe

Zhiming Wang,Cuo Zhou,Shunwei Wu,Chunyan Sun

Fluorescence analysis technology and ion imprinting technology are combined to prepare a copper ion fluorescence sensor. Carbon quantum dots (CQDs), with a quantum yield of 79%, were synthesized by a hydrothermal process using citric acid as the carbon source. The prepared CQDs, acting as the fluorophore, were grafted onto the surface of an SBA-15 mesoporous molecular sieve by an amidation reaction. Then, the fluorescent sensor CQDs@Cu-IIP was prepared using a surface imprinting technique with the modified SBA-15 as the substrate, copper ions as a template, tetraethoxysilane as the crosslinker, and 3-aminopropyl-3-ethoxysilane as the functional monomers. The sensor showed strong fluorescence from CQDs and high selectivity due to the presence of Cu(II)-IIP. After the detection conditions were optimized, the fluorescence intensity of the sensor had good linearity with Cu(II) concentration in a linear range of 0.25–2 mg/L and 3–10 mg/L. This CQDs@Cu-IIP was applied to the determination of traces Cu(II) in real water samples and good recoveries of 99.29–105.42% were obtained. The present study provides a general strategy for fabricating materials based on CQDs for selective fluorescence detection of heavy metals. Keywords:carbon quantum dots;fluorescence quenching;ion-imprinted polymer;copper(II) ion

Silica nanoparticles doped with a europium(III) complex and coated with an ion imprinted polymer for rapid determination of copper(II)

Zheng Xudong,Pan Jianming,Gao Lin,Wei Xiao,Dai JiangDong,Shi WeiDong,Yan Yongsheng

We have prepared highly monodisperse silica spheres doped with an Eu(III) complex and coated them with a nanoshell of a copper-imprinted polymer. The silica cores have a diameter of 200 nm, and the nanoshells have a thickness of 10–30 nm. The nanoparticles were characterized by means of scanning electron microscopy and transmission electron microscopy. The material is shown to be a viable fluorescent probe for Cu(II) ions. The surface molecular imprint renders the nanoprobe sensitive and selective. It was successfully applied to the detection of Cu(II) and gave a linear fluorescent response in the 10–100 μM concentration range. Compared to other methods, this one features high selectivity, sensitivity, low cost and ease of operation. We assume that this assay is applicable to routine trace determination of Cu(II) in water or biological environments. By adopting the appropriate functional monomers, cross-linking agents and template ions, the method can potentially be used to fabricate a highly selective imprinted probe for other heavy and transition metal ions. The europium complex fluorescent probe detected the content of divalent copper by surface ion imprinting. The imprinted shell of the probe bound Cu(II) rapidly and selectively. Due to Cu(II) quenching of the fluorescent core, the fluorescence intensity decreases.

A Ferritin–Albumin–Cu Nanoparticle that Efficaciously Delivers Copper(II) Ions to a Tumor and Improves the Therapeutic Efficacy of Disulfiram

Nuo Xu,Yuan-Fan Yang,Long Chen,Jian Lin

The application of disulfiram can be traced back to the 1920s, from when it was utilized to facilitate alcohol abstinence by producing allergic reactions toward alcohol. In previous research, combination of disulfiram and copper ions has demonstrated significant anti-tumor activity. However, both disulfiram and copper ions lack the ability of specific targeting to tumor tissues and may lead to a high risk of side effects, especially for copper ion, which is a kind of toxic heavy metal. Herein, a ferritin–albumin–Cu nanoparticle (FHC NP) was introduced. The nanoparticle was synthesized by first covalently cross-linking ferritin and albumin and then encapsulating the copper ions in the ferritin nanocage. The nanoparticle showed good accumulation in a tumor and when combined with disulfiram exhibited good in vitro selectivity toward cancer cells and better in vivo anti-tumor activity. Herein, the nanoparticle invented here represented a good strategy to efficaciously deliver copper ions into tumors and thus improve the therapeutic efficacy of disulfiram in tumor therapy.

Identification of tanshinone I as a natural Cu(II) ionophore

Ya-Long Zheng,Yuan Ji,Yan Li,Shuai Yan,Xiao-Rong Ren,Wei Tang,Fang Dai,Bo Zhou

The development of Cu(II) ionophores for targeted disruption of aberrant redox homeostasis in cancer cells has been considered an appealing strategy in the field of anticancer research. This study presents the first identification of tanshinone I ( Ts1 ), a natural o -quinone, as a Cu(II) ionophore. Structure-activity relationship studies on tanshinones and mechanistic investigations reveal that the presence of Cu(II) effectively promotes the tautomerization of Ts1 from its diketo to keto-enol forms, thereby facilitating its sequential proton-loss Cu(II) chelation, and enabling it to function as a Cu(II) ionophore due to its structural features including the presence of an o -quinone moiety, a benzyl hydrogen, and a large conjugated system. The unique property allows Ts1 to preferentially induce copper accumulation in human hepatoma HepG2 cells over human umbilical vein endothelial cells, by releasing copper driven by reduced glutathione (GSH). This copper accumulation leads to a reduction in the GSH-to-oxidized glutathione ratio and the generation of reactive oxygen species, ultimately triggering apoptosis of HepG2 cells. The findings not only provide support for o -quinones as innovative types of anticancer Cu(II) ionophores, but also shed light on the previously unrecognized role of Ts1 as a potent Cu(II) ionophore for eradicating cancer cells by selectively disrupting their redox regulation programs, resembling a "Trojan horse".

An in-situ plasmonic spectroscopy based biosensor for detection of copper (II) ions highlighting analytical specifications

Zheng Dou,Wenjia Zhang,Qian Du,Guohua Liu

Whether the copper ion (Cu 2+ ) exposed to us functions as a necessary part of metabolism or toxicant for organs depends on subtle concentration difference of trace-level. Although miniature sensors have combined various recognition probes and signal transducer moieties to form novel sensing scheme, haunting limits on practicality or analytical specifications like sensitivity and selectivity remain thwarting high Cu 2+ detection performance with excellent cost-effectiveness. In this work, we report a Cu 2+ -specific biosensor integrating Deoxyribozyme (DNAzyme) and single-particle plasmon. The basic principle here is that severity of cleavage corresponding to various Cu 2+ concentration determines these elements’ aggregation capacity. The evaluation criteria is set up on the shift of dark-field Localized Surface Plasmon Resonance (LSPR) wavelengths caused by the binding with free AuNPs. Not only excellent analytical specifications like limit of detection (LOD) of 0.082 nM, broad dynamic range from 0.1 nM–5 μM, ultra-specificity are demonstrated but recovery rate with real sample and term of validity is also investigated to be 101.25 ± 9.15 % at 3 nM and longer than 9 days corroborating its potential for practical scenario. Therefore, the sensor is introduced as an attract alternative to remove hindrances for high sensing performance and to generalize on-chip cleavage-inducing spectral shift to heavy metal ions concentration measurement.

A selectively fluorescein-based colorimetric probe for detecting copper(II) ion

Jiangang Zhang,Li Zhang,Yanli Wei,Jun Ma,Shaomin Shuang,Zongwei Cai,Chuan Dong

A novel fluorescein derivative furfuraldehyde fluorescein hydrazone (FFH) has been synthesized by reacting fluorescein hydrazide with furfuraldehyde and characterized by 1 H NMR, 13 C NMR, MS and elemental analysis. Addition of Cu 2+ to the solution of FFH results in a rapid color change from colorless to yellow together with an obvious new band appeared at 502   nm in the UV–vis absorption spectra. This change is attributed to the spirocycle form of FFH opened via coordination with Cu 2+ in a 1:1 stoichiometry and their association constant is determined as 6.1   ×   10 4   L   mol −1 . Experimental results indicate that the FFH can provide a rapid, selective and sensitive response to Cu 2+ with a linear dynamic range 6.6–330   μmol/L. Common interferent ions do not show any interference on the Cu 2+ determination. It is anticipated that FFH can be a good candidate probe and has potential application for Cu 2+ determination in aqueous solution.

Copper (II) Ion-Modified Gold Nanoclusters as Peroxidase Mimetics for the Colorimetric Detection of Pyrophosphate

Yunjing Shi,Jinjie Wang,Kun Mu,Suqin Liu,Guang Yang,Min Zhang,Jingxia Yang

Copper (II) ions have been shown to greatly improve the chemical stability and peroxidase-like activity of gold nanoclusters (AuNCs). Since the affinity between Cu2+and pyrophosphate (PPi) is higher than that between Cu2+and AuNCs, the catalytic activity of AuNCs-Cu2+decreases with the introduction of PPi. Based on this principle, a new colorimetric detection method of PPi with high sensitivity and selectivity was developed by using AuNCs-Cu2+as a probe. Under optimized conditions, the detection limit of PPi was 0.49 nM with a linear range of 0.51 to 30,000 nM. The sensitivity of the method was three orders of magnitude higher than that of a fluorescence method using AuNCs-Cu2+as the probe. Finally, the AuNCs-Cu2+system was successfully applied to directly determine the concentration of PPi in human urine samples. Keywords:gold nanocluster;copper ion;pyrophosphate;keratin;colorimetric detection

A colorimetric assay for ultrasensitive detection of copper (II) ions based on pH-dependent formation of heavily doped molybdenum oxide nanosheets

Meijun Li,Xiaoxuan Huang,Haili Yu

Here we report a simple and low-cost colorimetric assay for ultrasensitive detection of copper (II) ions (Cu 2+ ) based on pH-dependent formation of plasmonic MoO 3-x nanosheets. The reaction between ascorbic acid (AA) and Cu 2+ gives birth to hydrogen ions, which remarkably promotes the reduction of MoO 3 nanosheets by AA to form oxygen vacancies-rich MoO 3-x nanosheets that increase the free carrier concentration, generating a strong local surface plasmon resonance (LSPR) absorption. The Cu 2+ -triggered LSPR is utilized for development of the colorimetric assay. Under the optimal experimental conditions, this colorimetric assay can be used for selective detection of Cu 2+ , and the limit of detection is 0.8 nM, which is lower than that of most of the reported methods. Additionally, the present colorimetric assay has also been successfully employed for Cu 2+ determination in real serum and human hair samples with satisfactory recoveries ranging from 96% to 108%.

Nanopore-Based Strategy for Sensing of Copper(II) Ion and Real-Time Monitoring of a Click Reaction

Liping Liu,Zhen Fang,Xiangjiang Zheng,Dongmei Xi

A straightforward yet efficient, nanopore-based strategy that enables the sensitive detection of copper(II) ion (Cu2+) and real-time monitoring of a click reaction is provided. Two single-stranded DNAs (ssDNAs) are designed to act as the preprobes, one being modified with an azide and the other an alkyne. The presence of Cu2+ induces the ligation of two ssDNAs via click reaction, leading to the formation of a forked DNA which can quantitatively generate characteristic current signatures when interacts with α-hemolysin (α-HL) nanopore. The assay facilitates a highly selective and sensitive measurement of Cu2+ without the need for labels or signal amplification. More importantly, this nanopore platform exhibits excellent performance in real-time monitoring of a copper(I) ion (Cu+)-catalyzed click reaction at the single-molecule level, by recording the current signals of the forked DNA generated by click chemistry. The proposed strategy is believed to play an important role in both nanopore sensing and characterization of chemistry reactions, especially coupling reactions.

Fluorescent sensing of mercury (II) and copper (II) ions based on DNA-templated Cu/Ag nanoclusters

Xiu-Fang Huang,Bing-Xue Ren,Chi-Fang Peng,Xin-Lin Wei,Zheng-Jun Xie

In this study, the fluorescence enhancement effect of guanine base on DNA-Cu/AgNCs was reported and fluorescent methods for the determination of Hg 2+ or Cu 2+ ion were established. The effect of adjacent and inserted guanine base (G) sequences in DNA template on the fluorescence intensity of DNA-Cu/AgNCs was investigated. It was found that G 5 sequence extended from the 5′ end of a typical C-riched DNA strand, 5′-CCCTTAATCCCC-3′ (DNA1), demonstrated fluorescence enhancement effect in the preparation of the DNA-Cu/AgNCs. Moreover, this G5-DNA-Cu/AgNCs can be used to determine Hg 2+ and Cu 2+ ions due to fluorescence quenching by the two metal ions while the DNA1-Cu/AgNCs only can determine Hg 2+ ions. Specific assays for Hg 2+ ions and Cu 2+ ions were established through masking Cu 2+ ions by EDTA and Hg 2+ ions by NaBH 4 , respectively. In Hg 2+ determination, a linear range from 5.0 nM to 40 nM was obtained with a detection limit of 1.0 nM. In Cu 2+ determination, a wide linear range from 0.01 µM to 5.0 µM was obtained with a detection limit of 5.0 nM. The recoveries of 89.6–106.8% for Cu 2+ ions and 90.6–106.2% for Hg 2+ ions were obtained in the measurements of Hg 2+ and Cu 2+ ions spiked lake samples, respectively. The results indicate the potential application of the DNA-Cu/AgNCs as a metal ions probes in real samples.

Poly(methacrylic acid)-graft-Ni3Si2O5(OH)4 multiwalled nanotubes as a novel nanosorbent for effective removal of copper(II) ions

Yu Zheng,Ping Zhang,Hairong Yue,Gang Xiang,Zexin Qian,Hongtai Li,Wei Jiang,Bin Liang,Simo O. Pehkonen,Shaojun Yuan

Ni 3 Si 2 O 5 (OH) 4 nanotubes (NTs) are synthesized by a facile one-pot hydrothermal reaction, and further functionalized by grafting of poly(methacrylic acid) (PMAA) brushes via distillation-precipitation polymerization. The resultant PMAA-grafted NTs are used as a novel nanosorbent to uptake copper(II) ions. Success in each functionalization step is ascertained by the characterization of TEM, FTIR, XPS, SEM and TGA. Batch adsorption results indicate that the adsorption of the pristine and PMAA-grafted silicate NTs is fairly fast and the equilibrium is established within 20   min, and the kinetics follow the pseudo-second-order mechanism, evidencing chemisorption as the rate-limiting step. The best interpretation for the equilibrium data is given by Langmuir isotherm, and the maximum adsorption capacity is 2.13   mmol   g −1 (136   mg   g −1 ) at pH 5 and 25   °C. The successive adsorption-desorption studies reveal that the PMAA-grafted silicate NTs are readily regenerated, and keep their adsorption and desorption efficiencies constant over five cycles. The Ni 3 Si 2 O 5 (OH) 4 - g -PMAA NTs could be directly reduced via hydrothermal reaction to achieve magnetic property for easy separation. With the inherent advantages of pH-sensitivity of PMAA, large surface area and magnetic properties of Ni 3 Si 2 O 5 (OH) 4 NTs, the PMAA-grafted silicate NTs offer a promising and highly efficient nanocomposite sorbent to remove heavy metal ions from aqueous solution.
