Ambient temperature copper-mediated living radical polymerization of acrylonitrile with Me6TREN as the reducing agent

Yun-Hai Yu,Xiao-Hui Liu,Di Jia,Bo-Wen Cheng,Yuan-Lin Ren,Fei-Jun Zhang,Hui-Na Li,Peng Chen,Shan Xie

Graphical A universal high reactive ligand, Me 6 TREN, was successfully explored as the reducing agent for the ambient temperature copper-mediated LRP of AN. The key to success is ascribed to the optimized Me 6 TREN amount and reaction conditions employed, producing ideal living free radical polymerization characteristics. The very high chain-end functionality of the resultant polymer has been confirmed by GPC and 1 H NMR analyses as well as the chain-extension reaction.

Coating Titania Nanoparticles with Epoxy-Containing Catechol Polymers via Cu(0)-Living Radical Polymerization as Intelligent Enzyme Carriers

Donghao Wang,Wenyi Ding,Kaiyue Zhou,Shutong Guo,Qiang Zhang,David M. Haddleton

Immobilization of enzyme could offer the biocatalyst with increased stability and important recoverability, which plays a vital role in the enzyme’s industrial applications. In this study, we present a new strategy to build an intelligent enzyme carrier by coating titania nanoparticles with thermoresponsive epoxy-functionalized polymers. Zero-valent copper-mediated living radical polymerization (Cu(0)-LRP) was utilized herein to copolymerize N-isopropylacrylamide (NIPAM) and glycidyl acrylate (GA) directly from an unprotected dopamine-functionalized initiator to obtain an epoxy-containing polymer with terminal anchor for the “grafting to” or “one-pot” modification of titania nanoparticles. A rhodamine B-labeled laccase has been subsequently used as a model enzyme for successful immobilization to yield an intelligent titania/laccase hybrid bifunctional catalyst. The immobilized laccase has shown excellent thermal stability under ambient or even relatively high temperature above the lower critical solution temperature (LCST) at which temperature the hybrid particles could be facilely recovered for reuse. The enzyme activity could be maintained during the repeated use after recovery and enzymatic degradation of bisphenol A was proven to be efficient. The photocatalytic ability of titania was also investigated by fast degradation of rhodamine B under the excitation of simulated sunlight. Therefore, this study has provided a facile strategy for the immobilization of metal oxide catalysts with enzymes, which constructs a novel bifunctional catalyst that will be promising for the “one-pot” degradation of different organic pollutants.

Emulsifier-Free Emulsion Polymerization-Based Copper-Mediated Molecularly Imprinted Polymer for Vitamin B1 Extraction

Le Wu,Yumeng Liu,Ziying Zhang,Liang Pan,Xinzhu Dong,Shun Feng,Chungu Zhang

Here, we present a copper-mediated magnetic molecularly imprinted polymer (Cu-mMIP) as dispersive solid-phase extracting material (dSPE) for fast, selective, and specific extraction of a metabolic biomarker vitamin B1 (VB1) in complex biological matrices. With emulsifier-free emulsion polymerization using styrene and itaconic acid as functional co-monomers, Cu 2+ as central atom, and Fe 3 O 4 nanoparticles (NPs) as core, the resulted Cu-mMIP addresses VB1's structural challenges (conformational flexibility, hydrophilicity) while enabling rapid magnetic separation (< 10 s). The Cu-mMIP demonstrates exceptional specificity for VB1, achieving an imprinting factor of 5.63 and a maximum adsorption capacity of 48.75 mg/g, 2.36-fold higher than non-copper counterparts. Adsorption equilibrium is attained within 20 min, driven by chemisorption via pseudo-second-order kinetics. Competitive binding assays reveal twofold selectivity for VB1 over its structural analog thiamine pyrophosphate and negligible interference from other B vitamins (VB2, VB9, and VB12). When applied to simulated human plasma, the method achieved a detection limit of 9 ng/mL ( S / N = 3) with recoveries of 84.3%–90.5% at three spiking levels (RSD < 5%, n = 3), effectively eliminating matrix interferences. This work establishes a scalable, high-efficiency platform for clinical nutrient analysis, combining molecular imprinting precision with metal coordination robustness, and advances separation science by addressing critical challenges in biomarker enrichment and high-throughput sample pretreatment.

Enzalutamide Sensitizes Castration-Resistant Prostate Cancer to Copper-Mediated Cell Death

Xiang Gao,Haolin Zhao,Jiao Liu,Min Wang,Zhihong Dai,Wenjun Hao,Yanlong Wang,Xiang Wang,Min Zhang,Pixu Liu,Hailing Cheng,Zhiyu Liu

Despite the initial efficacy of enzalutamide in castration-resistant prostate cancer (CRPC), inevitable resistance remains a significant challenge. Here, the synergistic induction of copper-dependent cell death (cuproptosis) in CRPC cells is reported by enzalutamide and copper ionophores (elesclomol/disulfiram). Mechanistically, enzalutamide treatment increases mitochondrial dependence in CRPC cells, rendering them susceptible to cuproptosis, as evidenced by specific reversal with the copper chelator tetrathiomolybdate. This susceptibility is characterized by hallmarks of cuproptosis, including lipoylated protein aggregation and iron-sulfur cluster protein instability. Interestingly, the mitochondrial matrix reductase, FDX1, specifically correlates with elesclomol sensitivity, suggesting a potential mechanistic divergence between the two copper ionophores. Notably, this synergistic effect extends beyond in vitro models, demonstrating efficacy in 22Rv1 xenografts, mouse Pten p53 knockout organoids. Importantly, enzalutamide significantly enhances copper ionophore-mediated cytotoxicity in enzalutamide-resistant cells. Collectively, these findings indicate that enzalutamide and copper ionophores synergistically induce cuproptosis, offering a promising therapeutic avenue for CRPC, potentially including enzalutamide-resistant cases.
