Inhibition of humic acid on copper pollution caused by chalcopyrite biooxidation

Jun Wang,Yuling Liu,Wen Luo,Xingxing Wang,Rui Liao,Shichao Yu,Maoxin Hong,Chunxiao Zhao,Baojun Yang,Yang Liu,Xueduan Liu,Guanzhou Qiu

Humic acid has the advantages of wide source, easy availability and environmental friendliness, which may be a good choice for inhibiting chalcopyrite biooxidation and alleviating copper pollution. However, there are few researches on the inhibitory effect and mechanism of humic acid on the biooxidation of chalcopyrite. In order to fill this knowledge gap, this study proposed and validated a novel method for inhibiting chalcopyrite biooxidation by means of humic acid. The results showed that the biooxidation of chalcopyrite could be effectively inhibited by humic acid, which consequently decreased the release of copper ions. Humic acid with a concentration of 120 ppm had the best inhibitory effect, which reduced the biooxidation efficiency of chalcopyrite from 40.7 ± 0.5 % to 29.3 ± 0.8 %. This in turn suggested that humic acid could effectively suppress the pollution of copper under these conditions. The analysis results of solution parameters, mineral surface morphology, mineral phases and element composition showed that humic acid inhibited the growth of Acidithiobacillus ferrooxidans , promoted the formation of jarosite and intensified the passivation of chalcopyrite, which effectively hindered the biooxidation of chalcopyrite, and would help to alleviate the pollution of copper.

Insight into the effect of potassium amyl xanthate on copper pollution caused by chalcopyrite bio-dissolution

Rui Liao,Jun Wang,Shichao Yu,Xin Sun,Shitong Liu,Baojun Yang,Guanzhou Qiu

Chalcopyrite has evolved into a crucial source of copper pollution, and xanthates inevitably coexisted with chalcopyrite in mine environments. However, the effect of xanthate on chalcopyrite bio-dissolution has not been illustrated yet. To fill this knowledge gap, the effect of potassium amyl xanthate (KAX) on copper release from chalcopyrite mediated by Acidithiobacillus ferrooxidans was investigated. The results revealed that KAX promoted chalcopyrite bio-dissolution, indicating more copper ions were released into the environment under these circumstances. KAX treatment groups displayed a lag period in ferrous ion oxidation, thereby providing a more favorable redox potential for chalcopyrite bio-dissolution. Bacterial adsorption experiments indicated the number of free cells increased with the addition of KAX, hence the Fe 3+ regeneration was accelerated. Elemental composition analyses indicated the addition of KAX was conducive to reducing the formation of passivation substance (S n 2− /S 0 ). Additionally, the reduced surface tension of solution in KAX treatment groups was beneficial for the oxidant diffusing into the pores and cracks of chalcopyrite. This study provided a better comprehension of the effect of xanthate on copper pollution caused by chalcopyrite bio-dissolution, as well as a step toward better guidance in preventing copper pollution at source.

Sodium alginate-based composite microspheres for controlled release of pesticides and reduction of adverse effects of copper in agricultural soils

Yu Du,Qizhen Zhang,Manli Yu,Bin Jiao,Fuliang Chen,Mingming Yin

Excessive copper (Cu) concentrations pose significant health risks to both plants and humans. In this study, sodium alginate (SA)-gelatin (GEL)-polyvinyl pyrrolidone (PVP)- embedded dinotefuran (DIN) microspheres were prepared using spray-drying technology . The loading content and encapsulation efficiency of optimal microspheres determined by physical modifications were 19.77% and 99.32%, respectively. In addition, the microspheres showed variable stimuli-responsive controlled release capacities in different temperatures and types of soil, as well as showed better control efficiency of larvae of Protaetia brevitarsis at pesticide application in the early stage, with the potential ability to control pest outbreaks at high temperatures. In addition, blank microspheres improved the growth and physiological activity of cucumber seedlings, reduced copper content in leaves, increased soil nutrient content, and prevented soil acidification . Further, the use of blank microspheres increased the relative abundance of soil beneficial functional bacteria communities, which mediate heavy metal (HM) immobilization/tolerance and promote plant growth. Redundancy analysis (RDA) and Spearman correlation analysis showed that these beneficial functional bacteria were mainly positively correlated with soil EC, A-N, and N–N. In summary, this study showed that the technique of combining physically modified carrier materials with pesticides has the potential to reduce Cu contamination in the surrounding agricultural soil during pesticide application, thereby reducing Cu uptake by crops.

Nighttime Warming Reduced Copper Concentration and Accumulation in Wheat Grown in Copper-Contaminated Soil by Affecting Physiological Traits

Xianghan Cheng,Feifei Liu,Peng Song,Xiaolei Liu,Qin Liu,Taiji Kou

The changes in biomass (including yield), copper (Cu) concentration, and the accumulation of wheat (Triticum aestivum. L) in response to soil Cu pollution under nighttime warming had still not been explored. Hence, this study was carried out, and these variations were analyzed from a physiological perspective. Pot trials were performed at two levels of ambient temperatures (no-warming (NT) and average nighttime warming of 0.28 °C (WT)) and two levels of soil Cu concentrations (control check without Cu application (CK) and 100 mg/kg Cu application (Cu)). Soil was collected from the carbonate cinnamon soil region of central China. The warming effects of the passive nighttime warming system were prominent, and the average increment was 0.28 °C. Antioxidant enzyme activities were promoted by warming (p< 0.05) and Cu. The highest yield was achieved in NT-Cu, mainly attributed to relatively strong root activity and photosynthesis caused by supplemental Cu, but the Cu concentration in its grains was close to the threshold (10 mg/kg) for Cu concentration in foodstuff and could present a potential food safety risk. Though nighttime warming did not increase the total biomass and yield of wheat, it decreased the Cu accumulation of wheat grown in Cu-contaminated soil, especially in grains. Moreover, WT-CK and WT-Cu increased the Cu concentration in the roots and glumes and reduced the Cu concentration in grains by 13.09% and 55.84%, respectively, probably because of a lower transpiration rate. Among them, the Cu concentration of grains in WT-Cu was the lowest and significantly lower than other applications. Our findings reveal that nighttime warming has the potential to reduce the Cu risk of grains in wheat grown in the Cu-contaminated carbonate cinnamon soil region of central China and could then provide a theoretical reference for risk assessment of food quality for wheat subjected to dual stress from nighttime warming and Cu pollution.

Remediation of copper contaminated sediments by granular activated carbon-supported titanium dioxide nanoparticles: Mechanism study and effect on enzyme activities

Zhenzhou Yin,Lei Song,Hongwei Song,Kai Hui,Zhipeng Lin,Qian Wang,Lili Xuan,Zehao Wang,Wenjian Gao

After much effort, the remediation of heavy metal contaminated sediments still remains physically hard and technically challenging issue to resolve. In this study, granular activated carbon-supported titanium dioxide nanoparticles (GAC-TiO 2 NPs) are synthesized to remedy heavy metal copper (Cu) contaminated sediments. The concentration and chemical speciation of Cu in overlying water, interstitial water and contaminated sediments are fully assessed to examine the remediation effect of GAC-TiO 2 NPs. The GAC-TiO 2 NPs are separated from GAC-TiO 2 NPs-remedied sediments and characterized by X-ray photoelectron spectra (XPS), which reveals the mechanism of GAC-TiO 2 NPs remedy Cu Contaminated sediments. The results show that after 35 days adding 20% GAC-TiO 2 NPs to contaminated sediments, the Cu concentration in the overlying water and interstitial water decreases 89.47% and 83.52%, respectively, and the exchangeable fraction (F-1) of Cu in sediments decreases from 43.91% to 7.49%. The percentage of residual fraction (F-4) increases sharply from 42.79% to 80.30%. XPS results show that hydroxyl (–OH) plays an important role in the remediation process. The synergistic effects of pH, phosphorus concentration and organic matter (OM) content on the remediation effect are explored. When the pH value is 8, phosphorus concentration is 0.32 mg/L and OM content is 151.2 g/kg, adding 20% GAC-TiO 2 NPs achieves the best remediation effect on Cu contaminated sediment. Biological enzyme-activity experiments prove that GAC-TiO 2 NPs not only reduce the bioavailability and biotoxicity of Cu, but also effectively suppress the negative effects of granular activated carbon (GAC) on enzyme activities. All these results indicate that GAC-TiO 2 NPs is an environmentally friendly remediation material for Cu contaminated sediments with high-potential applications.

A proteomic-based investigation of potential copper-responsive biomarkers: Proteins, conceptual networks, and metabolic pathways featuring Penicillium janthinellum from a heavy metal-polluted ecological niche

Xin Feng,Jian Xu,Yu Liang,Guo-Li Chen,Xian-Wei Fan,You-Zhi Li

Graphical Our work not only provides the potential protein biomarkers indicating the Cu pollution, target metabolic steps for engineering the Cu-tolerant fungi during bioremediation, but also presents clues for insight into heavy metal tolerance mechanisms of other eukaryotes. Filamentous fungi-copper (Cu) interactions are very important in the formation of natural ecosystems and the bioremediation of heavy metal pollution. However, important issues at the proteome level remain unclear. We compared six proteomes from Cu-resistant wild-type (WT) Penicillium janthinellum strain GXCR and a Cu-sensitive mutant (EC-6) under 0, 0.5, and 3 mmol/L Cu treatments using iTRAQ. A total of 495 known proteins were identified, and the following conclusions were drawn from the results: Cu tolerance depends on ATP generation and supply, which is relevant to glycolysis pathway activity; oxidative phosphorylation, the TCA cycle, gluconeogenesis, fatty acid synthesis, and metabolism are also affected by Cu; high Cu sensitivity is primarily due to an ATP energy deficit; among ATP generation pathways, Cu-sensitive and Cu-insensitive metabolic steps exist; gluconeogenesis pathway is crucial to the survival of fungi in Cu-containing and sugar-scarce environments; fungi change their proteomes via two routes (from ATP, ATP-dependent RNA helicases (ADRHs), and ribosome biogenesis to proteasomes and from ATP, ADRHs to spliceosomes and/or stress-adapted RNA degradosomes) to cope with changes in Cu concentrations; and unique routes exist through which fungi respond to high environmental Cu. Further, a general diagram of Cu-responsive paths and a model theory of high Cu are proposed at the proteome level. Our work not only provides the potential protein biomarkers that indicate Cu pollution and targets metabolic steps for engineering Cu-tolerant fungi during bioremediation but also presents clues for further insight into the heavy metal tolerance mechanisms of other eukaryotes.

Response of microbial communities based on full-scale classification and antibiotic resistance genes to azithromycin and copper combined pollution in activated sludge nitrification laboratory mesocosms at low temperature

Yu-Xi Gao,Xing Li,Jun-Ru Zhao,Zhong-Xing Zhang,Xiao-Yan Fan

This study aimed to investigate the short-term response of abundant-rare genera and antibiotic resistance genes (ARGs) to azithromycin (AZM, 0.05–40 mg/L) and copper (1 mg/L) combined pollution in activated sludge nitrification system at low temperature. Nitrification was as expected inhibited in stress- and post-effects periods under AZM concentration higher than 5 mg/L. Abundant and rare taxa presented dissimilar responses based on full-scale classification. Conditionally rare or abundant taxa (CRAT) were keystone taxa. Relative abundance of ammonia-oxidizing archaea increased, and three aerobic denitrifying bacteria ( Brevundimonas , Comamonas and Trichococcus ) were enriched (from 9.83% to 68.91% in total). Ammonia nitrogen assimilating into Org-N and denitrification may be nitrogen pathways based on predict analysis. 29 ARGs were found with more co-occurrence patterns and high concentration of AZM (greater than 5 mg/L) caused their proliferation. Importantly, expect for some abundant taxa, rare taxa, potential pathogens and nitrogen-removal functional genera were the main potential hosts of ARGs.

Photoelectrochemical Determination of Cu2+ Using a WO3/CdS Heterojunction Photoanode

Jun Wang,Yining Pan,Liangxing Jiang,Min Liu,Fangyang Liu,Ming Jia,Jie Li,Yanqing Lai

Copper ions are not only physiologically essential for life but also hazardous materials causing a series of neurodegenerative diseases. Photoelectrochemical (PEC) detection has attracted a large amount of focus as a potential strategy to develop Cu2+ ion sensors. However, relatively low photocurrent signals with poor antidisturbance ability and the limited concentration range have prevented its practical applications. Here, we designed a WO3/CdS heterojunction photoanode for the PEC determination of Cu2+ in aqueous solution through a simple two-step chemical bath deposition method. The obtained WO3/CdS photoanode had a nanoplate morphology and showed an enhanced photoresponsivity with a photocurrent density of 1.5 mA/cm2 at 1.23 V versus RHE under illumination. Naturally, it exhibited a low detection limit (0.06 μM) and wider range (0.5 μM to 1 mM) for Cu2+ PEC detection first, suggesting that the WO3/CdS heterojunction photoanode is a promising tool to monitor copper pollution in natural environments.

Versatile, Oxygen-Insensitive Surface-Initiated Anionic Polymerization to Prepare Functional Polymer Brushes in Aqueous Solutions

Chong Liu,Fang Cheng,Bo Liu,Dongdong Gao,Gang Cheng,Chunmei Li,Huanan Wang,Wei He

Surface-initiated polymerization is an attractive approach to achieve desired interfacial compositions and properties on a wide range of substrates and surfaces. Due to mild reaction conditions, multiple surface-initiated polymerization methods, such as atom-transfer radical polymerization (ATRP), reversible addition–fragmentation chain-transfer polymerization, and so forth, have been developed and studied in academia and industry. However, the current methods require the combination of metal catalysts, special initiators, and oxygen removal. Herein, we developed a surface-initiated carbanion-mediated anionic polymerization (SI-CMAP), which can be conducted in aqueous solutions in the presence of oxygen without the need for metal catalysts. Zwitterionic 2-(N-3-sulfopropyl-N,N-dimethyl ammonium)ethyl methacrylate (SBMA) was selected as a model monomer to develop and demonstrate this strategy. The vinyl sulfone (VS) groups displayed on substrate surfaces reacted with N-methylimidazole (NMIM), which was used as the in situ initiator. The polymerization mechanism was extensively studied from many aspects at room temperature, including the changes in reaction conditions, factors affecting the polymerization extent, and substrate surfaces. We also demonstrated the compatibility of this method with a broad spectrum of monomers ranging from SBMA to other acrylates and acrylamides by using glycine betaine as a reaction additive. This method was also evaluated for the preparation of polymer-coated nanoparticles. For polymer-coated silica nanoparticles, their hydrodynamic diameter, copper contamination, and effects of salt and protein concentrations were compared with SI-ATRP in parallel. SI-CMAP in aqueous solutions in air and the absence of metal catalysts make this method sustainable and cost-effective. We believe that SI-CMAP can be readily adapted to the industrial surface coating and large-scale nanoparticle preparation under mild conditions.
