The coronavirus disease 2019 (COVID-19) pandemic caused by severe acute respiratory coronavirus 2 (SARS-CoV-2) is still raging all over the world. Hence, the rapid and sensitive screening of the suspected population is in high demand. The nucleocapsid protein (NP) of SARS-CoV-2 has been selected as an ideal marker for viral antigen detection. This study describes a lateral flow immunoassay (LFIA) based on colloidal gold nanoparticles for rapid NP antigen detection, in which sensitivity was improved through copper deposition-induced signal amplification. The detection sensitivity of the developed LFIA for NP antigen detection (using certified reference materials) under the optimized parameters was 0.01 μg/mL and was promoted by three orders of magnitude to 10 pg/mL after copper deposition signal amplification. The LFIA coupled with the copper enhancement technique has many merits such as low cost, high efficiency, and high sensitivity. It provides an effective approach to the rapid screening, diagnosis, and monitoring of the suspected population in the COVID-19 outbreak.Keywords:SARS-CoV-2;nucleocapsid protein;signal amplification;copper deposition

Surface metal deposition is an important method for tuning the optical property of plasmonic nanoparticles. In this work, we tried to deposit Cu shell on the surface of gold nanorods (Au NRs) by dihydronicotinamide adenine dinucleotide (NADH)-mediated reaction. Cu 2+ were reduced to Cu 0 through the gold-catalyzed oxidization of NADH. The deposition of Cu shell onto the Au NRs surface resulted in the obvious red-shift of longitudinal localized surface plasmon resonance peak. And the red-shift of longitudinal peak increased with the concentrations of NADH and Cu 2+ . The TEM images showed that a transparent layer structure with a different contrast around Au NRs was clearly displayed. And this transparent layer structure with the low contrast was generally considered to be pure Cu shell. However, the results from energy dispersive spectrometer (EDS) and X-ray photoelectron spectroscopy (XPS) indicated that the transparent layer was not just pure Cu composition, but contained NADH/NAD + and Cu 2+ components. This grown complex dielectric shell resulted in the reduced surface enhanced Raman scattering (SERS) activity of Au NRs. Based on the decreased SERS intensity, we demonstrated a SERS method for Cu 2+ detection using R6G as Raman reporter. The decreased SERS intensity had a linear correlation with the concentration of Cu 2+ in the range of 10 −6 –10 −2 M, with a detection limit of 3.89 × 10 −6 M. This study provides new insights about Cu deposition on Au NRs, which will further promote the spectral sensing applications with gold-catalyzed Cu 2+ reduction on plasmonic nanoparticles by NADH.

The fabrication of patterned metal films is essential for a wide range of applications. Here, a novel solution-processing method is presented to achieve the area-selective deposition of copper on aluminum substrates via an in situ Al/CuSO 4 /Graphite voltaic cell template. And the copper ions preferentially deposit in the graphite-covered area due to the formed latent image of the potential difference between the graphite-covered area and the blank area on the aluminum surface. Benefiting from the low adhesive force of graphite, the formed copper layer can be easily peeled off and transferred to a flexible substrate to obtain the patterned conductive metal films. By studying the electrochemical properties of different surfaces and the in situ microscopic observation, four stages of electroplating process are proposed. More importantly, a positive correlation between the deposition speed and graphite coverage density is confirmed. This further enables a regional control of deposition thickness in addition to area-selective deposition, which means a 3D controllable metal deposition. Other forms of electroactive carbon are also demonstrated to replace graphite in this method, which offers a versatile and accessible technique to fabricate large-area flexible patterned metal films with defined feature size and controlled resolution from microscale to macroscale.

O-GlcNAcylation is a very important post-translational modification of nucleocytoplasmic proteins. Aberrant O-GlcNacylation has been found in a variety of human diseases including many types of cancers, diabetes, and Alzheimer's disease. However, analysis of protein O-GlcNacylation still remains a great challenge due to its low levels and labile nature. Herein, a highly sensitive and reliable colorimetric method is developed by use of wheat germ agglutinin (WGA) lectin for specific recognition of O-GlcNAc and gold nanoparticle (AuNP)-catalyzed copper deposition for signal amplification. In presence of the target molecule, AuNP is specifically introduced onto the sensing surface and then catalyzes the copper deposition. An Au@Cu core–shell structure is produced from the deposition. The obtained copper shell on AuNP is dissolved by FeCl3 due to the oxidation–reduction reaction between copper and Fe3+, resulting in Cu2+ and Fe2+, which subsequently form a red coordination complex in the presence of bathophenanthrolinedisulfonic acid disodium salt (BPT2−). Thus, the amount of the red complex is proportional to the level of O-GlcNAc. Qualitative O-GlcNAc detection is realized by direct readout with the naked eye, while the quantitative analysis is achieved by measurement of the absorbance using a UV-Vis spectrophotometer. With increasing time, more and more copper is deposited on the surface of AuNP, thus achieving significant signal amplification. Under optimal conditions, an ultralow limit of detection (LOD) down to 0.02 pg mL−1 is achieved with GlcNAc-conjugated BSA as a model analyte. The developed platform was also successfully applied to analyze O-GlcNAcylation levels in prostate cancer cell line PC-3 and normal cell line RWPE-1. The results obtained by the proposed method are in good agreement with the data from protein microarray assays, demonstrating its great potential for sensitive and reliable O-GlcNAcaylation detection in fundamental research and in cancer diagnosis.

Cuproptosis is an emerging programmed cell death, displaying great potential in cancer treatment. However, intracellular copper content to induce cuproptosis is unmet, which mainly ascribes to the intracellular pumping out equilibrium mechanism by copper exporter ATP7A and ATP7B. Therefore, it is necessary to break such export balance mechanisms for desired cuproptosis. Mediated by diethyldithiocarbamate (DTC) coordination, herein a strategy to efficiently assemble copper ions into polydopamine nanostructure (PDA-DTC/Cu) for reprogramming copper metabolism of tumor is developed. The deposited Cu 2+ can effectively trigger the aggregation of lipoylated proteins to induce cuproptosis of tumor cells. Beyond elevating intracellular copper accumulation, PDA-DTC/Cu enables to break the balance of copper metabolism by disrupting mitochondrial function and restricting the adenosine triphosphate (ATP) energy supply, thus catalytically inhibiting the expressions of ATP7A and ATP7B of tumor cells to enhance cuproptosis. Meanwhile, the killed tumor cells can induce immunogenic cell death (ICD) to stimulate the immune response. Besides, PDA-DTC/Cu NPs can promote the repolarization of tumor-associated macrophages (TAMs ) to relieve the tumor immunosuppressive microenvironment (TIME). Collectively, PDA-DTC/Cu presented a promising “one stone two birds” strategy to realize copper accumulation and inhibit copper export simultaneously to enhance cuproptosis for 4T1 murine breast cancer immunotherapy.
