The phytotoxicity of aluminum (Al) to plants is well known. Auxin accumulation and reactive oxygen species (ROS) burst induced by Al toxicity are the key factors in root growth inhibition. Yucasin, an auxin synthesis inhibitor, effectively ameliorates Al phytotoxicity in tomato seedlings. However, the physiological mechanisms by which yucasin alleviates Al phytotoxicity in tomatoes remain elusive. Here, we examined the regulatory mechanisms of yucasin involved in tomato seedling growth under Al conditions through phenotypic, plant physiology analysis, and cellular experiments. Exogenous indole-3-acetic acid (IAA) application increased Al accumulation in tomato seedling roots, while yucasin decreased Al accumulation. Yucasin application reduced Al-induced ROS accumulation, lipid peroxidation, and cell death, enhanced root viability, and promoted tomato seedling root growth. Further, yucasin enhanced the antioxidant enzyme activities of superoxide dismutase, catalase, and peroxidase in plants under Al conditions. The results suggest that yucasin improves the scavenging capacity of ROS by maintaining the activities of antioxidative enzymes. This study elucidates the physiological mechanism by which yucasin alleviates Al phytotoxicity, highlighting its potential to enhance plant tolerance under acidic Al conditions.

Melatonin has emerged as an essential molecule in plants, due to its role in defence against metal toxicity. Aluminium (Al) and cadmium (Cd) toxicity inhibit rapeseed seedling growth. In this study, we applied different doses of melatonin (50 and 100 µ m ) to alleviate Al (25 µ m ) and Cd (25 µ m ) stress in rapeseed seedlings. Results show that Al and Cd caused toxicity in rapeseed seedling, as evidenced by a decrease in height, biomass and antioxidant enzyme activity. Melatonin increased the expression of melatonin biosynthesis-related Brassica napus genes for caffeic acid O-methyl transferase ( BnCOMT ) under Al and Cd stress. The genes BnCOMT-1 , BnCOMT-5 and BnCOMT-8 showed up-regulated expression, while BnCOMT-4 and BnCOMT-6 were down-regulated during incubation in water. Melatonin application increased the germination rate, shoot length, root length, fresh and dry weight of seedlings. Melatonin supplementation under Al and Cd stress increased superoxide dismutase, catalase, peroxidase, ascorbate peroxidase, proline, chlorophyll and anthocyanin content, as well as photosynthesis rate. Both Cd and Al treatments significantly increased hydrogen peroxide and malondialdehyde levels in rapeseed seedlings, which were strictly counterbalanced by melatonin. Analysis of Cd and Al in different subcellular compartments showed that melatonin enhanced cell wall and soluble fractions, but reduced the vacuolar and organelle fractions in Al- and Cd-treated seedlings. These results suggest that melatonin-induced improvements in antioxidant potential, biomass, photosynthesis rate and successive Cd and Al sequestration play a pivotal role in plant tolerance to Al and Cd stress. This mechanism may have potential implications in safe food production.

Aluminum (Al) toxicity represents a significant constraint on crop growth in acidic soils. Al stress can stimulate the biosynthesis of melatonin (MT) in rice ( Oryza sativa L.). The exogenous application of 20 μM MT markedly reduces Al accumulation in root tips and effectively mitigates Al stress. By enhancing the levels of root hemicellulose and down-regulating the expression of two genes sensitive to aluminum rhizotoxicity ( OsSTAR1 and OsSTAR2) , which function to obscure Al binding sites within the cell wall, MT significantly improves the fixation of Al by the cell wall. Furthermore, MT diminishes cytoplasmic entry of Al by reducing expression levels of nramp aluminum transporter 1 gene ( OsNRAT1 ), while inducing expression of an aluminum-sensitive gene ( OsALS1 ), thereby promoting vacuolar compartmentation to lower cytoplasmic Al concentration. Additionally, MT substantially decreases Nitric oxide (NO) accumulation induced by Al in roots. The alleviation effect exerted by MT on Al toxicity can be replicated through administration of the NO scavenger carboxyl-PTIO (c-PTIO). Moreover, the Al-sensitive phenotype exhibited in the osgsnor mutant, coupled with the absence of mitigation by MT, further substantiates that MT alleviates Al toxicity in rice primarily via regulation of NO content, potentially relying on NO signal transduction pathways. In summary, MT plays a pivotal role in modulating rice resistance to Al toxicity and stimulates multiple pathways to enhance tolerance against this abiotic stressor. This study provides new insights into understanding mechanisms underlying rice resistance to abiotic stresses.

Increasing aluminum (Al) use and its effects on aquatic systems have been a global issue, however the Al impacts on submerged plants and their ecological functions were poorly understood. Aquatic simulation experiments were performed to study Al-toxicity on the germination and seedling morphological and physiological characteristics of Vallisneria natans , and investigate their synergistic effect on nitrogen (N), phosphorus (P) change and microbial community in sediment. The seeds germination characteristics, growth and physiological parameters of seedlings, including root activity, were significantly affected by alum treatments and the inhibition levels increased with Al 3+ concentration. The Al accumulation in roots and leaves were significantly different. Al 3+ concentration above 0.3 mg/L showed toxic to V. natans . TN, TP, IP, Fe/Al-P contents in sediments varied markedly under co-existence of Al and V. natans . Additionally, the relative abundance of sediment microbial community related to N, P cycle was effected. Results concluded that the increasing aquatic Al-concentration inhibits growth and propagation of submerged plants and the ecological restoration effect, and exerts synergistic effect with submerged plants on N, P components in sediments. Such findings were helpful for Al ecological evaluation, and were instructive for the submerged plants restoration in shallow eutrophic lakes with Al input.

Soil acidification is a problem widely occurring worldwide, which severely threaten food security and agricultural sustainability. Calcium lignosulfonate (CaLS), a cheap and ecofriendly compound, is used for the first time to amend acid soil by utilizing its unique organic and inorganic functional moieties simultaneously. Both column leaching and incubation experiments were conducted to investigate the comparative effects of CaLS (four rates at 5, 10, 15, 20 g kg −1 ) and compared with conventional amendments, including gypsum (5 g kg −1 ), lignin (5 g kg −1 ), L + G (each at 5 g kg −1 ), and control. The soil pH, exchangeable acidity and base cations, organic carbon, and different Al fractions were determined to unravel the ameliorative performance and mechanism of the treatments. Regardless of application modes and dosages, the results demonstrated that CaLS incorporation significantly increased soil pH, exchangeable Ca 2+ , cation exchange capacity, and organic carbon and decreased the contents of exchangeable acidity, especially exchangeable Al 3+ . The ameliorative mechanism was that amendment material led to the displacement of H + and Al 3+ off soil colloids by Ca 2+ . These released H + and Al 3+ which complexed with lignosulfonate anions into soluble organo-Al were all quickly leached from the soil column. The CaLS addition enhanced the transformation of exchangeable Al 3+ and low-to-medium organo-Al complexes into highly stable organically bound fractions and immobilized into the soil. The complexing of CaLS functional groups with Al 3+ impeded Al 3+ from undergoing hydrolysis to produce more H + . As an environmental-friendly material, CaLS can be a promising amendment for soil acidity and Al toxicity amelioration.

Brassinosteroids (BRs) are growth-promoting hormones that exhibit high biological activities across various plant species. BRs shield plants against various abiotic stresses. In the present study, the effect of BRs against aluminum (Al) toxicity was investigated through seed priming with 24-epibrassinolide (0.01 μM) in two different rice cultivars. BRs application was found effective in confronting plants from Al toxicity (400 μM). The rice seeds primed with BRs showed enhancement in seed germination energy, germination percentage, root and shoot length, as well as fresh and dry weight under Al-absence and Al-stressed conditions as compared to water-priming. Especially under Al stress, BRs priming promoted the growth of rice seedlings more obviously. Al toxicity significantly increased the Al contents in seedling root and shoot, as well as the MDA concentration, H 2 O 2 production, and the activities of antioxidative enzymes including ascorbate peroxidase, catalase, and peroxidase. Meanwhile, the photosynthetic pigments of seedling reduced under Al stress. When compared to sensitive cultivar (CY-927), these modifications were more obvious in the tolerant variety (YLY-689). Surprisingly, BRs were able to alleviate the Al injury by lowering MDA and H 2 O 2 level and increasing antioxidant activities and photosynthetic pigments under Al stress. The results on antioxidant activities were further validated by gene expression study of SOD-Cu-Zn , SOD-Fe 2 , CATa , CATb , APX02 , and APX08 . It suggested that BRs were responsible for the mitigation of Al stress in rice seedlings by inducing antioxidant activities with an effective response to other seed growth parameters and reduced Al uptake under induced metal stress.

Soil acidification is a worldwide eco-environmental problem detrimental to plant growth and threatening food security. In this study, calcium poly(aspartic acid) (PASP-Ca) and calcium poly-γ-glutamic acid (γ-PGA-Ca) were obtained through cation exchange and used to mitigate soil acidity owing to high solubility and complexing capability. Three rates at 6.7, 13.4, and 20.1 g kg −1 , denoted as PASP-Ca1, PASP-Ca2, and PASP-Ca3, and γ-PGA-Ca (7.4 g kg −1 ) were surface-applied and compared with conventional lime (CaCO 3 , 2.5 g kg −1 ) along with control in two soil layers (top soil 0–10 cm, subsoil 10–20 cm). After leaching, various soil properties and aluminum fractions were measured to assess their ameliorative performance and mechanisms. Although lime achieved the highest soil pH (6.91) in the topsoil followed by PASP-Ca and γ-PGA-Ca (pH: 5.57–6.33), it had less effect on subsoil increase (5.3) vs. PASP-Ca and γ-PGA-Ca (pH: 5.44–5.74). Surface-applied PASP-Ca demonstrated efficiency in elevating soil pH and reducing exchangeable acidity, mainly as exchangeable Al 3+ , whereas γ-PGA-Ca addition superiorly improved soil pH buffering capacity (pHBC). Moreover, PASP-Ca and γ-PGA-Ca addition improved organic carbon by 34.4–44.9%, available P by 4.80–20.71%, and cation exchange capacity (CEC) by 6.19–29.2%, thus greatly enhanced soil fertility. Ca 2+ from polyAA-Ca promoted the displacement of exchangeable Al 3+ or H + from soil colloid, which were subsequently complexed or protonated and facilitated leaching. Additionally, the transformation into stable organo-aluminum fractions via complexation inhibited further hydrolysis. Under PASP-Ca or γ-PGA-Ca addition, the saturation of aluminum in cation exchange complex was reduced 2.91–7.81% compared to the control without addition amendments. Thus, PASP-Ca and γ-PGA-Ca can serve as potent ameliorants to alleviate soil acidity and aluminum toxicity for sustainable agricultural development.
