Rapid, low-cost, and in-situ detection of illegal residues on aquatic product surfaces remains a significant challenge. Herein, we developed a versatile surface-enhanced Raman scattering (SERS) sensor by integrating instant interfacial self-assembled silver nanostar film onto flexible polytetrafluoroethylene filter membrane (AgNS/PTFE) for sensitive detection of malachite green (MG) and crystal violet (CV) in crayfish. The entire process, from room-temperature substrate preparation to detection completion, takes less than 20 min. The AgNS/PTFE flexible substrate not only provides excellent hydrophobicity for signal enhancement with minimal sample volumes (2 μL) but also ensures good uniformity, resulting in a homogeneous and dense distribution of SERS-active hot spots with superior anti-interference properties and mechanical durability. By simple “drop/wipe” operations and data visualization treatment, this method achieved detection limits as low as 10 pg/mL for MG and 7 pg/mL for CV, with in-situ analysis in spiked and actual crayfish samples, demonstrating significant potential for food safety applications.

The detection performances of noble metal-based surface enhanced Raman spectroscopy (SERS) devices are determined by the compositions and geometries of the metal nanostructures, as well as the substrates. In the current study, long spiky Au-Ag alloy nanostars were synthesized, and both core diameters and spike lengths were controlled by Lauryl sulfobetaine concentrations (as the nanostructure growth skeleton). The long spiky star geometries were confirmed by transmission electron micrograph images. Elements energy dispersive spectrometer mapping confirmed that Au and Ag elements were inhomogeneously distributed in the nanostructures and demonstrated a higher Ag content at surface for potential better SERS performance. Selected synthesized spiky nanostars were uniformly assembled on multi-mode silica fiber for probe fabrication by silanization. The SERS performance were characterized using crystal violet (CV) and rhodamine 6G (R6G) as analyte molecules. The lowest detection limit could reach as low as 10−8M, with a 6.23 × 106enhancement factor, and the relationship between analyte concentrations and Raman intensities was linear for both CV and R6G, which indicated the potential qualitative and quantitative molecule detection applications. Moreover, the fiber probes also showed good reproducibility and stability in the ambient atmosphere.

Certain diuretics have been identified as potential contaminants in pharmaceutical and food products, posing a risk to food safety and public health. In this study, a surface-enhanced Raman spectroscopy (SERS) detection method based on Silver nanostars (Ag NSs) was developed for the detection of motion-prohibited diuretics in food and biological samples. Ag NSs synthesized via a simple solution-based method exhibited a star-like morphology with high-density "hot spot", achieving an enhancement factor of 1.0 × 10 5 —3 orders of magnitude higher than that of traditional spherical silver nanoparticles. A highly sensitive SERS detection system was established for bumetanide (limit of detection: 7.08 × 10 −9 g/mL), triamterene (1.13 × 10⁻⁸ g/mL), and dorzolamide (1.04 × 10⁻⁸ g/mL) for the first time. Characteristic fingerprint peaks enabled accurate molecular identification. The method was validated in milk and urine with recoveries of 94.2 %–104.9 %, and relative standard deviation (RSD) < 7 %. Results were consistent with those obtained by HPLC-HRMS/MS, confirming the method's reliability. The proposed system offers advantages of rapid detection (<10 min), low cost, and minimal matrix interference, enabling efficient multi-residue analysis in complex samples. This work provides a promising tool for food safety monitoring and anti-doping surveillance, contributing to effective regulatory enforcement.

In surface-enhanced Raman scattering (SERS) measurements, the creation of “hot spots” and the effective concentration of analytes in these areas are crucial for enhancing detection sensitivity. In this work, a novel composite with highly SERS activity, Ag nanospheres decorated Ag nanostar/ZIF-8 composite (Ag NS/ZIF-8/Ag) was achieved via first coating the Ag nanostars with the metal-organic framework ZIF-8, known for its strong molecular adsorption capabilities, followed by the electrostatic self-assembly of Ag nanospheres. The composites produce numerous hotspots due to the abundant plasmonic nanogaps, including the interstitial space between the Ag nanostar and Ag nanospheres, as well as internanogaps between neighboring Ag nanospheres, resulting in outstanding SERS performance. It was successfully applied for the quantitative and simultaneous detection of diquat and thiram in fruit juice, achieving detection limits of 5.0 × 10 −8 mol/L and 1 × 10 −8 mol/L, respectively, with recovery rates ranging from 86.6 % to 118.4 %. Notably, under UV light exposure, the composite was able to degrade pesticides efficiently within 80 min. Additionally, its reusability was demonstrated by sustaining strong SERS activity over five continuous detection cycles. These results emphasize the capability of Ag NS/ZIF-8/Ag composites for sensitive pesticide detection via SERS.
