Large-scale preparation and multi-dimensional characterization of high-purity mycotoxin deoxynivalenol in rice culture inoculated with Fusarium graminearum

Zhiyong Zhao,Suquan Song,Na Liu,Qinxiong Rao,Jiafa Hou,Sarah De Saeger,Aibo Wu

Food safety monitoring and toxicological research of mycotoxins are still in need of large quantities of high-purity deoxynivalenol (DON). To attain this purpose, a rapid, economical and reproducible purification method was developed for large-scale production of DON from rice culture inoculated with a DON-producing Fusarium graminearum strain JYH. The inoculated rice culture was first extracted with acetonitrile–water (84/16, v/v). The extracts were evaporated to dryness on a rotary evaporator after ethyl acetate partitioning and then dissolved in water followed by the final purification procedure through preparative high performance liquid chromatography and montmorillonite treatment. A combined approach of ultraviolet spectrometry (UV), ultra high performance liquid chromatography (UHPLC), mass spectrometry (MS) and nuclear magnetic resonance (NMR) spectroscopy was applied for the multi-dimensional characterization of the target compound. As a result, the recovery of DON from the crude extract to the final product was up to 70%. An amount of 150 mg DON with the desirable purity of 98.93% could be obtained from 100 g of rice culture, which possessed identical immunochemical characteristics compared to a certified commercial DON standard. This proposed strategy might act as a valuable reference to obtain rather expensive compounds in a straightforward way.

Trans-2-decenal inhibits Alternaria alternata through disruption of redox homeostasis and membrane integrity

Hamza Tariq,Peiyu Chen,Gang Wang,Siyuan Xie,Lanning Wang,Jingkun Lv,Fei Wang,Muhammad Saqib Bilal,Abdur Rashid Khan,Nasir Ahmed Rajput,Danyu Shen,Daolong Dou

BACKGROUND Alternaria alternata is a globally distributed plant pathogen infecting >400 plant species. Trans -2-decenal is a notable plant-derived secondary metabolite with strong volatility and antifungal properties, although its specific activity and mode-of-action against A. alternata remain unclear. RESULTS Toxicity assays showed that trans -2-decenal inhibited A. alternata with a median inhibitory concentration (IC 50 ) of 12.23 mg L −1 . Treated with trans -2-decenal resulted in hyphal abnormalities, reduced spore production and suppressed spore germination. Mechanistically, it disrupted fungal cell membrane integrity, as confirmed by propidium iodide staining and increased leakage of DNA and soluble proteins in conductivity assays. Further investigations revealed that trans -2-decenal disrupted redox homeostasis by inducing reactive oxygen species (ROS) accumulation. DCFH-DA fluorescence staining showed elevated ROS levels, whereas antioxidant enzyme assays revealed dose-dependent changes, including decreased superoxide anion (O 2 . − ) and catalase (CAT) activity, and significantly increased hydrogen peroxide (H 2 O 2 ), superoxide dismutase (SOD) and malondialdehyde (MDA) levels, collectively indicating oxidative stress and damage. Transcriptome analysis identified 2319 differentially expressed genes (DEGs), with gene ontology (GO) enrichment highlighting inhibition of pathways associated with oxidative stress response, transmembrane transport and ribosome biogenesis. Molecular docking further suggested interactions between trans -2-decenal and key antioxidant enzyme and membrane transporter of A. alternata . In vivo assays showed that 32 mg L −1 trans -2-decenal fumigation achieved protective and curative efficacies of 97.14% and 96.54%, respectively, against yam leaf spot disease. CONCLUSION Trans -2-decenal inhibited A. alternata through a multitarget synergistic mechanism, providing theoretical support for its development as a plant-derived biofumigant and a promising tool for the green management of A. alternata -induced diseases. © 2025 Society of Chemical Industry.

Early identification of fungal leaf blight disease (Alternaria alternate) on Platycladus orientalis plants by using gas chromatography-ion mobility spectrometry

Chengyu Zheng,Qin'an Zhou,Wenqing Shao,Jing Zhang,Jun Wang

Platycladus orientalis (L.) is a typical urban greening tree in Asia, but the young leaves of P. orientalis are   vulnerable to pathogenic fungi with hidden early   signs. In this study, we investigated the volatile organic compounds (VOCs) emissions of the Alternaria alternate infected P. orientalis plants, for the first time, using the gas chromatography-ion mobility spectrometry (GC-IMS) combined with chemometric approaches. Results showed that the A. alternate infection could reduce the VOCs release of P. orientalis leaves. The α-pinene and β-pinene emissions were down to 70% and 55% of controls on day 10, respectively. The destructive inoculated plants responded more to the A. alternate , both   in the VOCs emissions and the fungal growth. The (Z)-3-hexenol, n-hexanol, α-pinene, and β-pinene can serve as potential markers for infection discrimination, and their IMS signal intensities were calibrated with corresponding volume concentrations ranging from 0 to 250 μg/L. The PLS-DA model based on GC-IMS fingerprints can correctly classify at least 83% of samples in infection durations after fungal inoculation for the first 10 days. The PLSR model can predict the fungal colony counts in the P. orientalis needles (R c 2 >0.97 and R p 2 >0.92) with good robustness, whose RMSEP was lower than 0.68 (×10 6 CFU g −1 ) in predict set. In a word, the GC-IMS-based approach had the potential to evaluate A. alternate infection in P. orientalis , and the variation of potential markers can provide reference to its fungal infection monitor with   millisecond response in the forest.

Inactivation mechanism of Alternaria alternata by dielectric barrier discharge plasma and its quality control on fresh wolfberries

Yuhang Du,Shuna Mi,Huihui Wang,Fangwei Yang,Hang Yu,Yunfei Xie,Yahui Guo,Yuliang Cheng,Weirong Yao

Fresh wolfberry is a super fruit with excellent nutritive and medicinal values, but highly perishable and difficult to store. In this research, dielectric barrier discharge plasma (DBD) was used to control the spoilage and mycotoxin of Alternaria alternata ( A. alternata ), which frequently and heavily contaminated fresh wolfberries. Results showed that DBD treatment could inactivate A. alternata with a maximum reduction of 2.26 log CFU/mL (700V,480s). Its spore inactivation behavior most fitted the Log-logistic model (R 2  > 0.993). DBD disrupted A. alternata cell membrane integrity, oxidized membrane-associated substances and increased membrane permeability. The reactive oxygen species produced by DBD caused the accumulation of reactive oxygen metabolites in the spores and the inhibition of antioxidant enzyme activity. The average content of four mycotoxins on wolfberries could be reduced by 62.84% after the 60s treatment and storage for 6 days, while the decay rate decreased by 30.66%, and the firmness, TSS and DPPH free radical scavenging increased by 11.07%, 5.88% and 19.20%, respectively. Therefore, DBD could be used as a new technology to inactivate A. alternata , reduce its mycotoxin, and improve agricultural product quality.

Deciphering Alternaria metabolic responses in microbial confrontation via an integrated mass spectrometric targeted and non-targeted strategy

Ye Tian,Mohamed F. Abdallah,Marthe De Boevre,Kris Audenaert,Cheng Wang,Sarah De Saeger,Aibo Wu

Microbial confrontation is ubiquitously present in nature, such as mycoparasitism, competition and antibiosis between biocontrol agents and microbial pathogens. However, the internal metabolic responses of fungal pathogens under microbial interaction scenario have been scarcely investigated. In this study, we set up an integrated mycotoxin analysis and non-targeted metabolomics workflow to decipher metabolic changes of Alternaria pathogens when confronted with selected Trichoderma strains , as well as mycotoxin metabolization in the Trichoderma spp. Results demonstrated that Trichoderma spp. significantly influenced mycotoxin production and whole metabolome of Alternaria pathogens when in cocultivation, and one Trichoderma strain could metabolize alternariol into its hydroxylated form. These differential metabolites revealed fungal physiological alternations in various confrontation conditions . In all, a MS-based strategy was proposed to investigate microbial metabolic profiles under fungal/fungal and fungal/mycotoxin cocultivation, and this generic methodology would be significant for understanding the occurrence and change of food contaminants in microbial interactions.

Nature Nano-Barrier: HPMC/MD-Based Lactobacillus plantarum Pickering Emulsion to Extend Cherry Tomato Shelf Life

Youwei Yu,Tian Li,Shengwang Li,Silong Jia,Xinyu Yang,Yaxuan Cui,Hui Ma,Shuaishuai Yan,Shaoying Zhang

To improve the postharvest preservation of cherry tomatoes and combat pathogenic, both bacterial and fungal contamination (particularlyAlternaria alternata), a novel biodegradable coating was developed based on a water-in-water (W/W) Pickering emulsion system. The emulsion was stabilized byL. plantarum(Lactobacillus plantarum), with maltodextrin (MD) as the dispersed phase and hydroxypropyl methylcellulose (HPMC) as the continuous phase. Characterization of emulsions at varying concentrations revealed that the optimized W/W-PL^8 film exhibited superior stability, smooth morphology, and low water vapor permeability (WVP = 220.437 g/(m2·24 h)), making it a promising candidate for fruit and vegetable preservation. Furthermore, the coating demonstrated strong antioxidant activity, with scavenging rates of 58.99% (ABTS) and 94.23% (DPPH), along with potent antimicrobial effects, showing inhibition rates of 12.8% againstEscherichia coliand 23.7% againstStaphylococcus aureus. Applied to cherry tomatoes, the W/W-PL^8 coating significantly reduced respiration rates, minimized decay incidence, and maintained nutritional quality during storage. Remarkably, the coating successfully controlledAlternaria alternatacontamination, enhancing the storage duration of cherry tomatoes. These findings highlight the potential of W/W-PL^8 as an eco-friendly and functional packaging material for fresh produce preservation.

Engineering of halide methyltransferases for synthesis of SAE and its application in biosynthesis of ethyl vanillin

Gui-Ying Yang,Gao-Wei Zheng,Bu-Bing Zeng,Jian-He Xu,Qi Chen

Halide methyltransferase (HMT)-catalyzed synthesis of alkyl donors from S -adenosyl- L -homocysteine (SAH) and haloalkanes is a promising method for biocatalytic alkylation. However, the reported halide methyltransferases show limited catalytic capacity towards unnatural substrates such as ethyl iodide, which limits their application in biocatalytic ethylation. In this work, we performed structure-based directed evolution starting from the wild-type Aspergillus clavatus halide methyltransferase ( Ac HMT). The best variant, Ac HMT M3 , was identified with an activity of up to 421.5 mU mg –1 towards ethyl iodide, 38.7-fold higher than that of the wild type Ac HMT. Subsequently, molecular dynamics simulations provide structural insights into how mutations improve the catalytic activity towards iodoethane. Finally, Ac HMT M3 was utilized in one-pot cascade with the reported O -methyltransferase Rn COMT+Y200L to synthesize ethyl vanillin from 3,4-dihydroxybenzaldehyde (79% conversion). Our work demonstrated the potential of the engineered HMT in further biocatalytic alkylation.

An eco-friendly synthetic polyethylene glycol-coated copper nanoclusters for efficiently suppress Alternaria alternata in tobacco and safety evaluation

Zitong Yuan,Chuanxin Ma,Mengao Jia,Kun Qian

Overused agrochemicals threaten the environment and human health. Nanotechnology has shown great potential for maintaining or minimizing environmental issues in agriculture . Herein, a polyethylene glycol-coated copper nanoclusters (PEG@Cu NCs) has been developed for efficient utilization by tobacco. The NCs showed the superior antifungal activity against Alternaria alternata , achieving comparable activity using a dose almost 35-fold lower than that of 46% copper hydroxide water power (WP). More importantly, PEG@Cu NCs exhibited excellent therapeutic potential via xylem delivery. In the meantime, promising bioavailability of the NCs was achieved, which could be absorbed by tobacco seedlings via roots and foliar tissues (epidermal and mesophyll cells). In addition, the biosafety of PEG@Cu NCs was investigated in tobacco and earthworms. Both root exposure (1.27–3.17 mg L −1 ) and leaf exposure (1.27–6.33 mg L −1 ) to PEG@Cu NCs had no negative effects on tobacco growth. The acute toxicity of PEG@Cu NCs to earthworms was low, as determined by the 7 day-LC 50 value of 46.253 mg kg −1 and 14 day-LC 50 value of 30.187 mg kg −1 . Overall, such NCs are expected to be a competitive alternative to Cu-based fungicides in sustainable agriculture due to their superior antifungal properties , excellent biocompatibility, and high efficiency bioavailability.

Antifungal effects of citral against Alternaria alternata in postharvest pear fruit and its potential mechanism

Yuanyuan Chen,Yigui Zeng,Yi Li,Zimao Ye,Liulin Li,Zhiqin Zhou

Spot disease caused by Alternaria alternata ( A. alternata ) commonly occurs in postharvest pears, often resulting in severe rot and considerable economic losses. Here, citral, a natural compound, was found to exhibit potential inhibitory effects against A. alternata . The compound could markedly suppress mycelia growth, with a minimum inhibitory concentration of 0.156 g/L, and simultaneously reduce pear fruit decay. Further analysis revealed that citral altered the hyphal morphology of A. alternata and damaged the integrity of cellular membranes, leading to the release of nucleic acids and proteins. In addition, citral treatment was found to inhibit antioxidant enzyme activity, induce the accumulation of reactive oxygen species, and elevated the malondialdehyde level. It also resulted in a dose-dependent reduction in total cellular lipids, proteins, and sugars. Transcriptomic analysis revealed 1658 upregulated and 2741 downregulated genes in the citral treatment group. Among them, genes related to cell membrane lipids, such as those involved in lipid metabolism, glycerolipid metabolism, steroid biosynthesis, and sphingolipid biosynthesis, were notably enriched. Citral also disturbed fungal energy metabolism by affecting glycolysis/gluconeogenesis, the TCA cycle, and galactose metabolism pathways, which was also confirmed by metabolomic analysis. The results suggest that citral can effectively inhibit A. alternata growth by damaging membrane integrity, activating oxidative stress, and disturbing energy metabolism. Our findings provide new insights into understanding citral as a natural antifungal agent for controlling postharvest black spot disease in pears.

The combination of metabolome and transcriptome clarifies the inhibition of the Alternaria toxin accumulation by methyl ferulate

Shuai Ge,Ying Xie,Ke Ding,Saiqing Xu,Haishan Xu,Xia Chang,Huan Li,Rongrong Wang,Zisheng Luo,Yang Shan,Shenghua Ding

As one of the most typical pathogens in fruit postharvest diseases, Alternaria alternata ( A. alternata ) can produce Alternaria toxins (ATs) aggravating fruit decay and harming human health. In this study, ATs (tenuazonic acid, alternariol monomethyl ether, and alternariol) production was inhibited effectively by 200 and 8000 mg/L MF (methyl ferulate) in vitro and in vivo. 1-Octen-3-ol and 3-octanol were the potential iconic volatile organic compounds of ATs (R 2  > 0.99). MF induced oxidative stress, resulting in physiological and metabolic disorders, membrane lipid oxidation and cell damage. It decreased precursors and energy supply by disturbing amino acid metabolism, ABC transporters, citrate cycle, pentose and glucuronate interconversions to regulate ATs synthesis. MF down-regulated the genes related to ATs synthesis ( PksJ , AaTAS1 , and OmtI ), transport ( AaMFS1 and MFS ), and pathogenicity to affect ATs production and virulence. This study provided a theoretical basis for the control of ATs production.

2-hydroxy-3-phenylpropanoic acid suppressed the growth of Alternaria alternata through damaging cell membrane integrity and modulating reactive oxygen species metabolism

Jiaxin Liu,Canying Li,Chenyang Zhang,Rui Huang,Linhong Qu,Yonghong Ge

Black spot rot caused by Alternaria alternata is one of the major postharvest disease of apple fruit during logistic. This study evaluated in vitro inhibitory effect of 2-hydroxy-3-phenylpropanoic acid (PLA) at various concentrations on A. alternata and the possible mechanisms involved in its action. Results showed that different concentrations of PLA inhibited conidia germination and mycelial growth of A. alternata in vitro, and 1.0 g L −1 was the lowest effective concentration to suppress A. alternata growth. Moreover, PLA significantly reduced relative conductivity and increased malondialdehyde and soluble protein contents. PLA also increased H 2 O 2 and dehydroascorbic acid contents, but reduced ascorbic acid content. Additionally, PLA treatment inhibited catalase , ascorbate peroxidase , monodehydroascorbate acid reductase, dehydroascorbic acid reductase and glutathione reductase activities, whereas promoted superoxide dismutase activity. All these findings suggest that the possible mechanisms involved in the inhibitory effect of PLA on A. alternata included damaging the cell membrane integrity to cause electrolyte leakage and destroying reactive oxygen species balance.
