Wheat leaf rust is a destructive foliar disease of common wheat ( Triticum aestivum L.) worldwide. The most effective, economical s to control the disease is growing resistant cultivars with adult plant resistance (APR). The Chinese wheat lines W014204 and Fuyu 3 showed high leaf rust resistance in the field. To identify leaf rust APR genes in the two lines, two mapping populations with 215 and 163 F 2:3 lines from the crosses W014204/Zhengzhou 5389 and Fuyu 3/Zhengzhou 5389, respectively, were phenotyped for leaf rust severities during the 2010–2011, 2011–2012 and 2012–2013 cropping seasons in the field at Baoding, Hebei Province, China. A total of 1 215 SSR markers were used to identify the quantitative trait loci (QTLs) for leaf rust APR in the two populations. In the W014204/Zhengzhou 5389 population, three QTLs were detected and designated as QLr.hbu-1BL.1, QLr.hbu-2BS.1 and QLr.hbu-7DS, and explained 2.9–8.4, 11.5–38.3 and 8.5–44.5% of the phenotypic variance, respectively; all the resistance alleles at these loci were derived from W014204. In the Fuyu 3/Zhengzhou 5389 population, three QTLs, QLr.hbu-1BL.2, QLr.hbu-2BS.2 and QLr.hbu-7BL, explained 12.0–19.2, 22.3–38.9 and 4.1–4.3% of the phenotypic variance, respectively, and all resistance alleles were contributed by Fuyu 3. Based on chromosome positions of closely linked markers, both QLr.hbu-1BL.1 and QLr.hbu-1BL.2 are Lr46, and QLr.hbu-7DS is Lr34. QLr.hbu-7BL was mapped on chromosome 7BL near to Lr68 and they are likely the same gene. Based on chromosome positions, pedigree and field reactions, the two 2BS QTLs are different from all the known APR genes and are likely to be new APR QTL for leaf rust. These QTLs and their closely linked markers are potentially useful for improving leaf rust resistance in wheat breeding.

BACKGROUND Sclerotinia sclerotiorum , a pathogenic fungus of oilseed rape, poses a severe threat to the oilseed rapeseed industry. In this study, we evaluated the potential of the natural compound hinokitiol against S. sclerotiorum by determining its biological activity and physiological characteristics. RESULTS Our results showed that hinokitiol strongly inhibited the hyphae expansion of S. sclerotiorum , and its effective concentration of hyphae growing inhibition by 50% (EC 50 ) against 103 S. sclerotiorum strains varied from 0.36 to 3.45 μg/mL, with an average of 1.23 μg/mL. Hinokitiol possessed better protective efficacy than therapeutic effects, and it exhibited no cross-resistance between carbendazim. After treatment with hinokitiol, many vesicular protrusions developed on the mycelium with rough surface and thickened cell wall. Moreover, the cell membrane permeability and glycerol content increased, while the oxalic acid declined after hinokitiol treatment. In addition, hinokitiol induced membrane lipid peroxidation and improved the production of reactive oxygen species (ROS) in S. sclerotiorum . Importantly, real-time quantitative polymerase chain reaction showed that cell wall and ROS synthesis-related genes were significantly up-regulated after hinokitiol treatment. CONCLUSION This study revealed that hinokitiol has good biological activity against S. sclerotiorum and could be considered as an alternative bio-fungicide for the resistance management in controlling sclerotinia stem rot infected by S. sclerotiorum . These investigations provided new insights into understanding the toxic action of hinokitiol against pathogenic fungi. © 2024 Society of Chemical Industry.

BACKGROUND Microstegium vimineum (Trin.) A. Camus, commonly called stiltgrass, is a dominant weed in the United States and China. Although a lot of control approaches have been attempted, an economic, effective and practical measure has not been available to control the weed so far. RESULTS A serious rust disease of Microstegium vimineum was observed in three regions of Wenzhou city in China, from 2019 to 2021, with a disease incidence ranging from 82% to 97%. Typical rust disease symptoms on Microstegium vimineum were prominently visible during the early monsoon season (June–July), with chlorotic spots on the leaf surfaces. The morphological characterization of the strain WZ-1 which was isolated from the diseased leaves was consistent with Puccinia polliniicola . The virulence tests showed that the average disease index of Microstegium vimineum plants could reach 35% at 10 days post-inoculation. The host specificity of Puccinia polliniicola was tested on 64 plant species from 12 families and it did not cause any diseased symptoms on 24 major crops and 36 weeds, but slightly infected four gramineous weeds, Arthraxon hispidus , Polypogon fugax , Cynodon dactylon , and Microstegium ciliatum . However, newly-produced urediniospores were not observed on the slightly infected plants. The urediniospores of strain WZ-1 infected the Microstegium vimineum leaves by two main approaches: mycelium or appressorium invaded the stoma; and mycelium or appressorium directly invaded intercellular spaces. Field experiments showed that the rust disease naturally prevailed among Microstegium vimineum populations, causing severe rust disease symptoms on the leaf surface. The rust epidemic effectively controlled all of the target plants in the closed plot where the rust was released. CONCLUSION Puccinia polliniicola strain WZ-1 has great potential to be used as a classical biological control agent against Microstegium vimineum . © 2024 Society of Chemical Industry.

Microstegium vimineum (Trin.) A. Camus, commonly called stiltgrass, is an exotic and indigenous troublesome weed in the United States and eastern Asia, respectively. Between 2020 and 2022, a damaging disease was observed on M. vimineum plants at the foot of Daluo Mountain in Wenzhou City, China, with an incidence of 90 %. Typical rust disease symptoms were prominently visible during the early monsoon season (June to July), consisting of chlorotic spots with many orange or orange-yellow uredinia on the abaxial surfaces of the M. vimineum leaves. Based on morphological and molecular characteristics, the fungus was identified as Kweilingia divina (Syd.) Buriticá. This article describes the studies carried out on one strain of the fungus (WZ-2) to ascertain its potential as a biocontrol agent of the weed. Host specificity testing on 61 plant species from 15 families proved that 25 major crops and 36 weeds were insensitive to this fungus, with only two other gramineous weed species being slightly susceptible. However, urediniospores were not observed on these slightly infected plants. Further research is required to assess the pathogenicity and safety of various plant species to K. divina , especially bamboos, which have been identified as susceptible. Temperature and dew period experiments have shown that the fungus has a relatively wide tolerance for infection between 15 and 40 °C, and with only 4 h of dew exposure some uredinia appeared on artificially infected M. vimineum plants. A field trial proved that urediniospores from the strain WZ-2 could infect naturally occurring M. vimineum plants, causing severe disease symptoms and plant death. Thus, strain WZ-2 has great potential as an augmentative biological control agent against M. vimineum . K. divina is a heteroecious rust fungus that requires an alternate host to complete its life-cycle. What the alternative hosts are, and the impact of the alternate hosts on outbreaks of the disease requires further study.

Leaf rust caused by Coleosporium zanthoxyli is a destructive disease that threatens Zanthoxylum bungeanum plantations. To address this, screening for resistant cultivars and exploring their resistance mechanisms are crucial for effective disease management. However, the mechanisms underlying Z. bungeanum 's resistance remain unclear. In this study, we conducted in vivo and in vitro experiments to screen for the resistant cultivar Qiujiao (QJ) and the susceptible cultivar Fengxian Dahongpao (FD). Subsequently, the resistance mechanisms of Z. bungeanum were analyzed by integrating transcriptome and metabolome, the result showed the flavonoid pathway served as the primary metabolic pathway for QJ to confer resistance against C. zanthoxyli . Upon infection with C. zanthoxyli , the key genes ( PAL, CHI , and HCT ) and enzymes (PAL, C4H, and 4CL) involved in the flavonoid pathway were upregulated in QJ, and the total flavonoid content was increased in QJ. Additionally, eight flavonoids from this pathway demonstrated inhibitory effects on C. zanthoxyli spore germination, with naringenin and phloretin exhibiting the stronger activity. Collectively, the flavonoid pathway plays a dominant role in the resistance of Z. bungeanum leaves against C. zanthoxyli .
