In this study, we investigated cooked hams to understand the relationship between display life and quality changes, as well as the microbiological characteristics of refrigerated storage for up to 90 days. Initially, the L* and a* values exhibited an initial decline with a gradual restitution to levels insignificantly disparate from those Day 15 hams. In contrast, the b* and h° values demonstrated an ascending pattern, indicating an augmentation in yellow intensity during the storage. Furthermore, there was an observable escalation in the C* value following 45 days of storage, implying an amplified luminosity and vividness of color during the latter phase of the storage period. Analyses of TBARS , TVB-N, and other physicochemical indexes indicated that the hams underwent deterioration during storage. Whereas, UV–vis and fluorescence spectroscopy illustrated that the pigment in cooked hams changed from nitrosylheme (NO-heme) to Zn-protoporphyrin IX (ZnPP) during storage. The NO-heme content decreased from 41.08 ± 0.44 to 12.76 ± 0.29 mg/kg, while the ZnPP content increased from 12.19 ± 0.41 to 89.44 ± 1.57 mg/kg. The structure of the bacterial community in the ham samples changed over the storage period. Towards the end of the shelf life, the microbial population was dominated by Lactic acid bacteria (LAB), particularly Latilactobacillus (34.91%) and Leuconostoc (19.73%). Redundancy analysis and Pearson correlation revealed a positive correlation between Latilactobacillus and Leuconostoc with TBARS , TVB-N, ZnPP content, and negative correlations with Aw and NO-heme.

Caffeic acid is an antioxidant that has been widely been related to the health benefits of people in recent years. In this paper, the amino side chains of chitosan (CS) were modified with protoporphyrin IX by amide cross-linking, and then Zn ions were chelated. The properties of metalloporphyrin-preparing functionalized multi-walled carbon nanotubes (MWCNTs) and Zn ions chelated by protoporphyrin IX composites were used as sensitive-selective electrochemical biosensors for the determination of caffeic acid. The morphology and structure of nanocomposite Zn–PPIX–CS–MWCNTs were observed by X-ray spectroscopy mapping (EDX mapping), transmission electron microscopy (TEM), and Fourier-transform infrared spectroscopy (FTIR). The electrochemical behaviors of Zn–PPIX–CS–MWCNT-modified glassy carbon (GC) electrodes were evaluated by cyclic voltammetry (CV) and differential pulse voltammetry (DPV). The results show that the modified electrode had good electrocatalytic activity towards caffeic acid with a wide linear range of 0.0008–1.6 mM, an excellent sensitivity of 886.90 µAmM−1cm−1, and a detection limit of 0.022 µM. In addition, the caffeic acid sensor had excellent reproducibility, stability, and selectivity to various interfering substances. Therefore, the modified electrode prepared by this experiment can also be applied to electrochemical sensors of other substances.

Sonodynamic therapy (SDT) induces reactive oxygen species (ROS) to kill tumor cells. Heme oxygenase-1 (HO-1), as an important antioxidant enzyme, resists killing by scavenging ROS. Zinc protoporphyrin (ZnPP) not only effectively inhibits HO-1 activity, but also becomes a potential sonosensitizer. However, its poor water solubility limits its applications. Herein, we developed an improved water-soluble method. It was proved that pegylated zinc protoporphyrin-mediated SDT (PEG-ZnPP-SDT) could significantly enhance ROS production by destroying the HO-1 antioxidant system in ovarian cancer. Increased ROS could cause mitochondrial membrane potential collapse, release cytochrome c from mitochondria to the cytoplasm, and trigger the mitochondrial–caspase apoptotic pathway. In conclusion, our results demonstrated that PEG-ZnPP-SDT, as a novel sonosensitizer, could improve the antitumor effects by destroying the HO-1 antioxidant system. It provided a new therapeutic strategy for SDT to treat cancers, especially those with higher HO-1 expression. Keywords:pegylated zinc protoporphyrin;sonodynamic therapy;reactive oxygen species;apoptosis;antitumor effects

Background The activation of the cyclic guanosine monophosphate-adenosine monophosphate synthase-stimulator of interferon genes (cGAS-STING) signaling pathway has attracted great attention for its ability to up-regulate innate immune response and thus enhance cancer immunotherapy. However, many STING agonists limit the further advancement of immunotherapy due to weak tumor responsiveness or low activation efficiency. The responsive and effective activation of cGAS-STING signaling in tumors is a highly challenging process. Methods In this study, a manganese-based nanoplatform (MPCZ NPs) was constructed that could responsively and efficiently generate more manganese ions (Mn 2+ ) and reactive oxygen species (ROS) to activate cGAS-STING signaling pathway. Briefly, manganese dioxide (MnO 2 ) was loaded with zinc protoporphyrin IX (ZPP) molecule and coated by polydopamine (PDA) embedded with NH 4 HCO 3 to obtain MPCZ NPs. Additionally, MPCZ NPs were evaluated in vitro and in vivo for their antitumor effects by methyl thiazolyl tetrazolium (MTT) assay and TUNEL assays, respectively. Results In this system, tumor responsiveness was achieved by exogenous (laser irradiation) and endogenous (high levels GSH) stimulation, which triggered the collapse or degradation of PDA and MnO 2 . Moreover, the release of Mn 2+ augmented the cGAS-STING signaling pathway and enhanced the conversion of hydrogen peroxide (H 2 O 2 ) to hydroxyl radical (·OH) under NIR laser irradiation. Furthermore, the release of ZPP and the elimination of GSH by MPCZ NPs inhibited HO-1 activity and prevented ROS consumption, respectively. Conclusions This adopted open source and reduce expenditure strategy to effectively generate more ROS and Mn 2+ to responsively activate cGAS-STING signaling pathway, providing a new strategy for improving immunotherapy.

Nitrites, commonly used as coloring agents in traditional meat processing, can generate carcinogenic N-nitrosamines. This study developed a microbial fermentation-based coloring technology to replace nitrites. A fermented sausage model included five treatments: control; nitrite-added (50 mg/kg); and three treatments inoculated with food-grade probiotics ( Lactiplantibacillus plantarum YR07, Latilactobacillus sakei L48, Latilactobacillus curvatus WQ). Colorimetric analysis showed that on day 7 the YR07 treatment showed significantly higher redness (a* 9.17 ± 0.10) than control (7.38 ± 0.10), L48 (7.52 ± 0.35) and WQ (6.76 ± 0.14) treatments ( p < 0.05), approaching the nitrite-treated treatment (10.55 ± 0.05). UV-Vis spectroscopy confirmed characteristic nitrosylheme in nitrite-treated sausages, whereas zinc protoporphyrin (ZnPP) dominated probiotic-inoculated treatments. Fluorescence spectroscopy further demonstrated sustained highest intensity in YR07 during processing. ZnPP content on day 7, with the YR07 treatment showing significantly higher levels (5.16 ± 0.09 mg/kg) compared to the control (1.98 ± 0.04 mg/kg) and nitrite-treated treatments (0.50 ± 0.03 mg/kg) ( p < 0.05). Ferrochelatase activity in YR07-inoculated sausages (134.54 ± 2.96 U/g) significantly exceeded ( p < 0.05) both the control (102.01 ± 1.09 U/g) and nitrite-treated treatments (9.75 ± 0.72 U/g), demonstrating nitrite's inhibitory effect on the enzyme. HPLC analysis on day 7 showed that YR07-inoculated (13.32 ± 1.36 mg/kg) and nitrite-treated sausages (10.53 ± 49 mg/kg) contained less heme than the control (19.65 ± 0.31 mg/kg), suggesting heme may serve as substrate for ZnPP biosynthesis. This study not only provides a feasible microbial alternative to nitrite but also elucidates the potential mechanism of ZnPP formation.

The practical application of Lithium-sulfur (Li-S) batteries is significantly inhibited by (i) the notable ‘shuttle effect’ of lithium polysulfides (LiPS), (ii) the corrosion of the lithium interface, and (iii) the sluggish redox reaction kinetics. The functional separator in the Li-S battery has the potential to provide the perfect solution to these problems. Herein a triple-layer multifunctional PVDF-based nanofiber separator, which contains GoTiN/PVDF layer on the top and bottom and ZnTPP/PVDF layer on the middle, is designed. The polarity and porous structure of this multifunctional separator can greatly improve the wettability of electrolytes and enhance the transportation of Li + . With the zinc-based porphyrin framework (ZnTPP) structure, this separator has a strong chemisorption and LiPS conversion ability, which greatly prevent the ‘shuttle effect’. Consequently, the designed multilayer separator showed excellent electrochemical performance. As a result, the cell with GoTiN@ZnTPP@GoTiN nanofiber membrane displayed an initial discharge capacity of 1180 mAh/g with a benign capacity retention of 65.9% at 0.5C and high coulombic efficiency of more than 98.5% after 100 cycles. Even at 2C, it can still release a capacity of 798 mAh/g. Moreover, the remarkable capacity of 591 mAh/g could be achieved with a high sulfur load of 5.76 mg/cm 2 under a current density of 0.1C. Based on these merits, this novel and scalable multifunctional separator is a promising candidate to replace the conventional PP separator for advanced Li-S batteries to deal with various challenges.

Myelin sheath is essential for the maintenance of neural system functions. The mediator complex (MED) is associated with neurodegenerative diseases. Mutation of the MED subunit Med20 leads to abnormal brain development, but whether Med20 modulates myelination remains unknown. In this study, we report that Med20 is an important modulator of myelination in the peripheral nervous system. Loss of Med20 in Schwann cells (SCs) induces ferroptosis, thus impairing myelination. The functions of Med20 in SCs are mediated by its downstream target DDB1. DDB1 controls SC ferroptosis by regulating Hmox1 through a “ DDB1-UHRF1-BACH1-Hmox1 ” transcription regulation axis and its direct interaction and subsequent simultaneous ubiquitination of the HO-1 protein. HO-1 inhibitor ZnPP or ferroptosis inhibitor Fer-1 is sufficient and efficient for antagonizing ferroptosis in SCs and restoring myelination in Med20 -deficient mice. The results of our study reveal that Med20 is an important regulator in the network that controls myelination in the peripheral nervous system.

BACKGROUND:Deep vein thrombosis (DVT) is a prevalent peripheral vascular disorder associated with abnormal epigenetic processes and altered gene expression in endothelial cells. Accumulating evidence has demonstrated that NAT10 (N-acetyltransferase 10)–mediated N4-acetylcytidine modification exerts unique roles in ferroptosis, but its roles are still elusive in DVT.METHODS:To explore the potential mechanism of NAT10 and ferroptosis on thrombogenesis, we used NAT10 and GPX4 (glutathione peroxidase 4) knockout mice as an in vivo model, and utilized techniques, such as RNA immunoprecipitation, acRIP-qPCR, N4-acetylcytidine Dot Blotting assay, and Western blotting, for detailed molecular analysis.RESULTS:GPX4 is a pivotal gene that suppresses ferroptosis. Utilizing endothelial cell–specific GPX4 conditional knockout mice (GPX4fl/flCdh5-Cre+), we proved that ferroptosis in endothelial cells promotes the formation of thrombosis. Previous evidence indicates that NAT10 overexpression induces ferroptosis and downregulates GPX4 expression. Here, we found that NAT10 expression was elevated in DVT mice, and silencing of NAT10 markedly attenuated ferroptosis both in vitro and in vivo. Furthermore, endothelial cell–specific knockout of NAT10 (NAT10fl/flCdh5-Cre+) demonstrated a reduction in endothelial ferroptosis, thereby inhibiting both the formation and progression of DVT. Mechanistic studies indicated that NAT10 facilitated the N4-acetylcytidine modification of HMOX1 (heme oxygenase 1), which enhanced its mRNA stability, leading to the accumulation of ferrous ions, and exacerbating endothelial dysfunction in DVT.CONCLUSIONS:Collectively, our data elucidate that downregulation of NAT10 mitigates endothelial ferroptosis and prevents DVT formation and progression by modulating HMOX1 expression, which offers a potential novel strategy for the prevention and treatment of thrombosis in DVT.

Cadmium (Cd), a prevalent environmental contaminant, has attracted widespread attention due to its serious health hazards. Ferroptosis is a form of iron-dependent oxidative cell death that contributes to the development of various kidney diseases. However, the mechanisms underlying the occurrence of ferroptosis in Cd-induced renal tubular epithelial cells (TECs) have not been fully elucidated. Hereby, both in-vitro and in-vivo experiments were established to elucidate this issue. In this study, we found that Cd elicited accumulation of lipid peroxides due to intracellular ferrous ion (Fe 2+ ) overload and glutathione depletion, contributing to ferroptosis. Inhibition of ferroptosis via chelation of Fe 2+ or reduction of lipid peroxidation can significantly mitigate Cd-induced cytotoxicity. Renal transcriptome analysis revealed that the activation of heme oxygenase 1 (HO-1) was closely related to ferroptosis in Cd-induced TECs injury . Cd-induced ferroptosis and resultant TECs injury are significantly alleviated due to HO-1 inhibition, demonstrating the crucial role of HO-1 in Cd-triggered ferroptosis. Further studies showed that accumulation of lipid peroxides due to iron overload and mitochondrial ROS (mtROS) generation was responsible for HO-1-triggered ferroptosis in Cd-induced cytotoxicity. In conclusion, the current study demonstrates that excessively upregulating HO-1 promotes iron overload and mtROS overproduction to trigger ferroptosis in Cd-induced TECs injury, highlighting that targeting HO-1-mediated ferroptosis may provide new ideas for preventing Cd-induced nephrotoxicity .

Although immune checkpoint inhibitors (ICIs) have been widely applied to treat non-small cell lung cancer (NSCLC), a significant proportion of patients, especially those with spinal metastasis (NSCLC-SM), are insensitive to anti-programmed death 1 (PD-1)/programmed death ligand 1 (PD-L1) ICIs. A drug delivery nano-controller of PD-L1 that targets NSCLC-SM can solve this problem, however, none have been developed to date. In this study, it is shown that integrin β 3 ( β 3-int) is strongly upregulated in NSCLC-SM. Its inhibitor RGDyK promotes PD-L1 ubiquitination, indicating the potential application of RGDyK as a new PD-L1 inhibitor in nano-controller and a targeting peptide for NSCLC-SM treatment. According to the synergistic effect of photodynamic therapy and ICIs on T-cell activation through the release of tumor antigens, RGDyK-modified and zinc protoporphyrin (ZnPP)-loaded mesoporous silicon nanoparticles (ZnPP@MSN-RGDyK) are fabricated. The ZnPP@MSN-RGDyK nanoparticles precisely target β 3-int to inhibit PD-L1, exhibiting high photodynamic therapy efficiency, and excellent immunotherapeutic effects in an NSCLC-SM mouse model. Collectively, the findings indicate that ZnPP@MSN-RGDyK is a promising immunotherapeutic agent for treating NSCLC-SM.
