An "ON-OFF" fluorescent sensor based on a novel zinc-based flower-like structured metal-organic framework for sequential detection of deferasirox and tigecycline

Xiaowen Xu,Hui Lin,Bixia Lin,Lingyi Huang,Pingping Wu,Youjia Wu,Liying Huang

Deferasirox (DEF) is essential for patients with thalassemia requiring long-term transfusion therapy. Tigecycline (TIGE) is a first-line drug for the clinical treatment of complex, severe bacterial infections. The two drugs can be coordinated to treat Pseudomonas aeruginosa infections. Easy and efficient techniques for monitoring these two drugs in biological samples are few. Metal–organic framework (Zn-MOF) prepared from zinc nitrate hexahydrate and dithioglycolic acid has a flower structure. Interestingly, Zn-MOF can cause DEF to aggregate on it and induce DEF luminescence. The principle may be that Zn-MOF limits the vibration and rotation of DEF to avoid its nonradiative jump, which triggers aggregation-induced emission (AIE) and exhibits intense fluorescence. Further investigation revealed that TIGE could decompose Zn-MOF, thus alleviating the inhibitory effect of Zn-MOF on DEF and reducing the fluorescence intensity of DEF@Zn-MOF. A DEF/TIGE detection biosensor was created based on the fluorescence "turn-on" effect of Zn-MOF on DEF and the fluorescence "turn-off" effect of TIGE on DEF@Zn-MOF. The proposed technique was subsequently used to identify DEF/TIGE levels in pharmaceuticals and human plasma. The mean values for the percentage of the labeled amount of DEF/TIGE in DEF dispersible tablets/TIGE injection were 104.5 and 104.9%, respectively. The detection limits for the fluorescence detection of DEF and TIGE were 3.6 and 1.2 nM, respectively. This fluorescence assay is the first application of MOF to the simultaneous detection of DEF and TIGE and has the advantages of rapid sensitivity and high selectivity , providing a new strategy for drug detection.

Study of bioactive 3D-printed scaffolds incorporating zinc-based MOF for bone defect repair and anti-inflammatory applications

Yu Han,Xiufeng Jia,Yinwei Yang,Pei Guo,Chao Li,Yunze Zhang,Li Yin,Bo Jia,Haitao Wang

The development of bioactive scaffolds capable of orchestrating osteogenesis while modulating inflammatory responses remains a critical challenge in bone tissue engineering. To address this unmet need, we engineered a multifunctional composite scaffold through precise integration of poly(lactic-co-glycolic acid) (PLGA), type I collagen, zinc-imbued metal-organic frameworks (Zn-MOFs), and macrophage chemotactic factor (MCF) via extrusion-based 3D printing. Systematic biological assessments in vitro revealed the scaffold's remarkable osteoinductive capacity, evidenced by significant upregulation of alkaline phosphatase (ALP) activity (2.78±0.10 fold increase vs control, p<0.05) and enhanced collagen type I deposition (10.79±1.12 fold increase vs control, p<0.05) within 14 days of culture. In vivo implantation in rat femoral defect models demonstrated superior bone regeneration outcomes, with micro-CT analysis showing bone volume/ total volume (BV/TV) 31.39±3.04% compared to 16.44±3.26% in control (P<0.05). The Zn-MOF component mediated sustained release of Zn 2+ ions (cumulative release: 89.6±9.08% over 18 days). This dual-functional system synergistically combines MCF-driven cellular recruitment with Zn 2+ -mediated immunomodulation, creating a pro-regenerative microenvironment. Our findings not only demonstrate the feasibility of 3D-printed multi-material constructs for complex tissue engineering applications but also establish a new paradigm for developing smart biomaterials that concurrently address biological regeneration and inflammatory control.

Safety assessment of copper- and zinc-based metal-organic frameworks (MOFs) for embryonic development and adult fish of Oryzias melastigma

Mohammad Sadiq Nikzad,Jiangbing Qiu,Guixiang Wang,Xue Wang,Natnael Sisay Demo,Aifeng Li

The increasing prevalence of harmful algal blooms (HABs), specifically those resulted by toxigenic Alexandrium spp., notably threats human health and aquatic ecosystem. Metal organic frameworks (MOFs), particularly copper- and zinc-based MOFs have emerged as promising materials to control HABs. However, their application safety needs to be assessed because of poor knowledge of their ecotoxicity to marine organisms. In this study, the toxicity of Cu-MOF and Zn-MOF was evaluated to Oryzias melastigma at embryonic and adult stages. The Cu-MOF exhibited a concentration-dependent mortality and hatching rate at concentrations ≥ 2.0 mg L −1 . The Zn-MOF exhibited similar effects when the exposure doses ≥ 80 mg L −1 . Additionally, adult fish after exposure to Cu-MOF showed significant mortality rate at higher concentrations (≥ 4.0 mg L −1 ), beside elevated oxidative stress markers. In contrast, Zn-MOF did not show mortality and oxidative stress although bioaccumulation of Zn 2 + occurred in tissues. Based on the safety assessment of MOFs to marine medaka, the dosages for application of Cu-MOF (≤ 1.0 mg L −1 ) and Zn-MOF (≤ 20 mg L −1 ) are safe for controlling A. minutum -forming HABs. This study hints that the application of MOFs in mitigation of HABs should be carefully evaluated to balance their benefits in contrast to ecological risks.

A water-stable and biofriendly 2D anionic zinc(II) metal–organic framework for drug delivery

Yang Zhang,Jian Wang

A water-stable 2D Zn-based MOF with chemical formulae [(Me 2 NH 2 ) 4 Zn 2 (FDC) 4 ] n ( 1 ) (FDC = 2,5-furandicarboxylate) was synthesized and structurally characterized. TGA and PXRD results show high thermal and water stability for 1 . 1 exhibits a 2D (4,4) net with 1D channel of 7.2 × 7.2 Å 2 along the c axis. Desolvated 1 adsorbs around 11.4 wt% 5-Fluorouracil and the cumulative release value of 5-Fluorouracil at 48 h was about 96.5% and 99% in PBS (pH = 7.4 and pH = 6.0) at 37 °C, respectively. The in vivo Zebrafish toxicity tests and in vitro MTT assays of 1 at different concentrations results reveal that 1 is nontoxic.

An Inhibitor-Loaded LDH- and MOF-Based Bilayer Hybrid System for Active Corrosion Protection of Aluminum Alloys

Zhe Zhang,Juping Wang,Maria Serdechnova,Valeryia Kasneryk,Zheng Zhang,Carsten Blawert,Hao Wang,Mikhail L. Zheludkevich,Fei Chen,You Zhang

A novel inhibitor-loaded bilayer hybrid system based on the LDH inner layer and MOF outer layer is designed on an aluminum alloy 2A12 surface to improve corrosion performance. The hybrid film system covers the inherent cavities and intercrystalline defects of the LDH film using the affinity between the LDH and the MOF compounds. The results demonstrate that the LDH-inhI precursor film is entirely covered by new Zn-based MOF microrods. The LDH-inhI precursor film is partially dissolved and recrystallized in favor of MOF crystal growth to strengthen the binding adhesion between LDH and MOF films. The LDH-inhI/MOF-inhII bilayer film shows significantly enhanced corrosion resistance through the synergistic action of LDH and MOF nanocontainers doped with different corrosion inhibitors (vanadates, 2,5-furandicarboxylic acid, and benzotriazoles). Due to the multiple loadings of the MOF film and the sustained-release of the LDH film, this method provides an effective approach to developing new anticorrosion systems and enhancing both the barrier ability and active corrosion protection performance of LDH-based conversion treatments.

Zinc-based metal organic framework with antibacterial and anti-inflammatory properties for promoting wound healing

Chen Yuting,Cai Jinhong,Liu Dachang,Liu Shuhan,Lei Doudou,Zheng Li,Wei Qingjun,Gao Ming

The synergistic effect of antibacterial and anti-inflammatory is needed to overcome the problem of wound healing difficulties. Based on the favorable antibacterial and anti-inflammatory effect of zinc ions (Zn2+) and the physicochemical properties of metal organic frameworks (MOFs), we prepared nanosized zinc-based MOF: Zn-BTC with the ability to slowly release Zn2+. In cellular levels, Zn-BTC possessed lower toxicity to fibroblasts and enhanced capacity of cell proliferation and migration. It also had good bactericidal effect on multiple drug-resistant bacteria by reducing 41.4% MRSA and 47.2% Escherichia coli. In addition, Zn-BTC also displayed the ability of lowering the expression of antioxidant genes: superoxide dismutase 1, superoxide dismutase 2 and interleukin 6, and enhancing the expression of wound healing genes: transforming growth factors-β and type I collagen. Finally, it also demonstrated that Zn-BTC could effectively improve the skin wound healing of SD rats and had no toxicity on major organs. The favorable biocompatibility, antibacterial and anti-inflammatory properties of Zn-BTC gave a new insight of designing novel MOFs for promoting skin wound healing.
