Zinc-Chelating Mechanism of Sea Cucumber (Stichopus japonicus)-Derived Synthetic Peptides

Xiaoyang Liu,Zixu Wang,Fawen Yin,Yuxin Liu,Ningbo Qin,Yoshimasa Nakamura,Fereidoon Shahidi,Chenxu Yu,Dayong Zhou,Beiwei Zhu

In this study, three synthetic zinc-chelating peptides (ZCPs) derived from sea cucumber hydrolysates with limited or none of the common metal-chelating amino-acid residues were analyzed by flame atomic absorption spectroscopy, circular dichroism spectroscopy, size exclusion chromatography, zeta-potential, Fourier transform infrared spectroscopy, Raman spectroscopy and nuclear magnetic resonance spectroscopy. The amount of zinc bound to the ZCPs reached maximum values with ZCP:zinc at 1:1, and it was not further increased by additional zinc presence. The secondary structures of ZCPs were slightly altered, whereas no formation of multimers was observed. Furthermore, zinc increased the zeta-potential value by neutralizing the negatively charged residues. Only free carboxyl in C-terminus of ZCPs was identified as the primary binding site of zinc. These results provide the theoretical foundation to understand the mechanism of zinc chelation by peptides.Keywords:zinc-chelating peptide;sea cucumber;chelation mechanism

Isolation and identification of zinc-chelating peptides from sea cucumber (Stichopus japonicus) protein hydrolysate

Xiaoyang Liu,Zixu Wang,Jing Zhang,Liang Song,Deyang Li,Zixuan Wu,Beiwei Zhu,Yoshimasa Nakamura,Fereidoon Shahidi,Chenxu Yu,Dayong Zhou

BACKGROUND Zinc is known to play an essential role in the biological activities in the human body. In this study, a zinc-chelating peptide (ZCP) produced by Alcalase-assisted hydrolysis of the body wall of sea cucumber was isolated and identified. The ZCP was purified stepwise by ultrafiltration, anion-exchange chromatography, and gel filtration chromatography, in conjunction with ultraviolet–visual (UV–visual) spectrophotometry, which was used to analyze each purified fraction. RESULTS Analysis of the purified ZCP revealed that its zinc-chelating ability was 33.31%. Analysis of isothermal titration calorimetry suggested that the binding of ZCP and zinc (N ≈ 2) was endothermic, with weak binding affinity. Fourier transform infrared spectroscopy spectra (FTIR) indicated that carboxylic and amide groups in ZCP were the primary binding sites of Zn. Sequencing the result by ultra-performance liquid chromatography-quadrupole/time of flight mass spectrometry (UPLC-Q-TOF-MS/MS) showed that a representative ZCP had the sequence WLTPTYPE with a molecular weight of 1005.5 Da. CONCLUSION These results provide a promising foundation for the production of zinc supplements from sea-cucumber-derived ZCPs. © 2019 Society of Chemical Industry

Hydrophilic Food-Borne Nanoparticles from Beef Broth as Novel Nanocarriers for Zinc

Jiaxin Geng,Xunyu Song,Xuedi Zhang,Shanshan Tie,Lin Cao,Mingqian Tan

Food-borne nanoparticles (FNs) may be used as nanocarriers for metal ion chelation in micronutrient supplements. In this paper, the preparation and characterization of hydrophilic FNs were reported from beef broth cooked with a pressure cooker at 117 °C for different periods (30, 50, and 70 min) and their potential application as nanocarriers for zinc was investigated. The broth FNs are quasi-spherical with good water solubility and ultrasmall size, which can emit a strong sapphire color under 365 nm ultraviolet irradiation. X-ray photoelectron spectroscopy (XPS) analysis showed that there are carboxyl, amino, and hydroxyl groups on the FNs, which are useful for Zn(II) chelation. The vibration band of C═O at 1688 cm–1 in the infrared spectrum of FNs shifted to 1718 cm–1 after binding with Zn(II) ions, suggesting the participation of the carbonyl group in Zn(II) ion chelation. The appearance of Zn2p XPS peaks, at 1021.6 and 1045 eV for Zn(II)–FNs, clearly demonstrated the formation of Zn–O between the FNs and zinc ions. Biodistribution of FNs and the Zn(II)–FN complex in normal rat kidney cells demonstrated that they could easily enter normal rat kidney cells. A downfield was found for the signals of Zn(II)–FNs in 1H nuclear magnetic resonance spectroscopy and strongly suggested the binding of Zn(II) ions to FNs through carboxylic acid, hydroxyl, and amine groups. In addition, no obvious cytotoxicity was found for Zn(II)–FNs compared to zinc (ZnSO4) and commercial zinc gluconate. The results revealed that the FNs from beef broth may have a potential as nanocarriers for zinc chelation.

A novel zinc chelate complex containing both phosphorus and nitrogen for improving the flame retardancy of low density polyethylene

Zhenhu Cao,Yan Zhang,Pingan Song,Yuanzheng Cai,Qi Guo,Zhengping Fang,Mao Peng

A novel metal chelate complex containing phosphorus, nitrogen and zinc (II) ion was synthesized and used as the flame retardant of low density polyethylene (LDPE). The zinc chelate complex was synthesized by reacting zinc acetate with the ligand of tetraethyl (1,2-phenylenebis(azanediyl)) bis (2-hydroxylphenylmethylene) diphosphonate (TEPAPM). The chemical structure of the target Zn-TEPAPM was confirmed by FTIR, 1 H NMR, 13 C NMR and 31 P NMR spectra. The flame retardancy and thermal behavior of LDPE containing various amount of Zn-TEPAPM were investigated by limiting oxygen index test, thermogravimetric (TG) analysis and cone calorimetry . The results show that Zn-TEPAPM can greatly increase the thermal stability, the char formation and smoke suppression ability of LDPE. The TG curves show that even when the filler content of Zn-TEPAPM is as low as 1   wt% in LDPE, the onset degradation temperature of LDPE is increased from 429   °C to 442   °C, and the maximum degradation temperature is increased from 469   °C to 488   °C. Also, a reduction of 32% for the peak heat release rate (PHRR) is obtained in the cone test. Moreover, Zn-TEPAPM is demonstrated to be a very effective synergist of ammonium polyphosphate (APP). When 1   wt% of Zn-TEPAPM was introduced into the LDPE/APP (mass ratio 80/19) blend, the PHRR value is reduced by 32%, compared with that of LDPE/APP blend without Zn-TEPAPM, and the char layer becomes more compact and intact.

Preparation, characterization and evaluation of L-theanine - zinc chelate: increasing stability, antioxidant, and antidiabetic activity

Pan Xiaoya,Wang Hongyue,Guo Keshuang,Zhang Tong,Li Xinnan,Zhang Xiangrong

The study aimed to prepare the chelate of L-theanine (LT) with zinc (Zn) and evaluate its stability, antioxidant and antidiabetic activity. The chelation mechanism between Zn 2+ and LT was investigated by ultraviolet-visible, Fourier transform infrared, scanning electron microscopy/energy dispersive spectrometer and 1 H nuclear magnetic resonance spectroscopy. L-theanine-Zn chelate (LT-Zn) was obtained using LT to Zn citrate in a mass ratio of 3:1 at pH 6 and 70 °C for 180 min with the chelation efficiency of 61.86 ± 1.26%. The results revealed the amino nitrogen atoms and carboxyl oxygen atoms in LT chelated with Zn ions, and the mass of the chelate was confirmed by liquid chromatography with tandem mass spectrometry. Scanning electron microscopy with the element analyzed by energy dispersive spectroscopy showed that the morphology of LT-Zn consisted of irregular granular particles with rough surface. The LT-Zn exhibited higher solubility and improved stability under 100 °C, various pH condition and different concentrations of glucose and NaCl ( p  < 0.05) than that of sulfate Zn and Zn gluconate. Compared to sulfate Zn and Zn gluconate, the LT-Zn showed better stability during simulated gastrointestinal digestion. The chelate also exhibited antioxidant activity, as well as an inhibitory effect on α-glucosidase and α-amylase. Overall, the study provided a novel approach to prepare Zn nutraceuticals with antioxidant and antidiabetic properties.

The synthesis and properties of an intumescent zinc-chelated organometallic flame retardant

Tianyu Liu,Ying Tao,Xiuyuan Ni

Organometallic flame retardants (OFRs) are currently attracting intensive attention. Here, we synthesized a new zinc-chelated OFR, ZnDSi, which was incorporated with phosphorus, silicon, and nitrogen functional moieties. The resulting ZnDSi exhibited strong intumescent properties upon heating and demonstrated high thermal stability, with a decomposition temperature (T d, 5 % ) of 349 °C, making it suitable for the melt-processing of most engineering plastics. Comparative experiments demonstrated that the chelated zinc converted the properties of the resultant OFR, including its intumescence, thermostability and hydrophobicity. The ZnDSi was tested in polyamide 66 (PA66) for the flame-retardant properties. Incorporating 10 wt% of ZnDSi into PA66 attained a limiting oxygen index of 29.1 %, accompanied by the reduction of the peak heat release rate by 35.0 % and total heat release by 28.2 % when compared to the pure PA66 sample, according to the cone calorimeter tests. Mechanism investigation was conducted to probe into the interactions between zinc and organic components through a custom apparatus for pyrolytic gas analysis, and it was found that zinc could reduce the soot release by suppressing the formation of the soot precursors during the pyrolysis. This work contributes to a deeper understanding of how metal elements function in OFR systems, which may conduce to developing OFRs with advanced performance.

The preservative potential of Octopus scraps peptides−Zinc chelate against Staphylococcus aureus: Its fabrication, antibacterial activity and action mode

Zheng Fang,Liangzong Xu,Yanlan Lin,Xixi Cai,Shaoyun Wang

A new type of zinc-chelating peptide was fabricated from Octopus scraps protein hydrolysate and its antibacterial activity against Staphylococcus aureus was characterized. Results of Fluorescence spectra and Fourier Transform Infrared Spectrometer (FT-IR) demonstrated that the amino nitrogen atoms and the oxygen atoms belonging to the carboxylate groups were the primary chelating sites for zinc ions. The zinc-peptide chelate exerted remarkably antimicrobial ability towards S. aureus , whose minimum inhibitory concentration (MIC) was 1.56 mg/mL. To investigate the antibacterial mechanism, changes of bacterial morphology, cell constituents, membrane potential, intracellular reactive oxygen species (ROS) level and intracellular enzyme activity were measured. Results revealed that Zn-peptide exerted its bactericidal activity by damaging bacterial cell membranes and cell walls, increasing the cell permeability which resulted in structural lesions and release of cell components. Besides, Zn-peptide induced S. aureus cell death via ROS triggered pathway. These findings further promote the development of Zn-peptide complexes, suggesting that Zn-peptide chelate has potential as natural bacteriostatic agent in food processing industry.

Gastrointestinal digestion and absorption characterization in vitro of zinc-chelating hydrolysate from scallop adductor (Patinopecten yessoensis)

Jiatong Sun,Xiaoyang Liu,Zixu Wang,Fawen Yin,Huilin Liu,Yoshimasa Nakamura,Chenxu Yu,Dayong Zhou

BACKGROUD Zinc (Zn) is an essential catalytic element in the human health system but its absorption in the intestinal system can be strongly affected by gastrointestinal (GI) digestion. In this study, the food-derived potential Zn carrier, scallop adductor hydrolysates (SAHs), was produced and characterized. RESULTS During temporary storage at 4 °C, SAH decreased in Zn-chelating capacity in the aqueous phase, whereas the SAH–Zn complex exhibited high stability. Moreover, the secondary structure of SAH had no significant alteration. Zn morphologically altered the surface structures of SAH, which was involving in carboxyl group of SAH. Results of in vitro GI digestion suggested that the SAH–Zn maintained good stability in GI system and only proportion of high molecular weight cleaved. In addition, SAH could successfully carry and transport Zn while the fluorescence staining revealed free Zn accumulation inside the tissue. Finally, three representative absorbed peptides (around 600 Da) were identified and synthesized. Three synthetic peptides exhibit higher Zn-chelating capacity than SAH and could also successfully transported through the intestine. CONCLUSION This study provided a theoretical basis for the investigation of digestion and absorption of marine animal-derived peptides as Zn carriers. © 2021 Society of Chemical Industry.

Characterisation of zinc-chelating peptides derived from scallop (Patinopecten yessoensis) mantle and their intestinal transport in everted rat sacs

Xiaoyang Liu,Jiatong Sun,Yue Li,Xuening Yu,Fawen Yin,Deyang Li,Pengfei Jiang,Yoshimasa Nakamura,Dayong Zhou

Summary Recently, food-derived peptides were frequently studied and used as metal ion chelator against negative effects of trace element absorption induced by various human gastrointestinal environments. In this study, the scallop mantle hydrolysates (SMHs) were produced and characterised to study their intestinal promotion of zinc absorption. During production of hydrolysates, the results of degree of hydrolysis (DH) and proteins solubility suggested that the alcalase was suitable for the production of peptides from scallop mantle proteins where the chelating conditions were settled (30 °C, 10 min and 3 mg mL −1 ). The molecular distribution of SMH was mainly 1–5 kDa and chelation of zinc with SMH (involved in carboxyl and amine group of SMH) triggered no significant alteration on the secondary structure of SMH. Zinc morphologically altered the surface structures of SMH. Moreover, SMH could successfully carry and transport zinc while the fluorescence staining indicated free zinc accumulated inside the tissue of everted rat sacs. Finally, several representative transported peptides (intact or cleaved) were identified, such as FTGEPGPSGPT, AINDPFIDL and ALPHAILRI. This study provided a theoretical basis for the investigation of absorption of marine animal-derived peptides as zinc carriers.

Liu Shihao,Zang Chunxiu,Zhang Jiaming,Tian Shuang,Wu Yan,Shen Dong,Zhang Letian,Xie Wenfa,Lee Chun-Sing

Here, this work presents an air-stable ultrabright inverted organic light-emitting device (OLED) by using zinc ion-chelated polyethylenimine (PEI) as electron injection layer. The zinc chelation is demonstrated to increase the conductivity of the PEI by three orders of magnitude and passivate the polar amine groups. With these physicochemical properties, the inverted OLED shows a record-high external quantum efficiency of 10.0% at a high brightness of 45,610 cd m −2 and can deliver a maximum brightness of 121,865 cd m −2 . Besides, the inverted OLED is also demonstrated to possess an excellent air stability (humidity, 35%) with a half-brightness operating time of 541 h @ 1000 cd m −2 without any protection nor encapsulation.

Zinc deficiency reverses biofilm azole resistance in Candida albicans

Yingzhe Wang,Shigan Ye,Yuan Deng,Yingting Huang,Xiaoliang Zhu

Biofilm formation is one of the causes of azole resistance in Candida albicans . Although zinc is an essential trace element involved in biofilm regulation, its specific mechanistic role remains unclear. Here, we systematically evaluated the effects and mechanisms of zinc deficiency on biofilm formation and drug resistance. Intracellular zinc deficiency was induced using the zinc chelator N,N,N′,N′-tetrakis(2-pyridylmethyl)ethylenediamine (TPEN) and a CSR1 knockout strain, as confirmed using zinquin fluorescence. Biofilm formation and susceptibility were assessed using standardized microdilution techniques, including sessile minimum inhibitory concentration (sMIC) determinations via the XTT reduction assay, while drug interactions were assessed using a checkerboard assay. Efflux pump activity was measured using a Rhodamine 6G assay, and transcriptomic analysis was performed to elucidate underlying mechanisms. Pathogenicity was validated using a Galleria mellonella infection model. The results show that zinc deficiency inhibited biofilm development at all stages. Low-concentration TPEN (5µM) reduced the sMIC of fluconazole by more than 16-fold and ultimately reversed its azole resistance. This effect was mechanistically associated with the downregulation of key biofilm-related transcription factors and multidrug efflux pumps, as revealed by transcriptomic analysis, which also indicated that zinc deficiency triggered ribosomal remodeling and activated glucose metabolism. Survival analysis in the G. mellonella infection model confirmed that zinc deficiency reduced the overall pathogenicity of C. albicans biofilms. These results validate zinc homeostasis as a novel therapeutic strategy against drug-resistant and recurrent fungal infections, especially those involving biofilms.

Establishment of oyster protein hydrolysate-chitosan oligosaccharide-zinc delivery system and its characterization, digestion & absorption in-vitro and in-vivo

Xuening Yu,Shijie Dou,Xiaoyang Liu,Fawen Yin,Deyang Li,Pengfei Jiang,Dayong Zhou

Zinc deficiency highly influences human health while the effects of some zinc supplements (peptide-zinc complex) would be negatively attenuated by human gastrointestinal environments. Therefore, it is necessary to develop the embedding methods to improve the stability and absorption effects of zinc supplements. In this study, the chitosan oligosaccharide (COS) was used to react with oyster protein hydrolysates (OPH) for the formation of new zinc carrier. The selected conditions for preparing the OPH-COS-Zn complex were 4 mg/mL, 30°C and 80 min by testing the zinc-chelating capacity. The main binding sites of OPH-COS complex with zinc were presumed to be involved in amino, carboxyl and carbonyl groups. The OPH-COS-Zn delivery system showed high zinc ion release rate (57.04%) by offering high soluble zinc amounts (61.07%) and remained stable in gastrin phase but partially degraded in intestinal phase in gastrointestinal digestion in-vitro . Furthermore, the delivery system was successfully absorbed and carried zinc ions to the serosal side (0.3027 mg/L) in everted rat gut sacs model. The OPH-COS-Zn delivery system showed better in-vivo temporary dynamic absorption of zinc (7.63 μmol/dL) than OPH-Zn (7.07 μmol/dL) and ZnSO 4 (6.09 μmol/dL), showing slow-release effects of zinc ions. This experiment provided the theoretical foundation for further study of embedding effects of peptide-based zinc supplements.
